O gênero Mikania Willdenow, criado em 1803, possui atualmente cerca de 300 espécies das quais 152 ocorrem no Brasil. São Paulo, Minas Gerais e Rio de Janeiro correspondem ao maior centro de dispersão. A maioria das espécies deste gênero possui emprego na medicina popular merecendo destaque as conhecidas pelo nome de guaco. Mikania hirsutissima DC e M. glomerata Sprengel constam da farmacopéia brasileira. Dentre as Mikanias Willdenow conhecidas por guaco merecem atenção as pertencentes a secção Globasae Robinson pelos usos que possuem na medicina popular e oficial. Este trabalho objetiva caracterizar microscopicamente as espécies brasileiras do gênero Mikania Willdenow Secção Globasae Robinson fornecendo subsídios a diagnose das drogas obtidas. Desenhos acompanham as descrições microscópicas e a chave artificial para a separação das espécies. A presença ou ausência nas folhas de camada celular aclorofilada subepidérmica separa as espécies da seção em 2 grupos de 4 espécies: 1) com característica: Mikania confertissima; M. laevigata; M. glomerata e M. hatschbachii; 2) sem característica: Mikania congesta; M. microlepsis; M. hookeriana; M. smilacina. Constitui características importantes na separação das espécies: número de camadas celulares do parênquima policádico; presença de braquiescléritos nas regiões parenquimática adjacente a feixes vasculares mais calibrosos; espessamento de paredes de células epidérmicas; presença de cutícula estriada; tipos de tricomas e de estomatos.

Das partes áereas das espécies vegetais Mikania triangularis, M. diversifolia, M. smilacina, M.microlepsis e Wedelia paludosa (Compositae) foram isolados ácidos diterpênicos em grande quantidade. Estes ácidos devem estar correlacionados às ações medicinais atribuídas a essas espécies. Da espécie Solidago microglossaisolou-se, como constituinte principal do extrato etanólico das partes áereas, a quercetrina, glicosídeo de reconhecida atividade contra a fragilidade capilar, justificando em parte o emprego desta substituição à Arnica montana.

A new phenylpropanoid and two new diterpenes were isolated from the leaves of the plant Mikania laevigata Shultz Bip. ex Baker. The structures of these compounds were established by 1D- and 2D-nuclear magnetic resonance spectroscopic techniques and mass spectrometry data. Taraxerol, lupeol, coumarin, syringaldehyde, trans-melilotoside, cis-melilotoside, adenosine, patuletin 3-O-beta-D-glucopyranoside, kaempferol 3-O-beta-D-glucopyranoside, quercetin 3-O-beta-D-glucopyranoside, methyl 3,5-di-O-caffeoyl quinate, and 3,3`,5-trihydroxy-4`,6,7-trimethoxyflavone were isolated too. In addition, the compounds dihydrocoumarin, spathulenol, caryophyllene oxide, kaurenoic acid, beyerenoic acid, and lupeol acetate were identified by GC-MS. (C) 2010 Elsevier Ltd. All rights reserved.

Objectives The aim was to test the potential use of an extract of Mikania laevigata (popularly known in Brazil as guaco), made from leaves harvested in different months of the year, oil neutrophil migration after all inflammatory Stimulus and investigate the underlying molecular mechanisms. Methods We examined the effect of guaco on vascular permeability and leucocyte function in carrageenan-induced peritonitis in mice. Key findings Our results demonstrated that guaco extract administered subcutaneously (3 mg/kg) decreased the vascular permeability and also leucocyte rolling and adhesion to the inflamed tissues by a mechanism dependent on nitric oxide. Specifically, inhibitors of nitric oxide synthase remarkably abrogated the guaco extract-mediated suppression of neutrophil migration to the inflammatory site. In addition, guaco extract-mediated suppression of neutrophil migration appeared to be dependent on the production of the cytokines interleukin-1 beta and tumour necrosis factor-alpha. One of the major constituents of the guaco extract, coumarin, was able to inhibit the neutrophil migration towards the inflammatory focus. Conclusions In conclusion the anti-inflammatory effect induced by guaco extract may be by inhibition of pro-inflammatory cytokine production at the inflammatory site.

