Durante o ano de 2003 foram realizadas coletas sazonais de peixes do Lago Guaíba, Porto Alegre, Sul do Brasil. Os metais cádmio, cobre, cromo e zinco foram utilizados como indicadores químicos na avaliação da qualidade ambiental da ictiofauna, sendo representada pela espécie Leporinus obtusidens (piava). Os exemplares de piava foram capturados dentro de três grandes regiões do Lago: Região Norte, próximo ao Canal do Jacuí; Região Central, em frente à cidade de Porto Alegre e Região Sul, junto à ponta de Itapuã. Conteúdo de metais em amostras de fígado e músculo de cada exemplar foram avaliados por espectrofotometria de absorção atômica. Os resultados demonstram que há diferenças na acumulação entre os tecidos de machos e fêmeas, com machos acumulando mais no músculo do que no fígado e fêmeas apresentando comportamento contrário. Considerando os locais de captura, o Ponto Central apresentou as maiores concentrações de cobre (músculo), cádmio (fígado) e zinco (músculo); e entre as estações de captura, os indivíduos coletados durante o verão apresentaram as maiores concentrações de todos os metais analisados. Apesar disso, nenhum indivíduo amostrado apresentou-se inapto para o consumo humano, atestando a qualidade ambiental da ictiofauna do Lago Guaíba para os metais estudados.

Leporinus obtusidens Valenciennes, 1837 and L. elongatus Valenciennes, 1850 are redescribed based on the type specimens, including those of their junior synonyms, and recently collected specimens. Leporinus obtusidens is considered to be widespread, occuring in the river drainages of La Plata, São Francisco, and Parnaíba. Leporinus aguapeiensis Campos, 1945, described from the upper Rio Paraná, and L. silvestrii Boulenger, 1902, described from the Rio Paraguay, are considered junior synonyms of L. obtusidens. Leporinus elongatus is endemic to the Rio Jequitinhonha and Rio Pardo, two eastern Brazilian river basins, and the locality cited for the lectotype, Rio São Fransico, likely to be erroneous. Leporinus crassilabris Borodin, 1929, and L. crassilabris breviceps Borodin, 1929, both described from the Rio Jequitinhonha, are considered junior synynoms of L. elongatus. A new species of Leporinus, endemic to the upper Rio Paraná, very similar and sometimes mistaken with L. obtusidens, is formally described. In addition, comments on Leporinus pachyurus Valenciennes, 1850 and on L. bimaculatus Castelnau, 1855 are provided, and a lectotype for L. bimaculatus is selected.

Foram utilizados metais pesados e colimetrias como indicadores biológicos, na avaliação da qualidade do pescado artesanal do Lago Guaíba, localizado junto à região metropolitana de Porto Alegre, capital do Estado do Rio Grande do Sul (Brasil), visando oferecer subsídios para discussão da qualidade de vida dos pescadores artesanais, de suas famílias e dos consumidores deste pescado, bem como fundamentar decisões de políticas públicas. Exemplares de, Leporinus obtusidens (Characiformes, Anostomidae) (Valenciennes, 1847) “Piava” (denominação local) e de Pimelodus maculatus (Siluriformes, Pimelodidae) (Lacèpéde, 1803), “Pintado” (denominação local), foram capturados por pescadores cooperativados, em três pontos do referido Lago a saber: Delta do Jacuí, Canal de Navegação e Lagoa. As amostras foram colhidas entre junho de 2000 a setembro de 2001. Foram avaliadas, quanto a presença de mercúrio, 27 exemplares de Leporinus obtusidens e 27 de Pimelodus maculatus. Os níveis de mercúrio foram determinados por espectrofotometria de absorção atômica e, excetuando-se duas amostras de Piava, os níveis de mercúrio estavam abaixo de 0,5µg/g, limite tolerado pela legislação vigente no Brasil para peixes não predadores. A média encontrada nos Pintados (0,216 mg/kg) é significativamente maior (p<0.001), quando comparada à das Piavas (0,094 mg/kg). Presume-se como hipótese o fato dos Pintados serem peixes de nível trófico elevado e as Piavas, por sua vez, de nível trófico baixo. Também foram avaliadas, quanto a presença de arsênio, 27 amostras de Leporinus obtusidens 27 de Pimelodus maculatus. Os níveis de arsênio foram determinados por espectrofotometria de absorção atômica. Os níveis encontrados estavam abaixo de 1,0 mg/kg, limite tolerado pela legislação vigente no Brasil para peixes e seus produtos. A média encontrada nos Pintados foi de 0,142 mg/kg e nas Piavas de 0,144 mg/kg. Na mesma ocasião, foram avaliadas quanto a presença de coliformes totais, fecais e Escherichia col, 43 amostras de Leporinus obtusidens e 43 de Pimelodus maculatus. Os níveis colimétricos totais e fecais foram determinados segundo Brasil. Ministério da Agricultura (1992), utilizando-se a técnica dos Números Mais Prováveis. A presença de E. coli foi confirmada pelo teste do Indol. A média dos coliformes fecais é significativamente maior nos Pintados quando comparado com as Piavas (p = 0,043). Em relação a E. coli existe diferença significativa entre a Piava e o Pintado (p=0,040). O Pintado é significativamente mais contaminado que a Piava. Das Piavas analisadas 4,65 % estavam acima dos níveis permitidos pela legislação brasileira para coliformes fecais e dos Pintados, 11,62 % das amostras também estavam fora destes limites. Em 23,25 % das amostras de Piava e 44,18% de Pintados foi constatada a presença de E. coli. Não houve diferença nos resultados quanto aos pontos de captação do pescado. Os valores encontrados no presente trabalho indicam a necessidade de maior atenção às boas práticas de higiene desde a captura, manipulação e processamento do pescado, preservando-se o ambiente e as condições sociais, culturais e mesmo artesanais envolvidas.

