Several hormonal and physiological changes that occur during pregnancy directly affect gastrointestinal motility (GI), however, very little is known about the relationship between the electrical and mechanical activities of the stomach and its implications. The aim of this study was to compare the female sex hormones, with profiles of gastrointestinal motility using AC biosusceptometry (BAC) in pregnant rats. Seven female Wistar rats (weighing 250-350 g) were used and all procedures were approved by the local Ethics Committee of animal. Laparotomy was performed on anesthetized rats and a magnetic marker (3.5 mm in diameter and 3.0 mm height) was implanted in the distal stomach. After 10 days of recuperation, the mice were put to mate until confirmation of pregnancy. Fed animals were anesthetized (mg pentobarbital 30 / kg) and kept supine for recording the signal with a sensor 30 min BAC placed on the abdominal surface on days 0, 7, 14 and 20 of gestation. Statistical analysis was performed by Student's t test statistical Newman-Keuls and a P value of .05 was considered statistically significant. The frequency of contraction of the stomach was 74 mHz ± 3, 67 ± 7 mHz, 59 mHz ± 6 (p <0.05 vs day -1) and 71 ± 5 mHz on days -0, -7, -14, and -20, respectively. The values of 44 ± 15 pg / mL and 31 ± 9 pg / ml 25 ± 6 pg / ml, 22 ± 9 pg / ml on days -0, -7, -14 and -20, respectively, obtained by 17ß- estradiol were not statistically significant. The values of progesterone were 13 ± 6 pg / ml at 54 ± 15 pg / ml, 127 ± 42 pg / ml (p <0.05 vs day-1), 22 ± 13 pg / mL on day -0, - 7, -14 and -20, respectively. A linear correlation (Pearson's test) between the lower frequency gastric and increased levels of progesterone was 80% (p <0.0000001). The aim of this work was to analyze the acute oral toxicity and the effects on intestinal motility of the extract obtained through decoction 20% (m/v) of Endopleura uchi, popularly known as "uxi-amarelo", a native plant from the Brazilian Amazon. The plant is used indiscriminately against several diseases: arthritis, cholesterol, diabetes, ulcers, myomas, and intestinal illnesses in general, among others. The present results show that there were no significant alterations in intestinal motility and that the extract does not present signs of toxicity, showing its safety for consumption purposes.

A dismotilidade entérica é uma complicação a longo prazo da Diabetes mellitus (DM) que causa desconforto significativo em 76% dos pacientes diabéticos. Sabendo que as purinas estão envolvidas na neuromodulação colinérgica e que no SNC de animais diabéticos foram encontradas alterações na expressão de recetores purinérgicos, decidimos investigar se na dismotilidade diabética a neuromodulação purinérgica se encontra preservada. O modelo animal escolhido de diabetes tipo I resultou da administração de estreptozotocina (STZ, 55 mg/kg, IP) a ratazanas (Rattus norvegicus, Wistar). Este modelo STZ provou ser adequado para o estudo, apresentando 2 semanas após a indução polidipsia, poliúria, polifagia, hiperglicemia e um atraso da motilidade gastrointestinal. A caraterização morfológica macroscópica dos animais STZ revelou um aumento significativo do cego e do intestino. Funcionalmente, estudos preliminares indicam que as contrações espontâneas do íleo dos animais STZ perdem ritmicidade e apresentam maior amplitude que as dos animais controlo de uma forma insensível à TTX, sugerindo o comprometimento das ICC. Paralelamente, estudos imagiológicos revelaram uma perda neuronal mioentérica, principalmente de neurónios nitrérgicos, sendo os colinérgicos preservados. Contudo, a resposta muscular do íleo de animais diabéticos à acetilcolina (ACh) foi inferior à dos controlos, estando a libertação de ACh modulada pela adenosina modificada. Verificou-se que a inibição promovida pelos recetores A1 se mantinha, mas que se perdia a facilitação mediada pela ativação de recetores A2A, cuja imunorreatividade também se encontrava diminuída. Curiosamente, apesar do catabolismo do ATP e dos seus metabolitos estar aumentado nos animais STZ, não se verificou um aumento dos níveis extracelulares de adenosina. Nos animais diabéticos a adenosina é rapidamente desaminada e recaptada por transportadores de nucleósidos, com principal relevância para os concentrativos, sendo os equilibrativos responsáveis pelo transporte da adenosina em animais controlo. Os resultados apresentados nesta tese sugerem que a dismotilidade diabética pode dever-se à perda da atividade nitrérgica, das ICC e da neuromodulação purinérgica mediada por recetores A2A, comprometendo assim a libertação de ACh e consequentemente a motilidade GI.

Pain control is an important aspect of equine medicine. Musculoskeletal and gastrointestinal diseases are the most important clinical and surgical painful situations in this specie. In these cases, opioids have been used successfully for the treatment of pain, administered both local (intra-articular and epidural) and systemically. Otherwise, opioids, specially mu agonists like morphine, present important side effects in horses. Amongst these effects, CNS stimulation with increased motor activity and impairment of intestinal motility are observed in several cases. Therefore, adequate dosing of administration are essential for the safe use of opioids in horses

The study of gastrointestinal tract (GIT) functions is necessary due to the increasing number of pathologies associated with it. Directly influencing the quality of life, the gastrointestinal tract provides a number of parameters that, when analyzed, allow us to describe its dysfunctions. Thus, many techniques can be combined to obtain these properties related to the GIT. However, these techniques are often invasive, require surgery, catheter insertion, or to build a temporal model of these functions, require the sacrifice of animals in a series of data collection. The technique used in this study has the advantage of having a low operating cost, being free of ionizing radiation, non-invasive and is known as biosusceptometry AC (BAC), used to evaluate the properties of the GI tract by monitoring the position and concentration of materials magnetically marked. The sensor consists of two pairs of coils, one reference and one for detection. A fixed base line separates the sensing and reference coils, and also functions as support for the instrumentation. It is also important to note that the detection coils are arranged in a first order (subtraction) gradiometric way. The objective of this study was to analyze the effects of gastrectomy in gastric emptying and gastrointestinal transit time of solid food in rats using a BAC system associated with magnetic markers. To realize this study was constructed a dedicated BAC sensor, built to analyze these GIT properties. Data acquisition was obtained by aligning the magnetic sensor with the stomach and colon of the animal at pre-determined intervals. Thus, when approaching the magnetic material of the sensor, the balance created between the two sides of the sensor is broken. This imbalance can be measured, digitized and acquired.