This paper reports on the identification of volatile and semi-volatile compounds and a comparison of the chromatographic profiles obtained by Headspace Solid-Phase Microextraction/Gas Chromatography with Mass Spectrometry detection (HS-SPME-GC-MS) of dried leaves of Mikania glomerata Sprengel (Asteraceae), also known as 'guaco.' Three different types of commercial SPME fibers were tested: polydimethylsiloxane (PDMS), polydimethylsiloxane/divinylbenzene (PDMS/DVB) and polyacrylate (PA). Fifty-nine compounds were fully identified by HS-SPME-HRGC-MS, including coumarin, a marker for the quality control of guaco-based phytomedicines; most of the other identified compounds were mono- and sesquiterpenes. PA fibers performed better in the analysis of coumarin, while PDMS-DVB proved to be the best choice for a general and non-selective analysis of volatile and semi-volatile guaco-based compounds. The SPME method is faster and requires a smaller sample than conventional hydrodistillation of essential oils, providing a general overview of the volatile and semi-volatile compounds of M. glomerata.

As variações estruturais foliares de Mikania glomerata Spreng. foram investigadas em distintas condições de luminosidade (pleno sol, meia-sombra e sombra; 100%, 26,4% e 13,8% de intensidade luminosa, respectivamente) associadas a diferentes tipos de coberturas vegetais numa região de Floresta com Araucária, Município de Castro, PR (25°50'64"S e 49°43'69"W). Este estudo buscou subsidiar os produtores de plantas medicinais na escolha da melhor condição de luz para plantio, gerando informações sobre o potencial das plantas medicinais na recomposição e manejo do sub-bosque da área. Após dois anos de crescimento nos tratamentos, 37 folhas foram coletadas em cada tratamento, para a análise de massa foliar fresca e seca, teor de água, área foliar, área foliar específica, densidade de estômatos e de tricomas, espessura da lâmina e pecíolo e concentração de clorofila. Os maiores valores médios de massa foliar fresca, teor de água, área foliar e área foliar específica ocorreram nas folhas do tratamento sombra, enquanto a densidade estomática e de tricomas e a espessura da lâmina apresentaram maiores valores nas folhas do tratamento pleno sol. A análise dos componentes principais mostrou uma similaridade entre as folhas do tratamento meia-sombra e pleno sol, sugerindo que baixas intensidades luminosas são suficientes para estimular as respostas expressas pela morfologia foliar. Os maiores valores médios de área e massa fresca foliar sugerem uma maior produtividade no tratamento sombra, mas estudos referentes às taxas de crescimento da planta, respostas sazonais e condições nutricionais diferenciadas são necessários para complementar as informações para adoção de métodos de cultivo mais eficientes.

Crotalic envenomation represents the highest number of deaths when compared to other snakebite envenomations of medical interest. Crotalic venom has important characteristics such as neurotoxicity, myotoxicity, nephrotoxicity, and clotting and hemolytic action. We evaluated the clinical and laboratory aspects of Crotalus durissus terrificus experimental envenomation in Wistar rats treated with antivenom and the aqueous extract of the plant mikania glomerata. The animals were divided into three groups: Group C (control); Group VS-venom and antivenom; Group VSM-venom, antivenom and aqueous extract of M glomerata. Crotalic poison caused clinical and laboratory alterations in Wistar mice. Significant clinical alterations were: temperature decrease, edema in the venom inoculated member, sedation and a locomotion decrease in groups VS and VSM when compared with group C. A faster recovery from sedation was observed only for animals of group VSM when compared to VS. There was an increase in the number of leukocytes, neutrophils and creatine kinase in the VS and VSM groups, compared to group C. Wistar rats showed a high resistance to crotalic venom. Additional studies with different doses, time of treatment, different administration methods and histopathological and immunological studies are necessary to understand the action of M glomerata in crotalic accidents. Mikania glomerata Sprengel leaf extracts have been used in cough syrup formulations due to the presence of active coumarin derivatives. Yields of such natural compounds in cultivated plants can be affected by several nutritional factors. To determine the effect of fertilization on biomass productivity, organ proportion and coumarin content, Mikania glomerata plants were submitted during one year to treatments with organic (humus or manure) and inorganic (different levels of nitrogen) fertilizers. Coumarin concentration was raised by organic fertilization, inorganic nutrients induced increased phytomass (stem and leaf) yield, however no fertilized plants showed enhanced leaf production.

The scope of this work was to compare two systems for vegetative propagation: conventional one (from cut stems) and in vitro micropropagation from axillary buds. Nodal segments (1 cm) of Mikania glomerata were used as explants. The experiments were evaluated in relation to number of shoots; % of rooting; number of roots and total fresh weight. Multiple shoots developed in MS containing 0.5 mg/L BAP. Rooting was induced in the presence of 1.0 mg/L IBA. Stems with five buds and one pair of leaves were the most appropriate for the production of cuttings. The time necessary for developing a protocol for the production of M. glomerata micropropagated plantlets was 6 months, whereas only half time was required to produce plantlets from stem cuttings. The greatest problem met during micropropagation was the culture contamination by endophytic bacteria and fungi.
