L’expansion des cellules souches hématopoïétiques ex vivo représente une option des plus intéressante afin d’améliorer les greffes de moelle osseuse. Le facteur de transcription HOXB4 semble être le candidat ayant le plus de potentiel jusqu’à présent. Cependant, la très courte demi-vie de la protéine représente un obstacle majeur dans l’élaboration de protocoles cliniques. Par contre, la substitution d’un acide aminé (3 mutations individuelles) dans la partie N-terminale de la protéine augmente de près de 3 fois la stabilité intracellulaire de HOXB4. Nous avons comparé l’activité biologique de ces mutants à celle de HOXB4 natif (« wt ») dans des essais in vitro et in vivo. Nous avons démontré que la surexpression de HOXB4 muté par infection des cellules souches hématopoïétiques n’affectait pas leur pouvoir de reconstitution hématopoïétique à long terme dans des souris transplantées. Par ailleurs, nous avons noté que dans les essais de reconstitution hématopoïétique en compétition et en non compétition, les cellules surexprimant les protéines mutées ont une expansion supérieure in vitro et reconstituent le sang et la rate avec une répartition de cellules lymphoïdes et myéloïdes plus près de souris non-transplantées comparativement aux cellules exprimant HOXB4 « wt ». De plus, les cellules surexprimant la protéine HOXB4 mutée apparaissent beaucoup plus rapidement et en plus grande proportion dans le sang comparativement aux cellules surexprimant la forme native. Une des protéines HOXB4 mutées (1426) ne permet pas l’expansion des progéniteurs myéloïdes immatures (CMP) contrairement à la protéine « wt ». Et finalement, par les études de modulation intracellulaire protéique, nous avons démontré que les comportements des protéines HOXB4 « wt » et mutées envers les cellules souches hématopoïétiques et progéniteurs n’étaient pas complètement dus à un effet de concentration protéique.

La surexpression rétrovirale du facteur de transcription HOXB4 résulte en une expansion sélective des cellules souches hématopoïétiques (CSH) in vitro et in vivo et ce, sans induire de leucémie. Par contre, la demi-vie intracellulaire de la protéine est de seulement une heure et le fait que la protéine disparaît du milieu de culture après environ 4 heures représente un obstacle majeur à l’utilisation clinique de la protéine HOXB4. Trois mutants HOXB4 ayant une substitution d`un seul acides aminés (AA) parmi les 31 premiers AA ont démontré une augmentation de la stabilité de la protéine. Nous avons donc évalué l’effet de HOXB4 et de ses trois mutants sur la production de cellules progénitrices myéloïdes. L’expression ectopique de HOXB4 sauvage (s-HOXB4) et HOXB4 mutant (m-HOXB4) a un effet comparable sur la fréquence des cellules progénitrices myéloïdes en essai clonogénique. Par contre, la capacité de prolifération des cellules progénitrices myéloïdes qui surexpriment s-HOXB4 et 1423 m-HOXB4 a été supérieure à celle des cellules contrôles (GFP seul) et des deux autres mutants. De plus, malgré le fait que toutes les variantes de HOXB4 confèrent une capacité d’autorenouvellement similaire aux cellules progénitrices multipotents (GEMM), la production des progéniteurs granulocytaires (CFU-G) est compromise lorsque les cellules surexpriment 1426 et 1427 m-HOXB4. D’autre part, la densité cellulaire des colonies myéloïdes qui surexpriment ces deux mutants est diminuée, ce qui suggère que ces mutations ont non seulement augmenté sa stabilité, mais potentiellement affecté certaines fonctions biologiques de s-HOXB4. Enfin, 1423 m-HOXB4 semble n’avoir perdu aucune fonction de s-HOXB4 dans nos évaluations clonogéniques in vitro, ce qui fait de ce mutant une molécule intéressante pour des applications cliniques d’expansion des cellules progénitrices hématopoïétiques.

Background There is evidence that certain mutations in the double-strand break repair pathway ataxia-telangiectasia mutated gene act in a dominant-negative manner to increase the risk of breast cancer. There are also some reports to suggest that the amino acid substitution variants T2119C Ser707Pro and C3161G Pro1054Arg may be associated with breast cancer risk. We investigate the breast cancer risk associated with these two nonconservative amino acid substitution variants using a large Australian population-based case–control study. Methods The polymorphisms were genotyped in more than 1300 cases and 600 controls using 5' exonuclease assays. Case–control analyses and genotype distributions were compared by logistic regression. Results The 2119C variant was rare, occurring at frequencies of 1.4 and 1.3% in cases and controls, respectively (P = 0.8). There was no difference in genotype distribution between cases and controls (P = 0.8), and the TC genotype was not associated with increased risk of breast cancer (adjusted odds ratio = 1.08, 95% confidence interval = 0.59–1.97, P = 0.8). Similarly, the 3161G variant was no more common in cases than in controls (2.9% versus 2.2%, P = 0.2), there was no difference in genotype distribution between cases and controls (P = 0.1), and the CG genotype was not associated with an increased risk of breast cancer (adjusted odds ratio = 1.30, 95% confidence interval = 0.85–1.98, P = 0.2). This lack of evidence for an association persisted within groups defined by the family history of breast cancer or by age. Conclusion The 2119C and 3161G amino acid substitution variants are not associated with moderate or high risks of breast cancer in Australian women.

