A importância da patologia de sementes reside no fato de que aproximadamente 90% das culturas utilizadas para a alimentação são propagadas por semente. Dentre essas, nove são consideradas de importância primordial: soja, trigo, arroz, milho, feijão, amendoim, sorgo, cevada e beterraba açucareira.Todas essas culturas podem ser afetadas por patógenos muito agressivos transmitidos através da semente. Assim, o teste de sanidade de semente pode ser considerado como "medicina preventiva", tanto nos programas de quarentena quanto no sistema de produção de semente certificada. Nesta publicação, em sua primeira parte, são abordados os principais aspectos da patologia de sementes, como os seus históricos no mundo e no Brasil, os diferentes métodos utilizados e os fatores que podem causar variação nos resultados dos testes. Em sua segunda parte, são discutidos, em detalhe, os principais patógenos causadores de doenças na cultura da soja que são transmitidos pela semente. Dentre esses, destacam-se, Phomopsis sp. e Fusarium semitectum, causadores de problemas de germinação no laboratório quando ocorrem chuvas durante as fases de maturação e colheita da semente (podridão de semente); Diaporthe phaseolorum f.sp. meridionalis (Phomopsis meridionalis) (cancro da haste); Colletotrichum truncatum (antracnose); Cercospora kikuchii (mancha púrpura); Cercospora sojina (mancha olho-de-rã); Sclerotinia sclerotiorum (podridão branca); Sclerotium rolfsii (tombamento e morte de plantas); Macrophomina phaseolina (podridão de carvão); Rhizoctonia solani (tombamento) e Aspergillus spp. (A. flavus) que, além de ser considerado fungo de armazenagem, é responsável pela podridão da semente no solo, quando a semeadura é feita em solos com baixa disponibilidade de água, sem o tratamento da semente com fungicida. Finalmente é discutida a importância do tratamento de semente de soja com fungicidas, cuja tecnologia, desde a safra 2001/02, vem sendo utilizada em mais de 93% da área semeada com soja no Brasil.

We describe the genetic transformation of the mycelial tissue of Diaporthe phaseolorum, an endophytic fungus isolated from the mangrove species Laguncularia racemosa, using Agrobacterium tumefaciens-mediated transformation (ATMT). ATMT uses both the hygromycin B resistant (hph) gene and green fluorescent protein as the selection agents. The T-DNA integration into the fungal genome was assessed by both PCR and Southern blotting. All transformants examined were mitotically stable. An analysis of the T-DNA flanking sequences by thermal asymmetric interlaced PCR (TAIL-PCR) demonstrated that the disrupted genes in the transformants had similarities with conserved domains in proteins involved in antibiotic biosynthesis pathways. A library of 520 transformants was generated, and 31 of these transformants had no antibiotic activity against Staphylococcus aureus, an important human pathogen. The protocol described here, using ATMT in D. phaseolorum, will be useful for the identification and analysis of fungal genes controlling pathogenicity and antibiotic pathways. Moreover, this protocol may be used as a reference for other species in the Diaporthe genus. This is the first report to describe Agrobacterium-mediated transformation of D. phaseolorum as a tool for insertional mutagenesis.

Endophytic fungi are considered a rich source of active compounds resulting from their secondary metabolism. Fungi from marine environment grow in a habitat with unique conditions that can contribute to the activation of metabolic pathways of synthesis of different unknown molecules. The production of these compounds may support the adaptation and survival of the fungi in the marine ecosystem. Mangroves are ecosystems situated between land and sea. They are frequently found in tropical and subtropical areas and enclose approximately 18.1 million hectares of the planet. The great biodiversity found in these ecosystems shows the importance of researching them, including studies regarding new compounds derived from the endophytic fungi that inhabit these ecosystems. 3-hydroxypropionic acid (3-HPA) has been isolated from the mangrove endophytic fungus Diaporthe phaseolorum, which was obtained from branches of Laguncularia racemosa. The structure of this compound was elucidated by spectroscopic methods, mainly 1D and 2D NMR. In bioassays, 3-HPA showed antimicrobial activities against both Staphylococcus aureus and Salmonella typhi. The structure of this antibiotic was modified by the chemical reaction of Fischer-Speier esterification to evaluate the biologic activity of its chemical analog. The esterified product, 3-hydroxypropanoic ethyl ester, did not exhibit antibiotic activity, suggesting that the free carboxylic acid group is important to the pharmacological activity. The antibiotic-producing strain was identified with internal transcribed spacer sequence data. To the best of our knowledge, this is the first report of antibacterial activity by 3-HPA against the growth of medically important pathogens.

