1985年贲昆龙等首次发现人和猴T淋巴细胞能与树鼩(Tupaia belangeri)的红细胞(TRBC)形成亲和力极强的玫瑰花结,它与绵羊红细胞(SRBC)玫瑰花结(E花结)明显不同。例如,TRBC经神经氨酸酶处理之后,结花率明显下降,TRBC与T淋巴细胞所形成的玫瑰花结,经45℃保温,仍不受影响,为了进一步探讨TRBC受体和SRBC受体(CD2),以及与其它T细胞表面分化搞原(CD)的系。我用某些抗人T细胞CD的单克隆抗体(McAb)对人和猴淋巴细胞进行玫瑰花结抑制试验,抗原调变和共调变(Antigenic modulation or co-modulation)实验,并且研究了TRBC受体在其它免疫细胞和某些人类和长臂猿细胞系的分布。结果表明,TRBC与外周血E~+-PBL形成玫瑰花结的百分率为88.8%。而E~--PBL仅为4.16%。TRBC受体存在于所有被试T细胞系(CEM, H33JHJA1, Jurkat, MLA-144, Molt-3, Molt-4, Molt-4 clone-8 和PEER),但不存在于外周血粒细胞,B细胞,以及B细胞系的绝大多数细胞表面(Daudi, Raji和Reh)。分布于全T细胞的CD3, TCR, CD5, CD6和CD7的相McAb OKT3, T108(F1), T136(F101-15), T149(M-T604)和T152(7G5)均不能调变和共调变TRBC受体。猴和人外周血淋巴细胞与TRBC玫瑰花结的形成,不被T11.1 McAb OKT11所阻断,相反,OKT11显著地阻断猴和人外周血淋巴细胞E玫瑰花结的形成,最大抑制率分别为49.3%和77.7%。在世界各地10个实验室送交第四次国际人白细胞化化抗原讨论会待鉴定的13个CD2 McAb中,除T089 (39C1.5)因抗体量不够未作实验外,对其他12种McAb都进行试验,T081 (x/3),T082 (GLB-T11.2/1),T083 (GLB-T11.1/1),T085 (RPA-2.10),T1088 (0-275)和T092 (M-T201)对TRBC玫瑰花结和E玫瑰花结都呈现明显的阻断作用,T084 (F110.08),T091 (AICD2.1)以及已知参数CD2 McAb-T086 (D-66 clonel)和T087 (GT-2)都不阻TRBC玫瑰花结的形成,亦不调变TRBC受体,而对E玫瑰花结则有不同程度阻断效应,并且调变E变体,使E玫瑰花结形成细胞百分率下降。T090 (6F10.3)和T198 (JOR-T2)即不阻TRBC玫瑰花结的形成,也不抑制E玫瑰花结的形成。由此可见,TRBC受体分布于全T细胞,它不同于已知的全T细胞表面分化抗原CD2 (gp50), CD3/TCR复合物,CD5, CD6和CD7。CD2分子不与TRBC玫瑰花结的形成,也不是介导E玫瑰花结的唯一分子。至少有二个或三个以上的蛋白质与E玫瑰花结和TRBC玫瑰花结的形成有关,其中有的分子为E受体和TRBC受体所共有。TRBC受体很有可能包括新的T淋巴细胞分化抗原。

Monoclonal antibodies of the OKT series were used to identify T lymphocytes (OKT3+) and their inducer (OKT4+) and suppressor-cytotoxic (OKT8+) subsets in the peripheral blood mononuclear cells (PBMC) of 32 healthy old-aged people more than 70 years old (16 men and 16 women) compared to 47 adults (29 men, 18 women) less than 40 years old. The absolute lymphocyte count in the peripheral blood was not significantly influenced by age or sex. Both the proportions and the absolute numbers of T3+ and T4+ cells were significantly lower in aged than in young participants. The proportions but not the absolute counts of OKT8+ cells were higher in the elderly. Most interesting is the influence of sex and these parameters. Old women have normal numbers and proportions of T3+, T4+ and T8+ cells when compared to young women. The latter have a significantly higher proportion of T8+ cells than young adult males. Old men have a striking reduction of both the numbers and proportions of OKT3+ and OKT4+ cells when compared with young men and with women. In addition, old men have an elevated proportion, but a normal absolute number, of OKT8+ cells. The responses of PBMC to phytohaemagglutinin extent (PHA), concanavalin A (Con A) and pokeweed mitogen (PWM) are reduced to the same extent in ageing male and female subjects when compared to young adults. In the older group, the magnitude of the lymphocyte response to PHA and Con A but not to PWM is negatively correlated with the proportions of OKT8+ cells. Surprisingly, these correlations are observed only in old women but not in old men. The latter finding excludes the possibility that the age-associated decline of the lymphocyte response to T cell mitogens is secondary to an imbalance between T4+ and T8+ lymphocytes.

