The morphological characteristics of seedlings and the anatomical characteristics of vegetative organs and floral scape of P. hilairei (Eriocaulaceae) were analyzed to investigate the ecological and taxonomic implications for the species. P. hilairei is a perennial plant with an aerial stem and it grows in recently decomposed stony, sandy soils of the Serra do Cipo, in the State of Minas Gerais, Brazil. It is found both on wet and dry soils, exposed to constant winds, high light intensities and extreme daily fluctuations of temperature. The results show that the seeds have operculum and the initial seedling development stages are the same for other Eriocaulaceae. During the germination period, the undifferentiated axis of the embryo becomes apparent. The observation of the development of this axis, shows first, the growth of the leaves and later that of the adventitious root. Attention is focused on the apical stem region which presents sterelis lateral branching with adventitious roots in its base which will develop into paraclades (system of florescences). Both of them rise out of the pericycle and characterize its vegetative growth. The occurrence of a special leaf substomatal chamber may be related to an efficient protection for the gaseous exchange and may be considered an important characteristic of the Eriocaulaceae.

Macrófagos são conhecidos por exercerem uma importante função de mecanismo de defesa quando estas células são estimuladas com produtos naturais e produtos bacterianos (dentre outros). Uma variedade de citocinas e compostos químicos são liberados para induzir sistema de defesa fundamental. Entre outros peróxido de hidrogênio (H2O2) tem sido identificado como moléculas tendo multifunções. Entretanto está bem estabelecido que (H2O2) está envolvido em inúmeros processos fisiológicos, como por exemplo, neurotransmissão, relaxamento da musculatura lisa ou regulação imune. Para a determinação de peróxido de hidrogênio (H2O2) em macrófago peritoneal em camundongos, determinou-se a ação imunomodulatória de extratos (etanólico e etanólico 70%) obtidos de quatro espécies do gênero Paepalanthus (Eriocaulaceae) na concentração de 10 mg/mL. Os estratos etanólicos 70 % de capítulos de P. Hilairei, P. robustus, P. vellozioides e P. speciocus apresentaram maior liberação de (H2O2) do que os outros extratos etanólicos.

Paepalanthus subgenus Xeractis (Eriocaulaceae) comprises 28 recognized species endemic to the Espinhaco Range, in Minas Gerais state, Brazil. Most species of the subgenus are restricted to small localities and critically endangered, but still in need of systematic study. The monophyly of the subgenus has already been tested, but only with a few species. Our study presents the first phylogenetic hypothesis within the group, based on morphology. A maximum parsimony analysis was conducted on a matrix of 30 characters for 30 terminal taxa, including all species of the subgenus and two outgroups. The biogeographical hypotheses for the subgenus were inferred based on dispersal-vicariance analysis (DIVA). The analysis provided one most-parsimonious hypothesis that supports most of the latest published subdivisions (sections and series). However, some conflicts remain concerning the position of a few species and the relationships between sections. The distribution and origin(s) of microendemism are also discussed, providing the ground for conservation strategies to be developed in the region. In this article we describe and illustrate the new species Paepalanthus hirtellus (Eriocaulaceae, Paepalanthoideae). The species is, as far as known, restricted to Pico do Itambe State Park, in the state of Minas Gerais, Brazil. Paepalanthus hirtellus occurs on the campos rupestres of the Espinhaco Range. We compare it with P. lombensis and P. chrysophorus, the two morphologically most similar species. We provide additional comments on the morphological variability, habitat, geographic distribution and we provide its conservation status.

Paepalanthus is the largest genus in Eriocaulaceae, comprising about 400 species distributed mainly throughout the Neotropics. Through phylogenetic studies it has been demonstrated that the genus is polyphyletic, although many of its infrageneric categories are monophyletic. In an attempt to clarify the nomenclature and classification of Paepalanthus, we present a taxonomic survey of Paepalanthus section Diphyomene. This group consists of 10 species restricted to South America and is defined by inflorescences being arranged in the form of a tribotryum with terminal dibotryum, a terminal basic unit and pherophylls subtending the lateral dibotrya. Further important distinguishing characteristics are dimerous flowers, pistillate flowers with dolabriform sepals, bifid stigmatic branches and absent staminodes, and staminate flowers with an elongated anthophore. We hereby propose 19 new synonyms, six lectotypifications, one new status, one neotypification and one epitypification.

