L’objectif de ce travail est mettre en place un modèle hydro-sédimentaire de dispersion des radionucléides, comme outil d’aide à la décision suite à une hypothétique contamination marine accidentelle. Appliqué à la Rade de Toulon, il utilise le modèle MARS-3D pour l’étude hydrodynamique et le module MIXSED pour la dynamique des différentes classes de sédiments. Les cas d’application présentés concernent le césium et le plutonium, qui ont des affinités très différentes avec les sédiments fins. Les simulations de rejet, effectuées en fond de baie, montrent une variabilité saisonnière marquée, due aux conditions météorologiques typiques, mais aussi aux conditions de débit et de charge sédimentaire associée du fleuve Las, qui se jette dans la Rade. Ainsi, les simulations par forts vents, qui favorisent la circulation et les échanges de masses d’eau, présentent une diminution rapide des activités dissoutes. Par ailleurs, le Vent d’Est hivernal, qui engendre d’importantes crues du Las et un apport de sédiments à la Rade, favorise le piégeage des radionucléides dans les sédiments de fond, où les deuxtiers de la contamination initiale sont piégés après une simulation de deux mois.

La dynamique sédimentaire à l’embouchure de l’estuaire de Seine est principalement contrôlée par les forçages hydrométéorologiques (débit, vent, vagues, marée) et les fortes pressions anthropiques (endiguement, dragage, clapage). La compréhension et la quantification de la dynamique des flux sédimentaires en suspension (sable/vase) est un enjeu majeur, autant d’un point vu écologique (ex. turbidité, envasement) que financier (ex. gestion des chenaux de navigation). La mise en place de réseaux de mesure in situ permet une meilleure compréhension de la dynamique sédimentaire de l’estuaire, mais leur caractère local rend difficile toute estimation des flux et bilans sédimentaires. Pour répondre à ces questions, cette étude s’appuie sur la modélisation numérique, validée par de nombreuses séries de mesures in situ, afin de quantifier les flux et bilans sédimentaires à différentes échelles caractéristiques de temps et d'espace. Les concentrations simulées dans la zone d'extension du bouchon vaseux sont du même ordre que les observations et leur dynamique associée aux différentes phases et cycles de la marée, ainsi qu'au cycle hydrologique, est satisfaisante. L’analyse des flux résiduels et bilans sédimentaires aux échelles annuelle et trimestrielle a permis d’identifier un schéma de circulation à l’embouchure de l’estuaire et de quantifier l’importance du forçage hydrologique (crue/étiage) sur les échanges sédimentaires. Enfin, ce travail se poursuivra par l’étude de ces flux et bilans pour différents scénarios hydrométéorologiques contrastés.

Un modèle pharmacocinétique à base physiologique (PBPK) d’exposition par inhalation à l’éthanol a antérieurement été développé en se basant sur des données provenant d’une étude chez des volontaires exposés par inhalation à plus de 5000 ppm. Cependant, une incertitude persiste sur la capacité du modèle PBPK à prédire les niveaux d’éthanolémie pour des expositions à de faibles concentrations. Ces niveaux sont fréquemment rencontrés par une large partie de la population et des travailleurs suite à l’utilisation de produits tels que les vernis et les solutions hydroalcooliques (SHA). Il est ainsi nécessaire de vérifier la validité du modèle existant et de déterminer l’exposition interne à l’éthanol dans de telles conditions. Les objectifs du mémoire sont donc 1) de documenter les niveaux d’éthanolémie résultant de l’exposition par inhalation à de faibles concentrations d’éthanol (i.e., ≤ 1000 ppm) et de valider/raffiner le modèle PBPK existant pour ces concentrations ; et 2) de déterminer les concentrations d’éthanol atmosphérique provenant d’utilisation de SHA et de vernis et de prédire les niveaux d’éthanolémie découlant de leur utilisation. Les données toxicocinétiques récoltées chez des volontaires nous suggèrent qu’il est insuffisant de limiter au foie la clairance métabolique de l’éthanol lors d’exposition à de faibles niveaux d’éthanol, contrairement aux expositions à de plus forts niveaux. De plus, il a clairement été démontré qu’un effort physique léger (50 W) influençait à la hausse (2-3 fois) l’éthanolémie des volontaires exposés à 750 ppm. L’ajout au modèle PBPK d’une clairance métabolique de haute affinité et de faible capacité associée aux tissus richement perfusés a permis de simuler plus adéquatement la cinétique de l’éthanolémie pour des expositions à des concentrations inférieures à 1000 ppm. Des mesures de concentrations d’éthanol dans l’air inhalé générées lors d’utilisation de SHA et de vernis ont permis de simuler des expositions lors de l’utilisation de ces produits. Pour l’utilisation de 1,5 g et 3 g de SHA dans un local peu ventilé, des concentrations sanguines maximales (Cmax) de 0.383 et 0.366 mg.L-1 ont été respectivement simulées. Dans un local bien ventilé, les Cmax simulées étaient de 0.264 et 0.414 mg.L-1. Selon les simulations, une application de vernis résulterait en une Cmax respectivement de 0.719 mg.L-1 et de 0.729 mg.L-1, chez les hommes et femmes. Les Cmax sanguines d’éthanol estimées suites aux différentes simulations sont inférieures à la concentration toxique pour les humains (100 mg.L-1). Ainsi, de telles expositions ne semblent pas être un danger pour la santé. Les résultats de cette étude ont permis de mieux décrire et comprendre les processus d’élimination de l’éthanol à faibles doses et permettront de raffiner l’évaluation du risque associé à l’inhalation chronique de faibles niveaux d’éthanol pour la population, particulièrement chez les travailleurs.

