Estudos relacionados à morfo-anatomia de frutos e sementes de espécies florestais são relevantes para a compreensão de vários fenômenos ligados ao comportamento dessas espécies. O presente trabalho é uma contribuição ao estudo das características morfológicas e anatômicas de sementes de Podocarpus lambertii Klotz. e Podocarpus sellowii Klotz.. Para as observações do desenvolvimento dos estróbilos, foram efetuadas visitas quinzenais às áreas de coleta, a partir do início da formação dos estróbilos até a maturação das sementes. A cada visita, foram retiradas amostras para análises de microscopia estereoscópica e fotônica. O estróbilo feminino de P. lambertii e P. sellowii é constituído pela semente e epimácio, presos a um delgado pedúnculo. As sementes de P. lambertii e P. sellowii são globosas, possuem tegumento com estrutura espessa, constituído de três camadas celulares. O endosperma é abundante e de consistência gelatinosa; o embrião é do tipo linear e cotiledonar.

Leaf and wood plasticity are key elements in the survival of widely distributed plant species. Little is known, however, about variation in stomatal distribution in the leaf epidermis and its correlation with the dimensions of conducting cells in wood. This study aimed at testing the hypothesis that Podocarpus lambertii, a conifer tree, possesses a well-defined pattern of stomatal distribution, and that this pattern can vary together with the dimensions of stem tracheids as a possible strategy to survive in climatically different sites. Leaves and wood were sampled from trees growing in a cold, wet site in south-eastern Brazil and in a warm, dry site in north-eastern Brazil. Stomata were thoroughly mapped in leaves from each study site to determine a spatial sampling strategy. Stomatal density, stomatal index and guard cell length were then sampled in three regions of the leaf: near the midrib, near the leaf margin and in between the two. This sampling strategy was used to test for a pattern and its possible variation between study sites. Wood and stomata data were analysed together via principal component analysis. The following distribution pattern was found in the south-eastern leaves: the stomatal index was up to 25 higher in the central leaf region, between the midrib and the leaf margin, than in the adjacent regions. The inverse pattern was found in the north-eastern leaves, in which the stomatal index was 10 higher near the midrib and the leaf margin. This change in pattern was accompanied by smaller tracheid lumen diameter and length. Podocarpus lambertii individuals in sites with higher temperature and lower water availability jointly regulate stomatal distribution in leaves and tracheid dimensions in wood. The observed stomatal distribution pattern and variation appear to be closely related to the placement of conducting tissue in the mesophyll.

Here we propose a protocol for embryogenic cultures induction, proliferation and maturation for the Brazilian conifer Podocarpus lambertii, and investigated the effect of abscisic acid (ABA) and glutathione (GSH) supplementation on the maturation phase. ABA, zeatin (Z) and salicylic acid (SA) endogenous levels were quantified. Number of somatic embryos obtained in ABA-supplemented treatment was signifi- cant higher than in ABA-free treatment, showing the relevance of ABA supplementation during somatic embryos maturation. Histological analysis showed the stereotyped sequence of developmental stages in conifer somatic embryos, reaching the late torpedo-staged embryo. GSH supplementation in maturation culture medium improved the somatic embryos number and morphological features. GSH 0 mM and GSH 0.1 mM treatments correlated with a decreased ABA endogenous level during maturation, while GSH 0.5 mM treatment showed constantlevels. Alltreatments resulted in decreased Z endogenous levels, supporting the concept that cytokinins are important during the initial cell division but not for the later stages of embryo development. The lowest SA levels found in GSH 0.5 mM treatment were coincident with early embryonic development, and this treatment resulted in the highest development of somatic embryos. Thus, a correlation between lower SA levels and improved somatic embryo formation can be hypothesized

