Biocompatibility is a major challenge for successful application of many biomaterials. In this study the ability to coat chemically and enzymatically activated poly(L-lactic acid) (PLA) membranes with heat denatured human serum albumin to improve biocompatibility was investigated. PLA membranes hydrolyzed with NaOH or cutinase and then treated with 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide, hydrochloride (EDAC) as a heterobifunctional cross-linker promoted the coupling single bondCOOH groups on PLA membranes and single bondNH2 groups of heat denatured human serum albumin. This resulted in increased hydrophilicity (lowest water contact angles of 43° and 35°) and highest antioxidant activity (quenching of 79 μM and 115 μM tetramethylazobisquinone (TMAMQ) for NaOH and cutinase pretreated membranes, respectively). FTIR analysis of modified PLA membranes showed new peaks attributed to human serum albumin (amide bond, NH2 and side chain stretching) appearing within 3600–3000 cm−1 and 1700–1500 cm−1 (Fig. 3). MTT studies also showed that osteoblasts-like and MC-3T3-E1 cells viability increased 2.4 times as compared to untreated PLA membranes. The study therefore shows that this strategy of modifying the surfaces of PLA polymers could significantly improve biocompatibility.

This study investigated the microbial action in soil on poly(L-lactic acid) (PLLA) and polyvinyl chloride (PVC) films and a PLLA/PVC 7 : 3 blend, using Fourier transform infrared spectroscopy (FTIR), contact angle and scanning electron microscopy (SEM). The films (50 mu m) were obtained from the evaporation of dichloromethane solutions and buried in soil columns, in controlled conditions, for 120 days. The results showed that the surface of the PLLA films and blend became 18 and 31% more hydrophilic, respectively. The morphology of the films also changed after 120 days of microbial treatment, particularly that of the PLLA phase in the blend, confirmed by structural and conformational changes in the FTIR CO region at 12001000 cm1 and an increase in the relative intensity of the band at 1773 cm1, which was attributed to C O group vibration due to a rotational isomer in the interlamellar region (semi-ordered region). Created periodontal defects in dogs were randomly assigned for experimental (Guidor bioresorbable membranes) or control (conventional therapy) treatment the results showed that the new connective issue attachment was significantly greater in test sites than in controls. This new attachment averaged 2.79 +/- 0.74 mm and 1.47 +/- 0.20 mm at test and control sites, respectively (P < 0.05). Epithelial downgrowth was also reduced in the test sites (P < 0.05). No differences in bone response were found. The bioresorbable barrier was effective in blocking gingival epithelial downgrowth and connective tissue proliferation, promoting new attachment according to the principles of guided tissue regeneration.

Polylactic acid (PLA) is a bio-derived, biodegradable polymer with a number of similar mechanical properties to commodity plastics like polyethylene (PE) and polyethylene terephthalate (PETE). There has recently been a great interest in using PLA to replace these typical petroleum-derived polymers because of the developing trend to use more sustainable materials and technologies. However, PLA¿s inherent slow crystallization behavior is not compatible with prototypical polymer processing techniques such as molding and extrusion, and in turn inhibits its widespread use in industrial applications. In order to make PLA into a commercially-viable material, there is a need to process the material in such a way that its tendency to form crystals is enhanced. The industry standard for producing PLA products is via twin screw extrusion (TSE), where polymer pellets are fed into a heated extruder, mixed at a temperature above its melting temperature, and molded into a desired shape. A relatively novel processing technique called solid-state shear pulverization (SSSP) processes the polymer in the solid state so that nucleation sites can develop and fast crystallization can occur. SSSP has also been found to enhance the mechanical properties of a material, but its powder output form is undesirable in industry. A new process called solid-state/melt extrusion (SSME), developed at Bucknell University, combines the TSE and SSSP processes in one instrument. This technique has proven to produce moldable polymer products with increased mechanical strength. This thesis first investigated the effects of the TSE, SSSP, and SSME polymer processing techniques on PLA. The study seeks to determine the process that yields products with the most enhanced thermal and mechanical properties. For characterization, percent crystallinity, crystallization half time, storage modulus, softening temperature, degradation temperature and molecular weight were analyzed for all samples. Through these characterization techniques, it was observed that SSME-processed PLA had enhanced properties relative to TSE- and SSSP-processed PLA. Because of the previous findings, an optimization study for SSME-processed PLA was conducted where throughput and screw design were varied. The optimization study determined PLA processed with a low flow rate and a moderate screw design in an SSME process produced a polymer product with the largest increase in thermal properties and a high retention of polymer structure relative to TSE-, SSSP-, and all other SSME-processed PLA. It was concluded that the SSSP part of processing scissions polymer chains, creating defects within the material, while the TSE part of processing allows these defects to be mixed thoroughly throughout the sample. The study showed that a proper SSME setup allows for both the increase in nucleation sites within the polymer and sufficient mixing, which in turn leads to the development of a large amount of crystals in a short period of time.

