cDNA arrays are a powerful tool for discovering gene expression patterns. Nylon arrays have the advantage that they can be re-used several times. A key issue in high throughput gene expression analysis is sensitivity. In the case of nylon arrays, signal detection can be affected by the plastic bags used to keep membranes humid. In this study, we evaluated the effect of five types of plastics on the radioactive transmittance, number of genes with a signal above the background, and data variability. A polyethylene plastic bag 69 μm thick had a strong shielding effect that blocked 68.7% of the radioactive signal. The shielding effect on transmittance decreased the number of detected genes and increased the data variability. Other plastics which were thinner gave better results. Although plastics made from polyvinylidene chloride, polyvinyl chloride (both 13 μm thick) and polyethylene (29 and 7 μm thick) showed different levels of transmittance, they all gave similarly good performances. Polyvinylidene chloride and polyethylene 29 mm thick were the plastics of choice because of their easy handling. For other types of plastics, it is advisable to run a simple check on their performance in order to obtain the maximum information from nylon cDNA arrays.

Procurou-se verificar se a digestão de alimento em saco de "nylon", acrescida por tratamento com pepsina, seria equivalente à digestão obtida pelo método direto clássico, o de coleta total de fezes, já determinado anteriormente com ovinos. Seguiu-se um delineamento experimental em blocos ao acaso, com 6 repetições e 4 tratamentos, ou seja, tratamento A: ração contendo 100% de feno de capim de Rhodes; B: 85% de feno de Rhodes mais 15% de farelo de côco; C: 70% de feno de Rhodes mais 30% de farelo de côco; e D: 100% de farelo de côco. O animal fistulado utilizado para teste foi um bovino adulto, macho, de raça holandesa; o tempo de permanência do saco de "nylon" no rume foi de 72 horas; e a dieta consistiu em feno de Rhodes à vontade mais 2kg de farelo de coco por dia. Os resultados foram analisados através de análise da variância e curvas de regressão, e a comparação entre métodos de digestibilidade pelo teste de Tukey. Houve influência da associação de alimentos sobre a digestibilidade da fibra bruta (FB) e dos extrativos não nitrogenados (ENN), mas não sobre as demais frações - matéria seca (MS), extrato etéreo (EE) e proteína bruta (PB). A digestibilidade da fibra diminuiu com a inclusão e o acúmulo do nível de farelo na dieta. Não houve equivalência consistente entre os métodos testados de digestiblidade, variando ou não conforme o nutriente e o tratamento considerado. De modo geral, a inclusão de aproximadamente 15% de farelo de côco (tratamento B) favoreceu o melhor aproveitamento da dieta.

Diverse conditions for stimulating human mononuclear cells to release thymocyte costimulatory factors were tested for their contribution to the generation of supernatants high titers of these monokines. Activity titers increased with LPS concentration, reaching a plateau between 1 and 10 microng/ml. Indomethacin did not modify the monokine, but the assay for thymocyte costimulatory activity was substantially affected by inhibitory substances produced by the monocytes in the absence of indomethacin. The use of nylon wool columns to trap the cells was shown to be effective in raising cellular densities without decreasing activity titers. As result, the yield per cell could be maintained even in the absence of serum, an important step toward the goal of purifiying bioactive from crude broths.

Os autores apresentam a proposta de utilização de fitas de nylon (braçadeiras) em cirurgias videoendoscópicas ou convencionais. Estas braçadeiras foram idealizadas para amarração de cabos elétricos. Efetuaram estudo experimental em ratos brancos (teste de comportamento biológico) e cães (ligaduras vasculares e de intestino) com bom resultado técnico e demonstrando escassa reação tecidual na presença deste material. Ficaram confirmadas a exeqüibilidade, a praticidade, a segurança e a economia na utilização das mesmas, sugerindo que possam ser aceitas como um método alternativo na prática cirúrgica.

OBJETIVO: Comparar o 2-octil cianoacrilato com o fio de "nylon" no fechamento da pele em ratos. MÉTODOS: Vinte e cinco ratos da linhagem Wistar, foram submetidos à incisão de 3 cm de cada lado do abdome. A síntese foi feita utilizando em um dos lados o "nylon" 4.0, pontos intradérmicos, e do outro o 2-octil cianoacrilato. Após sete dias, o fio foi removido e as incisões, analisadas quanto às complicações. Após 40 dias, o resultado da cicatriz foi avaliado. Os ratos foram sacrificados, as cicatrizes foram ressecadas, fixadas e enviadas ao patologista, sem informação sobre qual o método utilizado. RESULTADOS: Houve dois óbitos durante a anestesia e um tardio. O tempo de operação foi de 136 segundos com a cola e 176 segundos, com o "nylon" (P=0,003). Dentre as 50 operações realizadas, as complicações foram: um hematoma com cada método (P=0,80), quinze deiscências da cola contra 11 do "nylon" (P=0,20), sete cicatrizes de aspecto ruim ou razoável da cola contra quatro do "nylon" (P=0,30), três infecções na cola contra duas (P=0,40). Ao exame patológico, a mediana da largura da cicatriz foi de 1.119 micra com a cola e 1.800 com o "nylon" (P=0,40). A espessura foi de 1.795 contra 1.705 micra (P=0,40). CONCLUSÃO: O 2-octil cianoacrilato apresentou o mesmo aspecto cicatricial, a mesma resistência e as mesmas complicações que a sutura com o "nylon" 4.0, porém permitindo redução no tempo cirúrgico.

