A cDNA encoding the chloroplast/mitochondrial form of glutathione reductase (GR:EC 1,6,4,2) from pea (Pisum sativum L.) was used to map a single GR locus, named GORI. In two domesticated genotypes of pea (cv, Birte and JI 399) it is likely that the GORI locus contains a single gene. However, in a semi-domesticated land race of pea sequences were detected but closely related sets of GR gene sequences were in JI 281 represent either a second intact gene or a partial or pseudogene copy. A GR gene was cloned from ev. Birte, sequenced and its structure analysed. No features of the transcription or structure of the gene suggested a mechanism for generating any more than one form of . From these data plus previously published biochemical evidence was suggested a second, distinct gene encoding for the cytosolic form of GR should be present in peas. The GORI-encoded GR mRNA can be detected in all main organs of the plant and no alternative spliced species was present which could perhaps account for the generation of multiple isoforms of GR. The mismatch between the number of charge-separable isoforms in pea and the proposed number suggests that different GR isoforms arise by some form of post-transnational modification.

An important factor in crop production is the nutrient liberation matching the growth rates, so these combined effects together with plant development can reach the best results. In this work, the times of nitrogen application (topdressing) were tested in the rate of 120 kg ha(-1) of N, applied at 20 (T20), 25 (T25), 30 (T30), 35 (T35) and 40 (T40) days after emergency in eight cultivars of pea (Pisum sativum L) (Maria, Kodama, Mikado, Flavia, Luiza, Amelia, Dileta and Marina). Here, the aim was to evaluate the effect of these treatments on yield and seed quality in Cerrado Region. After harvest, the seeds were submitted to the quality tests (germination tests, electric conductivity and emergency under field conditions). The results showed that the peas crops are not influenced by the N timing application. Amongst the eight cultivars studied, the best productivity were for Mariana, Mikado, Amelia, Maria and Flavia and the lowest yield was for Kodama cultivar. The cultivar Mikado showed the best results for the quality tests.

The objective was to determine the distribution in pea seedling of GA (by thin layer chromatography) and IAA (by HPLC). Three samples of 30 7/day-old seedlings were sectioned into: apical meristem + plumular hook; stem elongation zone; stem non-elongation zone; cotyledons; root non-elongation zone; root tip. The material was frozen in liquid N and stored at -80 degrees C. The stem elongation zone, the apical meristem+plumular hook and the root tip had low IAA, whereas the stem non-elongation zone and root non-elongation zone had high IAA content. The stem elongation zone and apical meristem plumular hook had high GA, while root non-elongation zone had the lowest GA content.

A ervilha é cultivada no Brasil Central, durante o inverno seco, exigindo para o pleno êxito da cultura o uso da irrigação. Assim, o presente trabalho se propõe a avaliar os efeitos do potencial da água no solo nos índices fisiológicos da análise quantitativa de crescimento de plantas de ervilha (Pisum sativum L.). O experimento foi montado em casa de vegetação, em solos de textura argilosa, com delineamento experimental inteiramente casualizado, com quatro tratamentos decorrentes de potenciais mínimos da água no solo (-33, -100, -200 e -1500 kPa) em três repetições, cada qual contendo duas plantas de ervilha, cultivar Caprice. Os resultados obtidos evidenciaram que a redução do potencial água no solo, induziu o decréscimo na área foliar, sem interferir no comportamento da razão de massa foliar, razão de área foliar, taxa assimilatória líquida e taxa de crescimento relativo.

The impact of heat stress on the functioning of the photosynthetic apparatus was examined in pea (Pisum sativum L.) plants grown at control (25 °C; 25 °C-plants) or moderately elevated temperature (35 °C; 35 °C-plants). In both types of plants net photosynthesis (Pn) decreased with increasing leaf temperature (LT) and was more than 80% reduced at 45 °C as compared to 25 °C. In the 25 °C-plants, LTs higher than 40 °C could result in a complete suppression of Pn. Short-term acclimation to heat stress did not alter the temperature response of Pn. Chlorophyll a fluorescence measurements revealed that photosynthetic electron transport (PET) started to decrease when LT increased above 35 °C and that growth at 35 °C improved the thermal stability of the thylakoid membranes. In the 25 °C-plants, but not in the 35 °C-plants, the maximum quantum yield of the photosystem II primary photochemistry, as judged by measuring the Fv/Fm ratio, decreased significantly at LTs higher than 38 °C. A post-illumination heat-induced reduction of the plastoquinone pool was observed in the 25 °C-plants, but not in the 35 °C-plants. Inhibition of Pn by heat stress correlated with a reduction of the activation state of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco). Western-blot analysis of Rubisco activase showed that heat stress resulted in a redistribution of activase polypeptides from the soluble to the insoluble fraction of extracts. Heat-dependent inhibition of Pn and PET could be reduced by increasing the intercellular CO2 concentration, but much more effectively so in the 35 °C-plants than in the 25 °C-plants. The 35 °C-plants recovered more efficiently from heat-dependent inhibition of Pn than the 25 °C-plants. The results show that growth at moderately high temperature hardly diminished inhibition of Pn by heat stress that originated from a reversible heat-dependent reduction of the Rubisco activation state. However, by improving the thermal stability of the thylakoid membranes it allowed the photosynthetic apparatus to preserve its functional potential at high LTs, thus minimizing the after-effects of heat stress.

