The efficiency with which a small beam trawl (1 x 0.5 m mouth) sampled postlarvae and juveniles of tiger prawns Penaeus esculentus and P, semisulcatus at night was estimated in 3 tropical seagrass communities (dominated by Thalassia hemprichii, Syringodium isoetifolium and Enhalus acoroides, respectively) in the shallow waters of the Gulf of Carpentaria in northern Australia. An area of seagrass (40 x 3 m) was enclosed by a net and the beam trawl was repeatedly hand-hauled over the substrate. Net efficiency (q) was calculated using 4 methods: the unweighted Leslie, weighted Leslie, DeLury and Maximum-likelihood (ML) methods. The Maximum-likelihood is the preferred method for estimating efficiency because it makes the fewest assumptions and is not affected by zero catches. The major difference in net efficiencies was between postlarvae (mean ML q +/- 95% confidence limits = 0.66 +/- 0.16) and juveniles of both species (mean q for juveniles in water less than or equal to 1.0 m deep = 0.47 +/- 0.05), i.e. the beam trawl was more efficient at capturing postlarvae than juveniles. There was little difference in net efficiency for P, esculentus between seagrass types (T, hemprichii versus S. isoetifolium), even though the biomass and morphologies of seagrass in these communities differed greatly (biomasses were 54 and 204 g m(-2), respectively). The efficiency of the net appeared to be the same for juveniles of the 2 species in shallow water, but was lower for juvenile P, semisulcatus at high tide when the water was deeper (1.6 to 1.9 m) (0.35 +/- 0.08). The lower efficiency near the time of high tide is possibly because the prawns are more active at high than low tide, and can also escape above the net. Factors affecting net efficiency and alternative methods of estimating net efficiency are discussed.

Juvenile tiger prawns (Penaeus semisulcatus De Haan and P. esculentus Haswell) show a strong association with vegetated habitats and are rarely caught on non-vegetated areas. This pattern of distribution may be caused by postlarvae selecting vegetation when they settle, or to differences in post-settlement mortality in different habitats. In this study, we examined whether the postlarvae and early juvenile stages of P. semisulcatus would distinguish between seagrass (Zostera capricorni Aschers) without epiphytes, artificial seagrass and bare substratum in the laboratory. The responses of prawns reared from the egg to different stages of postlarval and juvenile development were tested to determine whether, and when, each size class showed a response to a particular habitat. Five size classes of postlarvae (average carapace lengths [CL] of 1.2, 1.4, 1.6, 1.7 and 2.0 mm) were offered a choice between Z. capricorni and bare sand. Small size classes of postlarvae either did not respond to Z. capricorni (1.2 and 1.6 mm CL), or were more abundant on bare substratum than Z. capricorni. In contrast, the largest size classes of postlarvae (1.7 and 2.0 mm CL) were more abundant on Z. capricorni during the day but not at night. The behaviour of postlarvae changed markedly at a size of 1.7 mm CL (22 days from the first nauplius): smaller postlarvae frequently swam in the water column; 1.7 and 2.0 mm CL postlarvae spent much more. time resting on the substrate and perched on seagrass leaves. This size at which postlarvae first respond to seagrass during the day, and show mainly benthic behaviour, is similar to the size at which they are found on shallow seagrass beds in northern Australia. Large postlarvae (2.7 mm CL) and juveniles (4.1 mm CL) both were more abundant on artificial seagrass than bare sand during the day but not at night, indicating that they respond to structured habitats. When large postlarvae (2.4 mm CL) and juveniles (3.5 mm CL) were offered a choice between Z. capricorni without epiphytes and artificial seagrass, they were more abundant on the Z. capricorni, which suggests that chemical cues from seagrass may explain some of the responses of P. semisulcatus to seagrass. A study of the reproductive physiology of P. (P). semisulcatus wasundertaken as this information is an essential prerequisite for brood stock development for hatchery operations, and the results are embodied in this thesis. The thesis is presented in seven chapters. The protein, lipid and carbohydrate contents of ovary were estimated for the different vitellogenic phases. The protein, lipid, carbohydrate and cholesterol contents were estimated in testes and spermatophore.The thesis present the results of induced maturation experiments by eyestalk ablation, CNS extract injection and UV rays application on immature female prawns.

