The plants belonging to Pfaffia genus are used in folk medicine to treat gastric disturbances. This study examined the effects of an aqueous extract of Pfaffia glomerata (Spreng) Pedersen (AEP) on the gastrointestinal tract. Wistar rats were pretreated orally (p.o.) with the AEP (125, 250, 500 and 1000 mg.kg(-1)) before induction of ulcers by hypothermic restraint stress (HRS, 3 h restraint stress at 4 degreesC), ethanol (ET, 70%; 0.5 ml/animal; p.o.) or indomethacin (IND, 20 mg.kg(-1); s.c.). Control animals received water (C) or ranitidine (60 mg.kg(-1)) p.o. The AEP protected rats against HRS and ET-induced ulcers, but was not able to protect the gastric mucosa against IND-induced ulcers. When injected into the duodenal lumen, the AEP reduced total acidity and both basal and histamine-stimulated acid secretion in pylorus-ligated rats. In addition, gastric secretion from AEP-treated animals exhibited increased concentrations of nitrite and nitrate. Treatment of animals with L-NAME (120 mg.kg(-1), p.o.) prevented both the reduction of total acidity and the increase in NO, levels promoted by AEP treatment. In conclusion, AEP effectively protected the gastric mucosa and inhibited gastric acid secretion in rats, probably by involving the histaminergic pathway and an enhanced production of nitric oxide in the stomach. O trabalho teve como objetivo verificar a influência da adubação orgânica em 6 épocas de crescimento na produção de β-ecdisona por plantas de Pfaffia glomerata. O experimento foi conduzido na Fazenda Santo Antonio do Araquá, distrito de Catâneo Ângelo, município de São Manuel, São Paulo, Brasil. Utilizou-se o delineamento de blocos ao acaso, num esquema fatorial 5x6, com quatro repetições, considerando-se 8 plantas úteis por parcela. Os blocos foram constituídos de 6 épocas de crescimento (60, 120, 180, 240, 300 e 360 dias após a germinação) e de 5 doses de esterco de galinha curtido [testemunha (sem adubação), 15, 30, 45 e 60 t ha-1]. Após cada colheita, as raízes das plantas foram secas em estufa com circulação de ar forçada a 40ºC e pesadas para posterior extração do β-ecdisona, seguindo metodologia desenvolvida por Magalhães (2000). Os resultados foram submetidos à análise de variância e ao teste de separação de médias de Scott Knott, todos a 5% de probabilidade. Quando ocorreu interação os resultados foram avaliados usando-se análise de regressão polinominal. O teor de β-ecdisona não foi influenciado pelas doses de adubo e nem pela época do crescimento das plantas. Porém a quantidade total de β-ecdisona por raiz foi influenciada pela época de crescimento, sendo que aos 360 dias após a emergência ocorreu uma maior quantidade do princípio ativo em todos os tratamentos. Apesar de não diferir estatisticamente dos demais tratamentos, aos 360 dias após a emergência das plantas, o tratamento 30 t ha-1 foi o que proporcionou maior quantidade de β-ecdisona.

Interviews were conducted with fafia (Pfaffia glomerata (Spreng.) Pedersen) collectors from the municipal districts of Querencia do Norte and Porto Rico, where enterprises and buyers of this plant are concentrated. The results allowed us to conclude that both the adults and children make collection. Each adult collects from 50 to 150 kg of roots/day, depending on the collection area, for about 8 months/year. Collections mostly occur all over the year, but the activity becomes more intense from May to August. All families are not exclusively dedicated to fafia collection and also develop other rural activities. They have been collecting fafia for 2 to 13 years, indicating that an intensive exploitation has been present in the region for over a decade. During collection, no plant part is used for replanting the species. The roots are commercialised by regional buyers. The price of the roots varies from US $ 0.07 to US $ 0.13/kg and average gain is about US $ 2,055/family/year, representing a considerable profit for the collectors.

