Two new crosses involving four races (races 7, 16, 17, and 25) of the soybean root and stem rot pathogen Phytophthora sojae were established (7/16 cross; 17/25 cross). An F-2 Population derived from each cross was used to determine the genetic basis of avirulence towards 11 different resistance genes in soybean. Avirulence was found to be dominant and determined by a single locus for Avr1b, 1d, 1k, 3b, 4, and 6, as expected for a simple gene-for-gene model. We also observed several cases of segregation, inconsistent with a single dominant gene being solely responsible for avirulence, which suggests that the genetic background of the different crosses can affect avirulence. Avr4 and 6 cosegregated in both the 7/16 and 17/25 crosses and, in the 7/16 cross, Avr1b and 1k were closely linked. Information from segregating RAPD, RFLP, and AFLP markers screened on F-2 progeny from the two new crosses and two crosses described previously (a total of 212 F-2 individuals, 53 from each cross) were used to construct an integrated genetic linkage map of P. sojae. This revised genetic linkage map consists of 386 markers comprising 35 RFLP, 236 RAPD, and 105 AFLP markers, as well as 10 avirulence genes. We have used suspension-cultured parsley cells (Petroselinum crispum) and an oligopeptide elicitor derived from a surface glycoprotein of the phytopathogenic fungus Phytophthora megasperma f.sp. glycinea to study the signaling pathway from elicitor recognition to defense gene activation. Immediately after specific binding of the elicitor by a receptor in the plasma membrane, large and transient increases in several inorganic ion fluxes (Ca2+, H+, K+, Cl-) and H2O2 formation are the first detectable plant cell responses. These are rapidly followed by transient changes in the phosphorylation status of various proteins and by the activation of numerous defense-related genes, concomitant with the inactivation of several other, non-defense-related genes. A great diversity of cis-acting elements and trans-acting factors appears to be involved in elicitor-mediated gene regulation, similar to the apparently complex nature of the signal transduced intracellularly. With few exceptions, all individual defense responses analyzed in fungus-infected parsley leaves have been found to be closely mimicked in elicitor-treated, cultured parsley cells, thus validating the use of the elicitor/cell culture system as a valuable model system for these types of study.

Surveys of commercial soybean fields, disease nurseries, and trial plots of soybean were conducted throughout eastern Australia between 1979 and 1996, and 694 isolates of Phytophthora sojae were collected and classified into races. Fourteen races, 1, 2, 4, 10, 15, and 25, and eight new races, 46 to 53, were identified, but only races 1, 4, 15, 25, 46, and 53 were found in commercial fields. Races 1 and 15 were the only races found in commercial fields in the soybean-growing areas of Australia up until 1989, with race 1 being the dominant race. Race 4 was found in central New South Wales in 1989 on cultivars with the Rps1a gene, and it is now the dominant race in central and southern New South Wales. Races 46 and 53 have only been found once, in southern New South Wales, and race 25 was identified in the same region in 1994 on a cultivar with the Rps1k gene. Only races 1 and 15 have been found in the northern soybean-growing regions, with the latter dominating, which coincides with the widespread use of cultivars with the Rps2 gene. Changes in the race structure of the P. sojae population from commercial fields in Australia follow the deployment of specific resistance genes.

Stem inoculation of clonally propagated lucerne genotypes was used to assess levels of host species and genotype specialisation in Phytophthora medicaginis. A quantitative assessment of pathogenic aggressiveness of 29 P. medicaginis isolates (from lucerne and chickpea) on 9 different clonally propagated lucerne genotypes revealed no significant difference in aggressiveness between isolates from lucerne and those from chickpea on all of the lucerne genotypes. This supported previous studies which showed that P. medicaginis isolates from lucerne and chickpea were indistinguishable using random amplified polymorphic DNA (RAPD) analysis. Analysis of pathogenic aggressiveness towards individual lucerne genotypes revealed, for the first time, specificity of individual P. medicaginis isolates. This has implications for breeding for resistance to P. medicaginis in lucerne, where screening should be done using the widest range of pathogen specificity obtainable.

Phytophthora capsici has been identified from two new hosts in Queensland, Custard Apple (Annona squamosa) and Mandevilla sp. This is the first record of P. capsici in Queensland and only the second record of the pathogen. in Australia. It is also the first report of the pathogen on these hosts.

Phytophthora-resistant lucerne cultivars do not always perform well under conditions of high disease pressure in the field. To determine whether resistance expression remains stable under different infection intensities, tetraploid and diploid lucerne genotypes, genotypically defined for their reactions to Phytophthora medicaginis, were clonally propagated, and the influence of different reproducible inoculum levels (0 . 5 and 5 . 0 g dry weight mycelium/kg dry weight potting mix), the period of exposure to these levels (10-60 days), and temperature (16/22 degrees C and 24/30 degrees C) on disease expression was determined in controlled environments. Generally, expression of resistance by resistant genotypes, remained stable under these conditions. Biotic (e.g. Aphanomyces eutiches) or abiotic factors other than P. medicaginis may be responsible for the poorer than expected performance under field conditions in some instances, or the percentage of resistant plants in some cultivars currently classified as resistant is insufficient to provide buffering against productivity reductions under severe epidemics. Further research is needed to clarify the situation.