The Atlantic rainforest has the second highest biodiversity in Brazil. It has been shrinking rapidly in area as a result of intensive deforestation, and only 7% of the original cover now remains, as isolated patches or in ecological reserves. In order to obtain new information on the distribution of the Atlantic rainforest during the Quaternary, we examined herbarium data to locate relevant populations and extracted DNA from fresh leaves from 26 populations. The present-day distribution of endemic Podocarpus populations shows that they are widely dispersed across eastern Brazil, and that the expansion of Podocarpus recorded in single Amazonian pollen records may have originated from either western or eastern populations. Genetic analysis enabled us to determine the boundaries of their regional expansion: northern and central populations of P. sellowii appeared between 5 degrees and 15 degrees S some 16,000 years ago; populations of P lambertii or sellowii have appeared between 15 degrees and 23 degrees S at different times since the last glaciation at least; and P lambertii appeared between 23 degrees and 30 degrees S during the recent expansion of Araucaria forests. The combination of botanical, pollen, and molecular analyses proved to be a rapid means of inferring distribution boundaries for sparse populations and their regional evolution within tropical ecosystems. Today the rainforest refugia we identified have become hotspots that are crucial to the survival of the Atlantic forest under unfavourable climatic conditions and, as such, offer the only possible opportunity for this type of forest to expand in the event of future climate change.

In Ergaenzung des 1. Teils des Katalogs der Baumpollen Sued-brasiliens werden die drei einzigen hier vorkommenden Gymnospermaearten beschrieben: Araucaria angustifolia, Podocarpus sellowii und P. lamberti. Die erste besitzt runde, mit zwei Ausbuchtungen versehene Pollen und eine komplex gebaute Exine. Beide Podocarpusarten aehneln einander sehr. Bezueglich ihrer Groesse koennen sie nicht voneinander geschieden werden. Die beste Art der Unterscheidung besteht in der Oberflaechenstruktur der Pollenkoerper: P. sellowii hat eine feine, engmaschige, anaehernd netzartige oberflaeche (entsprechend dem "reticulate-ornate" = Typ), waehrend P. lamberti grobe Erhebungen (entsprechend dem "ornate" = typ) vorweist.

(Aspectos ecológicos de um trecho de floresta de brejo em Itatinga, SP: florística, fitossociologia e seletividade de espécies). No presente trabalho estudou-se um trecho de floresta de brejo localizada na fazenda Santa Irene, de propriedade da EUCATEX Ltda., no município de Itatinga-SP (48º38'52,2"W e 23º17'41,6"S), com uma área de 2 ha. O levantamento fitossociológico foi realizado através do método de censo de 1 ha do remanescente florestal, onde foram amostrados todos os indivíduos com PAP (perímetro à altura do peito) > a 15 cm. Foram amostrados 1310 indivíduos, pertencentes a 28 famílias, 34 gêneros e 39 espécies do estrato arbustivo-arbóreo. As famílias que apresentaram os maiores valores de IVC foram Clusiaceae (53,49), Burseraceae (41,30), Euphorbiaceae (31,42). Podocarpaceae (9,04), Caesalpiniaceae (6,99), Myrsinaceae (6,38), Melastomataceae (5,75), Anacardiaceae (5,70), Arecaceae (4,51) e Styracaceae (4,44). Estas 10 famílias somaram 74,50% do IVC total. As espécies de maior destaque em IVC foram Calophyllum brasiliense (53,49), Protium almecega (41,30), Pera obovata (14,46), Podocarpus sellowii (9,04), Hyeronima alchorneoides (8,68), Copaifera langsdorffii (6,99), Rapanea guianensis (5,72), Tapirira guianensis (5,70), Miconia ligustroides (5,62) e Sebastiania serrata (4,71). Tais espécies somadas perfazem 77,84% do IVC total. O índice de diversidade de Shannon (H') obtido foi de 2,751 nats/indivíduos. Baseado em critérios florísticos e fitossociológicos, as espécies amostradas foram divididas em dois grupos: a) espécies peculiares, com os subgrupos de peculiares exclusivas e não exclusivas e b) complementares, com os subgrupos de áreas secas e indiferentes.

MP [4-(3′,3′-dimethylallyloxy)-5-methyl-6-methoxyphthalide] was obtained from liquid culture of Pestalotiopsis photiniaeisolated from the Chinese Podocarpaceae plant Podocarpus macrophyllus. MP significantly inhibited the proliferation of HeLa tumor cell lines. After treatment with MP, characteristic apoptotic features such as DNA fragmentation and chromatin condensation were observed in DAPI-stained HeLa cells. Flow cytometry showed that MP induced G1 cell cycle arrest and apoptosis in a dose-dependent manner. Western blotting and real-time reverse transcription-polymerase chain reaction were used to investigate protein and mRNA expression. MP caused significant cell cycle arrest by upregulating the cyclin-dependent kinase inhibitor p27KIP1 protein and p21CIP1 mRNA levels in HeLa cells. The expression of p73 protein was increased after treatment with various MP concentrations. mRNA expression of the cell cycle-related genes, p21CIP1, p16INK4a and Gadd45α, was significantly upregulated and mRNA levels demonstrated significantly increased translation ofp73, JunB, FKHR, andBim. The results indicate that MP may be a potential treatment for cervical cancer.

