In previous studies it has been established that resistance to superoxide by Neisseria gonorrhoeae is dependent on the accumulation of Mn(II) ions involving the ABC transporter, MntABC. A mutant strain lacking the periplasmic binding protein component (MntC) of this transport system is hypersensitive to killing by superoxide anion. In this study the mntC mutant was found to be more sensitive to H2O2 killing than the wild-type. Analysis of regulation of MntC expression revealed that it was de-repressed under low Mn(II) conditions. The N. gonorrhoeae mntABC locus lacks the mntR repressor typically found associated with this locus in other organisms. A search for a candidate regulator of mntABC expression revealed a homologue of PerR, a Mn-dependent peroxide-responsive regulator found in Gram-positive organisms. A perR mutant expressed more MntC protein than wild-type, and expression was independent of Mn(II), consistent with a role for PerR as a repressor of mntABC expression. The PerR regulon of N. gonorrhoeae was defined by microarray analysis and includes ribosomal proteins, TonB-dependent receptors and an alcohol dehydrogenase. Both the mntC and perR mutants had reduced intracellular survival in a human cervical epithelial cell model.

The regulatory mechanisms by which hydrogen peroxide (H2O2) modulates the activity of transcription factors in bacteria (OxyR and PerR), lower eukaryotes (Yap1, Maf1, Hsf1 and Msn2/4) and mammalian cells (AP-1, NRF2, CREB, HSF1, HIF-1, TP53, NF-κB, NOTCH, SP1 and SCREB-1) are reviewed. The complexity of regulatory networks increases throughout the phylogenetic tree, reaching a high level of complexity in mammalians. Multiple H2O2 sensors and pathways are triggered converging in the regulation of transcription factors at several levels: (1) synthesis of the transcription factor by upregulating transcription or increasing both mRNA stability and translation; (ii) stability of the transcription factor by decreasing its association with the ubiquitin E3 ligase complex or by inhibiting this complex; (iii) cytoplasm-nuclear traffic by exposing/masking nuclear localization signals, or by releasing the transcription factor from partners or from membrane anchors; and, (iv) DNA binding and nuclear transactivation by modulating transcription factor affinity towards DNA, co-activators or repressors, and by targeting specific regions of chromatin to activate individual genes. We also discuss how H2O2 biological specificity results from diverse thiol protein sensors, with different reactivity of their sulfhydryl groups towards H2O2, being activated by different concentrations and times of exposure to H2O2. The specific regulation of local H2O2 concentrations is also crucial and results from H2O2 localized production and removal controlled by signals. Finally, we formulate equations to extract from typical experiments quantitative data concerning H2O2 reactivity with sensor molecules. Rate constants of 140 M-1s−1 and ≥ 1.3 × 103 M-1s−1 were estimated, respectively, for the reaction of H2O2 with KEAP1 and with an unknown target that mediates NRF2 protein synthesis. In conclusion, the multitude of H2O2 targets and mechanisms provides an opportunity for highly specific effects on gene regulation that depend on the cell type and on signals received from the cellular microenvironment.

O feijão representa importante fonte de proteína e pode auxiliar na modulação dos níveis de glicose e lipídios sangüíneos. Objetivou-se avaliar o valor protéico e funcional de variedades de feijão. Para a qualidade protéica foram usados 7 grupos (n = 6) de ratos recém desmamados: um recebeu dieta aprotéica por 14 dias e os demais, dietas contendo 9-10% de proteína como caseína ou como um dos feijões (Ouro Branco, Pérola, BRS Radiante, Diamante Negro e Talismã). No segundo experimento, 4 grupos (n = 10) de ratos adultos receberam dietas de caseína, Ouro Branco, Diamante Negro e Talismã por 28 dias. Ouro Branco apresentou maior Coeficiente de Eficiência Protéica (PERr = 61,18%) do que os demais e o BRS Radiante o menor (PERr = 44,60%). Quanto à Razão Protéica Líquida (NPR) e à digestibilidade, não houve diferença (p > 0,05) entre os grupos, variando de 67,05 a 78,19% e de 78,70 a 84,88%, respectivamente. As dietas de Diamante Negro e Talismã promoveram redução da glicemia de 30 e 26%, respectivamente, em relação à caseína, não sendo, porém, significativa; não houve diferença (p > 0,05) entre os grupos quanto aos níveis séricos de colesterol e triacilgliceróis. Assim, de acordo com as necessidades apresentadas pelas populações, suas demandas nutricionais e funcionais podem ser supridas por diferentes feijões.

La escritura (literacy) y las matemáticas (numeracy) constituyen los códigos más relevantes de las comunidades occidentales y desarrolladas. La escritura y las matemáticas son, de hecho, dos códigos secretos desarrollados sociohistóricamente por la humanidad. La alfabetización requiere adquirir un código secreto basado en normas y unos géneros discursivos de contenido, forma y estilo para saber escribir y leer, en la ponencia se desarrolla todo este aprendizaje de las matemáticas, mostrando ideas para dar una dimensión más discursiva a las matemáticas.

A synthetic version of the metal-regulated gene A (mrgA) promoter from Bacillus subtilis, which in this Gram-positive bacterium is negatively regulated by manganese, iron, cobalt, or copper turned out to promote high level of basal gene expression that is further enhanced by Co(II), Cd(II), Mn(II), Zn(II), Cu(II), or Ni(II), when cloned in the Gram-negative bacterium Cupriavidus metallidurans. Promoter activity was monitored by expression of the reporter gene coding for the enhanced green fluorescent protein (EGFP), and cellular intensity fluorescence was quantified by flow cytometry. Expression levels in C. metallidurans driven by the heterologous promoter, here called pan, ranged from 20- to 53-fold the expression level driven by the Escherichia coli lac promoter (which is constitutively expressed in C. metallidurans), whether in the absence or presence of metal ions, respectively. The pan promoter did also function in E. coli in a constitutive pattern, regardless of the presence of Mn(II) or Fe(II). In conclusion, the pan promoter proved to be a powerful tool to express heterologous proteins in Gram-negative bacteria, especially in C. metallidurans grown upon high levels of toxic metals, with potential applications in bioremediation.
