OBJETIVO: Determinar os desfechos maternos e perinatais em gestantes com o líquido amniótico diminuído segundo o índice de líquido amniótico (ILA). MÉTODOS: Realizou-se estudo de coorte com 176 pacientes admitidas na enfermaria de alto risco do Instituto de Medicina Integral Prof. Fernando Figueira (IMIP. O líquido amniótico foi mensurado pelo índice de líquido amniótico , sendo classificado como diminuído, quando entre 5,1 e 7,9 cm; oligohidrâmnio moderado, entre 3,1 e 5,0 cm; e grave, quando menor ou igual a 3,0 cm. Para se determinar a diferença entre os três grupos das variáveis categóricas estudadas, foram utilizados o teste de chi-quadrado e exato de Fisher, quando pertinentes, e, para as variáveis numéricas, utilizou-se o teste de Mann Whitney, em um nível de significância de 5%. RESULTADOS: As malformações fetais ocorreram mais frequentes quando o oligohidrâmnio foi grave, enquanto as síndromes hipertensivas foram associadas ao oligohidrâmnio moderado. Observou-se semelhança entre os três grupos em relação à rotura prematura das membranas e outras causas. O líquido amniótico reduzido foi encontrado com maior frequência quando a idade gestacional do diagnóstico foi ≥32ª semana. Em relação aos desfechos perinatais, a incidência de índice de Apgar <7 no 1ºe 5ºminuto do óbito perinatal, da icterícia neonatal e da hipoplasia pulmonar foi mais elevada na presença do oligohidrâmnio moderado a grave. CONCLUSÕES: As causas e os desfechos maternos e perinatais em gestantes com líquido amniótico reduzido varia em relação a sua classificação pelo ILA, estando o oligohidrâmnio grave associado aos desfechos perinatais adversos e às malformações fetais.

The Northern Snake Range (Nevada) represents a spectacular example of a metamorphic core complex and exposes a complete section from the mylonitic footwall into the hanging wall of a fossil detachment system. Paired geochronological and stable isotopic data of mylonitic quartzite within the detachment footwall reveal that ductile deformation and infiltration of meteoric fluids occurred between 27 and 23 Ma. Ar-40/Ar-39 ages display complex recrystallization-cooling relationships but decrease systematically from 26.9 +/- 0.2 Ma at the top to 21.3 +/- 0.2 Ma at the bottom of footwall mylonite. Hydrogen isotope (delta D) values in white mica are very low (-150 to -145 %) within the top 80-90 m of detachment footwall, in contrast to values obtained from the deeper part of the section where values range from -77 to -64 %, suggesting that time-integrated interaction between rock and meteoric fluid was restricted to the uppermost part of the mylonitic footwall. Pervasive mica-water hydrogen isotope exchange is difficult to reconcile with models of Ar-40 loss during mylonitization solely by volume diffusion. Rather, we interpret the Ar-40/Ar-39 ages of white mica with low-delta D values to date syn-mylonitic hydrogen and argon isotope exchange, and we conclude that the hydrothermal system of the Northern Snake Range was active during late Oligocene (27-23 Ma) and has been exhumed by the combined effects of ductile strain, extensional detachment faulting, and erosion.

We report the molecular characterization of a novel reiterated family of transcribed oligo(A)-terminated, interspersed DNA elements in the genome of Trypanosoma cruzi. Steady-state level of transcripts of this sequence family appeared to be developmentally regulated, since only in the replicative forms the parasite showed expression of related sequences with a major band around 3 kb. The presence of frame shifts or premature stop codons predicts that transcripts are not translated. The sequence family also contains truncated forms of retrotransposons elements that may become potential hot spots for retroelement insertion. Sequences homologous to this family are interspersed at many chromosomes including the subtelomeric regions.