Intact chloroplasts were isolated from mature pea (Pisum sativum L.) leaves in order to study the degradation of several stromal proteins in organello. Changes in the abundances of ribulose-1,5-bisphosphate carboxylase/oxygenase (EC 4.1.1.39), phosphoribulokinase (EC 2.7.1.19), glutamine synthetase (EC 6.3.1.2) and ferredoxin-dependent glutamine:α-ketoglutarate aminotransferase (glutamate synthase; EC 1.4.7.1) were detected by sodium dodecyl sulfate-polyacrylamide gel electrophoresis followed by Coomassie-staining of the gels or immunoblotting using specific antibodies for the different enzymes. Degradation of several stromal proteins was strongly stimulated when intact chloroplasts were incubated in the light in the presence of dithiothreitol. Since free radicals may artificially accumulate in the chloroplast under such conditions and interfere with the stability of stromal proteins, the general relevance of these processes remains questionable. In the absence of light, proteolysis proceeded slowly in isolated chloroplasts and was not stimulated by dithiothreitol. Inhibition by ethylenediaminetetraacetic acid (EDTA), 1,10-phenanthroline or excess zinc ions as well as the requirement for divalent cations suggested that a zinc-containing metalloprotease participated in this process. Furthermore, light-independent degradation of ribulose-1,5-bisphosphate carboxylase/oxygenase and phosphoribulokinase was enhanced in chloroplasts isolated from leaves in which senescence was accelerated by nitrogen starvation. Our results indicate that light-independent stromal protein degradation in intact chloroplasts may be analogous to proteolysis that occurs in intact leaves during senescence.

Se realizó el estudio electroforético de proteínas seminales totales de 13 cultivares de arveja (Pisum sativum L.) en geles de poliacrilamida con Sodio Dodecil Sulfato. Se logró fraccionar 53 bandas proteicas, de las cuales 36 mostraron diferencias en sus movilidades. De esta manera se obtuvieron modelos electroforéticos diferentes para cada cultivar y mediante análisis de conglomerados (método UPGMA) se obtuvo un dendrograma que mostró la distinción entre cultivares en dicho carácter, permitiendo expresar las diferencias entre ellos.

The nutritive value of transgenic peas expressing an a-amylase inhibitor (alpha-Ail) was evaluated with broiler chickens. The effects of feeding transgenic peas on the development of visceral organs associated with digestion and nutrient absorption were also examined. The chemical composition of the conventional and the transgenic peas used in this study were similar. In the two feeding trials, that were conducted normal and transgenic peas were incorporated into a maize-soybean diet at concentrations up to 500 g kg(-1). The diets were balanced to contain similar levels of apparent metabolisable energy (AME) and amino acids. In the first trial, the birds were fed the diets from 3 to 17days post-hatching and with levels of transgenic peas at 250 g kg(-1) or greater there was a significant reduction in body weight but an increase in feed intake resulting in deceased feed conversion efficiency. In the second trial, in which the birds were fed diets containing 300 g kg(-1) transgenic peas until 40 days of age, growth performance was significantly reduced. It was also demonstrated that the ileal starch digestibility coefficient (0.80 vs 0.42) was significantly reduced in the birds fed transgenic peas. Determination of AME and ileal digestibility of amino acids in 5-week-old broilers demonstrated a significant reduction in AME (12.12 vs 5.08 MJ kg(-1) DM) in the birds fed the transgenic peas. The AME value recorded for transgenic peas reflected the lower starch digestibility of this line. Real digestion of protein and amino acids was unaffected by treatment. Expression of a-Ail in peas did not appear to affect bird health or the utilisation of dietary protein. However, the significant reduction in ileal digestion of starch in transgenic peas does reduce the utility of this feedstuff in monogastric diets where efficient energy utilisation is required.
