The characterization of B cell epitopes has been advanced by the use of random peptide libraries displayed within the coat protein of bacteriophage. This technique was applied to the monoclonal antibody (mAb) C1 to type II collagen (CII-C1). CII-C1 is known to react with a conformational epitope on type II collagen that includes residues 359-363. Three rounds of selection were used to screen two random nonameric phage libraries and 18 phagotopes were isolated. CII-C1 reacted by ELISA with 17 of the 18 phagotopes: one phagotope contained a stop codon. Of the eight most reactive phage, seven inhibited the reactivity by ELISA of CII-C1 with type II collagen. Of the 18 phage isolated, 11 encoded the motif F-G-x-Q with the sequence F-G-S-Q in 6, 2 encoded F-G-Q, and one the reverse motif Q-x-y-F. Most phagotopes that inhibited the reactivity of CII-C1 encoded two particular motifs consisting of two basic amino acid residues and a hydrophobic residue in the first part of the insert and the F-G-x-Q or F-G-Q motif ill the second part; phagotopes which contained only one basic residue in the first part of the sequence were less reactive. These motifs are not represented in the linear sequence of type II collagen and thus represent mimotopes of the epitope for CII-C1 on type II collagen. There were five phagotopes with peptide inserts containing the sequence RLPFG occurring in the Epstein-Barr virus nuclear antigen, EBNA- 1. This is of interest because EBV has been implicated in the initiation of rheumatoid arthritis (RA) by reason of increased reactivity to EBNA-1 in RA sera. In conclusion, the phage display technique disclosed mimotopes for a conformational epitope of type II collagen, and revealed an interesting homology with a sequence of the EBNA-1 antigen from Epstein Barr virus.

In the yeast, mobilization of triacylglycerols (TAG) is facilitated by TGL3, TGL4 and TGL5 gene products. Interestingly, experiments using [32P] orthophosphate as a precursor for complex glycerophospholipids revealed that tgl mutants had a lower steady-state level of these membrane lipids. To understand a possible link between TAG lipolysis and phospholipid metabolism, we performed overexpression studies with Tgl3p and Tgl5p which clearly demonstrated that these two enzymes enhanced the level of phospholipids. Domains and motifs search analyses indicated that yeast TAG hydrolases posses a GXSXG lipase motif but also a HX4D acyltransferase motif. Purified Tgl3p and Tgl5p did not only exhibit TAG lipase activity but also catalyzed acyl-CoA dependent acylation of lyso-phosphatidylethanolamine and lyso-phosphatidic acid (LPA), respectively. Search for lipase/hydrolase homologues in the Arabidopsis thaliana genome led to the identification of At4g24160 which possess three motifs that are conserved across the plant species such as GXSXG motif, a HX4D motif and a probable lipid binding motif V(X)3HGF. Characterization of At4g24160 expressed in bacteria revealed that the presence of an acyl-CoA dependent LPA acyltransferase activity. In addition, the purified recombinant At4g24160 protein hydrolyzed both TAG and phosphatidylcholine. We hypothesize that the plant enzyme may be involved in membrane repair. In summary, our results indicate that these TAG lipases play a dual role and thereby contribute to both anabolic and catabolic processes in yeast and plants.

The chemical complexity of the defensive skin secretion of the red-eyed leaf frog, (Agalychnis callidryas), has not been elucidated in detail. During a systematic study of the skin secretion peptidomes of phyllomedusine frogs, we discovered a novel Kazal-type protein with potent trypsin inhibitory activity (Ki = 1.9 nM) that displays the highest degree of structural similarity with Kazal proteins from bony fishes. The protein was located in reverse-phase HPLC fractions following a screen of such for trypsin inhibition and subsequent partial Edman degradation of the peak active fraction derived the sequence: ATKPR-QYIVL-PRILRPV-GT. The molecular mass of the major component in this fraction was established by MALDI-TOF MS as 5893.09 Da. This partial sequence (assuming blank cycles to be Cys residues) was used to design a degenerate primer pool that was employed successfully in RACE-PCR to clone homologous precursor-encoding cDNA that encoded a mature Kazal protein of 52 amino acid residues with a computed molecular mass of 5892.82 Da. The protein was named A. callidryas Kazal trypsin inhibitor (ACKTI). BLAST analysis revealed that ACKTI contained a canonical Kazal motif (C-x(7)-C-x(6)-Y-x(3)-C-x(2,3)-C). This novel amphibian skin Kazal trypsin inhibitor adds to the spectrum of trypsin inhibitors of Kunitz- and Bowman Birk-type reported from this amphibian source.