L’immunosuppression optimale après greffe d’organe solide est une balance délicate et propre à chaque individu entre le risque de rejet et les risques liés à une surexposition au traitement immunosuppresseur. L’évaluation de la fonction résiduelle des lymphocytes T après stimulation par un mitogène (pharmacodynamie effective) devrait permettre de mesurer l’effet direct des médicaments immunosuppresseurs sur leur cible. Nous avons étudié différents paramètres de pharmacodynamie effective chez 34 receveurs pédiatriques de greffe d’organes solides traités par tacrolimus et mycophénolate. Les tests proposés dans ce travail sont adaptés au milieu pédiatrique et à une réalisation en temps réel. La quantification du CD25 parmi les CD4 activés par l’OKT3 permet de distinguer deux groupes de patients selon leur degré d’immunosuppression. L’âge médian est plus bas et la concentration plasmatique médiane en MPA plus élevée dans le groupe de patients plus fortement immunosupprimés. L’étude des paramètres immunologiques pouvant influencer la réponse (sécrétion des interleukines, proportion des sous-populations lymphocytaires CD4, CD8, T naïfs et Trég) ainsi que l’étude du pouvoir de restauration de la fonction lymphocytaire par l’Il-2, la guanosine ou la xanthosine, ne permettent pas de mieux comprendre les variabilités interindividuelles observées. Ces résultats devront être confirmés sur une cohorte plus grande de patients afin de juger de leur intérêt en pratique clinique.

As características clínicas da hepatite C em transplantados renais ainda não estão bem definidos. A punção biópsia hepática (PBH) é o padrão-ouro para o diagnóstico do grau de lesão nesses pacientes, O presente estudo objetivou avaliar o papel da citologia hepática obtida por meio de punção aspirativa com agulha fina (PAH) no diagnóstico das lesões hepáticas em transplantados renais com hepatite crônica pelo vírus C. Avaliou-se, também, a freqüência e a gravidade das lesões hepáticas nesses pacientes e características clínico-laboratoriais correlacionadas com a gravidade da doença. Vinte e oito pacientes foram submetidos à PBH e à PAH e os resultados comparados. A PAH utilizou a técnica de Hayry e von Willebrand calculando-se o Incremento Corrigido Total e o número de células imunoativadas (CIA). Dezoito pacientes (66,6%) apresentaram, na PBH, ausência de lesões, lesões mínimas ou lesões não caracterizadas como hepatite crônica; 6 pacientes (22,2%) apresentaram hepatite crônica persistente ou lobular e 3 pacientes (11,1%) tiveram hepatite crônica ativa. Não se verificou associação estatisticamente significativa entre os resultados da PAH e os da histologia (erro β = 0,82). A idade dos pacientes mostrou associação significativa com o diagnóstico histológico, sendo a idade média dos pacientes com lesões mínimas igual a 37,50 (±10,1) anos, com hepatite crônica persistente igual a 51,33 (±6,2) anos e com hepatite crônica ativa igual a 58,33 (±12,7) anos (p=0,0015 - ANOVA). O uso de OKT3 associou-se à presença de lesões de hepatite crônica (p=0,0420). A presença de rejeições agudas, um maior número de rejeições e ter recebido pulsos de metilprednisolona associou-se a lesões histológicas de menor gravidade. As demais variáveis não apresentaram relação significativa com o diagnóstico da PBH. Na análise multivariada, apenas a idade manteve-se como fator independentemente associado à presença de lesões de hepatite crônica. Conclui-se que a citologia hepática obtida mediante punção aspirativa com agulha fina não demonstrou utilidade para diagnosticar ou estabelecer o nível de gravidade ou de evolução da doença hepática em transplantados renais com hepatite crônica pelo vírus C, embora essa possibilidade não possa ser totalmente excluída. A imunossupressão e as rejeições mostraram resultados contraditórios, não estando ainda clara a sua relação com o dano hepático provocado pelo vírus C. A idade foi o único fator consistentemente associado à maior gravidade das lesões histológicas nesses pacientes.