Paepalanthus sect. Diphyomene has inflorescences arranged in umbels. The underlying bauplan seems however to be more complex and composed of several distinct subunits. Despite appearing superficially very similar, the morphology and anatomy of the inflorescences can supply useful information for the understanding of the phylogeny and taxonomy of the group. Inflorescences of Paepalanthus erectifolius, Paepalanthus flaccidus, Paepalanthus giganteus, and Paepalanthus polycladus were analyzed in regard to branching pattern and anatomy. In P. erectifolius, P. giganteus and P. polycladus the structure is a tribotryum, with terminal dibotryum, and with pherophylls bearing lateral dibotrya. In P. flaccidus, the inflorescence is a pleiobotryum, with terminal subunit, and without pherophylls. Secondary inflorescences may occur in all species without regular pattern. Especially when grown in sites without a pronounced seasonality, the distinction between enrichment zone (part of the same inflorescence) and new inflorescences may be obscured. The main anatomical features supplying diagnostic and phylogenetic information are as follows: (a) in the elongated axis, the thickness of the epidermal cell walls and the cortex size; (b) in the bracts, the quantity of parenchyma cells (c) in the scapes, the shape and the presence of a pith tissue. Therefore, P. sect. Diphyomene can be divided in two groups; group A is represented by P. erectifolius, P. giganteus and P. polycladus, and group B is represented by P. flaccidus. The differentiation is based in both, inflorescence structure and anatomy. Group A presents a life cycle and anatomical features similar to species of Actinocephalus. Molecular trees also point that these two groups are closely related. However, inflorescence morphology and blooming sequence are different. Species of group B present an inflorescence structure and anatomical features shared with many genera and species in Eriocaulaceae. The available molecular and morphology based phylogenies still do not allow a precise allocation of the group in the bulk of basal species of Paepalanthus collocated in P. sect. Variabiles. Five new species of Paepalanthus section Diphyomene are described and illustrated: P. brevis, P. flexuosus, P. longiciliatus, P. macer, and P. stellatus. Paepalanthus brevis, similar to P. decussus, is easily distinguished by its short reproductive axis, and pilose and mucronate leaves. Paepalanthus flexuosus, morphologically related to P. urbanianus, possesses a distinctive short and tortuous reproductive axis. Paepalanthus longiciliatus, morphologically similar to P. weddellianus, possesses long trichomes on the margins of the reproductive axis bracts, considered a diagnostic feature. Paepalanthus macer shares similarities with P. amoenus, differing by its sulfurous capitula and adpressed reproductive axis bracts. Paepalanthus stellatus also has affinity with P. decussus, but possesses unique, membranaceous, reproductive-axis bracts and a punctual inner-capitulum arrangement of pistillate flowers. Four of the described species are narrowly distributed in the state of Goias, whereas P. brevis is endemic to Distrito Federal. All are considered critically endangered. Detailed comparisons of these species are presented in tables. Comments on phenology, distribution, habitat and etymology, along with an identification key, are provided.

The first isocoumarin isolated from the methylene chloride extract of Paepalanthus bromelioides, named paepalantine (isocoumarin 1), was found to have antimicrobial activity; but, it is mutagenic clastogenic and cytotoxic. Two other isocoumarins, paepalantine-9-O-beta-D-glucopyranoside (isocoumarin 2) and paepalantine-9-O-beta-D-allopyranosyl(1-->6) glucopyranoside (isocoumarin 3) were isolated from the ethanolic extract. A fourth new isocoumarin, also isolated from the methylene chloride extract of the capitula of P. bromelioides, was characterized as an 8-8' dimer of paepalantine and denominated isocoumarin 4. The abilities of isocoumarins 2, 3 and 4 to induce mutations in Salmonella typhimurium strains TA97a, TA98, TA100 and TA102 were investigated. Mutagenic activity was observed in strain TA97a treated with isocoumarin 2 in the presence of S9 mixture. The substitution of H at position 9 by glucose or glucose-allose caused reductions in the mutagenic activities of paepalantine, indicating this to be an important site for these properties. Quinones constitute an important class of naturally occurring compounds. They are found in plants, fungi and bacteria. Large number of quinones has been associated with antitumor, antibacterial, antimalarial and antifungal activities. In this work we describe the isolation, structure determination and the cytotoxic index of a new 1,4-naphthoquinone isolated from the capitula of Paepalanthus latipes.

A naphthopyranone dimer, named planifolin, was isolated from a methylene chloride extract of the capitula of Paepalanthus planifolius Koern. The molecule (C31H26O10) appeared to be made up of two monomeric portions, semi-vioxanthin and paepalantine (an isocoumarin), linked by an ether bond, and it may possess several kinds of biological activity that can be related to its polyphenolic structure. Short-term tests that detect genetic damage can afford the information needed to evaluate carcinogenic risks of chemicals to humans. The Ames test, recommended for testing the mutagenicity of chemical compounds with potential pharmacological application, was used in the present study. The mutagenic activity was evaluated in Salmonella typhimurium strains TA100, TA98, TA102 and TA97a and the cytotoxic effect in McCoy cells. The in vitro cytotoxicity of planifolin to McCoy cells, tested in microculture with neutral red, showed a significant cytotoxic index (CI50) of 12.83 mu g/mL. Planifolin showed mutagenic activity for TA100, TA98 and TA97a. The results indicate that this new naphthopyranone dimer causes mutations by substitution and by addition and deletion of bases in the sequence of DNA. Moreover, its mutagenic potential was increased by metabolic activation. (c) 2005 Elsevier Ltd. All rights reserved.

The isocoumarins (1-50 mu M) paepalantine (9,10-dihydroxy-5,7-dimethoxy-1H-naptho(2,3c)pyran-1-one), 8,8 '-paepalantine dimer, and vioxanthin isolated from Paepalanthus bromelioides, were assessed for antioxidant activity using isolated rat liver mitochondria and non-mitochondrial systems, and compared with the flavonoid quercetin. The paepalantine and paepalantine dimers, but not vioxanthin, were effective at scavenging both 1,1-diphenyl-2-picrylhydrazyl (DPPH*) and superoxide (O-2(-)) radicals in non-mitochondrial systems, and protected mitochondria from tert-butylhydroperoxide-induced H2O2 accumulation and Fe2+ -citrate-mediated mitochondrial membrane lipid peroxidation, with almost the same potency as quercetin. These results point towards paepalantine, followed by paepalantine dimer, as being a powerful agent affording protection, apparently via O-2(-) scavenging, from oxidative stress conditions imposed on mitochondria, the main intracellular source and target of those reactive oxygen species. This strong antioxidant action of paepalantine was reproduced in HepG2 cells exposed to oxidative stress condition induced by H2O2. (C) 2007 Elsevier Ltd. All rights reserved.