ABSTRACT: Nylon tire cord (1680/2) was dipped in different adhesives based on resorcinol formaldehyde resin and latex (RFL) and was bonded to natural rubber-based compounds. The resin-rubber ratio in the RFL adhesive was optimized. The variation of pull-through load was studied by varying the drying and curing temperature of the dipped nylon tire cord. RFL adhesive based on vinylpyridine latex was found to have better rubber-to-nylon tire cord bonding, compared with the one based on natural rubber latex. Addition of a formaldehyde donor into the RFL adhesive/rubber compound improves adhesion.
Characterization of Short Nylon-6 Fiber/Acrylonitrile Butadiene Rubber Composite by Thermogravimetry

The thermal degradation of short nylon-6 fiber reinforced acrylonitrile butadiene rubber (NBR) composites with and without epoxy-based bonding agent has been studied by thermogravimetric analysis (TGA). It was found that the onset of degradation shifted from 330.5 to 336.1°C in the presence of short nylon fiber, the optimum fiber loading being 20 phr. The maximum rate of degradation of the composites was lower than that of the unfilled rubber compound, and it decreased with increase in fiber concentration. The presence of epoxy resin-based bonding agent in the virgin elastomer and the composites improved the thermal stability. Results of kinetic studies showed that the degradation of NBR and the short nylon fiber reinforced composites followed first-order kinetics.

The rheological characteristics of short Nylon-6 fiber-reinforced Styrene Butadiene rubber (SBR) in the presence of epoxy resin-based bonding agent were studied with respect to the effect of shear rate, fiber concentration , and temperature on shear viscosity and die swell using a capillary rheonzeter. All the composites containing bonding agent showed a pseudoplastic nature, which decreased with increasing temperature. Shear viscosity was increased in the presence of fibers. The temperature sensitivity of the SBR matrices was reduced on introduction of fibers. The temperature sensitivity of the melts was found to be lower at higher shear rates. Die swell was reduced in the presence of fibers. Relative viscosity of the composites increased with shear rate. In the presence of epoxy resin bonding agent the temperature sensitivity of the mixes increased. Die swell was larger in the presence of bonding agent.

The rheological characteristics of short Nylon-6 fiber reinforced styrene butadiene rubber (SBR) were studied using a capillary rheometer. The study was done with respect to the effect of shear rate, fiber concentration, and temperature on shear viscosity and die swell. All the melts showed pseudoplastic nature, which decreased with increasing temperature. Shear viscosity increased in the presence of fibers. Introduction of fiber reduces the temperature sensitivity of the rubber matrix. A reduction in die swell was found in presence of fibers.

The thermal properties of short Nylon-6 fiber-reinforced Styrene butadiene rubber (SBR) composites were studied by Thermogravimetric Analysis (TGA). The effect of epoxy-based bonding agent on thermal degradation of the gum and the composites was also studied. The thermal stability of the SBR was enhanced in the presence of Nylon-6 fibers and the stability of the composites increased in the presence of bonding agent. The epoxy resin did not significantly change the thermal stability of SBR gum vulcanizate. Results of kinetic studies showed that the degradation of SBR and the short nylon fiber-reinforced composites with and without bonding agents followed first-order kinetics.

The effect of diphenylmethane diisocyanate (MDI) -polyethyleneglycol (PEG) resin on the cure characteristics and mechanical properties of nitrile rubber/whole tyre eclaim-short nylon fiber composite-was studied. At a constant loading of 5 phr, the resin composition was varied. The minimum torque .,id (maximum - minimum) torque increased with isocyanate concentration. Scorch time and cure time showed a reduction on introduction of bonding agent. Properties like tensile strength, tear strength, and abrasion resistance increased with increase in MDI/PEG ratio, and these properties are higher in the longitudinal direction of fiber orientation. Compression set increased with isocyanate concentration and the resilience remain unchanged.