Purpose: Our objective was to relate immunological data for healthy but sedentary elderly women to aerobic power, strength, and mood state. Methods: We measured peak aerobic power and one-repetition maximum strength along with mood (depression and fatigue), quality of life and carbohydrate intake on 42 women aged 60-77 years. Standard immunological techniques determined natural killer cell count and cytotoxic activity (NKCA), proliferative responses to phytohemaglutinin and OKT3, various lymphocyte subpopulations (CD3(+), CD3(-)CD19(+), CD56(+), CD4(+), CD8(+), CD56(dim) and CD56(bright)), and markers of activation, maturation, down-regulation and susceptibility to apoptosis (CD25(+), CD28(+), CD45RA(+), CD45RO(+), CD69(+), CD95(+), HLA-DR+). Results: Correlations of immune parameters with aerobic power and strength were very similar for absolute and relative immunological data. In the group as a whole, the only correlation with aerobic power was -0.35 (relative CD4(+)CD69(+) count), but in subjects with values <22.6 mL kg(-1) min(-1) correlations ranged from -0.57 (relative CD4(+)CD45RO(+)) to 0.92 (absolute CD56(dim)HLA-DR+). In terms of muscle strength, univariate correlation coefficients ranged from -0.34 (relative and absolute CD3(+)CD4(+)CD8(+)) to +0.48 (absolute CD3(+)HLA-DR+.) and +0.50 (absolute CD8(+)CD45RA(+)CD45RO(+)). Neither NKCA nor lymphocyte proliferation were correlated with aerobic power or muscle strength. Although mood state and quality of life can sometimes be influenced by an individual's fitness level, our multivariate analyses suggested that depression, fatigue and quality of life were more important determinants of immune profile than our fitness measures. Conclusions: Psychological changes associated with aging may have a substantial adverse effect upon the immune system, and immunological function may be enhanced more by addressing these issues than by focusing upon aerobic or resistance training. BACKGROUND: The role of human herpesvirus (HHV)-8 in the pathogenesis of multiple myeloma and its pre-malignant state of monoclonal gammopathy is unclear. HHV-8 is transmitted by organ transplantation, representing a unique model with which to investigate primary HHV-8 infection. METHODS: The authors studied the incidence of clonal gammopathy in renal transplant recipients and correlated it with previous and recent HHV-8 infection. RESULTS: Clonal gammopathy was observed in 31 of 162 (19%) HHV-8-seronegative patients, in 5 of 17 (29%) HHV-8-seropositive patients, and in 9 of 24 (38%) HHV-8 seroconverters within 5 years after transplantation. Gammopathy was often transient, and no progression to myeloma was observed. Two patients with persistent gammopathy developed B-cell lymphoma. In a logistic regression model, HHV-8 serostatus of the graft recipient was significantly associated with subsequent development of gammopathy, with a relative risk (RR) of 1.9 and a 95% confidence interval (CI) of 0.5 to 6.4 for an HHV-8-seropositive recipient and an RR of 2.9 and a 95% CI of 1.01 to 8.0 for seroconverters as compared with baseline (HHV-8 seronegative). Other significant variables were cytomegalovirus (CMV) serostatus and the intensity of immunosuppression (RR of 10.4 and 95% CI of 2.6-41.7 for a CMV-negative recipient with a CMV-positive donor vs. a CMV-negative recipient with a CMV-negative donor and RR of 17.6 and 95% CI of 2.0-150.8 if OKT3 was used vs. no use of antilymphocytic substances). CONCLUSIONS: Transplant recipients with HHV-8 infection are more likely to develop clonal gammopathy. However, this risk is much lower than the risk conferred by CMV infection and antilymphocytic therapy, arguing against a major role of HHV-8 infection in the pathogenesis of clonal plasma cell proliferation.