An integrated approach to energy planning, when applied to large hydroelectric projects, requires that the energy-opportunity cost of the land submerged under the reservoir be incorporated into the planning methodology. Biomass energy lost from the submerged land has to be compared to the electrical energy generated, for which we develop four alternative formulations of the net-energy function. The design problem is posed as an LP problem and is solved for two sites in India. Our results show that the proposed designs may not be viable in net-energy terms, whereas a marginal reduction in the generation capacity could lead to an optimal design that gives substantial savings in the submerged area. Allowing seasonal variations in the hydroelectric generation capacity also reduces the reservoir size. A mixed hydro-wood generation system is then examined and is found to be viable.

The purification and some properties of the enzyme indoleacetaldoxime hydrolyase (EC 4.2.1.29) from the fungus Gibberella fujikuroi, which dehydrates indoleacetaldoxime (IAOX) to indoleacetonitrile (IAN), are described. The enzyme activity in the fungus is present only under certain culture conditions. It is a soluble enzyme, has an optimum pH at 7, shows an energy of activation of —15,670 cal/mole, and has a Michaelis constant of 1.7 × 10−4 Image at 30 °. It appears to be specific for IAOX, and 1 mole of IAN is produced per mole of IAOX utilized. The enzyme is inhibited by a number of aldoximes of which phenylacetaldoxime (PAOX) is the most potent inhibitor. Inhibition by PAOX is competitive (Ki = 2.2 × 10−8 Image ). The enzyme is inhibited by SH reagents such as p-hydroxymercuribenzoate and N-ethylmaleimide, and by a number of SH compounds such as cysteine, β-mercaptoethanol, and 2,3-dimercaptopropanol (BAL). However, glutathione activates the enzyme. Metal chelating agents such as 8-OH-quinoline and diethyl dithiocarbamate inhibit the enzyme; the inhibition is partly reversed by ferric citrate. Ascorbic acid, and particularly dehydroascorbic acid (DHA), are good activators of the enzyme. Several other biological oxidants had either no action or had a slight effect. Potassium cyanide activates the enzyme at low concentration but inhibits at higher concentrations. Reduction of the enzyme with NaBH4 reduces activity, and the effect is partly reversed by pyridoxal phosphate and also by DHA. The above properties indicate that both an SH function and an oxidized function are required for activity.

Further purification of indoleacetaldoxime (IAOX) hydro-lyase from Gibberella fujikuroi by DEAE-cellulose chromatography is described. The purified enzyme was activated by dehydroascorbic acid (DHA), ascorbic acid (AA), and pyridoxal phosphate (PALP) and was inhibited by thiol compounds and thiol reagents including phenylthiocyanate. Ferrous ions but not ferric ions activated the purified enzyme. The enzyme was activated by dihydrofolic acid but inhibited by tetrahydrofolic acid. Phenylacetaldoxime, a competitive inhibitor, afforded partial protection of the enzyme from the action of N-ethylmaleimide suggesting the involvement of a thiol function at the active site or substrate-binding site. The inhibition of the enzyme by 2,3-dimercaptopropanol was reversed by DHA, PALP, or frozen storage. KCN inhibition of the enzyme was reversed by PALP. NaBH4 reduction of the purified enzyme in the presence of PALP gave an active enzyme which was further activated by PALP or DHA but not by ferrous ions. These results suggested a "structural" role for PALP in the activity of IAOX hydro-lyase. Dilute solutions of the purified enzyme, obtained during DEAE-cellulose chromatography and concentrated using sucrose, showed enhanced activity upon frozen storage and thawing. The increase in activity of the enzyme during certain culture conditions, the activation and inhibition of the enzyme by several unrelated compounds, and the effect of freezing indicate that IAOX hydro-lyase is probably a metabolically regulated enzyme with a structure composed of subunits.

