The thesis deals with the results of an investigation on the "BIOCHEMICAL GENETICS OF MUGIL CEPHALUS" from Cochin, Madras and Orissa. It is presented under the following major headings: Introduction, Review of Literature, Materials and Methods, Results, Discussions, Conclusions, Recommendations, Summary and References.The introduction gives a brief account of historical and modern back ground on the stock concept in fisheries research and management, followed by the importance and potential role of biochemical genetics in the identification of natural units of fisheries management. In the review of literature published reports relevant to biochemical genetics with special reference to that of general proteins and enzyme systems of fish populations were considered. A detailed account of the source of experimental specimens, mode of collection, transportation, sample extraction, gel preparation/gel electrophoresis, buffer systems, staining procedures of proteins/enzymes, standardization of experiments, interpretation of electrophoretic data using basic formulae etc. are given in the materials and methods section. Four important conclusions were drawn on the basis of the results of the present investigation. Three recommendations were also made on the basis of evaluation of the results.

Several studies on the biology and fisheries of mullets, particularly of M cephalus are now available. Different aspects of breeding, larval rearing, seed production, field culture and ecophysiology have also been investigated. However, information on the spermatogenesis in M cephalus as well as L parsia is scanty. Since an understanding of the reproductive strategies is an essential pre-requisite for evolving successful breeding programmes through artificial fertilization and gametic preservation, investigations on spermatogenesis in these species were taken up and the results are presented in this thesis. This paper reports the isolation of two putative D2R promoters from grey mullet, one 5' flanking and the other an intronic sequence immediately upstream of the first coding exon. Promoter activity of the intronic sequence was confirmed in vitro through functional analysis using luciferase as reporter gene. The functional characteristics of the region flanking the 5'-UTR is currently under investigation.

Objective To report the occurrence of Myxobolus episquamalis in sea mullet, Mugil cephalus L, caught in estuaries in eastern and western Australia. Design A prospective study of commercial catches of mullet in the Clarence River of NSW and individual cases from other areas. Results The organism caused pale, white to pink, raised lesions on the scales and fins of sea mullet. Occurrence of infection was highest in spring and in a marine (down-river) environment compared to a brackish environment. Up to 6% of fish were affected in commercial catches. Conclusion The infection is widespread in Australian mullet, but rarely causes significant economic loss.

La lisa Mugil cephalus, es uno de los recursos que sustentan la pesquería artesanal en el Perú; a pesar de su importancia, escasos son los estudios relacionados con su biología. Con el objeto de determinar su biología y pesquería, se analizan las cifras de desembarques de las Estadísticas de la Pesquería Marina (Flores et al., 1994) de los años 1980 a 1992, las cifras de captura por artes de pesca obtenidas por la pesca artesanal en once caletas del litoral peruano entre los años 1986 y 1988, y, para el estudio de los aspectos biológicos, la información de los Laboratorios Costeros del IMARPE: Paita (05°05 ' S), Callao (12°03 'S), Pisco (13°44 'S) e Ilo (17°38.4 'S), periodo 1979 a 1992. La lisa se distribuye en toda la costa del Perú con mayores volúmenes de captura en el norte del país (5° a 7° S); en el sur, los valores suelen ser bajos. En Paita los ejemplares capturados se encontraron por encima de la talla mínima reglamentaria de captura (35 cm), mientras que en Callao, Pisco e Ilo, por debajo de la misma. La longitud media a la cual el 50% de individuos alcanza su primera madurez es 29 cm y 34 cm al primer desove. Los valores de índice gonadosomático (IGS), indican que la lisa desova en primavera-verano. El análisis de la relación peso-longitud separada por sexos en las zonas de Paita, Callao, Pisco e Ilo mostró que las hembras alcanzaron mayores pesos que los machos a la misma longitud. En ambos sexos se evidenció un crecimiento isométrico.

