We report the interesting finding that crystallization of calcium carbonate (CaCO3) in the presence of dimyristoylphosphatidylglycerol (DMPG) vesicles by a simple gas diffusion method results in the formation of unusual microscopic CaCO3 spherules. The experimental results indicate that the as-prepared CaCO3 spherules, which have a complex macroporous structure, are predominantly vaterite. It is believed that DMPG vesicles play an important role in the process of crystallization, and the possible formation mechanism is proposed.

Previous results indicated that translation of four mitochondrion-encoded genes and one nucleus-encoded gene (COX4) is repressed in mutants (pgs1Delta) of Saccharomyces cerevisiae lacking phosphatidylglycerol and cardiolipin. COX4 translation was studied here using a mitochondrially targeted green fluorescence protein (mtGFP) fused to the COX4 promoter and its 5' and 3' untranslated regions (UTRs). Lack of mtGFP expression independent of carbon source and strain background was established to be at the translational level. The translational defect was not due to deficiency of mitochondrial respiratory function but was rather caused directly by the lack of phosphatidylglycerol and cardiolipin in mitochondrial membranes. Reintroduction of a functional PGS1 gene under control of the ADH1 promoter restored phosphatidylglycerol synthesis and expression of mtGFP. Deletion analysis of the 5' UTR(COX4) revealed the presence of a 50-nucleotide fragment with two stem-loops as a cis-element inhibiting COX4 translation. Binding of a protein factor(s) specifically to this sequence was observed with cytoplasm from pgs1Delta but not PGS1 cells. Using HIS3 and lacZ as reporters, extragenic spontaneous recessive mutations that allowed expression of His3p and beta-galactosidase were isolated, which appeared to be loss-of-function mutations, suggesting that the genes mutated may encode the trans factors that bind to the cis element in pgs1Delta cells.

The in vitro conversion of phosphatidylglycerophosphate (PGP) to phosphatidylglycerol (PG) involves at least two membrane bound phosphatases in Escherichia coli. The genes encoding these two PGP-phosphatases, pgpA and pgpB, are unique and map distally to min 10 and min 28 respectively. Although point mutations in either or both of these genes decrease the level of PGP phosphatase as assayed in vitro, and also result in a minor accumulation of the precursor, PGP, in the membrane, the mutations have no significant effect on the level of PG in the cell (Icho, T. and Raetz, C. R. H. (1983) J. Bact. 153, 722-730). This dilemma suggests that there remains a significant level of phosphatase activity in the pgpAand pgpB mutants which is sufficient to support normal PG metabolism in vivo, but it is not clear whether this activity is a consequence of a separate phosphatase, or due to "leakiness" of the point lesions in these genes. To address this problem, we have constructed null alleles of the two phosphatase genes, and characterized the effects of these mutations on PG metabolism. Our findings demonstrate that neither the pgpA nor the pgpB phosphatase gene is essential for cell viability. In addition, similar to the pgpA$\sp{-}$, pgpB$\sp{-}$ double point mutant, a strain containing both of the corresponding null alleles still retains enough phosphatase activity to maintain normal levels of PG in the membrane. These data demonstrate that there exists at least a third gene encoding a major biosynthetic phosphatase which is responsible for the in vivo conversion of PGP to PG, and calls into question the actual roles of the pgpA and the pgpB gene products in PG metabolism and cell function. ^

To investigate the role of phosphatidylglycerol (PG) in photosynthesis, we constructed a mutant defective in the CDP-diacylglycerol synthase gene from a cyanobacterium, Synechocystis sp. PCC6803. The mutant, designated as SNC1, required PG supplementation for growth. Growth was repressed in PG-free medium concomitantly with the decrease in cellular content of PG. These results indicate that PG is essential, and that SNC1 is defective in PG synthesis. Decrease in PG content was accompanied by a reduction in the cellular content of chlorophyll, but with little effect on the contents of phycobilisome pigments, which showed that levels of chlorophyll–protein complexes decreased without alteration of those of phycobilisomes. Regardless of the decrease in the PG content, CO2-dependent photosynthesis by SNC1 was similar to that by the wild type on a chlorophyll basis, but consequently became lower on a cell basis. Simultaneously, the ratio of oxygen evolution of photosystem II (PSII) measured with p-benzoquinone to that of CO2-dependent photosynthesis, which ranged between 1.3 and 1.7 in the wild type. However, it was decreased in SNC1 from 1.3 to 0.4 during the early growth phase where chlorophyll content and CO2-dependent photosynthesis were little affected, and then finally to 0.1, suggesting that PSII first lost its ability to reduce p-benzoquinone and then decreased in its level and actual activity. These results indicate that PG contributes to the accumulation of chlorophyll–protein complexes in thylakoid membranes, and also to normal functioning of PSII.