Dielectric barrier discharge (DBD) air plasma is a novel technique for in-package decontamination of food, but it has not been yet applied to the packaging material. Characterization of commercial polylactic acid (PLA) films was done after in-package DBD plasma treatment at different voltages and treatment times to evaluate its suitability as food packaging material. DBD plasma increased the roughness of PLA film mainly at the site in contact with high voltage electrode at both the voltage levels of 70 and 80 kV. DBD plasma treatments did not induce any change in the glass transition temperature, but significant increase in the initial degradation temperature and maximum degradation temperature was observed. DBD plasma treatment did not adversely affect the oxygen and water vapor permeability of PLA. A very limited overall migration was observed in different food simulants and was much below the regulatory limits. Industrial relevance: In-package DBD plasma is a novel and innovative approach for the decontamination of foods with potential industrial application. This paper assesses the suitability of PLA as food packaging material for cold plasma treatment. It characterizes the effect of DBD plasma on the packaging material when used for in-package decontamination of food. The work described in this research offers a promising alternative to classical methods used in fruit and vegetable industries where in-package DBD plasma can serve as an effective decontamination process and avoids any post-process recontamination or hazards from the package itself.

Poly(lactic acid) PLA, and poly(hydroxybutyrate) PHB, blends were processed as films and characterized for their use in food packaging. PLA was blended with PHB to enhance the crystallinity. Therefore, PHB addition strongly increased oxygen barrier while decreased the wettability. Two different environmentally-friendly plasticizers, poly(ethylene glycol) (PEG) and acetyl(tributyl citrate) (ATBC), were added to these blends to increase their processing performance, while improving their ductile properties. ATBC showed higher plasticizer efficiency than PEG directly related to the similarity solubility parameters between ATBC and both biopolymers. Moreover, ATBC was more efficiently retained to the polymer matrix during processing than PEG. PLA–PHB–ATBC blends were homogeneous and transparent blends that showed promising performance for the preparation of films by a ready industrial process technology for food packaging applications, showing slightly amber color, improved elongation at break, enhanced oxygen barrier and decreased wettability.