Inter-bedded volcanic and organic sediments from Erazo (Ecuador) indicate the presence of four different forest assemblages on the eastern Andean flank during the middle Pleistocene. Radiometric dates (40Ar–39Ar) obtained fromthe volcanic ash indicate that deposition occurred between 620,000 and 192,000 years ago. Examination of the organic sediment composition and the fossil pollen, wood and charcoal it contains provides insight into depositional environment, vegetation assemblage and fire history. The high organic content and abundance of macro fossils found throughout the sediment suggest that during the period of deposition the local environment was either a swamp or a shallow water body. The correlation of fire activity (peaks in charcoal abundance) with volcanic ash deposits through most of the record suggests that volcanoes were the main source of ignition. The low abundance of grass (typically b10%) throughout the sedimentary sequence along with the low abundance of other taxa indicative of open vegetation suggests the persistence of forest at Erazo. Four types of forest assemblage were identified (with the first taxa as the most dominant): i) Alnus-Arecaceae, ii) Miconia- Melastomataceae/Combretaceae-Moraceae/Urticaceae, iii) Arecaceae-Alnus, and iv) Podocarpus with Oreopanax sp. and Melastomataceae/Combretaceae. Changes in the forest floristic composition indicate high vegetation turnover and reassortment of taxa between upper and lower montane forests during the middle Pleistocene as well as the persistence of forest cover.

Puyasena et al. question our interpretation of climate-driven vegetation change on the Andean flank in western Amazonia during the middle Pleistocene and suggest that the use of Podocarpus spp. as a proxy of past climate change should be reassessed. We defend our assertion that vegetation change at the Erazo study site was predominantly driven by climate change due to concomitant changes recorded by multiple taxa in the fossil record.

A reconstruction of past environmental change from Ecuador reveals the response of lower montane forest on the Andean flank in western Amazonia to glacial-interglacial global climate change. Radiometric dating of volcanic ash indicates that deposition occurred ~324,000 to 193,000 years ago during parts of Marine Isotope Stages 9, 7, and 6. Fossil pollen and wood preserved within organic sediments suggest that the composition of the forest altered radically in response to glacial-interglacial climate change. The presence of Podocarpus macrofossils ~1000 meters below the lower limit of their modern distribution indicates a relative cooling of at least 5°C during glacials and persistence of wet conditions. Interglacial deposits contain thermophilic palms suggesting warm and wet climates. Hence, global temperature change can radically alter vegetation communities and biodiversity in this region.