The aim of this experiment is to evaluate the development in height of the plants of four accessions (2 from Parana - accession 1 and 2, one from São Paulo - accession 3, and one from Mato Grosso do Sul State- accession 4). The experimental design was made of randomized blocks, with four treatments (accessions) and four replications. In each plot were planted 24 seedlings. Five evaluations of the height were accomplished, with an interval of two months, starting from the second month after the planting of the seedlings. The results show a significant difference between accession 4 and other three accessions in first evaluation (141.42 % higher than accession 2, the lowest of three and 44.16 % higher than accession 3, the highest of three), demonstrating a higher vigour of this accession on initial stage of its development. In the second and third evaluation, all accessions have no statistical difference, demonstrating a similar level of development at intermedial stages of development. At 5 th evaluation, all accessions have almost the same result, with no statistical difference among them. This is a clear result of similar level of development of this species in these accessions, representing a stabilization of their height development at the end of first year. We concluded that fáfia possesses a strong initial growth, with visible difference among the accessions in the first months. From the eight month on, we observed a slight decrease in the height. This suggests the end of annual cycle.

O presente trabalho objetivou avaliar o efeito do pH do meio de cultivo sobre alguns parâmetros de crescimento da Pfaffia glomerata (Spreng.) Pedersen cultivada in vitro, bem como checar se o crescimento dos explantes altera o pH do meio ao longo do período de cultivo. Foram testados quatro tratamentos constituídos de distintos valores de pH (3,7; 5,0; 6,0 e 7,5) do meio de cultivo. O pH do meio de cultivo foi ajustado antes da inclusão do agar (6g L-1 - Merck) e da autoclavagem. Como fonte de explantes foram utilizadas segmentos nodais de plantas previamente estabelecidas in vitro em meio MS. Dos nove aos 15 dias após a inoculação (DAI) dos segmentos nodais, verificou-se maior número de raízes em pH 6,0 e o menor no pH 7,5. Aos 35 DAI, o comprimento da maior brotação e o número total de segmentos nodais por planta foram maiores em torno de pH 6,0. Aos 35 DAI, observou-se menor crescimento em biomassa de raízes em pH 3,7. Já a parte aérea apresentou menor biomassa em pH 7,5. Aos 35 DAI, a produção de matéria fresca e seca total da plântula foi maior em pH próximo a 6,0. Concluiu-se que valores de pH do meio de cultivo próximos a 6,0, ajustados antes da autoclavagem, são ideais para o crescimento da P. glomerata cultivada in vitro. Também se verificou que o crescimento da plântula modificou significativamente o pH do meio de cultivo.

Por meio do estudo de estruturas fúngicas ao microscópio estereoscópico e ótico e de uma caracterização em detalhes, inclusive com micrografias, foi possível identificar cinco espécies de fungos causadores de ferrugens da família Pucciniaceae associadas com sete espécies vegetais medicinais do horto Ervas & Matos da Universidade Federal de Lavras, em Minas Gerais (21º14' - latitude sul; 45º00' - longitude oeste). As espécies de ferrugem e seus respectivos hospedeiros foram: Puccinia lantanae em Lippia alba; P. leonotidicola em Leonotis nepetaefolia; P. menthae em Mentha spp. e M. arvensis; P. porophylli em Porophyllum ruderale; e Uromyces platensis sobre Pfaffia glomerata

Abstract In this work, a novel on-line process for production of food-grade emulsions containing oily extracts, i.e. oil-in-water (O/W) emulsions, in only one step is presented. This process has been called ESFE, Emulsions from Supercritical Fluid Extraction. With this process, emulsions containing supercritical fluid extracts can be obtained directly from plant materials. The aim in the conception of this process is to propose a new rapid way to obtain emulsions from supercritical fluid extracts. Nowadays the conventional emulsion formulation method is a two-step procedure, i.e. first supercritical fluid extraction for obtaining an extract; secondly emulsion formulation using another device. Other variation of the process was tested and successfully validated originating a new acronymed process: EPFE (Emulsions from Pressurized Fluid Extractions). Both processes exploit the supercritical CO2-essential oils miscibility, in addition, EPFE process exploits the emulsification properties of saponin-rich pressurized aqueous plant extracts. The feasibility of this latter process was demonstrated using Pfaffia glomerata roots as source of saponin-rich extract, water as extracting solvent and clove essential oil, directly extracted using supercritical CO2, as a model dispersed phase. In addition, examples of pressurized fluid-based coupled processes applied for adding value to food bioactive compounds developed in the past five years are reviewed.