Cette étude est destinée à la production et à la caractérisation des composites d’acide polylactique (PLA) et des fibres naturelles (lin, poudre de bois). Le moussage du PLA et ses composites ont également été étudiés afin d’évaluer les effets des conditions de moulage par injection et du renfort sur les propriétés finales de ces matériaux. Dans la première partie, les composites constitués de PLA et des fibres de lin ont été produits par extrusion suivit par un moulage en injection. L’effet de la variation du taux de charge (15, 25 et 40% en poids) sur les caractéristiques morphologique, mécanique, thermique et rhéologique des composites a été évalué. Dans la deuxième étape, la poudre de bois (WF) a été choisie pour renforcer le PLA. La préparation des composites de PLA et WF a été effectuée comme dans la première partie et une série complète de caractérisations morphologique, mécanique, thermique et l’analyse mécanique dynamique ont été effectués afin d’obtenir une évaluation complète de l’effet du taux de charge (15, 25 et 40% en poids) sur les propriétés du PLA. Finalement, la troisième partie de cette étude porte sur les composites de PLA et de renfort naturel afin de produire des composites moussés. Ces mousses ont été réalisées à l’aide d’un agent moussant exothermique (azodicarbonamide) via le moulage par injection, suite à un mélange du PLA et de fibres naturelles. Dans ce cas, la charge d’injection (quantité de matière injectée dans le moule: 31, 33, 36, 38 et 43% de la capacité de la presse à injection) et la concentration en poudre de bois (15, 25 et 40% en poids) ont été variées. La caractérisation des propriétés mécanique et thermique a été effectuée et les résultats ont démontré que les renforts naturels étudiés (lin et poudre de bois) permettaient d’améliorer les propriétés mécaniques des composites, notamment le module de flexion et la résistance au choc du polymère (PLA). En outre, la formation de la mousse était également efficace pour le PLA vierge et ses composites car les masses volumiques ont été significativement réduites.

The search for an ideal filler for soft tissue augmentation still continues. Because aging changes are continuous, temporary fillers should be preferred against permanent ones. Since 1999, the poly-L-lactic acid filler (PLA) has been marketed in Europe as Newfill. As a synthetic biocompatible polymer, PLA originally was used in suture materials and screws. In 2004, the U.S. Food and Drug Administration approved PLA under the name of Sculptra for the treatment of human immunodeficiency virus-related facial lipoatrophy. This study aimed to evaluate a 3-year follow-up investigation into the effect of PLA implant injection for the treatment of sunken nasolabial folds. Between October 2003 and February 2004, 10 women with a median age of 54 years (range, 43-60 years) were injected with polylactic acid hydrogel (Newfill) in the nasolabial fold area for aesthetic reasons. All the patients underwent three injections: one injection per month for 3 months. Evaluation of the results based on clinical examination and photography was performed at each session, at 6 months, and then 36 months after the third session. Injectable PLA was able to correct nasolabial folds successfully with a more lasting result than absorbable fillers commonly used in clinical practice, such as hyaluronic acid and collagen. Careful and standardized photographic documentation is indispensable.

Commercial stents, especially metallic ones, present several disadvantages, and this gives rise to the necessity of producing or coating stents with different materials, like natural polymers, in order to improve their biocompatibility and minimize the disadvantages of metallic ones. This review paper discusses some applications of natural-based polymers in stents, namely polylactic acid (PLA) for stent development and chitosan for biocompatible coatings of stents . Furthermore, some effective stent functionalization techniques will be discussed, namely Layer by Layer (LBL) technique.

Aim Primary implant stability can be compromised by overdrilling of the implant bed. Filling the gap between the implant and the bone with a highly viscous copolymer of polylactic and polyglycolic acid (PLA/PGA) might stabilize the implant and thus supply osseointegration. The aim of this study was to evaluate implants installed in overdrilled beds associated with PLA/PGA in rats tibia model by means of removal torque test and fluorochrome analysis. Materials and methods For this experiment two groups were selected: in the test group 0.4 mm overdrilled defects (2.0 in diameter and 3 mm long ) were produced in the right tibia of seven rats and implants were placed covered with PLA/PGA biomaterial to fill the gap; the control group was not overdrilled and the implants were placed without the biomaterial. Implants of 1.6 mm in diameter and 3 mm long where placed into all defects. Calcein, alizarin and oxytetracyclin were injected at 7, 15 and 21 postoperative days, respectively, and the animals were sacrificed at 35 postoperative day. Results The results showed that all the implants achieved osseointegration. There were no statistical significance differences in torque-reverse and fluorocrome analysis (P>0.05). Conclusion We can conclude that overdrilled defects filled with PLA/PGA did not disturb osseointegration in this experimental model. © ARIESDUE.