In this study we used electro-spray ionization mass-spectrometry to determine phospholipid class and molecular species compositions in bacteriophages PM2, PRD1, Bam35 and phi6 as well as their hosts. To obtain compositional data of the individual leaflets, phospholipid transbilayer distribution in the viral membranes was studied. We found that 1) the membranes of all studied bacteriophage are enriched in PG as compared to the host membranes, 2) molecular species compositions in the phage and host membranes are similar, and 3) phospholipids in the viral membranes are distributed asymmetrically with phosphatidylglycerol enriched in the outer leaflet and phosphatidylethanolamine in the inner one (except Bam35). Alternative models for selective incorporation of phospholipids to phages and for the origins of the asymmetric phospholipid transbilayer distribution are discussed. Notably, the present data are also useful when constructing high resolution structural models of bacteriophages, since diffraction methods cannot provide a detailed structure of the membrane due to high motility of the lipids and lack of symmetric organization of membrane proteins.

Structural specificity for the direct vesicle−vesicle exchange of phospholipids through stable molecular contacts formed by the antibiotic polymyxin B (PxB) is characterized by kinetic and spectroscopic methods. As shown elsewhere [Cajal, Y., Rogers, J., Berg, O. G., & Jain, M. K. (1996) Biochemistry 35, 299−308], intermembrane molecular contacts between anionic vesicles are formed by a small number of PxB molecules, which suggests that a stoichiometric complex may be responsible for the exchange of phospholipids. Larger clusters containing several vesicles are formed where each vesicle can make multiple contacts if sterically allowed. In this paper we show that the overall process can be dissected into three functional steps: binding of PxB to vesicles, formation of stable vesicle−vesicle contacts, and exchange of phospholipids. Polycationic PxB binds to anionic vesicles. Formation of molecular contacts and exchange of monoanionic phospholipids through PxB contacts does not depend on the chain length of the phospholipid. Only monoanionic phospholipids (with methanol, serine, glycol, butanol, or phosphatidylglycerol as the second phosphodiester substituent in the head group) exchange through these contacts, whereas dianionic phosphatidic acid does not. Selectivity for the exchange was also determined with covesicles of phosphatidylmethanol and other phospholipids. PxB does not bind to vesicles of zwitterionic phosphatidylcholine, and its exchange in covesicles is not mediated by PxB. Vesicles of dianionic phospholipids, like phosphatidic acid, bind PxB; however, this phospholipid does not exchange. The structural features of the contacts are characterized by the spectroscopic and chemical properties of PxB at the interface. PxB in intermembrane contacts is readily accessible from the aqueous phase to quenchers and reagents that modify amino groups. Results show that PxB at the interface can exist in two forms depending on the lipid/PxB ratio. Additional studies show that stable PxB-mediated vesicle−vesicle contacts may be structurally and functionally distinct from “stalks”, the putative transient intermediate for membrane fusion. The phenomenon of selective exchange of phospholipids through peptide-mediated contacts could serve as a prototype for intermembrane targeting and sorting of phospholipids during their biosynthesis and trafficking in different compartments of a cell. The protocols and results described here also extend the syllogistic foundations of interfacial equilibria and catalysis.