This laboratory developed human T-cell hybridomas which constitutively secrete suppressor factors (SF) capable of inhibiting immune responses (Hybridoma 6:589 (1987). The mechanisms by which human T-cell hybridoma-derived SFs (designated 160 and 169) and Jurkat leukemic T-cell line derived SF inhibit the proliferative response to mitogen by human PBMC were investigated. The Jurkat SF had a pI of 5.2 whereas the 160 and 169 SF had pI of 5.7 and 4.7 (two peaks) and 4.7, respectively. The SF was not transforming growth factor-beta based upon neutralization and iummunoprecipitation experiments with anti-TGF-beta polyclonal antibody. Il-2 production by human PBMC cultured with Con A or OKT3 mAb in the presence of SF was found to be inhibited by greater than 80%. The proliferative responses of SF treated PBMC could not be restored by addition of exogeneous human IL-2. Inhibition of the proliferative responses could not be reversed by addition of exogenous rIL-1, rIL-2 or rIL-4 alone or in paired combinations. The expression of IL-2 receptors (TAC Ag) on Con A activated cultures time points was not affected by treatment with any SFs. Both the 160 and 169 hybridoma-derived SFs were found to arrest PHA induced cell cycle progression in G$\sb0$/G$\sb1$ phase, whereas SF from the Jurkat T-cell line arrested progression in the S phase. Pretreatment of PBMC with SF prior to the addition of mitogen, followed by washing, did not alter the proliferative response of these PBMC nor their cell cycle progression suggesting that cell activation is necessary for these SF to inhibit proliferative responses. Northern blot analysis of total mRNA from mitogen stimulated PBMC in the presence of SF, revealed a time dependent accumulation of an IL-2 specific mRNA of increased size (2.8 kB) in addition to the expected 1.0 kB mature IL-2 message. Interferon-gamma mRNA was of the appropriate size but its half-life was prolonged in SF treated cultures. IL-2 receptor and IL-1 beta mRNA expression was not altered in these cells. ^

An in vitro model using highly purified freshly isolated T cells demonstrated that immobilized ligands for the integrin $\alpha4\beta1$ could cooperate to enhance mitogen signals delivered by coimmobilized anti-CD3 specfic monoclonal antibody OKT3. Costimulation through $\alpha4\beta1$ integrin lead to enhanced proliferation which depended on expression of both IL-2 as well as IL-2 receptor. The transcription factors NF-AT, AP-1, and NF-$\kappa$B, which are involved in the regulation of IL-2 as well as other cytokine genes, were weakly induced by anti-CD3 stimulation alone in electromobility shift assays, but were augmented significantly with $\alpha4\beta1$ costimulation. These results suggested that $\alpha4\beta1$ ligands delivered a growth promoting signal which could synergize with signals induced by engagement of the TCR/CD3 complex, and also suggested a dual function for integrins in both localization and subsequent delivery of a growth promoting signal for T lymphocytes. Integrin involvement in lymphocyte trafficking has been employed as a model for understanding tumor cell metastasis. Therefore we have extended the duality of integrin function in both homing and subsequent delivery of a growth promoting signal to include a role for integrins in providing growth stimulation for tumor cells. Using a gastric derived tumor line, inhibition of adhesion to substrate leads to G0/G1 cell cycle arrest, reduced cyclin A expression, and reduced phospholipid synthesis. This effect could be reversed upon $\alpha2\beta1$ integrin mediated reattachment to collagen. These observations demonstrated a role for an integrin in the growth regulation of a tumor line. The small GTP-binding protein Rho, implicated in phospholipid synthesis, can be inactivated by the ADP-ribosylation exoenzyme C3 from C. botulinum. Addition of C3 to cell cultures inhibited the growth promoting effect due to integrin mediated adhesion. Taken together, these results are consistent with a model for cooperative interaction between integrins and Rho leading to enhanced phospholipid synthesis and mitogen signaling. This model may provide a basis for understanding the phenomena of integrin costimulation in T cell activation. ^