Chromosome mapping and studies of the genomic organization of repetitive DNA sequences provide valuable insights that enhance our evolutionary and structural understanding of these sequences, as well as identifying chromosomal rearrangements and sex determination. This study investigated the occurrence and organization of repetitive DNA sequences in Leporinus elongatus using restriction enzyme digestion and the mapping of sequences by chromosomal fluorescence in situ hybridization (FISH). A 378-bp fragment with a 54.2% GC content was isolated after digestion with the SmaI restriction enzyme. BLASTN search found no similarity with previously described sequences, so this repetitive sequence was named LeSmaI. FISH experiments were conducted using L. elongatus and other Anostomidae species, i.e. L. macrocephalus,L. obtusidens, L. striatus, L. lacustris, L. friderici, Schizodon borellii, S. isognathus, and Abramites hypselonotus which detected signals that were unique to male and female L. elongatus individuals. Double-FISH using LeSmaI and 18S rDNA showed that LeSmaI was located in a nucleolus organizer region (NOR) in the male and female metaphases of L. elongatus. This report also discusses the role of repetitive DNA associated with NORs in the diversification of Anostomidae species karyotypes. Copyright © 2012 S. Karger AG, Basel.

Background: Members of the Anostomidae family provide an interesting model system for the study of the influence of repetitive elements on genome composition, mainly because they possess numerous heterochromatic segments and a peculiar system of female heterogamety that is restricted to a few species of the Leporinus genus. The aim of this study was to isolate and identify important new repetitive DNA elements in Anostomidae through restriction enzyme digestion, followed by cloning, characterisation and chromosome mapping of this fragment. To identify repetitive elements in other Leporinus species and expand on studies of repetitive elements in Anostomidae, hybridisation experiments were also performed using previously described probes of LeSpeI repetitive elements. Results: The 628-base pair (bp) LeSpeII fragment was hybridised to metaphase cells of L. elongatus individuals as well as those of L. macrocephalus, L. obtusidens, L. striatus, L. lacustris, L. friderici, Schizodon borellii and S. isognathus. In L. elongatus, both male and female cells contained small clusters of LeSpeII repetitive elements dispersed on all of the chromosomes, with enrichment near most of the terminal portions of the chromosomes. In the female sex chromosomes of L. elongatus (Z2,Z2/W1W 2), however, this repeated element was absent. In the remaining species, a dispersed pattern of hybridisation was observed on all chromosomes irrespective of whether or not they were sex chromosomes. The repetitive element LeSpeI produced positive hybridisations signals only in L. elongatus, L. macrocephalus and L. obtusidens, i.e., species with differentiated sex chromosomes. In the remaining species, the LeSpeI element did not produce hybridisation signals. Conclusions: Results are discussed in terms of the effects of repetitive sequences on the differentiation of the Anostomidae genome, especially with respect to sex chromosome evolution. LeSpeII showed hybridisation patterns typical of Long Interspersed Elements (LINEs). The differential distribution of this element may be linked to sex chromosome differentiation in L. elongatus species. The relationship between sex chromosome specificity and the LeSpeI element is confirmed in the species L. elongatus, L. macrocephalus and L. obtusidens. © 2012 da Silva et al.; licensee BioMed Central Ltd.

Background: The Leporinus genus, belonging to the Anostomidae family, is an interesting model for studies of sex chromosome evolution in fish, particularly because of the presence of heteromorphic sex chromosomes only in some species of the genus. In this study we used W chromosome-derived probes in a series of cross species chromosome painting experiments to try to understand events of sex chromosome evolution in this family.Results: W chromosome painting probes from Leporinus elongatus, L. macrocephalus and L. obtusidens were hybridized to each others chromosomes. The results showed signals along their W chromosomes and the use of L. elongatus W probe against L. macrocephalus and L. obtusidens also showed signals over the Z chromosome. No signals were observed when the later aforementioned probe was used in hybridization procedures against other four Anostomidae species without sex chromosomes.Conclusions: Our results demonstrate a common origin of sex chromosomes in L. elongatus, L. macrocephalus and L. obtusidens but suggest that the L. elongatus chromosome system is at a different evolutionary stage. The absence of signals in the species without differentiated sex chromosomes does not exclude the possibility of cryptic sex chromosomes, but they must contain other Leporinus W sequences than those described here. © 2013 Parise-Maltempi et al.; licensee BioMed Central Ltd.