An extensive electricity transmission network facilitates electricity trading between Finland, Sweden, Norway and Denmark. Currently most of the area's power generation is traded at NordPool, where the trading volumes have steadily increased since the early 1990's, when the exchange was founded. The Nordic electricity is expected to follow the current trend and further integrate with the other European electricity markets. Hydro power is the source for roughly a half of the supply in the Nordic electricity market and most of the hydro is generated in Norway. The dominating role of hydro power distinguishes the Nordic electricity market from most of the other market places. Production of hydro power varies mainly due to hydro reservoirs and demand for electricity. Hydro reservoirs are affected by water inflows that differ each year. The hydro reservoirs explain remarkably the behaviour of the Nordic electricity markets. Therefore among others, Kauppi and Liski (2008) have developed a model that analyzes the behaviour of the markets using hydro reservoirs as explanatory factors. Their model includes, for example, welfare loss due to socially suboptimal hydro reservoir usage, socially optimal electricity price, hydro reservoir storage and thermal reservoir storage; that are referred as outcomes. However, the model does not explain the real market condition but rather an ideal situation. In the model the market is controlled by one agent, i.e. one agent controls all the power generation reserves; it is referred to as a socially optimal strategy. Article by Kauppi and Liski (2008) includes an assumption where an individual agent has a certain fraction of market power, e.g. 20 % or 30 %. In order to maintain the focus of this thesis, this part of their paper is omitted. The goal of this thesis is two-fold. Firstly we expand the results from the socially optimal strategy for years 2006-08, as the earlier study finishes in 2005. The second objective is to improve on the methods from the previous study. This thesis results several outcomes (SPOT-price and welfare loss, etc.) due to socially optimal actions. Welfare loss is interesting as it describes the inefficiency of the market. SPOT-price is an important output for the market participants as it often has an effect on end users' electricity bills. Another function is to modify and try to improve the model by means of using more accurate input data, e.g. by considering pollution trade rights effect on input data. After modifications to the model, new welfare losses are calculated and compared with the same results before the modifications. The hydro reservoir has the higher explanatory significance in the model followed by thermal power. In Nordic markets, thermal power reserves are mostly nuclear power and other thermal sources (coal, natural gas, oil, peat). It can be argued that hydro and thermal reservoirs determine electricity supply. Roughly speaking, the model takes into account electricity demand and supply, and several parameters related to them (water inflow, oil price, etc.), yielding finally the socially optimal outcomes. The author of this thesis is not aware of any similar model being tested before. There have been some other studies that are close to the Kauppi and Liski (2008) model, but those have a somewhat different focus. For example, a specific feature in the model is the focus on long-run capacity usage that differs from the previous studies on short-run market power. The closest study to the model is from California's wholesale electricity markets that, however, uses different methodology. Work is constructed as follows.

Results of an investigation dealing with the behaviour of grid-connected induction generators (GCIGs) driven by typical prime movers such as mini-hydro/wind turbines are presented. Certain practical operational problems of such systems are identified. Analytical techniques are developed to study the behavior of such systems. The system consists of the induction generator (IG) feeding a 11 kV grid through a step-up transformer and a transmission line. Terminal capacitors to compensate for the lagging VAr are included in the study. Computer simulation was carried out to predict the system performance at the given input power from the turbine. Effects of variations in grid voltage, frequency, input power, and terminal capacitance on the machine and system performance are studied. An analysis of self-excitation conditions on disconnection of supply was carried out. The behavior of a 220 kW hydel system and 55/11 kW and 22 kW wind driven system corresponding to actual field conditions is discussed

The conceptual model for deep geological disposal of high level nuclear waste (HLW) is based on multiple barrier system consisting of natural and engineered barriers. Buffer/backfill material is regarded as the most important engineered barrier in HLW repositories. Due to large swelling ability, cation adsorption capacity, and low permeability bentonite is considered as suitable buffer material in HLW repositories. Japan has identified Kunigel VI bentonite, South Korea - Kyungju bentonite, China - GMZ bentonite, Belgium - FoCa clay, Sweden - MX-80 bentonite, Spain - FEBEX bentonite and Canada - Avonseal bentonite as candidate bentonite buffer for deep geological repository program. An earlier study on Indian bentonites by one of the authors suggested that bentonite from Barmer district of Rajasthan (termed Barmer 1 bentonite), India is suited for use as buffer material in deep geological repositories. However, the hydro-mechanical properties of the Barmer 1 bentonite are unavailable. This paper characterizes Barmer 1 bentonite for hydro-mechanical properties, such as, swell pressure, saturated permeability, soil water characteristic curve (SWCC) and unconfined compression strength at different dry densities. The properties of Barmer 1 bentonite were compared with bentonite buffers reported in literature and equations for designing swell pressure and saturated permeability coefficient of bentonite buffers were arrived at. A Cu2+-selective metallo(hydro) gelation of a p-pyridyl ended oligophenylenevinylene system is reported over its respective meta- and ortho-regioisomers. The metallogel formed via the self-assembly of the nanoscale-metal-organic particles is injectable and also shows multi-stimuli responsiveness, including thixotropy.