The impact of global positioning systems (GPS) and plotter systems on the relative fishing power of the northern prawn fishery fleet on tiger prawns (Penaeus esculentus Haswell, 1879, and P. semisulcatus de Haan, 1850) was investigated from commercial catch data. A generalized linear model was used to account for differences in fishing power between boats and changes in prawn abundance. It was found that boats that used a GPS alone had 4% greater fishing power than boats without a CPS. The addition of a plotter raised the power by 7% over boats without the equipment. For each year between the first to third that a fisher has been working with plotters, there is an additional 2 or 3% increase. It appears that when all boats have a GPS and plotter for at least 3 years, the fishing power of the fleet will increase by 12%. Management controls have reduced the efficiency of each boat and lowered the number of days available to fish, but this may not have been sufficient to counteract the increases. Further limits will be needed to maintain the desired levels of mortality.

Five species of commercial prawns Penaeus plebejus, P. merguiensis, P. semisulcatus/P. esculentus and M. bennettae, were obtained from South-East and North Queensland, chilled soon after capture and then stored either whole or deheaded on ice and ice slurry, until spoilage. Total bacterial counts, total volatile nitrogen, K-values and total demerit scores were assessed at regular intervals. Their shelf lives ranged from 10-17 days on ice and >20 days on ice slurry. Initial bacterial flora on prawns from shallower waters (4-15m) were dominated by Gram-positives and had lag periods around 7 days, whereas prawns from deeper waters (100m) were dominant in Pseudomonas spp. with no lag periods in bacterial growth. The dominant spoiler in ice was mainly Pseudomonas fragi whereas the main spoiler in ice slurry was Shewanella putrefaciens. Bacterial interactions seem to play a major role in the patterns of spoilage in relation to capture environment and pattern of storage

Siddeek et al. (1991) discussed very briefly some recent developments in Kuwait's shrimp fishery, including an important increase in landings of the main commercial species, Penaeus semisulcatus. This increase coincided with a marked fall in landings of the other important species, Metapenaeus affinis. They thought that these changes were caused by a reduction in effort combined with a more or less simultaneous favorable environmental change for P. semisulcatus and an unfavorable environmental change for M. affinis, but did not give any unequivocal evidence to support this conclusion. The results they reported are, nevertheless, very important and may be relevant to scientists and managers in other parts of the world.

This paper deals with the male reproductive organs of nine species of penaeid prawns; Penaeus penicillatus Alcock, P. merguiensis De Man, P. semisulcatus De Haan, Metapenaeus affinis (H.Milne Edwards), M. monoceros (Fabricius), M. stebbingi Nobili, Parapenaeopsis hardwickii (Miers), P. sculptilis (Heller) and P. stylifera (H. Milne Edwards). The male reproductive organs exhibited structural variations, which were more pronounced at generic level. These variations are mainly due to the type of spermatophore they possess. One species of each genus, that is, P. merguiensis, M. affinis and P. sculptilis were also studied histologically to examine the internal structure of the male reproductive organs. Spermatophores of the six species belonging to the genera Penaeus and Parapenaeopsis are also described and illustrated.