This article documents the addition of 512 microsatellite marker loci and nine pairs of Single Nucleotide Polymorphism (SNP) sequencing primers to the Molecular Ecology Resources Database. A Wolbachiapertence a um grupo de bactérias intracelulares, transmitidas maternalmente, que se encontram amplamente distribuídas nos artrópodes. Estes endossimbiontes encontram-se normalmente nos tecidos do sistema reprodutor dos artrópodes e têm a capacidade manipular a sua reproduçãode modo a garantir a sua transmissão à descendência e rápida dispersão na população.A capacidade de manipulação reprodutiva da Wolbachia tornou-a o alvo de diversos estudos para uma maior e melhor perceção da sua implicação em processos biológicos e evolutivos e por acreditar-se que esta bactéria é uma promissora ferramenta no controlo de populações de insetos que são pragas agrícolas. Os afídeos são um grupo de insetos associados às plantas que podem ter um efeito devastador nas culturas agrícolas e hortícolas pois não só retiram nutrientes às plantas como podem ser vetores de doenças. Embora durante muito tempo se pensasse que estes insetos não albergavam a Wolbachia estudos recentes mostram que são várias as espécies de afídeos infetados com esta bactéria. O principal objetivo deste trabalho é estudar a prevalência de infeção por Wolbachia assim como a caracterização das suas estirpes em amostras de afídeos dos Arquipélagos da Madeira e dos Açores. Neste estudo foram analisadas 545 amostras de afídeos, 361 provenientes do Arquipélago da Madeira e 184 dos Açores. Utilizando a técnica da “Polymerase Chain Reaction” (reação em cadeia da polimerase) amplificou-se o gene 16S rRNA (RNA ribossomal) e verificou-se que 32 destas amostras encontravam-se infetadas com Wolbachia sendo a maior parte das amostras infetadas provenientes dos Açores. Para determinar a estirpe que infeta estes afídeos utilizou-se a tipagem sequencial multilocus (MLST) com os genesglutamil-tRNA amidotransferase, subunidade B (gatB), citocromo c oxidase, subunidade I (coxA), proteína hipotética conservativa (hcpA) e proteína da divisão celular (ftsZ). A análise filogenética realizada para os diferentes genes mostrou que grande parte das amostras analisadas estão incluídas em dois dos novos supergrupos descobertos para Wolbachia, supergrupo M e N.Foi detetada a presença da mesma estirpe de Wolbachia, supergrupo N, em duas espécies diferentes de afídeos, Neophyllaphis podocarpi e Aphis spiraecola da mesma planta hospedeira, Podocarpus macrophyllus. Esta infeção reforça a ideia de que a Wolbachia não recorre só a transmissão vertical para se difundir na população mas utiliza também a transmissão horizontal. A deteção e caracterização das estirpes de Wolbachia é essencial para um maior entendimento sobre a sua origem e forma de disseminação. Esta informação é importante para desenvolvimento de estratégias de controlo de pestes recorrendo a estes endossimbiontes.

A dinâmica da vegetação oeste da Amazônia durante os últimos 42000 anos AP foi estudada por pólen, fácies sedimentares, datação por 14C, δ13C e C/N. Dois testemunhos foram coletados, o primeiro próximo da cidade de Humaitá no sul do Estado do Amazonas e o segundo nas proximidades de Porto velho, norte de Rondônia. Os pontos de amostragem estão localizados em uma região coberta por campos naturais e vegetação de floresta tropical, respectivamente. Os sedimentos depositados são predominantemente compostos por areia compacta, lama e areia com estratificação heterolítica, lama laminada e compacta representando sedimentos acumulados em um canal ativo, planície aluvial e lago em ferradura representando os ambientes sedimentares. Nessa configuração, as condições subaquáticas são desenvolvidas em um ambiente de baixa energia, favorecendo localmente a preservação da comunidade de grãos de pólen de vegetação herbácea e de floresta glacial representado principalmente por Alnus, Drymis, Hedyosmum, Podocarpus e Weinmannia com ocorrência entre > 42.033 – 43.168 cal anos AP e 34.804 – 35.584 cal anos AP. A vegetação arbórea e herbácea formam um ecótono que persiste do Holoceno Inferior ao Médio, enquanto a assembleia de vegetação adaptada ao frio se extingue. Os resultados desse trabalho sugerem a presença de uma significativa população de plantas de origem glacial nas planícies baixas do leste da Amazônia antes do Máximo Glacial Pleistocênico, as quais, na atualidade estão restritas aos Andes (2000-3000 m), sugerindo que nesse intervalo de tempo podem ter ocorrido temperaturas com valores inferiores aos que foram propostos para essa região.

Pollen and stable carbon (d13C) and hydrogen (dD) isotope ratios of terrestrial plant wax from the South Atlantic sediment core, ODP Site 1085, is used to reconstruct Miocene to Pliocene changes of vegetation and rainfall regime of western southern Africa. Our results reveal changes in the relative amount of precipitation and indicate a shift of the main moisture source from the Atlantic to the Indian Ocean during the onset of a major aridification 8 Ma ago. We emphasise the importance of declining precipitation during the expansion of C4 and CAM (mainly succulent) vegetation in South Africa. We suggest that the C4 plant expansion resulted from an increased equator-pole temperature gradient caused by the initiation of strong Atlantic Meridional Overturning Circulation following the shoaling of the Central American Seaway during the Late Miocene.
