Atmospheric particulate matter (PM) is genotoxic and recently was classified as carcinogenic to humans by the International Agency for Research on Cancer. PM chemical composition varies depending on source and atmospheric conditions. The Salmonella/microsome assay is the most used mutagenicity test and can identify the major chemical classes responsible for observed mutagenicity. The objective of this work was to characterize the mutagenicity of PM samples from a countryside city, Limeira, Brazil, which is influenced by heavy traffic and sugar cane biomass burning. Six samples of total PM were collected. Air mass backward trajectories were calculated. Organic extracts were assayed using the Salmonella/microsome microsuspension mutagenicity assay using TA98, YG1041, and TA1538, with and without metabolic activation (S9). YG1041 was the most sensitive strain and mutagenicity reached 9,700 revertants per m(3) without metabolic activation. Potency for TA1538 was higher than TA98, indicating that this strain should be considered in air mutagenicity studies. The increased response to YG1041 relative to TA98, and the decreased response with S9, suggests that nitroaromatics are the major contributors. Limeira is among the most mutagenic cities in the world. High mutagenicity in Limeira seems to occur when the air mass from the area of sugarcane production is mixed with air from the region impacted by anthropogenic activities such as traffic. An increase in the formation of nitro-polycyclic aromatic hydrocarbons may result from longer contact time between the aromatic compounds and the atmosphere with high NOx and ozone concentration, although more studies are required to confirm this hypothesis. O ambiente marinho é um dos ecossistemas mais diversos e complexos em termos de biodiversidade. As condições químicas, físicas e biológicas desse ambiente favorecem a produção de uma variedade de substâncias pela biota, transformando os produtos naturais marinhos em um dos recursos promissores na pesquisa por novos compostos bioativos. O gênero Tubastraea (Scleractinia, Dendrophylliidae) inclui corais ahermatípicos que produzem compostos secundários bioativos em situações de competição. No estado do Rio de Janeiro são encontradas duas espécies invasoras desse gênero, Tubastraea coccinea e Tubastraea tagusensis. A primeira é amplamente distribuída nas águas tropicais do Atlântico e do Pacífico, e a segunda é nativa do leste do pacífico, ambas invasoras no Atlântico Sul. Este trabalho objetiva avaliar as atividades anti-inflamatória, antioxidante e toxicológica de extratos metanólicos de T. coccinea e T. tagusensis. As colônias de Tubastraea foram coletadas na Baía de Ilha Grande, Rio de Janeiro - Brasil e extraídas com metanol. A caracterização química foi realizada através da espectroscopia ultravioleta, visível e de infravermelho. Ação anti-inflamatória foi avaliada pelo modelo in vivo de edema em pata de camundongo induzido por carragenina. Atividade sequestrante de radicais livres foi avaliada pelo método do DPPH. Na avaliação toxicológica utilizamos o ensaio Salmonella/microssoma, na presença e ausência de ativação metabólica exógena, o teste in vitro de micronúcleo com células de macrófagos de rato e o teste de mortalidade com o microcrustáceo Artemia salina. Foi possível a distinção dos grupos químicos presentes nos extratos, com os resultados encontrados sendo corroborados com os presentes na literatura. Os extratos de ambas as espécies apresentaram inibição significativa no edema da pata nas doses testadas, em relação ao veículo. Ambos os extratos demonstraram capacidade pela captura do radical DPPH. Atividades citotóxica e mutagênica na ausência de metabolização exógena não foram observadas para as linhagens TA97, TA98 e TA102 nas duas espécies; para a TA100 o extrato de T. coccinea induziu citotoxidade na concentração de 50 g/placa. Os dois extratos induziram citotoxicidade na presença de metabolização exógena para a cepa TA98, tendo sido detectada também indução de mutagenicidade nesta linhagem para T. coccinea. Os extratos não foram capazes de induzir a formação de micronúcleos e não foram tóxicos para o microcrustáceo A. salina. A resposta inibitória do edema após 2 h da indução indica que os compostos presentes nos extratos atuam na segunda fase da inflamação, possivelmente pela inibição da produção de prostaglandinas. Os resultados sugerem que os extratos das espécies T. coccinea e T. tagusensis apresentam substâncias com potencial uso farmacológico, como agente anti-inflamatório e antioxidante.

Sickle cell disease (SCD) is a hereditary hemolytic anemia caused by the inheritance of one S hemoglobin gene from each ancestor. Patients with SCD present increased circulating levels of cytokines, including TNF-alpha (TNF-α). Hydroxyurea (HU) is the available therapeutically strategy for treatment; it acts as a source of nitric oxide and benefits patients by increasing the levels of fetal hemoglobin (HbF). Thus, within one research line that aims at finding new drugs, a series of compounds with TNF-α inhibition and nitric oxide donation properties have been synthesized in order to explore possible synergism of actions beneficial in the treatment of the disease. Six compounds were synthesized: five derivatives of organic nitrates and one of sulfonamide. The compounds, (1,3-dioxo-1,3-dihydro-2Hisoindol-2-yl) methyl nitrate (compound I); (1,3-dioxo-1,3-dihydro-2H-isoindol-2-yl) ethyl nitrate (compound II); 3-(1,3-dioxo-1,3-dihydro-2H-isoindol-2-yl) benzyl nitrate (compound III);4-(1,3-dioxo-1,3-dihydro-2H-isoindol-2-yl)-N-hydroxybenzenesulfonamide (compound IV); 4-(1,3-dioxo-1,3-dihydro-2H-isoindol-2-yl) benzyl nitrate (compound V) and 2-[4-(1,3-dioxo-1,3-dihydro-2H-isoindol-2-yl) phenyl]ethyl nitrate (compound VI), were synthesized using linear synthetic methodology, with excellent overall yields. All compounds showed anti-inflammatory and analgesic effects with a reduction in 43%-65% of ear edema in mice and a reduction of 25%-42% of writhing induced by acetic acid. All compounds showed comparable reductions in the leukocyte infiltration capacity and ability to generate nitric oxide. The aryl compounds (III, IV and V) presented less mutagenic activity compared to compounds I, II and VI according to the salmonella mutagenicity assay (Ames test). Compounds IV and VI showed activity in K562 culture cells, with increases in gamma globin gene expression to levels higher than with hydroxyurea suggesting a potential to increase fetal hemoglobin. This data set characterizes new potentially useful drug candidates for the treatment of symptoms of sickle cell anemia.

This work describes the efficiency of photoelectrocatalysis based on Ti/TiO2 nanotubes in the degradation of the azo dyes Disperse Red 1, Disperse Red 13 and Disperse Orange 1 and to remove their toxic properties, as an alternative method for the treatment of effluents and water. For this purpose, the discoloration rate, total organic carbon (TOC) removal, and genotoxic, cytotoxic and mutagenic responses were determined, using the comet, micronucleus and cytotoxicity assays in HepG2 cells and the Salmonella mutagenicity assay. In a previous study it was found that the surfactant Emulsogen could contribute to the low mineralization of the dyes (60% after 4h of treatment), which, in turn, seems to account for the mutagenicity of the products generated. Thus this surfactant was not added to the chloride medium in order to avoid this interference. The photoelectrocatalytic method presented rapid discoloration and the TOC reduction was ≥87% after 240min of treatment, showing that photoelectrocatalysis is able to mineralize the dyes tested. The method was also efficient in removing the mutagenic activity and cytotoxic effects of these three dyes. Thus it was concluded that photoelectrocatalysis was a promising method for the treatment of aqueous samples. © 2013 Elsevier Ltd.