Fourteen species of penaeid shrimps with commercial value in Batan Bay and Tigbauan-Guimbal waters were identified as follows: Penaeus monodon, P.semisulcatus, P.japonicus, Metapenaeus ensis, M.burkenroadi, M.endeavouri, Metapenaeopsis palmensis, M.stridulans, Trachypenaeus fulvus, and Parapenaeus longipes. Among the 14 penaeids, P.semisulcatus, M.ensis and M.palmensis were found to be the dominant species within each genus. There are seven existing fishing gears for shrimping in the Batan Bay and Tigbauan-Guimbal waters: fish corrals, lift net, filter net, gill net, skimming net, baby trawler and commercial trawler. In general, female penaeids are larger than males. The largest P.monodon female measured was 81 mm in carapace length with 23 g in body weight. The largest male measuring 59 mm in carapace length with 119 g of body weight was caught in Batan Bay. Judging from spermatozoa occurrence on both sexes of P.monodon, the biological minimum size for male is 37 mm in carapace length and 49 mm for female. A total of 133 Penaeus postlarvae obtained from the offshore were identified by comparison with those reared in the laboratory. The postlarvae of P.japonicus-latisulcatus complex were quite dominant (60 . 2%), followed by P.semisulcatus (18 . 0%), and P.merguiensis-indicus complex (17 . 3%). The number of P.monodon postlarvae was relatively small (4 . 5%). The modal carapace length of P.monodon postlarvae from the offshore was 1 . 3 mm with three or four dorsal and no ventral spines on the rostrum, while P.monodon fry from the shoreline had 2 . 3 mm with five or six dorsal and one or two ventral spines.

Diagnostic characters for the identification of postlarval Penaeus appearing along the coastal waters of the Philippines are reviewed and categorized, based on materials from the wild and laboratory. Presently there are 7 adult known species belonging to the genus Penaeus around Panay Island and its adjacent waters, namely: P. monodon, P. semisulcatus, P. merguiensis, P. indicus, P. japonicus , P. canaliculatus, and P. latisulcatus.

The study was undertaken to ascertain the commercially important species of penaeid prawns caught in Batan Bay, Philippines and their abundance in fish corrals. A total of 12 species were commercially caught: Penaeus monodon, P. semisulcatus, P. merguiensis, P. indicus, P. latisulcatus, P. japonicus, P. canaliculatus, Metapenaeus ensis, M. endeavouri, M. dalli, M. elegans, and Trachypenaeus fulvus.

Green tiger prawn, Penaeus sentisulcatus is one of the commercial species of Persian Gulf, which is distributed from north to Strait of I Iormoze. Concerning its role in fisheries economic, various research projects on stock assessment, biology and aquaculture has been conducted. This research is targeted the identification of various populations of green tiger prawn in northern waters of Persian Gulf. The area has been divided to five regions from north to south named; Bahrakan, Boushehr, Tangestan, Motaaf and Strait of Hormoz. In each region, numbers of sampling stations trawled, and live shrimp species carried in containers equipped with air pump, to coastal laboratories in Boushehr and Bandar Abbass Fisheries Research Centers. Biometeric, morphometeric and merestic measures for 45 factars done, and peices of muscles, eye and ovary tissues dissected, and stored in liquid nitrogen. Protein extraction, and polyacrylamid gel electrophoresis by SDS-PAGE technique for tissues samples conducted. Data of 45 morphometeric and merestic characteristics analyzed by principal component analysis (PCA), and clustering analysis methods. The results of analysis showed that, the populations of Bahrakan and Mota.af regions are differentiated, while population of Boushehr and Tangestan regions were mixed, and named as a single population. The analysis of electrophoretic data also confirmed this result, and showed a distinct population in Strait of Hormoz. Therefore, this research illustrated four distinct populations for P. semisulcatus in northern area of Persian Gulf, named Bahrakan (north of Boushehr), Boushehr and Tangesta.n (adjacent), Motaaf and it's south, and Strait of Hormoz. Study of morphometeric characteristics of carapace factors, genital organs, antenna and life cycle of samples of different regions resulting identification of a subspecies, which is named Penaeus seinisuleatus persicus.