The identification of Diaporthe (anamorph Phomopsis) species associated with stem canker of sunflower (Helianthus annuus) in Australia was studied using morphology, DNA sequence analysis and pathology. Phylogenetic analysis revealed three clades that did not correspond with known taxa, and these are believed to represent novel species. Diaporthe gulyae sp. nov. is described for isolates that caused a severe stem canker, specifically pale brown to dark brown, irregularly shaped lesions centred at the stem nodes with pith deterioration and mid-stem lodging. This pathogenicity of D. gulyae was confirmed by satisfying Koch's Postulates. These symptoms are almost identical to those of sunflower stem canker caused by D. helianthi that can cause yield reductions of up to 40% in Europe and the USA, although it has not been found in Australia. We show that there has been broad misapplication of the name D. helianthi to many isolates of Diaporthe ( Phomopsis) found causing, or associated with, stem cankers on sunflower. In GenBank, a number of isolates had been identified as D. helianthi, which were accommodated in several clades by molecular phylogenetic analysis. Two less damaging species, D. kochmanii sp. nov. and D. kongii sp. nov., are also described from cankers on sunflower in Australia.

Tem sido atribuído a vários patógenos reduções na qualidade de sementes de soja. Com o objetivo de avaliar o transporte dos fungos pelas sementes provenientes de plantas de soja, artificialmente inoculadas em condições de campo com dois isolados de Colletotrichum dematium var. truncata e um isolado de Phomopsis sojae, e seu efeito sobre a qualidade sanitária e fisiológica das mesmas, quatro variedades de soja (BRS 133, MSoy 6101, Conquista e Liderança) foram manualmente semeadas no campo, e as plantas obtidas foram inoculadas com C. dematium var. truncata isolado 1 e isolado 2, e P. sojae. A testemunha permaneceu sem inoculação. As sementes foram colhidas manualmente e submetidas a testes para avaliação da sanidade, teor de água, peso de 100 sementes, germinação em papel e emergência em areia. A desinfestação das sementes com hipoclorito de sódio a 1% por 3 minutos diminuiu a incidência dos patógenos nas mesmas. As sementes apresentaram uma alta infecção por Fusarium spp.. Phomopsis sojae foi encontrada em todos os tratamentos, e sua incidência nas sementes esteve relacionada com a incidência de Fusarium sp.. Há diferença na patogenicidade entre isolados de C. dematium var. truncata, sendo o isolado 1 o mais patogênico, com destaque para a variedade Liderança (25% de infecção por C. dematium var. truncata). Fusarium sp. influenciou negativamente a germinação de sementes da variedade MSoy 6101, independente do tratamento, e C. dematium var. truncata afetou negativamente a germinação das sementes das variedades Conquista e Liderança. Os valores de emergência em areia apresentaram-se significativamente superiores à germinação em papel, para as variedades MSoy 6101, Conquista e Liderança.

Inclusions of sp-hybridised, trans-polyacetylene [trans-(CH)x] and poly(p-phenylene vinylene) (PPV) chains are revealed using resonant Raman scattering (RRS) investigation of amorphous hydrogenated carbon (a-C:H) films in the near IR – UV range. The RRS spectra of trans-(CH)x core Ag modes and the PPV CC-H phenylene mode are found to transform and disperse as the laser excitation energy ћωL is increased from near IR through visible to UV, whereas sp-bonded inclusions only become evident in UV. This is attributed to ћωL probing of trans-(CH)x chain inhomogeneity and the distribution of chains with varying conjugation length; for PPV to the resonant probing of phelynene ring disorder; and for sp segments, to ћωL probing of a local band gap of end-terminated polyynes. The IR spectra analysis confirmed the presence of sp, trans-(CH)x and PPV inclusions. The obtained RRS results for a-C:H denote differentiation between the core Ag trans-(CH)x modes and the PPV phenylene mode. Furthermore, it was found that at various laser excitation energies the changes in Raman spectra features for trans-(CH)x segments included in an amorphous carbon matrix are the same as in bulk trans-polyacetylene. The latter finding can be used to facilitate identification of trans-(CH)x in the spectra of complex carbonaceous materials.