An interspecific cross was attempted between two homothallic species of Phytophthora, P. sojae and P. vignae. From 1640 single-oospore cultures isolated, DNA was extracted from 800, and two interspecific F-1 hybrids (F(1)1121 and F(1)1426) were putatively identified using RAPD markers. The true hybrid nature of these F-1 hybrids was confirmed using additional AFLP analysis. Single- zoospore cultures were generated for each F-1 hybrid and one single-zoospore culture of each was used in pathogenicity and virulence tests. Both F-1 hybrids were pathogenic to soybean and cowpea, causing symptoms including lesions, wilting and death of susceptible soybean and cowpea cultivars. However, the aggressiveness of the F-1 hybrids was reduced and was substantially more variable when compared with that of the parental isolates on their respective hosts. The F-1 hybrids were reisolated from infected seedlings and their hybrid nature confirmed using RAPD and AFLP analysis. These results provide a basis for further research aimed at obtaining an increased understanding of the genetics of host specificity in the Oomycetes.

A cross between two different races (race 7 x race 25) of the soybean root and stem rot pathogen Phytophthora sojae was analyzed to characterize the genomic region flanking two cosegregating avirulence genes, Anur4 and Anur6. Both genes cosegregated in the ratio of 82:17 (avirulent:virulent) in an F-2 population, suggestive of a single locus controlling both phenotypes. A chromosome walk was commenced from RAPD marker OPE7.1C, 2.0 cM distant from the Anur4/6 locus. Three overlapping cosmids were isolated which included genetic markers that flank the Anur4/6 locus. The chromosome walk spanned a physical distance of 67 kb which represented a genetic map distance of 22.3cM, an average recombination frequency of 3.0kb/cM and 11.7-fold greater than the predicted average recombination frequency of 35.3 kb/cM for the entire P. sojae genome. Six genes (cDNA clones) expressed from the Anur4/6 genomic region encompassed by the cosmid contig were identified. Single nucleotide polymorphisms and restriction fragment length polymorphisms showed these six genes were closely linked to the Anur4/6 locus. Physical mapping of the cDNA clones within the cosmid contig made it possible to deduce the precise linkage order of the cDNAs. None of the six cDNA clones appear to be candidates for Anur4/6. We conclude that two of these cDNA clones flank a physical region of approximately 24 kb and 4.3 cM that appears to include the Anur4/6 locus. (C) 2003 Elsevier Inc. All rights reserved.

O antagonismo de Pseudomonas putida biovar A (C1-1B), P. putida biovar B (Santa Bárbara), P. fluorescens (C2-8C e RA2), Bacillus subtilis (OG e RC2) e Flavobacterium sp. (CIS/NA) contra Phytophthora parasitica e P. citrophthora , agentes da podridão radicular dos citros, foi avaliado através da inibição do crescimento micelial (cultura pareada) e redução na percentagem de infecção da doença em mudas de citros (tratamento de sementes com rizobactérias). Na seleção preliminar, 33 isolados bacterianos foram testados. Sementes de citros pré-germinadas foram tratadas por imersão nas suspensões das bactérias (10(9) ufc/ml), e plantadas em tubetes contendo solo natural infestado com o fitopatógeno (50 ml de suspensão/ kg de solo). A avaliação da percentagem de infecção foi efetuada após 15 dias. In vitro, os isolados bacterianos RC2, OG, CIS/NA e C1-1B foram os mais ativos inibidores do crescimento micelial de Phytophthora. Em condições de casa de vegetação, todos os isolados proporcionaram redução na percentagem de infecção da doença em todos os ensaios realizados. Promoção de crescimento de plantas foi verificada pela inoculação de plântulas com as linhagens OG, RC2, CiS/Na e C1-1B.

Foi avaliado o comportamento de 14 porta-enxertos para o limão 'Eureka km 47' [Citrus limon (L.) Burm. f.], clone nucelar, em experimento instalado em 1991, na Fazenda Fittipaldi, Araraquara-SP. Os porta-enxertos foram: tangelos 'Orlando' e 'Seminole' (C. reticulata Blanco x C. paradisi Macfad.), tangerinas 'Cleópatra' (C. reshni hort. ex Tanaka) e 'Sunki' [C. sunki (Hayata) hort. ex Tanaka], limões 'Cravo' (C. limonia Osbeck), 'Rugoso da África' (C. jambhiri Lush.) e 'Volkameriano Catania 2' (C. volkameriana V. Ten. & Pasq.), laranja 'Caipira DAC' [C. sinensis (L.) Osbeck], laranja 'Azeda Doble Cálice' (C. aurantium L.), trifoliatas 'EEL'e 'Kryder 8-5' [Poncirus trifoliata (L.) Raf.], citrange 'Morton' [P. trifoliata (L.) Raf. x C. sinensis (L.) Osbeck], C. karna Raf. e C. pennivesiculata (Lush.) Tanaka. As produções foram avaliadas no período de 1998 a 2002. As maiores produções médias em peso de frutos estiveram relacionadas aos porta-enxertos C. pennivesiculata e tangerina 'Cleópatra' (>220 kg/planta). Entre os porta-enxertos que induziram as mais baixas produções, estão C. karna, os dois tangelos, os dois trifoliatas e a 'Caipira DAC' (<180 kg/planta). As características de qualidade apresentadas pelos frutos e referentes ao ano de 2002 indicaram não haver diferenças expressivas devido à influência dos tratamentos. São também apresentados dados relativos à suscetibilidade das plantas à gomose de Phytophthora, no período estudado.