对虾病害在世界范围内肆虐,给水产养殖和沿海农村经济造成了重大损失。在水产养殖的实践中快速检测水产动物的病害并及时采取隔离等措施对于控制病害尤为重要,其中关键的环节就是快速检测出病害,并在对虾免疫机制上寻找对虾疾病防治的有效方法。研究表明当对虾等甲壳动物受到外界病原刺激时,极微量的微生物多糖就可以激活proPO系统。激活过程中涉及和产生一系列活性物质,如黑色素、酚氧化酶原激活因子(PPA)、模式识别蛋白(BGBP、PGBP、LGBP、LBP)及其膜上受体和A2巨球蛋白等,它们可通过多种方式参与防御反应,包括提供调理素,促进血细胞吞噬作用,形成结节或包囊以及介导凝集和凝固,产生杀菌物质并且黑色素化。黑色素常常在节肢动物的体表形成黑色斑点,形成的色素沉着对机体起到保护作用。所以,酚氧化酶原激活的级联反应是节肢动物免疫的关键因素。本论文研究开发了以环等温介导技术(LAMP)为基础的检测对虾白斑病毒(WSSV)和鳗弧菌(V. anguillarum)的快速检测方法。并从对虾对病害的免疫机制为切入点,从中国明对虾体内克隆了酚氧化酶原(PrpPO)和丝氨酸蛋白酶FcSP3这两个免疫系统中重要的基因,分析了它们的分子结构特征,组织分布及应答鳗弧菌病原刺激的表达变化模式。 建立的对虾常见病害对虾白班病毒(WSSV)和鳗弧菌(V. anguillarum)的LAMP检测方法,经过实验比对和Blast检索,发现本研究中使用的引物,比已经报导的LAMP方法或者PCR方法具有更宽的检测范围(更低的假阴性)。检测WSSV的LAMP方法使用病毒的VP28基因设计引物,而鳗弧菌的检测方法使用empA基因设计引物。在方法中,首次提出加入UNG酶和dUTP的措施来预防污染,在实际检测中非常有效。LAMP方法与PCR检测方法的灵敏性比较也进行了研究,二者灵敏性相当。 依据中国明对虾血液cDNA文库提供的部分片段信息,结合SMART-RACE技术,克隆了酚氧化酶原(PrpPO)基因,通过序列比对分析发现,PrpPO基因cDNA全长为3040 bp,其中开放阅读框2061 bp,编码686个氨基酸,其中推测的信号肽为12个氨基酸。推测的序列与斑节对虾(P. monodon)同源性为93%,与短钩对虾(P. semisulcatus.)同源性为92%。real time RT-PCR实验结果表明, ProPO在血细胞中的相对表达量最高,肝胰脏中表达量最低。弧菌刺激实验中注射弧菌,刺激了血细胞和淋巴器官中的ProPO mRNA显著增加,说明在血细胞和淋巴器官中存在快速反应的ProPO通路。而ProPO mRNA量在淋巴器官中在时间上早于血液中升至最高,说明该动物在在病原刚开始入侵的时候先有淋巴器官发挥主要的免疫作用,随着时间推移血细胞便变成主要的免疫器官。 根据中国明对虾肝胰脏cDNA文库提供EST信息,经过SMART-RACE克隆了一个丝氨酸蛋白酶FcSP3基因,通过序列比对分析发现,该丝氨酸蛋白酶基因cDNA全长为1622 bp,其中开放阅读框1431 bp,编码477个氨基酸,其中推测的信号肽为22个氨基酸。推测的序列与疟蚊的丝氨酸蛋白酶(A. gambiae)同源性为33%,与丽蝇蛹集金小蜂的酚氧化酶原激活因子(N. vitripennis)同源性为32%,与东北大黑鳃金龟的酚氧化酶原激活因子(H. diomphalia)同源性为34%。淋巴器官中PPAⅡ表达量约为血液中表达量的47560倍,肝胰脏中的FCSP3表达量为血细胞表达量的6226倍。鳗弧菌注射对虾后,淋巴器官中刺激组和对照组FcSP3的mRNA量在刺激后6小时显著降低,但是刺激组的表达量明显高于对照组。刺激组的血细胞与肝胰脏中FcSP3 mRNA的相对表达量增高。而病原刺激后的血液与肝胰脏中的FcSP3 mRNA的增长趋势也在时间上先与ProPO mRNA。这说明FcSP3对ProPO有正调控的作用,但这个调控有一个时间差,并且在不同组织中有不同的调控效率。