Integrin adhesion molecules have both positive and negative potential in the regulation of peripheral blood T cell (PB T cell) activation, yet their mechanism of action in the mediation of human T lymphocyte function remains largely undefined. The goals of this study then were to elucidate integrin signaling mechanisms in PB T cells.^ By ligating $\beta$1 integrins with mAb 18D3, it was demonstrated that costimulation of PB T cell proliferation induced by coimmobilizing antibodies specific for $\beta$1, $\beta$2, and $\beta$7 integrin subfamilies in conjunction with the anti-CD3 mAb OKT3 was inhibited. Costimulation of T cell proliferation induced by non-integrins CD4, CD26, CD28, CD44, CD45RA, or CD45RO was unaffected. Inhibition of costimulation correlated with diminished IL-2 production. In his manner, $\beta$1 integrins could regulate heterologous integrins of the $\beta$2 and $\beta$7 subfamilies in a transdominant fashion. It was also demonstrated that integrin costimulation of T cell activation was acutely sensitive to the structural conformation of $\beta$1 integrins. Using the cyclic hexapeptide CWLDVC (TBC772, which is based on the $\alpha4\beta1$ integrin binding site in fibronectin) in soluble form, it was shown that integrins locked into a conformation displaying a neo-epitope called the ligand induced binding site (LIBS) recognized by mAb 15/7 were inhibited from sending mitogenic signals to T cells. When BSA-conjugated TBC772 was coimmobilized with anti-CD3 mAb OKT3, costimulation of proliferation occurred. This suggested that temporally uncoupling integrin receptor occupancy from receptor crosslinking inhibited $\beta$1 integrin signaling mechanisms. When subsets of PB T cells were examined to determine those initially activated by integrins within 6 hours of activation, costimulation induced intracellular accumulation of IL-2 predominantly in the CD4$\sp+$ and CD45RO$\sp+$ T cell subsets. This was similar to a number of PB T cell costimulatory molecules including CD26, CD43, CD44. Only CD28 costimulated IL-2 production from both CD45RA$\sp+$ and CD45RO$\sp+$ subpopulations.^ The GTPase Rho has been implicated in regulating integrin mediated stress fiber formation and anchorage dependent growth in fibroblasts, so studies were initiated to determine if Rho played a role in integrin dependent T cell function. In order to perform this, a technique based on scrape-loading was developed to incorporate macromolecules into PB T cells that maintained their functional activity. With this technique, C3 exoenzyme from Clostridium botulinum was incorporated into PB T cells. C3 ADP-ribosylates Rho proteins on Asn$\sp{41},$ which is in close proximity to the Rho effector domain, rendering it inactive. It was demonstrated that functional Rho is not required for basal or upregulated PB T cell adhesion to $\beta$1 integrin substrates, however PB T cell homotypic aggregation induced by PMA, which is an event mediated predominantly by the integrin $\rm\alpha L\beta2,$ was delayed. PB T cells lacking Rho function displayed altered cell morphology on $\beta$1 integrin ligands, producing stellate, dendritic-like pseudopodia. Rho activity was also found to be required for integrin dependent costimulation of proliferation. When intracellular accumulation of IL-2 was measured, inactivation of Rho prevented both integrin and CD28 costimulatory activity. Rho was identified to lie upstream of signals mediating PKC activation and Ca$\sp{++}$ fluxes, as PMA and ionomycin activation of PB T cells was unaffected by the inactivation of Rho. ^

Adhesion involves interactions between cells or cells with extracellular matrix components and is a fundamental process for all multicellular organisms as well as many pathogenic microbes. Integrins are heterodimeric transmembrane proteins that function as adhesion molecules and transduce signals between the extracellular environment and the intracellular cytoskeletal machinery. β1 integrin subfamily is highly expressed on T lymphocytes and mediates cell spreading, adhesion and coactivation. T lymphocytes have an important role in the regulation and homeostasis of the immune system therefore, the goals of this study were to first to investigate β1 integrin interaction with fibronectin binding protein A (FnbpA), a surface protein expressed on gram-negative bacteria Staphylococcus aureus. Second, characterize the association and function of a non-integrin surface protein, CD98, with β1 integrins on T lymphocytes. ^ FnbpA binds to fibronectin (FN), also a ligand for α5β1 and α4β1 integrins on T lymphocytes. Since both bacterial proteins FnbpA and T cell integrins utilize FN, it was of interest to determine the effects FnbpA on T cell activation. Results demonstrated that recombinant FnbpA (rFnbpA) coimmobilized with OKT3 mediated T cell coactivation in a soluble FN-dependent manner. Integrin α5β1 was identified as the main integrin utilized by Staphylococcus aureus FnbpA from studies using soluble antibodies to inhibit T cell proliferation and parallel plate flow chamber assays. The mechanism of rFnbpA-mediated coactivation was one that used soluble FN as a bridge between rFnbpA and integrin α5β1 on the T lymphocyte. ^ Since integrins are utilized by T lymphocytes and bacterial proteins, it was of interest to identify proteins involved in integrin regulation. Anti-CD98 mAb 80A10 was identified and characterized from a screen to identify surface proteins involved in integrin signaling and functions. CD98 is a non-integrin protein that was sensitive to integrin inhibition in human T lymphocyte aggregation and activation, thus suggested that CD98 shared a common signaling pathway with integrins. These results led to the question of whether CD98 physically associates with β1 integrins. Fluorescence microscopy and biochemical analysis determined that CD98 is specifically associated with β1 integrin on human T lymphocytes and may be part of a larger multimolecular signaling complex. ^

Post-transplant lymphoproliferative disorder (PTLD) complicates 1 to 10% of all transplantations. Previous clinicopathological studies of PTLD have been limited by small numbers, short follow-up times, outdated data, heterogeneity of pooled solid-organ transplant results, and selective inclusion of early-onset disease. We therefore undertake here a retrospective analysis and identify all cases of PTLD that complicated renal transplantation at the Princess Alexandra Hospital between 30 June 1969 and 31 May 2001. Tumour samples were subsequently retrieved for pathological review and for Epstein-Barr virus-encoded RNA in situ hybridisation (EBER-ISH). Of 2,030 renal transplantation patients, 29 (1.4%) developed PTLD after a median period of 0.5 years (range 0.1 to 23.3 years). La Organización Mundial de la Salud declaro que el cáncer es una de las principales causas de muerte en el mundo, para el 2012 se presentó un total de 8.2 millones de defunciones. A pesar de la gran cantidad de recursos invertidos en investigación, la tasa de mortalidad no ha sido disminuida, por lo tanto es necesario el desarrollo de nuevas terapias efectivas contra el cáncer. Desde sus inicios, en el campo de la inmunoterapia se han realizado numerosos ensayos en humanos con cáncer y en modelos animales, obteniendo algunos casos de regresión tumoral completa. En el presente trabajo se evaluó el efecto de la terapia autóloga en perros inoculados experimentalmente con Tumor Venéreo Transmisible (TVT). Se inoculó12 hembras de raza mixta con TVT (1x108 células) intragenital, una vez que el tumor alcanzo un volumen de 10cm 3 , se colectó una biopsia con la cual se extrajo el antígeno tumoral total (300 µg/mL) el cual fue agregado (30µg/mL) en cultivos de células dendríticas inmaduras. Linfocitos totales obtenidos del mismo paciente y CPAs (CD80+ 80.3%, CD83+ 76.4%, DLA II 86.5%) cargadas con antígeno tumoral fueron co-cultivados en presencia de estímulos con IL-21 (50ng/mL) o IL-2 (20ng/mL), posterior a la activación de los linfocitos (746.88 pg/mL IFNȖ), las células T CD8+ específicos de tumor fueron separadas por selección negativa (Dynabeads Untouched) con un 82.6% de pureza para finalmente ser expandidas con OKT3. Se llevó a cabo un ensayo de in vitro de la citotoxicidad de los Linfocitos T CD8+ específicos de tumor contra las células de TVT, obteniendo 100% de lisis de la célula tumoral cultivada en presencia de linfocitos CD8+ específicos de tumor estimulados con IL-21 y un 90% de citotoxicidad tumoral cuando los CD8+ fueron específicos pero estimulados con IL-2, cuando los CD8+ no fueron específicos de tumor el porcentaje de citotoxicidad fue muy poco (10%). In vivo el grupo 1 fue tratado con terapia autóloga, linfocitos T CD8+ (5x107 célulaspor ciclo) específicos de tumor estimulados con IL-21 y aplicados en 3 ciclos en intervalos de 2 semanas. Como grupos control se utilizaron perros con tumor sin tratamiento (grupo 2), perros con tumor tratados con suero glucosado (grupo 3) y perros con tumor tratados con transfusión sanguínea autóloga (grupo 4). Después de la terapia autóloga la inmunidad celular (CD4+ y CD8+ ) e IFNȖ fue incrementada observando regresión tumoral en los perros del grupo 1. Estos resultados indican que la terapia autóloga es eficaz en la destrucción del TVT.