We present the first evaluation of a novel molecular assay, the Speed-oligo Direct Mycobacterium tuberculosis (SO-DMT) assay, which is based on PCR combined with a dipstick for the detection of mycobacteria and the specific identification of M. tuberculosis complex (MTC) in respiratory specimens. A blind evaluation was carried out in two stages: first, under experimental conditions on convenience samples comprising 20 negative specimens, 44 smear- and culture-positive respiratory specimens, and 11 sputa inoculated with various mycobacterium-related organisms; and second, in the routine workflow of 566 fresh respiratory specimens (4.9% acid-fast bacillus [AFB] smear positives, 7.6% MTC positives, and 1.8% nontuberculous mycobacteria [NTM] culture positives) from two Mycobacterium laboratories. SO-DMT assay showed no reactivity in any of the mycobacterium-free specimens or in those with mycobacterium-related organisms. Compared to culture, the sensitivity in the selected smear-positive specimens was 0.91 (0.92 for MTC and 0.90 for NTM), and there was no molecular detection of NTM in a tuberculosis case or vice versa. With respect to culture and clinical data, the sensitivity, specificity, and positive and negative predictive values for the SO-DMT system in routine specimens were 0.76 (0.93 in smear positives [1.0 for MTC and 0.5 for NTM] and 0.56 in smear negatives [0.68 for MTC and 0.16 for NTM]), 0.99, 0.85 (1.00 in smear positives and 0.68 in smear negatives), and 0.97, respectively. Molecular misidentification of NTM cases occurred when testing 2 gastric aspirates from two children with clinically but not microbiologically confirmed lung tuberculosis. The SO-DMT assay appears to be a fast and easy alternative for detecting mycobacteria and differentiating MTC from NTM in smear-positive respiratory specimens.

The main objective of the present study is to have a detailed investigation on the gelation properties, morphology and optical properties of small π-conjugated oligomers. For this purpose we have chosen oligo(p-phenylenevinylene)s (OPVs), a class of molecules which have received considerable attention due to their unique optical and electronic properties. Though a large number of reports are available in the literature on the self-assembly properties of tailor made OPVs, none of them pertain to the design of nanostructures based on organogels. In view of this, we aimed at the creation of functional chromophoric assemblies of π-conjugated OPVs through the formation of organogels, with the objective of crafting nanoscopic assemblies of different size and shape thereby modulating their optical and electronic properties.In order to fulfill the above objectives, the design and synthesis of a variety of OPVs with appropriate structural variations were planned. The design principle involves the derivatization of OPVs with weak H-bonding hydroxymethyl end groups and with long aliphatic hydrocarbon side chains. The noncovalent interactions in these molecules were expected to lead the formation of supramolecular assembly and gels in hydrocarbon solvents. In such an event, detailed study of gelation and extensive analysis of the morphology of the gel structures were planned using advanced microscopic techniques. Since OPVs are strongly fluorescent molecules, gelation is expected to perturb the optical properties. Therefore, detailed study on the gelation induced optical properties as a way to probe the nature and stability of the selfassembly was planned. Apart from this, the potential use of the modulation of the optical properties for the purpose of light harvesting was aimed. The approach to this problem was to entrap an appropriate energy trap to the OPV gel matrix which may lead to the efficient energy transfer from the OPV gel based donor to the entrapped acceptor. The final question that we wanted to address in this investigation was the creation of helical nanostructures through proper modification of the OPV backbone With chiral handles.The present thesis is a detailed and systematic approach to the realization of the above objectives which are presented in different chapters of the thesis.

Gelation provides a unique medium, which often induces organization of molecules resulting in the modulation of their optical, morphological and electronic properties thereby opening a new world of fascinating materials with interesting physical properties at nano- meso- and macroscopic levels. Supramolecular gels based on linear π-systems have attracted much attention due to their inherent optical and electronic properties which find application in organic electronics, light harvesting and sensing. They exhibit reversible properties due to the dynamic nature of noncovalent forces. As a result, studies on such soft materials are currently a topic of great interest. Recently, researchers are actively involved in the development of sensors and stimuli-responsive materials based on self-assembled π-systems, which are also called smart materials. Amyloid fibrils are formed by a model surfactant-like peptide (Ala)10-(His)6 containing a hexahistidine tag. This peptide undergoes a remarkable two-step self-assembly process with two distinct critical aggregation concentrations (cac’s), probed by fluorescence techniques. A micromolar range cac is ascribed to the formation of prefibrillar structures, whereas a millimolar range cac is associated with the formation of well-defined but more compact fibrils. We examine the labeling of these model tagged amyloid fibrils using Ni-NTA functionalized gold nanoparticles (Nanogold). Successful labeling is demonstrated via electron microscopy imaging. The specificity of tagging does not disrupt the β-sheet structure of the peptide fibrils. Binding of fibrils and Nanogold is found to influence the circular dichroism associated with the gold nanoparticle plasmon absorption band. These results highlight a new approach to the fabrication of functionalized amyloid fibrils and the creation of peptide/nanoparticle hybrid materials.