The thesis embodies the results of the studies carried out on certain diseases affecting the commercially important penaeid prawns in the capture and culture fisheries of the southwest and southeast coasts of india during october, 1981 to april 1985.initially a survey is conducted to obtain information and to understand the commom diseases and abnormalities occurring in the penaeid prawms in nature and those farmed. A result of the survey then cases of diseases and abnormalities are reported. These include tumour like growth soft prawm syndrome, tail necrosis brown spot disease, red rostrum, ciliate infestation ,helminth parasitisation,metacercarial infestation and bopyrid infestation in the penaeid prawns such as penaeus indicus p. monodon , p. semisulcatus ,Metapenaeus dobsoni and m.affinis.The symptoms ,occurrence and incidence of each of the above cases are provided along with the information on environmental factors such as salinity,dissolved oxygen,temperature and ph of the water from the collection sites. The nature of the disease,the tissuse of the host that are affected by the infection or infestation or by the pathogen ,and the actors influencing the infection in each of the ten cases are studied histopathologically and discussed. In the light of the available published information, the control measures for the different diseases of penaeid prawns are presented and discussed.

The thesis presents the results of the studies carried out on certain diseases encountered in the larvae and postlarvae of penaeid prawns raised in the hatcheries at Cochin, Madras and Mandapam Camp during September 1985- April 1988. In the preliminary survey carried out to understand the common diseases occurring in the penaeid larvae and postlarvae, seven cases of diseases and abnormalities were encountered. These included ciliate infestation, Nit_zschia closteriurn infestation, parasitic protozoaninfection, parasitic dinoflagellate infection, appendage necrosis, heteromorphic eye and abnormal eggs and deformed nauplii .The clinical signs, seasonal occurrence and incidence of each of the above cases were provided along with the information on environmental factors such as salinity, dissolved oxygen, temperature and pH of the rearing medium. The Thésis is presented in nine chapters. Chapter 1 surveys the literature on the diseases of penaeid larvae, postlarvae and adult prawns from India and abroad. This is followed by a chapter on the material and methods employed during the present investigation. In the third chapter, seven cases of diseases and abnormalities encountered in the larvae and postlarvae of Penaeus indicus and p. semisulcatus during the survey carried out in the hatcheries located at different centres of Central‘ Marine Fisheries Research Institute are presented and discussed .A bacterium responsible for appendage necrosis was isolated and its taxonomy was studied. It was Gram-negative, fermentative and motile rod. It was sensitive to vibriostatic compound, 0/129. This bacterium was found to be a new isolate of vibrio on the basis of its morphological, biological, physiological and biochemical characters and comparison of these characters with those described for other related vibrios. This new isolate of vibrio was deposited in vibro Referrence Laboratory, Centres for Disease Control, Georgia, U.S.A. and coded as vibrio sp. 2448-88.

Penaeid prawns were sampled with a small seine net to test whether catches of postlarvae and juveniles in seagrass were affected by the distance of the seagrass (mainly Zostera capricorni) from mangroves and the density of the seagrass in a subtropical marine embayment. Sampling was replicated on the western and eastern sides of Moreton Bay, Queensland, Australia. Information on catches was combined with broad-scale spatial information on the distribution of habitats to estimate the contribution of four different categories of habitat (proximal dense seagrass, distal dense seagrass, proximal sparse seagrass, distal sparse seagrass) to the overall population of small prawns in these regions of Moreton Bay. The abundance of Penaeus plebejus and Metapenaeus bennettae was significantly and consistently greater in dense seagrass proximal to mangroves than in other types of habitat. Additionally, sparse seagrass close to mangroves supported more of these species than dense seagrass farther away, indicating that the role of spatial arrangement of habitats was more important than the effects of structural complexity alone. In contrast, the abundance of P. esculentus tended to be greatest in sparse seagrass distal from mangroves compared with the other habitats. The scaling up of the results from different seagrass types suggests that proximal seagrass beds on both sides of Moreton Bay provide by far the greatest contribution of juvenile M. bennettae and P. plebejus to the overall populations in the Bay.