The in vitro and in vivo degradation properties of poly(lactic-co-glycolic acid) (PLGA) scaffolds produced by two different technologies-therm ally induced phase separation (TIPS), and solvent casting and particulate leaching (SCPL) were compared. Over 6 weeks, in vitro degradation produced changes in SCPL scaffold dimension, mass, internal architecture and mechanical properties. TIPS scaffolds produced far less changes in these parameters providing significant advantages over SCPL. In vivo results were based on a microsurgically created arteriovenous (AV) loop sandwiched between two TIPS scaffolds placed in a polycarbonate chamber under rat groin skin. Histologically, a predominant foreign body giant cell response and reduced vascularity was evident in tissue ingrowth between 2 and 8 weeks in TIPS scaffolds. Tissue death occurred at 8 weeks in the smallest pores. Morphometric comparison of TIPS and SCPL scaffolds indicated slightly better tissue ingrowth but greater loss of scaffold structure in SCPL scaffolds. Although advantageous in vitro, large surface area:volume ratios and varying pore sizes in PLGA TIPS scaffolds mean that effective in vivo (AV loop) utilization will only be achieved if the foreign body response can be significantly reduced so as to allow successful vascularisation, and hence sustained tissue growth, in pores less than 300 mu m. As crescentes preocupações ambientais e a necessidade de um desenvolvimento sustentável tem proporcionado um grande interesse no estudo e desenvolvimento de materiais mais ecológicos e amigos do ambiente. No caso particular da indústria dos materiais compósitos, a utilização de fibras naturais de origem vegetal, em substituição das tradicionais fibras de vidro, tem aumentado significativamente nos últimos anos. Para além dos claros benefícios ecológicos, a utilização de fibras naturais em detrimento das fibras de vidro, possibilita também o fabrico de componentes com baixo peso, boas propriedades mecânicas, baixo custo, pouco abrasivos às ferramentas de produção e com boas propriedades de isolamento térmico e acústico. Contudo, existem também algumas limitações decorrentes da utilização de fibras naturais como reforço de materiais poliméricos, como exemplo, a possibilidade de emitirem odores e absorverem água, a falta de adesão entre as fibras e as matrizes e o facto de possuírem baixa resistência à temperatura. No presente trabalho, foram estudadas e analisadas as propriedades mecânicas de laminados de matriz termoendurecível de epóxido e poliéster, reforçados com várias camadas de tecido bidireccional de fibras de juta. Para além dos referidos laminados, foram também produzidos e estudados compósitos de matriz termoplástica biodegradável de PLA (ácido poliláctico), reforçados com fibras curtas de juta. Todos os compósitos produzidos foram sujeitos a ensaios de tracção e flexão e as suas propriedades foram comparadas. O tecido de juta utilizado como reforço dos compósitos fabricados, foi caracterizado através de vários ensaios, utilizados tipicamente na indústria têxtil. As propriedades extraídas destes ensaios, foram úteis para a previsão das propriedades mecânicas dos materiais compósitos fabricados.Por fim, foi realizada uma análise critica sobres todos os resultados extraídos dos ensaios efectuados.

Lipid nanocapsules (NCs) represent promising tools in clinical practice for diagnosis and therapy applications. However, the NC appropriate functionalization is essential to guarantee high biocompatibility and molecule loading ability. In any medical application, the immune system-impact of differently functionalized NCs still remains to be fully understood. A comprehensive study on the action exerted on human peripheral blood mononuclear cells (PBMCs) and major immune subpopulations by three different NC coatings: pluronic, chitosan and polyethylene glycol-polylactic acid (PEG) is reported. After a deep particle characterization, the uptake was assessed by flow-cytometry and confocal microscopy, focusing then on apoptosis, necrosis and proliferation impact in T cells and monocytes. Cell functionality by cell diameter variations, different activation marker analysis and cytokine assays were performed. We demonstrated that the NCs impact on the immune cell response is strongly correlated to their coating. Pluronic-NCs were able to induce immunomodulation of innate immunity inducing monocyte activations. Immunomodulation was observed in monocytes and T lymphocytes treated with Chitosan-NCs. Conversely, PEG-NCs were completely inert. These findings are of particular value towards a pre-selection of specific NC coatings depending on biomedical purposes for pre-clinical investigations; i.e. the immune-specific action of particular NC coating can be excellent for immunotherapy applications.