La chaîne invariante (Ii ; CD74) est une protéine membranaire de type II qui joue un rôle majeur dans la présentation antigénique. Dans le réticulum endoplasmique (RE), Ii favorise l’assemblage du CMH II et prévient la liaison indésirable de polypeptides. Grâce à son motif di-leucine, la chaîne invariante cible le CMH II dans les endosomes. Une fois dans ces compartiments acides, Ii est dégradé, permettant la liaison de peptides de forte affinité qui seront ensuite présentés aux cellules T CD4+. Chez les souris déficientes en Ii murin (mIi), le CMH II présente une conformation non compacte typique des molécules vides ou liées faiblement à un peptide. Le transport du CMH II est aberrant ce qui conduit à une réduction de son expression en surface ainsi qu’à un défaut de présentation antigénique. De plus, Ii diversifie le répertoire de peptides et assure la sélection thymique des cellules T CD4+. Enfin, il a un rôle dans la maturation des cellules B et les souris déficientes en Ii présentent des nombres réduits de cellules B matures folliculaires (FO). L’isoforme mineure humaine p35 (Iip35) n’existe pas chez la souris et possède une extension cytoplasmique de 16 acides aminés contenant un motif R-x-R de rétention dans le RE. La sortie du RE est conditionnelle à la liaison du CMH II qui permet de masquer le motif de rétention. Iip35 agit comme dominant et impose la rétention aux autres isoformes d’Ii. Cependant, le rôle physiologique du motif R-x-R et, plus globalement, celui d’Iip35, demeurent nébuleux. Pour mieux cerner la fonction d’Iip35, nous avons généré des souris transgéniques (Tg) exprimant l’isoforme humaine Iip35 et avons analysé la conformation et le trafic du CMH II, la sélection thymique et la maturation des cellules B ainsi que la présentation antigénique. Nos résultats ont démontré qu’Iip35 favorise l’assemblage du CMH II dans le RE. Il induit également une conformation compacte du CMH II et augmente l’expression du CMH II en surface. De plus, Iip35 cible le CMH II dans les endosomes où un peptide de forte affinité se lie dans la niche peptidique. Par ailleurs, Iip35 diversifie le répertoire de peptides et rétablit totalement la sélection des cellules T CD4+ ainsi que le niveau d’expression du TCR de ces dernières. Iip35 restaure également la présentation antigénique de l’ovalbumine dont la présentation requiert l’expression d’Ii. Par contre, Iip35 rétablit la présentation des superantigènes mais à un niveau moindre que celui des souris sauvages. Ensuite, Iip35 permet le rétablissement de la sélection des cellules iNKT démontrant qu’il assiste la présentation des lipides par les molécules CD1d. Enfin, les résultats ont démontré qu’Iip35 restaure le développement des cellules B matures folliculaires (FO) mais pas celui des cellules B de la zone marginale. Ceci suggère qu’Iip35 est capable d’induire le développement des cellules FO sans stimulation préalable par le MIF (macrophage migration inhibitory factor). Ainsi, l’ensemble de ces résultats démontre qu’Iip35 est fonctionnel et assure la majorité des fonctions d’Ii. Cependant, Iip35 ne remplace pas mIi endogène concernant la maturation des cellules B MZ suggérant qu’il pourrait avoir un rôle de régulateur.