O mamoeiro (Carica papaya) é uma das mais importantes fruteiras tropicais, e seus frutos contribuem com uma produção de 1,65 milhão de toneladas por ano para o Brasil. Um isolado de Phytophthora sp. obtido de frutos de mamoeiro da cultivar Havaí, em Alagoas, foi caracterizado morfologicamente e sua patogenicidade foi confirmada em frutos e mudas deste hospedeiro. Em meio suco V-8, os esporangióforos apresentaram-se delgados, simples ou pouco ramificados, enquanto os esporângios se apresentaram ovóides a elipsóides, proeminentemente papilados e um pedicelo curto na porção basal, medindo em média 37,68 x 27,52 mm. Através das características apresentadas pelo isolado e os sintomas induzidos, o agente causal foi identificado como Phytophthora palmivora. Apesar de essa doença já ocorrer em outros estados, como Bahia, Espírito Santo, São Paulo, Pernambuco e Pará, este é o primeiro relato de Phytophthora palmivora em mamoeiro no Estado de Alagoas.

Uma alternativa de manejo das doenças causadas por Phytophthora spp. é o uso de matéria orgânica. No presente trabalho foi avaliada a potencialidade do lodo de esgoto na indução de supressividade in vitro a P. nicotianae. O efeito do lodo de esgoto incorporado ao solo na sobrevivência de P. nicotianae foi avaliado mediante um experimento fatorial com dois fatores: doses de lodo de esgoto (0, 10, 20 e 40% p/p) e concentrações de inóculo [0, 10 ou 20 g de grãos de trigo (Triticum aestivum) colonizados kg-1]. Aos 21 dias, quando aumentaram as doses de lodo de esgoto, a sobrevivência de P. nicotianae e os pHs das misturas diminuíram, e as condutividades elétricas (CE) aumentaram. As correlações entre a CE e a sobrevivência do patógeno foram negativas e significativas (P>0,05). Para estudar o efeito dos compostos químicos envolvidos na supressividade, foram obtidos extratos em água, H2SO4 2N e KOH 0,4N de misturas de areia – lodo de esgoto (20% p/p), e foram acrescentados ao meio de cultura e seu efeito avaliado no crescimento das colônias de P. nicotianae. O extrato ácido (H2SO4 2N) do tratamento com 20% de lodo de esgoto inibiu significativamente (P>0,05) o crescimento da colônia do patógeno. O efeito biológico foi estudado mediante isolamento de microrganismos em meio de cultura e seleção por antagonismo. No bioensaio com plântulas de alfafa (Medicago sativa) destacaram-se os isolados F9.1 (Aspergillus sp.) e A12.1 (actinomiceto, não identificado); e no teste de culturas pareadas destacou-se um Trichoderma sp. e dois actinomicetos por antibiose, e um Trichoderma sp. e três Aspergillus sp. por hiperparasitismo.

Sete grupos de híbridos entre porta-enxertos elite de citros (Citrus sp.) e seus genitores foram estudados quanto a reação à infecção de tronco por Phytophthora parasitica, em plantas adultas no campo. Inocularam-se artificialmente, em duas posições do tronco, 132 plantas nucelares dos genitores e 486 híbridos entre limão (Citrus limonia) Cravo 'Limeira' (C), Poncirus trifoliata 'Davis A' (T), tangerina (C.sunki) 'Sunki' (S) e laranja Azeda (C. aurantium) 'São Paulo' (A). A classificação do grau de resistência à gomose de tronco em plantas adultas no campo foi possível somente quando baseada na média do tamanho de lesões de mais de 30 plantas nucelares, devido às grandes variações observadas em plantas individuais do mesmo clone, impossibilitando a seleção precoce de híbridos baseada em valores de plantas individuais. O trifoliata e a laranja Azeda mostraram-se bastante resistentes, desenvolvendo, em geral, lesões de tamanho reduzido. A tangerina Sunki e o limão Cravo desenvolveram lesões maiores, embora a tangerina Sunki tenha mostrado uma tendência em desenvolver lesões maiores que às do limão Cravo. Híbridos de trifoliata apresentaram, no geral, lesões intermediárias. Os híbridos de Azeda apresentaram lesões de tamanhos bastante variáveis, porém a maioria, com lesões grandes. Da mesma forma comportaram-se os híbridos recíprocos entre Sunki e Cravo.