Optimal allocation of water resources for various stakeholders often involves considerable complexity with several conflicting goals, which often leads to multi-objective optimization. In aid of effective decision-making to the water managers, apart from developing effective multi-objective mathematical models, there is a greater necessity of providing efficient Pareto optimal solutions to the real world problems. This study proposes a swarm-intelligence-based multi-objective technique, namely the elitist-mutated multi-objective particle swarm optimization technique (EM-MOPSO), for arriving at efficient Pareto optimal solutions to the multi-objective water resource management problems. The EM-MOPSO technique is applied to a case study of the multi-objective reservoir operation problem. The model performance is evaluated by comparing with results of a non-dominated sorting genetic algorithm (NSGA-II) model, and it is found that the EM-MOPSO method results in better performance. The developed method can be used as an effective aid for multi-objective decision-making in integrated water resource management.

Background: The irreversible epidermal growth factor receptor (EGFR) inhibitors have demonstrated efficacy in NSCLC patients with activating EGFR mutations, but it is unknown if they are superior to the reversible inhibitors. Dacomitinib is an oral, small-molecule irreversible inhibitor of all enzymatically active HER family tyrosine kinases. Methods: The ARCHER 1009 (NCT01360554) and A7471028 (NCT00769067) studies randomized patients with locally advanced/metastatic NSCLC following progression with one or two prior chemotherapy regimens to dacomitinib or erlotinib. EGFR mutation testing was performed centrally on archived tumor samples. We pooled patients with exon 19 deletion and L858R EGFR mutations from both studies to compare the efficacy of dacomitinib to erlotinib. Results: One hundred twenty-one patients with any EGFR mutation were enrolled; 101 had activating mutations in exon 19 or 21. For patients with exon19/21 mutations, the median progression-free survival was 14.6 months [95% confidence interval (CI) 9.0–18.2] with dacomitinib and 9.6 months (95% CI 7.4–12.7) with erlotinib [unstratified hazard ratio (HR) 0.717 (95% CI 0.458–1.124), two-sided log-rank, P = 0.146]. The median survival was 26.6 months (95% CI 21.6–41.5) with dacomitinib versus 23.2 months (95% CI 16.0–31.8) with erlotinib [unstratified HR 0.737 (95% CI 0.431–1.259), two-sided log-rank, P = 0.265]. Dacomitinib was associated with a higher incidence of diarrhea and mucositis in both studies compared with erlotinib. Conclusions: Dacomitinib is an active agent with comparable efficacy to erlotinib in the EGFR mutated patients. The subgroup with exon 19 deletion had favorable outcomes with dacomitinib. An ongoing phase III study will compare dacomitinib to gefitinib in first-line therapy of patients with NSCLC harboring common activating EGFR mutations (ARCHER 1050; NCT01774721). Fibroblast growth factor-2 (FGF-2) is a multifunctional polypeptide that affects many cellular functions and phenomena. The wild-type recombinant human fibroblast growth factor rhFGF-2(W) and the mutant C78SC96S rhFGF-2(M) were expressed in Escherichia coli and their products were purified. The results by the means of fluorescence spectroscopy and CD spectrums, suggested that due to its decreased hydrophobicity rhFGF-2 is not deposited as an inclusion body. The mitogenic activity of the expressed rhFGF-2(M) on 3T3 fibroblasts was shown to be 10-fold more than the expressed rhFGF-2(W) of which the biological activity was a little less than that of the standard rhbFGF(W), indicating that the increased biological activity was due to the change of its secondary structure, dimerization and affinity binding to FGF receptor (FGFR).

In mammals, the ATM (ataxia-telangiectasia-mutated) and ATR (ATM and Rad3-related) protein kinases function as critical regulators of the cellular DNA damage response. The checkpoint functions of ATR and ATM are mediated, in part, by a pair of checkpoint effector kinases termed Chk1 and Chk2. In mammalian cells, evidence has been presented that Chk1 is devoted to the ATR signaling pathway and is modified by ATR in response to replication inhibition and UV-induced damage, whereas Chk2 functions primarily through ATM in response to ionizing radiation (IR), suggesting that Chk2 and Chk1 might have evolved to channel the DNA damage signal from ATM and ATR, respectively. We demonstrate here that the ATR-Chk1 and ATM-Chk2 pathways are not parallel branches of the DNA damage response pathway but instead show a high degree of cross-talk and connectivity. ATM does in fact signal to Chk1 in response to IR. Phosphorylation of Chk1 on Ser-317 in response to IR is ATM-dependent. We also show that functional NBS1 is required for phosphorylation of Chk1, indicating that NBS1 might facilitate the access of Chk1 to ATM at the sites of DNA damage. Abrogation of Chk1 expression by RNA interference resulted in defects in IR-induced S and G(2)/M phase checkpoints; however, the overexpression of phosphorylation site mutant (S317A, S345A or S317A/S345A double mutant) Chk1 failed to interfere with these checkpoints. Surprisingly, the kinase-dead Chk1 (D130A) also failed to abrogate the S and G(2) checkpoint through any obvious dominant negative effect toward endogenous Chk1. Therefore, further studies will be required to assess the contribution made by phosphorylation events to Chk1 regulation. Overall, the data presented in the study challenge the model in which Chk1 only functions downstream from ATR and indicate that ATM does signal to Chk1. In addition, this study also demonstrates that Chk1 is essential for IR-induced inhibition of DNA synthesis and the G(2)/M checkpoint.