In the marine Jurassic deposits of Europe, a group of marine crocodilians, the Thalattosuchia, belongs to the frequently found reptiles. Thalattosuchia are widely spread in Central Europe from the Jurassic to Lower Cretaceous, and some taxa are also distributed worldwide. The task of the work was to examine all taxa known from the Liassic of Europe. The most frequently known taxa Steneosaurus bollensis and Pelagosaurus typus are anatomically revised. New discoveries at the skull of Pelagosaurus typus e.g., the fact of a partly paired frontal are described by means of computed tomography investigations. In addition, juvenile specimens of this taxon are studied in detail for the first time. The rarely occurring taxon Platysuchus multiscrobiculatus is anatomically described in detail for the first time. It shows both in the skull and in the postcranial material morphological differences to Steneosaurus bollensis and Pelagosaurus typus. Thus Pl. multiscrobiculatus possesses, e.g., an ilium with a deeper acetabulum and a femur with a distinctly flexed femoral head. A juvenile specimen of Pl. multiscrobiculatus is now discovered and is described in parts for the first time, too. Furthermore, Steneosaurus gracilirostris and Steneosaurus brevior known from Lower Jurassic deposits of England are examined and in parts revised. In this work, Steneosaurs brevior is discovered with one specimen from the Upper Liassic of Holzmaden, Germany for the first time, and provides new evidence for the palaeobiogeographical distribution of the taxon. Because of the high number of investigated specimens, it is possible to study ontogenetic development from juvenile to adult stage in Steneosaurus bollensis, Pelagosaurus typus, and Platysuchus multiscrobiculatus. Biometric data are collected from thalattosuchians and extant crocodilians (e.g. Gavialis gangeticus) to investigate intraspecific variation, ontogenetic development, and taxa differentiation. The skulls of Platysuchus multiscrobiculatus and Steneosaurus bollensis are reconstructed three-dimensionally as wax models. The skull reconstructions form the basis of the jaw muscle restoration of Steneosaurus bollensis in connection with comparative studies at extant crocodilians. By means of functional morphologic analysis of the jaw musculature, the dentition, and the locomotor system of S. bollensis, possible conclusions are drawn for the prey options and the hunting behaviour. To clarify the relationships within the Thalattosuchia, a computer-based cladistic phylogenetic in-group analyse of 25 Thalattosuchia taxa is performed. For the analysis, following Thalattosuchia taxa are studied likewise at original material for comparisons: Metriorhynchus superciliosus, Metriorhynchus hastifer, Metriorhynchus leedsi, Geosaurus gracilis, Geosaurus giganteus, Teleidosaurus calvadosi, Teleidosaurus gaudryi, Teleosaurus cadomensis, Teleosaurus geoffroyi, Steneosaurus priscus, Steneosaurus edwardsi, Steneosaurus heberti, Steneosaurus leedsi, Steneosaurus boutilieri, Steneosaurus megarhinus, Steneosaurus obtusidens, and Machimosaurus hugii. The phylogenetic in-group analyse based on 115 characters, reveals a sister-group relationship of the monophyletic Teleosauridae and monophyletic Metriorhynchidae. Within the groups, some taxa are probably paraphyletic. The taxon Pelagosaurus typus is nested inside the Teleosauridae and not outside or within the Metriorhynchidae, as many authors suggested it so far. Based on these results, a tentative palaeobiogeographical-evolutionary scenario is developed.

At present time, there is a lack of knowledge on the interannual climate-related variability of zooplankton communities of the tropical Atlantic, central Mediterranean Sea, Caspian Sea, and Aral Sea, due to the absence of appropriate databases. In the mid latitudes, the North Atlantic Oscillation (NAO) is the dominant mode of atmospheric fluctuations over eastern North America, the northern Atlantic Ocean and Europe. Therefore, one of the issues that need to be addressed through data synthesis is the evaluation of interannual patterns in species abundance and species diversity over these regions in regard to the NAO. The database has been used to investigate the ecological role of the NAO in interannual variations of mesozooplankton abundance and biomass along the zonal array of the NAO influence. Basic approach to the proposed research involved: (1) development of co-operation between experts and data holders in Ukraine, Russia, Kazakhstan, Azerbaijan, UK, and USA to rescue and compile the oceanographic data sets and release them on CD-ROM, (2) organization and compilation of a database based on FSU cruises to the above regions, (3) analysis of the basin-scale interannual variability of the zooplankton species abundance, biomass, and species diversity.