The pH-structure correlation of the products of aniline peroxydisulfate reaction was mainly investigated by resonance Raman spectroscopy. The reactions of aniline and ammonium peroxydisulfate were carried out in aqueous solutions of initial pH ranging from 4.9 to 13.2 and monomer/oxidant molar ratio of 4/1. For an initial pH of 4.9, the spectroscopic techniques showed that the emeraldine salt form of polyaniline (PANI-ES) is the main product, corroborating that the usual head-to-tail coupling mechanism is taking place. The resonance Raman spectra at 1064 nm exciting wavelength were useful to detect the emeraldine salt as a minor product for reactions at an initial pH of 5.3-11.5. The Raman spectra of the main product of the reaction at initial pH of 13.2 excited at 1064 and 413.1 nm showed new spectral features consistent with 1,4-Michael-type adducts of aniline monomers and 1,4-benzoquinone-monoimine unit. These compounds and their products of hydrolysis/oxidation are the predominant species for the reaction media of initial pH from 5.3 to 13.2. In order to get PANI with different nanoscale morphologies, a pH value of more than 0 or 1 was used in the aniline polymerization. The spectroscopic data obtained in this work reveal that head-to-tail coupling does not occur when aniline reacts at media pH higher than about 5. It is suggested that chemical structures of the products of aniline oxidation by an unusual mechanism are the driving force for the development of assorted morphologies. The complex of Brookhart Ni(α-diimine)Cl2 (1) (α-diimine = 1,4-bis(2,6- diisopropylphenyl)-acenaphthenediimine) has been characterized after impregnation on silica (S1) and MAO-modified silicas (4.0, 8.0 and 23.0 wts.% Al/SiO2 called S2, S3 and S4, respectively). The treatment of these heterogeneous systems with MAO produces some active catalysts for the polymerization of the ethylene. A high catalytic activity has been gotten while using the system supported 1/S3 (196 kg of PE/mol[Ni].h.atm; toluene, Al/Ni = 1000, 30ºC, 60 min and atmospheric pressure of ethylene). The effects of polymerization conditions have been tested with the catalyst supported in S2 and the best catalytic activity has been gotten with solvent hexane, MAO as cocatalyst, molar ratio Al/Ni of 1000 and to the temperature of 30°C (285 kg of PE/mol[Ni].h.atm). When the reaction has been driven according to the in situ methodology, the activity practically doubled and polymers showed some similar properties. Polymers products by the supported catalysts showed the absence of melting fusion, results similar to those gotten with the homogeneous systems by DSC analysis. But then, polymers gotten with the transplanted system present according to the GPC’s curves the polydispersity (MwD) varies between 1.7 and 7.0. A polyethylene blend (BPE/LPE) was prepared using the complex Ni(α-diimine)Cl2 (1) (α-diimine = 1,4-bis(2,6-diisopropylphenyl)-acenaphthenediimine) and {TpMs*}TiCl3 (2) (TpMs* = hydridobis(3-mesitylpyrazol-1-yl)(5-mesitylpyrazol-1-yl)) supported in situ on MAO-modified silica (4.0 wts. -% Al/SiO2, S2). Reactions of polymerization of ethylene have been executed in the toluene in two different temperatures (0 and 30°C), varying the molars fraction of nickel (xNi), and using MAO as external cocatalyst. To all temperatures, the activities show a linear variation tendency with xNi and indicate the absence of the effect synergic between the species of nickel and the titanium. The maximum of activity have been found at 0°C. The melting temperature for the blends of polyethylene produced at 0 °C decrease whereas xNi increases indicating a good compatibility between phases of the polyethylene gotten with the two catalysts. The melting temperature for the blends of polyethylene showed be depend on the order according to which catalysts have been supported on the MAO-modified silica. The initial immobilization of 1 on the support (2/1/S2) product of polymers with a melting temperature (Tm) lower to the one of the polymer gotten when the titanium has been supported inicially (1/2/S2). The observation of polyethylenes gotten with the two systems (2/1/S2 and 1/2/S2) by scanning electron microscopy (SEM) showed the spherical polymer formation showing that the spherical morphology of the support to been reproduced. Are described the synthesis, the characterization and the catalytic properties for the oligomerization of the ethylene of four organometallics compounds of CrIII with ligands ([bis[2-(3,5-dimethyl-1-pyrazolyl)ethyl]amine] chromium (III) chloride (3a), [bis[2-(3,5- dimethyl-l-pyrazolyl)ethyl]benzylamine] chromium (III) chloride (3b), [bis[2-(3,5-dimethyl-lpyrazolyl) ethyl]ether] chromiun(III)chloride (3c), [bis[2-(3-phenyl-lpyrazolyl) ethyl]ether]chromiun(III)chloride (3d)). In relation of the oligomerization, at exception made of the compounds 3a, all complex of the chromium showed be active after activation with MAO and the TOF gotten have one effect differentiated to those formed with CrCl3(thf)3. The coordination of a tridentate ligand on the metallic center doesn't provoke any considerable changes on the formation of the C4 and C6, but the amount of C8 are decrease and the C10 and C12+ have increased. The Polymers produced by the catalyst 3a to 3 and 20 bar of ethylene have, according to analyses by DSC, the temperatures of fusion of 133,8 and 136ºC respectively. It indicates that in the two cases the production of high density polyethylene. The molar mass, gotten by GPC, is 46647 g/mols with MwD = 2,4 (3 bar). The system 3c/MAO showed values of TOF, activity and selectivity to different α-olefins according to the pressure of ethylene uses. Himself that shown a big sensibility to the concentration of ethylene solubilized.