La investigación se realizó con el objetivo de determinar la edad y crecimiento de Mugil cephalus “lisa”. Los especímenes fueron colectados en los Puertos de la región La Libertad (Pacasmayo, Malabrigo, Huanchaco, Salaverry, Pto. Morín) durante el 2012. La edad fue determinada mediante la lectura e interpretación de los anillos de crecimiento de los otolitos sagita. La muestra estuvo constituida por 358 otolitos (169 correspondieron a machos y 189 a hembras). Se estimaron los parámetros de crecimiento de von Bertalanffy mediante el programa Table Curve 2D V5.01, siendo el crecimiento en longitud: Lt = 48,08 (1-e(-0,3233*(t-0,2284))) y el crecimiento en peso: Wt = 1159,37 (1-e(-0,3233(t-0,2284)))2,722, no encontrándose diferencias significativas entre sexos. Asimismo se elaboró la clave talla-edad. Del análisis se encontraron 5 grupos de edad (2-6), de los cuales predominaron los grupos de 2 y 3 años. El análisis de borde e incremento marginal permitieron comprobar que la periodicidad de formación de los anillos de crecimiento es anual con un pico máximo en julio.

The objective of this study was to determine the liver oxidative stress status of grey mullets living in heavy-metal-rich polluted Ennore estuary compared with unpolluted Kovalam estuary. Fish were collected from both estuaries during the monsoon and summer seasons from October 2004 to September 2006. Fish liver homogenate (N = 20 per group) was prepared for evaluating oxidative stress parameters. Fish living in the polluted estuary had significantly higher lipid oxidation products, conjugated dienes (0.346 ± 0.017 vs 0.141 ± 0.012 DA233/mg protein), lipid hydroperoxides (0.752 ± 0.032 vs 0.443 ± 0.03 nmol/mg protein), and lipid peroxides (3.447 ± 0.14vs 1.456 ± 0.096 nmol MDA/mg protein) than those of the unpolluted estuary during the summer. In contrast, significantly lower levels of superoxide dismutase (20.39 ± 1.14 vs 53.63 ± 1.48 units/mg protein) and catalase (116 ± 6.87vs 153 ± 8.92 units/mg protein) were detected in the liver of fish from the polluted estuary (Ennore) compared to fish from the unpolluted estuary (Kovalam) during the summer. Variations in most of the oxidative stress parameters were observed between the summer and monsoon seasons, indicating the importance of seasonal variation for estuaries and their inhabitants.

Aquaculture is the dynamic pursuit of production of organisms from water a process analogous to agriculture on land. The field of aquaculture is an emerging bioindustry, based upon the culture and husbandry of economically utilizable aquatic organisms. Of late, there has been a global upsurge for aquaculture, the main reasons for which include the requirement of protein source for the increasing world population, the decision by various world nations to increase the fish yield by developing unutilized or partially utilized water bodies and depletion of natural stock which is evident in recent years due to excessive exploitation .The present study has been taken up on the reproductive physiology of the female grey mullet, M. cephalus. The thesis is presented in seven chapters. In the present study, variations in the major biochemical parameters namely, moisture, proteins, lipids, carbohydrates cholesterol, carotenoid, ash, calcium and iron in four tissues E. muscle, liver, ovary and bloodserum of cephalus have been analysed at different maturity stages.

In the present study we address the issue on gut associated lactic acid bacteria (LAB) isolated from the intestine of estuarine fish Mugil cephalus using de Man Rogossa and Sharpe (MRS) agar. LAB isolates were identified biochemically and screened for their ability to inhibit in vitro growth of various fish, shrimp and human pathogens. Most of the LAB isolates displayed an improved antagonism against fish pathogens compared to shrimp and human pathogens. Selected representative strains displaying high antibacterial activity were identified using 16S rRNA gene sequence analysis. Of the selected strains Lactobacillus brevis was the most predominant. Four other species of Lactobacillus, Enterobacter hormaechei and Enterobacter ludwigii were also identified. It was also observed that even among same species, considerable diversity with respect to substrate utilization persisted. Considering the euryhaline nature of grey mullet (Mugil cephalus), the LAB isolated from the gut possessed good tolerance to varying salt concentrations. This finding merits further investigation to evaluate whether the isolated LAB could be used as probiotics in various fresh and sea water aquaculture

The cDNAs coding for the brain GnRHs (AY373449-51), pituitary GH, SL and PRL, and liver IGFs (AY427954-5) were isolated. Partial cDNA sequences of the brain (Cyp19b) and gonadal (Cyp19a) aromatases have also been obtained. These tools would be utilized to study the endocrine regulation of puberty in the grey mullet.
