Co-crystals of 4,4'-bipyridine and 4-hydroxybenzoic acid (1 : 2) show synthon polymorphism with the former being more stable. A 2 : 1 co-crystal is pseudopolymorphic within the same structural landscape with the structural roles of the two bipyridine N-atoms being distinct, as evidenced by mimicry by 4-phenylpyridine.

A range of novel chiral tellurium compounds having an azomethine functional group in the position ortho to tellurium has been synthesized by the reaction of the tellurium-containing aldehydes bis(o-formylphenyl) telluride (1) and o-(butyltelluro)benzaldehyde (4) with chiral amines (R)-(+)-(1-pheylethylamine) and (1R,2S)-(-)-norephedrine, respectively. The precursor aldehydes were prepared by using a reported procedure with slight but advantageous modifications. During the preparation of o-(butyltelluro)benzaldehyde, interesting side products, namely bis(o-formylphenyl) ditelluride ethylene acetal 5, bis(o-formylphenyl) tritelluride (6), and bis(o-formylphenyl) ditelluride (7) were isolated in moderate yields. The ditelluride 7 has been characterized by single-crystal X-ray diffraction studies. The liquid Schiff bases 10 and 11 were further characterized by derivatizing with liquid bromine. The title compound was obtained in excellent yield by reacting the Schiff base 11 with elemental bromine. Detailed NMR studies indicated the presence of a rigid environment for the hydroxyl group. Single-crystal X-ray determinations of the crystals obtained from the different batches indicated. the presence of the two pseudopolymorphic forms 13a and 13b, respectively. In the case of 13a there is one molecule of CH3CN as solvent of crystallization, whereas in 13b half a molecule of CH3CN per molecule of the title compound lies along the 2-fold axis. In 13a the hydroxyl hydrogen is hydrogen-bonded to the nitrogen of the solvent molecule, whereas in 13b it is hydrogen-bonded to the bromine of the neighboring molecule.

A dihydrate pseudopolymorph of bis(2,4-dihydroxyphenyl)methanone, C13H10O5 center dot 2H2O, (I), was obtained during polymorphism screening of hydroxybenzophenone derivatives. This structure, in which the molecule sits on a twofold axis, was compared with the known anhydrous form of (I) [Schlemper (1982). Acta Cryst. B38, 554559]. The role of water in the crystal assembly was established on the basis of the known monohydrate pseudopolymorph of 3,4-dihydroxybenzophenone [Landre, Souza, Correa, Martins & Doriguetto (2010). A utilização combinada de espectroscopia vibracional e de cálculos envolvendo a teoria do funcional de densidade (DFT) possibilita o estudo de ligações de hidrogénio em fase condensada, assim como a análise da estrutura molecular dos sistemas em estudo. Por um lado, a espectroscopia vibracional permite a detecção de associações moleculares, enquanto os métodos computacionais auxiliam na obtenção de informação referente aos mecanismos de associação, nomeadamente no que diz respeito à possível estrutura de dímeros e compostos de inclusão em ciclodextrinas e às energias de interacção e de inclusão. O estudo que originou a presente dissertação pretende contribuir para o reforço da aplicação de estudos espectroscópicos e computacionais na elucidação de diversos fenómenos químicos, com especial destaque para o papel desempenhado por interacções intermoleculares fracas na estrutura e propriedades de materiais moleculares. No âmbito desta tese foram investigados os seguintes tópicos: polimorfismo e pseudopolimorfismo em sólidos farmacêuticos, transições de fase em misturas binárias de ácidos gordos, inclusão em ciclodextrinas, interacção de compostos farmacêuticos com superfícies metálicas e formação de agregados de água em materiais híbridos orgânicos-inorgânicos. Os sistemas foram analisados utilizando a espectroscopia vibracional – particularmente a espectroscopia de difusão de Raman – como técnica fundamental. Para uma melhor caracterização de processos envolvendo transições de fase, foram efectuados estudos com variação de temperatura, variação de humidade relativa e substituição isotópica. O estudo da interacção com superfícies metálicas foi realizado por espectroscopia de Raman intensificada à superfície. Dada a complexidade dos sistemas em estudo, a informação obtida por espectroscopia vibracional foi complementada por resultados de cálculos mecânico-quânticos. Em particular, os cálculos DFT foram utilizados para a optimização de geometrias e previsão de frequências vibracionais de moléculas e associações moleculares, permitindo assim a análise e interpretação de espectros vibracionais e a caracterização da estrutura de materiais.

To optimize solubility of drugs, current strategies mainly focus on engineering and screening of smart crystal phases. Two salts of the anti-human immunodeficiency virus (HIV) drug lamivudinenamely, lamivudine hydrochloride and lamivudine hydrochloride monohydrate, were prepared in the course of screening the crystallization conditions of lamivudine duplex, an uncommon DNA-mimic, double-stranded helical structure made up of partially protonated drug pairs. Here, water solubilities of lamivudine hydrochloride, lamivudine hydrochloride monohydrate, and lamivudine duplex are reported. The aqueous solubility of this anti-HIV drug was significantly increased in both salts and also in lamivudine duplex in relation to the water solubility of lamivudine form II. In comparison with the lamivudine form II incorporated into therapeutic formulations, the drug solubility was increased at a temperature of 299 +/- 2 K by factors of 1.2, 3.3, and 4.5 in lamivudine hydrochloride, lamivudine hydrochloride monohydrate, and lamivudine duplex, respectively, demonstrating that this solid-state property of lamivudine can be improved by crystal engineering strategies. Solubility profiles were understood on the basis of structural and solventsolute interaction approaches. At last, correlations between solubility and crystal structures allowed for a rational approach to understand how this physicochemical feature could be enhanced by engineering new salts of the drug.