O Campo Socororo é considerado maduro, com dados antigos e de baixa qualidade. O campo não produziu nos volumes originalmente calculados, o que se traduz num campo com potencial para continuar produzindo. A presente pesquisa considera como fator importante a compreensão da origem e distribuição dos níveis de arenito capazes de conter hidrocarbonetos. Campos com predomínio de ambientes flúvio-deltaicos contêm reservatórios com corpos de arenito separados na vertical e na lateral pela presença de folhelhos. Essa disposição das camadas dificulta a correlação poço a poço desses reservatórios, o que leva à necessidade de incluir modelos geológicos. Para avaliar os resultados da interpretação dos perfis do campo Socororo, nos níveis Merecure (Oligoceno) e Oficina (Mioceno), foi feita uma correlação com os resultados de perfis de poço do Campo Budare, imediatamente a oeste de Socororo, e do Projeto Piloto da área Zuata, na faixa de óleo pesado do Orinoco. No modelo geológico obtido, é possível observar a separação entre os diferentes níveis-reservatório, mostrando que a unidade com maior volume de arenito encontra-se na porção inferior, identificada como “Formação Merecure” (Oligo-Mioceno). Na porção superior encontra-se a Formação Oficina, caracterizada por níveis de arenito pouco espessos e com diminuição da frequência dos arenitos para o topo. Tais características são observadas na correlação cronoestratigráfica de ciclos e zonas-reservatório dos campos Socororo e Budare e da área Zuata na Faixa do Orinoco.

Carbohydrates are a complex group of biomolecules with a high structural diversity. Their almost omnipresent occurrence has generated a broad field of research in both biology and chemistry. This thesis focuses on three different aspects of carbohydrate chemistry, synthesis, structure elucidation and the conformational analysis of carbohydrates. The first paper describes the synthesis of a penta- and a tetrasaccharide related to the highly branched capsular polysaccharide from Streptococcus pneumoniae type 37. In the second paper, the structure of the O-antigenic repeating unit from the lipopolysaccharide of E. coli 396/C1 was determined along with indications of the structure of the biological repeating unit. In addition, its structural and immunological relationship with E. coli O126 is discussed. In the third paper, partially protected galactopyranosides were examined to clarify the origin of an intriguing 4JHO,H coupling, where a W-mediated coupling pathway was found to operate. In the fourth paper, the conformation of methyl a-cellobioside is studied with a combination of molecular dynamics simulations and NMR spectroscopy. In addition to the expected syn-conformation, detection and quantification of anti-ø and anti-ψ conformers was also possible.