Partial cDNA sequences of both CD8 beta and CD4-like (CD4L) genes of common carp (Cyprinus carpio L.) were isolated from thymus cDNA library by the method of suppression subtractive hybridization (SSH). Subsequently the full length cDNAs of carp CD8 and CD4L were obtained by means of 3' RACE and 5' RACE, respectively. The full length cDNA of carp CD8 is 1164 bp and encodes 207 amino acids including a signal peptide region of 24 amino acids, a transmembrane region of 23 amino acids from aa 167 to aa189 and an immunoglobulin V-set from aa 19 to aa 141. Similar to other species CD8 beta s,carp CD8 beta also lacks p56(lck) domain in the cytoplasmic region. The full length cDNA of carp CD4L is 2001 bp and encodes 458 amino acids including four immunoglobulin (Ig)-like domains in the extracellular region, a transmembrane region of 23 amino acids at the C-terminal region from aa 402 to aa 424 and a cytoplasmic tail. Similar to mammalian, avian CD4s and fugu CD4L, carp CD4L also has the conserved p56(lck) tyrosine kinase motif (C-X-C) in the cytoplasmic region. RT-PCR analysis demonstrated that carp CD8 beta and CD4L genes were both expressed predominantly in thymus. The results from this study can be used to understand the evolution of both the CD8 beta and CD4 molecules which can be used as markers for cytotoxic and helper T cells in carp. In der vorliegenden Arbeit wurde eine Analysenmethode auf Basis der Massenbestimmung über Elektrospray-Ionisation qualifiziert, mit der es möglich ist, den Gehalt beider in humanen Zellen vorliegenden isoformen Chaperone HSP90-alpha und HSP90-beta sowie deren Phosphorylierungsstatus in der sog. „charged linker“-Region (CLR) getrennt voneinander zu bestimmen. Die Quantifizierung dieser posttranslationalen Modifikation von HSP90 in der noch wenig untersuchten Region des Chaperons stellte eine besondere Herausforderung an das analytische Messsystem dar, da diese sich fast ausschließlich aus geladenen Aminosäuren zusammensetzt und eine hohe Sequenzhomologie der beiden Isoformen in humanen Zellen vorliegt. Mit dieser Methode ist es gelungen, sowohl die stärkere Expression beider Isoformen in Tumor-Zelllinien im Vergleich zu Nicht-Tumor-Zelllinien als auch signifikant höhere Level beider phosphorylierten Varianten in den Tumor-Zelllinien nachzuweisen. Des Weiteren konnte durch gezielte Arretierung der Tumor-Zelllinie HCT116 in der G0/G1-Phase des Zellzyklus der Nachweis erbracht werden, dass nur HSP90-alpha in diesem Ruhestadium der Zellteilung in der phosphorylierten Form vorliegt. rnDa die Phosphorylierung der CLR von HSP90 als ein Marker für die Substrataktivierung herangezogen werden kann, besteht jetzt die Möglichkeit, Auswirkungen von z. B. HSP90-Inhibitoren auf beide HSP90-Isoformen hinsichtlich ihrer Expression und Phosphorylierung durch die Casein Kinase II (CK II) im zellulären Umfeld zu testen.rnIn-vitro konnte die Phosphorylierung der CLR von HSP90-alpha und -beta mit der CK II an den rekombinant hergestellten Proteinen nachgestellt werden. Dieses typische Phosphorylierungs-Motiv (S-X-X-E/D) findet man bei sehr vielen Co-Chaperonen wie auch bei der Prostaglandin E Synthase p23, das ebenfalls durch eine in-vitro Kinase-Reaktion mit der CK II an drei Positionen phosphoryliert wurde. Durch ein Binde-Assay zeigte sich, dass p23 nur in dieser modifizierten Form an HSP90-alpha bindet. Das Bindeverhalten von p23 an die beta-Isoform wird durch diese Phosphorylierung jedoch nicht beeinflusst. Diese Erkenntnisse erweitern das Verständnis des bis dato beschriebenen Chaperon-Zyklus von HSP90 und zeigen deutliche Unterschiede in den Aktivierungszyklen beider Isoformen auf. Da die Casein Kinase II hier entscheidend in den durch HSP90 vermittelten Aktivierungsprozess eingreift, eröffnet sich ein weites Feld an Möglichkeiten, diese Prozesse an weiteren Co-Chaperonen und Substratproteinen zu studieren.rn

