Parkia platycephala lectin 2 was purified from Parkia platycephala (Leguminosae, Mimosoideae) seeds by affinity chromatography and RP-HPLC. Equilibrium sedimentation and MS showed that Parkia platycephala lectin 2 is a nonglycosylated monomeric protein of molecular mass 29 407 +/- 15 Da, which contains six cysteine residues engaged in the formation of three intramolecular disulfide bonds. Parkia platycephala lectin 2 agglutinated rabbit erythrocytes, and this activity was specifically inhibited by N-acetylglucosamine. In addition, Parkia platycephala lectin 2 hydrolyzed beta(1-4) glycosidic bonds linking 2-acetoamido-2-deoxy-beta-D-glucopyranose units in chitin. The full-lengthamino acid sequence of Parkia platycephala lectin 2, determined by N-terminal sequencing and cDNA cloning, and its three-dimensional structure, established by X-ray crystallography at 1.75 angstrom resolution, showed that Parkia platycephala lectin 2 is homologous to endochitinases of the glycosyl hydrolase family 18, which share the (beta alpha)(8) barrel topology harboring the catalytic residues Asp125, Glu127, and Tyr182.

Histamine release induced by plant lectins was studied with emphasis on the carbohydrate specificity, external calcium requirement, metal binding sites, and mast cell heterogeneity and on the importance of antibodies bound to the mast cell membrane to the lectin effect. Peritoneal mast cells were obtained by direct lavage of the rat peritoneal cavity and guinea pig intestine and hamster cheek pouch mast cells were obtained by dispersion with collagenase type IA. Histamine release was induced with concanavalin A (Con A), lectins from Canavalia brasiliensis, mannose-specific Cymbosema roseum, Maackia amurensis, Parkia platycephala, Triticum vulgaris (WGA), and demetallized Con A and C. brasiliensis, using 1-300 µg/ml lectin concentrations applied to Wistar rat peritoneal mast cells, peaking on 26.9, 21.0, 29.1, 24.9, 17.2, 10.7, 19.9, and 41.5%, respectively. This effect was inhibited in the absence of extracellular calcium. The lectins were also active on hamster cheek pouch mast cells (except demetallized Con A) and on Rowett nude rat (animal free of immunoglobulins) peritoneal mast cells (except for mannose-specific C. roseum, P. platycephala and WGA). No effect was observed in guinea pig intestine mast cells. Glucose-saturated Con A and C. brasiliensis also released histamine from Wistar rat peritoneal mast cells. These results suggest that histamine release induced by lectins is influenced by the heterogeneity of mast cells and depends on extracellular calcium. The results also suggest that this histamine release might occur by alternative mechanisms, because the usual mechanism of lectins is related to their binding properties to metals from which depend the binding to sugars, which would be their sites to bind to immunoglobulins. In the present study, we show that the histamine release by lectins was also induced by demetallized lectins and by sugar-saturated lectins (which would avoid their binding to other sugars). Additionally, the lectins also released histamine from Rowett nude mast cells that are free of immunoglobulins.

Studies were carried out using 96hr static toxicity bioassay to determine the effect of lethal concentrations of extracts from two local plants Tephrosia vogelii and Parkia clappertoniana which are known fish poison, on a species of mud fish. Clarias gariepinus Phytochemical analysis of the plant extracts was done and the extract from T. vogelii was found to contain alkaloids, tannins and flavonoids, while the extract from P. clappertoniana was formed to contain alkaloids tannins and saponins. Experimental fish were exposed to test water separately polluted by varying concentrations of extraction of both plant species ranging from 0.50mgl super(-1), 1.50mgl super(-1), 2.50mgl super(-1), 3.0mgl super(-1), 5.00mgl super(-1), 10.00mgl super(-1) in the case of T. vogelii and 5.00mgl super(-1), 7.50mgl super(-1), 10.00mgl super(-1), 15.00mgl super(-1), 20.00mgl super(-1) and 30.00mgl super(-1) in the case of P. clappertaniana. Behavioural hispathological and heamatological examinations were made. Both plant extracts were found to have lethal effects at the higher concentrations, affecting the gills and the central nervous system as well as having a depressive effect on the total count and increasing platelet and white blood cell count. Symptoms of toxicosis observed include, initial inactivation agitated swimming, tumbling movement air gulping, increased opercular beat and period of quiescence/knockdown before death. Marked differences were also observed in the hematological and histopathological analysis of poisoned fish. Lower concentrations of the extracts had sub lethal effects on the fish, which manifested as zigzag movement air gulping increased opercular movement etc. None of these effects were observed in the control experiment

Seed moisture content is significant in the handling and processing of seeds. This work therefore determined the physical properties of Locust bean seeds as functions of seed moisture content in the moisture range of 5.9 – 28.2% dry basis. Mohsenin, Stepanoff and ASAE standard methods were used in determining the properties. Increases in seed dimensions vitz length = 10.2±1.0 – 11.3±0.9 mm; width = 8.5±0.8 – 9.1±0.6 mm; surface area = 191.2±24.6 – 208.3±26.3 mm2 ; geometric mean diameter = 7.78±0.49 – 8.12±0.03 and arithmetic mean diameter = 8.06±0.56 – 8.34±0.49 mm were recorded. Seed thickness = 5.49±0.43 – 5.26±0.62 mm; sphericity = 0.75±0.04 – 0.71±0.03; true density = 1251.96±55.5 - 1222±62.16 kgm-3 and porosity = 48.4±2.14 – 41.9±3.78 decreased. Static coefficient of friction increased on plywood (0.5±0.02 – 0.6±0.01), glass (0.4±0.05 – 0.5±0.01) and decreased on aluminium (0.5±0.02 – 0.5±0.04). A data of the physical properties of Locust bean; Parkia biglobosa was developed. This is useful for the design and development of equipment necessary for its handling and processing.