Die Zielsetzung der Arbeit war die Darstellung von Polyindenofluorenen PIF, einer neuen Klasse konjugierter Polymere, die aus Dihydroindenofluoren-Untereinheiten aufgebaut und formal an den Methylenbrücken über exocyclische Doppelbindungen verknüpft sind.Am Anfang stand die Ausarbeitung eines allgemeinen Synthesekonzeptes zum Aufbau von Polymeren mit sterisch gehinderten Doppelbindungen. Mit der Kupplung von geminalen Dichloriden mit Übergangsmetallverbindungen als Reduktionsmittel wurde eine vielversprechende neue Polyolefinierungsmethode gefunden. Diese Synthesemethode zeichnet sich durch eine einfache Durchführung, synthetisch leicht zugängliche Edukte und neutrale Reaktionsbedingungen aus. Es lassen sich selbst bei gespannten Olefinen sehr gute Ausbeuten erzielen. Drei strukturisomere Dihydroindenofluorene wurden als potentielle Strukturbausteine untersucht, die sich durch die Stellung der Methylenbrücken bezüglich des zentralen Phenylrings unterscheiden. Die resultierenden Polymerstrukturen wurden als para-, meta- und ortho-Poly(indenofluorene) bezeichnet.Das Poly(para-indenofluoren) pPIF wurde als ein hochmolekulares und strukturdefiniertes Polymer erhalten. Die über Molekulargewichtsanalyse (GPC) erhaltenen Polymerisationsgrade ergaben Werte von bis zu 55 Wiederholungseinheiten. Die längstwellige Absorptionsbande von pPIF liegt bei circa 800 nm, was einer Bandlücke von 1.45 eV enspricht. Darüberhinaus zeichnet sich pPIF durch eine hohe chemische Stabilität und sehr hohe Löslichkeit aus. Durch den kleinen Bandabstand ließen sich interessante nichtlineare-optische (NLO) Effekte und eine hohe Photoleitfähigkeit erwarten. Da diese Effekte von materialwissenschaftlichem Interesse sind, wurden detailierte Untersuchungen dieser Eigenschaften vorgenommen. Das Poly(meta-indenofluoren) mPIF konnte mit einem Polymerisationsgrad von circa 13 Wiederholungseinheiten dargestellt werden. Bedingt durch die meta-Verknüpfung, die keine Konjugation über den zentralen Phenylring erlaubt, liegt das Absorptionsmaximum, im Vergleich zu pPIF blauverschoben bei 437 nm.Bei Poly(ortho-indenofluoren) oPIF scheiterten alle Versuche zum Aufbau eines Polymers, was mit den, im Vergleich zu pPIF und mPIF, noch stärker gespannten Doppelbindungen zwischen den Indenofluorenen, sprich Untereinheiten, erklärt werden kann. An strukturdefinierten Modellverbindungen von pPIF konnte gezeigt werden, daß eine Konvergenz der elektronischen Eigenschaften beim Nonamer noch nicht erreicht ist. Oligomere mit konjugierten pi-Elektronensystemen sind für die Materialwissenschaften von großer Bedeutung. Die vielfältigen und umfangreichen Forschungen auf diesem Gebiet gründen im Potenzial dieser Substanzklassen, das im Bereich der Laserfarbstoffe, Leuchtdioden, Photoleiter, optische Schalter oder auch der molekularen Elektronik angesiedelt ist. Zu diesen gehören auch die in dieser Arbeit synthetisierten und untersuchten Phenylenethinylene. Die Herstellung der Oligomere erfolgt nach der Methode von Sonogashira und Hagihara. Dabei wird ein Halogenaren mit einer Alkinkomponente zur Reaktion gebracht. Als Katalysator dient dabei ein Gemisch aus Bis(triphenylphosphin-palladiumdichlorid), Kupfer-(I)-iodid und Triphenylphosphin. Verwendung fanden bei der Synthese zwei Arten von Schutzgruppen. Es handelt sich dabei einerseits um die Trimethylsilyl- und die Triisopropylsilyl-Funktion, die unabhängig voneinander in ein System eingeführt werden und selektiv wieder entfernt werden können. Die zweite Art sind die Halogene Brom und Iod, die aufgrund ihrer Eigenschaft vielmehr als 'dormant group' bezeichnet werden müssen. Eine Ethinylierung führt zunächst zur Substitution des Iod- und anschließend des Bromatoms. Die so erhaltenen Oligomere werden mit verschiedenen spektroskopischen Methoden untersucht. Besonderes Interesse liegt dabei auf der Bestimmung der effektiven Konjugationslänge (EKL). Damit ist es möglich, die Länge des konjugierten Systems zu bestimmen, das für die betreffenden Eigenschaften des entsprechenden Polymers maßgeblich ist. Das nichtlineare optische Verhalten der Oligomere wird mittels der Third-Harmonic-Generation-Methode (THG) gemessen. Die resultierende Größe, die Suszeptibilität 3. Ordnung, gibt Aufschluß über mögliche industrielle Anwendungen.