Cette thèse rapporte l’étude des propriétés physicochimiques des nanoparticles polymériques et leur impact sur l’interaction avec les cellules vivantes. Nous nous sommes tout spécialement attachés à étudier l’effet des propriétés adhésives et mécaniques des nanoparticules sur leur capacité de pénétration de la membrane cellulaire. Pour ce faire, nous avons tout d’abord utilisé des nanoparticules d’acide polylactique (PLA) fonctionnalisées en surface avec un ligand des sélectines E et P. Le greffage du ligand sur la particule s’est fait par une nouvelle méthode expérimentale garantissant la présence du ligand à la surface de la particule durant toute sa durée de vie. Cette méthode consiste à mélanger un polymère fonctionnalisé avec le ligand avec un autre polymère non fonctionnalisé. La présence du ligand à la surface des nanoparticules formées à partir de ce mélange de polymères a été confirmée par analyse ToF SIMS. Nous avons pu prouver que les particules possédant le ligand greffé à leur surface démontraient une capacité adhésive supérieure à leurs homologues non fonctionnalisés sur des cellules endothéliales HUVEC activées par différentes drogues. De plus, le captage des particules par les cellules HUVEC est modulé par le niveau d’expression des récepteurs selectine E et P et aussi par la quantité de ligand libre. Ces résultats montrent clairement que le greffage du ligand confère aux particules des propriétés adhésives accrues et spécifiques ce qui permet leur usage postérieure comme vecteur pharmaceutique capable de cibler un récepteur particulier à la surface d’une cellule. Nous avons aussi démontré que l’interaction entre les nanoparticules et la membrane cellulaire peut aussi être contrôlée aussi bien par les propriétés mécaniques de la cellule que de la nanoparticule. Dans une première étape, nous avons mesuré à l’aide de l’appareil de forces de surface l’élasticité de cellules macrophagiques déposées sur différents substrats. En contrôlant l’interaction entre la cellule et le substrat sur lequel elle repose nous avons montré qu’il était possible de modifier à ii volonté les propriétés mécaniques cellulaire. Une augmentation de l’élasticité cellulaire s’accompagne d’une augmentation systématique de l’internalisation de nanoparticules de PLA non fonctionnalisées. Ceci suggère un rôle prépondérant des propriétés mécaniques du cortex cellulaire dans le captage des nanoparticules de PLA. Dans une seconde étape, nous avons étudié l’effet des propriétés mécaniques des nanoparticules sur leur capacité de pénétration cellulaire. Pour ce faire, nous avons synthétisé des particules d’hydrogel dont l’élasticité était contrôlée par le degré d’agent réticulant inclus dans leur formulation. Le contrôle des propriétés mécaniques des nanoparticules a été confirmé par la mesure du module de Young des particules par microscopie de force atomique. L’impact des propriétés mécaniques de ces particules sur leur capacité de pénétration dans les cellules vivantes a été étudié sur des cellules macrophagiques de souris. Les résultats ont montré que la cinétique d’internalisation, la quantité de particules internalisées et le mécanisme d’internalisation dépendent tous du module de Young des nanoparticules. Aucune différence dans le trajet intracellulaire des particules n’a pu être observée malgré le fait que différentes voies d’internalisation aient été observées. Ce dernier résultat peut s’expliquer par le fait que les nanoparticules sont internalisées par plusieurs voie simultanément ce qui facilite leur accumulation dans les organelles digestives intracellulaires. Un modèle simple permettant d’expliquer ces résultats a été proposé et discuté.