Lipopolysaccharide (LPS) is an endotoxin, a potent stimulator of immune response and induction of LPS leads to acute lung injury (ALI)/acute respiratory distress syndrome (ARDS). ARDS is a life-threatening disease worldwide with a high mortality rate. The immunological effect of LPS with spleen and thymus is well documented; however the impact on membrane phospholipid during endotoxemia has not yet been studied. Hence we aimed to investigate the influence of LPS on spleen and thymus phospholipid and fatty acid composition by 32P]orthophosphate labeling in rats. The in vitro labeling was carried out with phosphate-free medium (saline). Time course, LPS concentration-dependent, pre- and post-labeling with LPS and fatty acid analysis of phospholipid were performed. Labeling studies showed that 50 mu g LPS specifically altered the major phospholipids, phosphatidylcholine and phosphatidylglycerol in spleen and phosphatidylcholine in thymus. Fatty acid analysis showed a marked alteration of unsaturated fatty acids/saturated fatty acids in spleen and thymus leading to immune impairment via the fatty acid remodeling pathway. Our present in vitro lipid metabolic labeling study could open up new vistas for exploring LPS-induced immune impairment in spleen and thymus, as well as the underlying mechanism.

Tobacco-specific nitrosamines (TSNA) have implications in the pathogenesis of various lung diseases and conditions are prevalent even in non-smokers. N-nitrosonornicotine (NNN) and 4-(methyl nitrosamino)-1-(3-pyridyl)-1-butanone (NNK) are potent pulmonary carcinogens present in tobacco product and are mainly responsible for lung cancer. TSNA reacts with pulmonary surfactants, and alters the surfactant phospholipid. The present study was undertaken to investigate the in vitro exposure of rat lung tissue slices to NNK or NNN and to monitor the phospholipid alteration by P-32]orthophosphate labeling. Phospholipid content decreased significantly in the presence of either NNK or NNN with concentration and time dependent manner. Phosphatidylcholine (PC) is the main phospholipid of lung and significant reduction was observed in PC similar to 61%, followed by phosphatidylglycerol (PG) with 100 mu M of NNK, whereas NNN treated tissues showed a reduction in phosphatidylserine (PS) similar to 60% and PC at 250 mu M concentration. The phospholipase A(2) assays and expression studies reveal that both compounds enhanced phospholipid hydrolysis, thereby reducing the phospholipid content. Collectively, our data demonstrated that both NNK and NNN significantly influenced the surfactant phospholipid level by enhanced phospholipase A(2) activity. (C) 2014 Elsevier Ltd. 以类囊体膜中唯一的阴离子型磷脂一磷脂酰甘油(PG)为研究对象,应用放氧测定和富立叶红外光谱等实验方法和技术手段,对PG与光系统II (PSII)之间的相互作用进行了研究。 研究表明,PG对PSII的放氧活性产生显著影响,具有明显的浓度效应。在低浓度(2~22 mg PG/mg Chl)时对PSII的放氧活性有明显的促进作用,而在高浓度(24~40 mg PG/mg Chl)下则表现出显著的抑制作用。 PG对PSII放氧活性的影响与其引起蛋白结构的改变密切相关。结果显示,PG的作用导致PSII颗粒中蛋白质二级结构的改变,主要表现为α-螺旋、β-折叠的增加和无规卷曲的减少。 不仅如此,红外光谱的分析还表明,PG还使蛋白酪氨酸残基中的酚基构象及其周围的微极性发生改变,即在红外光谱的1620—1500 cm-1,之间芳香环骨架的伸缩振动带向高频方向变化,其吸收强度也相应增加;在3500~3100 cm. -1间出现新的氢键吸收峰。 PG除能促进PSII的放氧活性以外,还对PSII表现出新的作用,即PG可以使PSII颗粒因缺钙而受抑制的放氧活性得到恢复;外加Ca2+可使PG表现出对缺钙PSII颗粒(dc。PSII)放氧活性的更大促进作用,且随Ca2+浓度的增加,促进作用也越显著。 PG的作用也使dc。PSII蛋白的结构发生了改变,导致蛋白二级结构中a-螺旋、p_折叠结构的增加和转角、无规卷曲成分的减少,即可使PSII颗粒因缺钙而改变的蛋白结构基本得到恢复。PG还能与Ca2+形成离子对似的配合物,而这种配合物的形成可以优化缺钙PSII颗粒的功能如放氧活性等。