As vespas sociais são predadoras de várias espécies de insetos e, portanto, o estudo de suas presas pode revelar seu potencial para programas de controle biológico de pragas. Durante o período de setembro de 2000 a janeiro de 2002, foram realizadas 70h de coleta de presas capturadas em doze ninhos de Polybia platycephala Richards, localizados em áreas urbanas do município de Juiz de Fora, MG. As presas capturadas por P. platycephala compreenderam cinco ordens de insetos: Diptera (33,4%), Lepidoptera (28,6%), Hemiptera (12,0%), Hymenoptera (9,4%) e Coleoptera (7,2%). O peso médio da carga protéica transportada pelas vespas foi 1,9 ±1,6 mg (n = 34, 0,3 - 6,2 mg), e a taxa média de proteína transportada por dia foi 22,8 mg. de acordo com os resultados, pode-se estimar a captura de 4.380 presas por ano por uma única colônia de P. platycephala. Desta forma, a espécie pode ser utilizada em programas de manejo em ambientes urbanos contribuindo para o controle de insetos pragas como larvas de pernilongos, lagartas desfolhadoras de plantas de jardins, pulgões e formas aladas de formigas.

Lectins are proteins important in various biological processes such as infection, cell differentiation and metastasis. The Parkia pendula lectin has been crystallized using the hanging-drop vapour diffusion method. X-ray diffraction data were collected using a Rigaku RU300 rotating anode generator and R-AXIS IV diffractometer. The cell parameters for P. pendula lectin are a=93.7 Angstrom b=161.1 Angstrom, c=80.0 Angstrom and space group C222. The maximum resolution was of 2.98 Angstrom. These data showed a R-sym=12.8%.

The present study investigates the chemical composition of the African plant Parkia biglobosa (Fabaceae) roots and barks by Liquid Chromatography - Electrospray Ionization and Direct Injection Tandem Mass Spectrometry analysis. Mass spectral data indicated that B-type oligomers are present, namely procyanidins and prodelphinidins, with their gallate and glucuronide derivatives, some of them in different isomeric forms. The analysis evidenced the presence of up to 40 proanthocyanidins, some of which are reported for the first time. In this study, the antiradical activity of extracts of roots and barks from Parkia biglobosa was evaluated using DPPH method and they showed satisfactory activities.

Os objetivos deste trabalho foram isolar, identificar e caracterizar a atividade alelopática de substâncias químicas produzidas por Parkia pendula. Os efeitos alelopáticos foram avaliados sobre a germinação de sementes e o desenvolvimento da radícula das plantas daninhas malícia (Mimosa pudica) e mata-pasto (Senna obtusifolia). O processo de isolamento das substâncias envolveu a extração com solvente em ordem crescente de polaridade, e a elucidação estrutural foi realizada via Ressonância Magnética Nuclear, espectro de COSY e de HETCOR. Os bioensaios foram desenvolvidos em condições controladas de 25 ºC de temperatura e fotoperíodo de 12 (germinação) e 24 horas (desenvolvimento da radícula). Foram isolados e identificados nas folhas da P. pendula os seguintes aleloquímicos: ácido 3,4,5-trimetoxibenzóico (S1), ácido 3,4-dimetoxibenzóico (S2) e o Blumenol A (S3). Comparativamente, S1 e S2 apresentaram maior atividade alelopática. Os efeitos promovidos sobre o desenvolvimento da radícula foram de maior magnitude do que aqueles verificados sobre a germinação das sementes. As substâncias isoladas mostraram baixo potencial inibitório da germinação das sementes, especialmente as sementes de mata-pasto. Os efeitos alelopáticos inibitórios estiveram positivamente associados à concentração das substâncias, embora em alguns casos esses efeitos não tenham correspondido às diferenças estatísticas.

The nutritional and amino acid analysis of raw and fermented seeds of Parkia biglobosa were carried out. Parameters investigated include moisture, crude protein, crude fat, ash, crude fibre and mineral contents; and the effect of the degree of fermentation on these parameters was also investigated. The amino acid compositions of all the samples were evaluated and amino acid quality determined by calculating amino acid scores and the predicted protein efficiency ratio (P-PER). Results showed that the proximate composition was significantly affected by fermentation, although there was little difference between the parameters for the partially fermented and completely fermented samples. Based on dry matter percentage, protein content was in the 39.77 – 43.74 % range while crude fibre ranged between 5.55 – 7.42 %. The ash content was lowest in the raw sample (2.34 %), while the fermented samples had ash contents between 4.27 and 8.33 % for the fully fermented and the partially fermented seeds, respectively. The fat content increased from 8.65 % in the raw seed to 24.4 % and 27.6 % for the partially and completely fermented samples, respectively. Results of the amino acid analysis showed that the partially fermented sample had the lowest quantities of all amino acids determined and had lysine as the limiting amino acid, whereas the raw and completely fermented samples had very similar amino acid profile with amino acid scores of 100, indicating that there are no limiting amino acids. All the samples were rich in essential amino acids. The P-PER also showed that the partially fermented sample had the lowest protein efficiency while the raw seed had the highest. Mineral contents generally increased from the raw, through the partially fermented, to the completely fermented seeds and results showed the samples to be good sources of potassium (K), calcium (Ca), manganese (Mn) and copper (Cu) in addition to being complementary sources of other metals. Locust bean seed does not accumulate lead and is, therefore, safe for consumption without the potential of food poisoning.