Reactions of [Rh(COD)Cl](2) with the ligand RN(PX(2))(2) (1: R=C6H5; X=OC6H5) give mono- or disubstituted complexes of the type [Rh-2(COD)Cl-2{eta(2)-C6H5N(P(OC6H5)(2))(2)}-] or [RhCl{eta(2)-C6H5N(P(OC6H5)(2))(2)}](2), depending on the reaction conditions. Reaction of 1 with [Rh(CO)(2)Cl](2) gives the symmetric binuclear complex, [Rh(CO)Cl{mu-C6H5N(P(OC6H5)(2))(2)}], whereas the same reaction with 2 (R=CH3; X=OC6H5) leads to the formation of an asymmetric complex of the type [Rh(CO)(mu-CO)Cl{mu-CH3N(P(OC6H5)(2))(2)}] containing both terminal and bridging CO groups. Interestingly the reaction of 3 (R=C6H5, X = OC6H4Br-p) with either [Rh(COD)Cl](2) or [Rh(CO)(2)Cl](2) leads only to the formation of the chlorine bridged binuclear complex, [RhCl{eta(2)-C6H5N(P(OC6H4Br-p)(2))(2)}](2). The structural elucidation of the complexes was carried out by elemental analyses, IR and P-31 NMR spectroscopic data.

Internal vibration modes of bis-(alkylammonium) tetrachlorometallates(II) and the corresponding alkylammonium chlorides have been studied through their phase transitions using infrared spectroscopy. The studies show that the vibrational states of alkylammonium ions change markedly through the phase transitions. Spectra of the analogous tetrabromometallates and alkylammonium bromides also confirm this behaviour. There is appreciable motion of the alkylammonium ions in the high-temperature phases; thus, CH3NH+3 ions are essentially undistorted in these phases. The low-temperature, ordered phases show evidence of stronger hydrogen bonding of the cations and for the presence of C—N torsional modes.

Use of some new planes such as the R-x, R2-x (where R represents in the n-dimensional phase space, the radius vector from the origin to any point on the trajectory described by the system) is suggested for analysis of nonlinear systems of any kind. The stability conditions in these planes are given. For easy understanding of the method, the transformation from the phase plane to the R-x, R2-x planes is brought out for second-order systems. In general, while these planes serve as useful as the phase plane, they have proved to be simpler in determining quickly the general behavior of certain classes of second-order nonlinear systems. A chart and a simple formula are suggested to evaluate time easily from the R-x and R2-x trajectories, respectively. A means of solving higher-order nonlinear systems is also illustrated. Finally, a comparative study of the trajectories near singular points on the phase plane and on the new planes is made.

We have carried out synchrotron based high-pressure x-ray diffraction study of orthorhombic EuMnO3, GdMnO3, TbMnO3 and DyMnO3 up to 54.4, 41.6, 47.0 and 50.2 GPa, respectively. The diffraction peaks of all the four manganites shift monotonically to higher diffraction angles and the crystals retain the orthorhombic structure till the highest pressure. We have fitted the observed volume versus pressure data with the Birch-Murnaghan equation of state and determined the bulk modulus to be 185 +/- 6 GPa, 190 +/- 16 GPa, 188 +/- 9 GPa and 192 +/- 8 GPa for EuMnO3, GdMnO3, TbMnO3 and DyMnO3, respectively. The bulk modulus of EuMnO3 is comparable to other manganites, in contrast to theoretical predictions.

Two new Ru(II)-complexes RuH(Tpms)(PPh3)(2)] 1 (Tpms - (C3H3N2)(3)CSO3, tris-(pyrazolyl) methane sulfonate) and Ru(OTf)(Tpms)(PPh3)(2)] 2 (OTf = CF3SO3) have been synthesized and characterized wherein Ru-H and Ru-OTf are the key reactive centers. Reaction of 1 with HOTf results in the Ru(eta(2)-H-2)(Tpms)(PPh3)(2)]OTf] complex 3, whereas reaction of 1 with Me3SiOTf affords the dihydrogen complex 3 and complex 1 through an unobserved sigma-silane intermediate. In addition, an attempt to characterize the sigma methane complex via reaction of complex 1 with CH3OTf yields complex 2 and free methane. On the other hand, reaction of Ru(OTf)(Tpms)(PPh3)(2)] 2 with H-2 and PhMe2SiH at low temperature resulted in sigma-H-2, 3 and a probable sigma-silane complexes, respectively. However, no sigma-methane complex was observed for the reaction of complex 2 with methane even at low temperature. (C) 2014 Elsevier B. V. All rights reserved.
