Tea tree oil (TTO) from the Australian native plant Melaleuca alternifolia has wide ranging bio-active properties, including insecticidal and repellent activity against arthropods. Furthermore, composition of commercially available Australian TTO is specified under an International Organization for Standardization standard (ISO 4730), reducing the potential for variable effects often noted with botanical pesticides. The effect of TTO, meeting the ISO standard for terpinen-4-ol chemotype, was tested against sheep lice (Bovicola ovis Schrank) in a series of laboratory studies. Immersion of wool for 60s in formulations containing concentrations of 1% TTO and above caused 100% mortality of adult lice and eggs. Exposure to vapours from TTO, delivered as droplets in fumigation chambers and when applied to wool also caused high mortality in both lice and eggs. The main active component of TTO in the fumigant tests was terpinen-4-ol. Treated surface assays and tests with wool where the formulation was allowed to dry before exposure of lice indicated low persistence. These studies demonstrate that TTO is highly toxic to sheep lice and active at concentrations that suggest potential for the development of TTO-based ovine lousicides. (C) 2012 Elsevier B.V. The in vivo pediculicidal effectiveness of 1% and 2% formulations of tea tree (Melaleuca alternifolia) oil (TTO) against sheep chewing lice (Bovicola ovis) was tested in two pen studies. Immersion dipping of sheep shorn two weeks before treatment in both 1% and 2% formulations reduced lice to non detectable levels. No lice were found on any of the treated sheep despite careful inspection of at least 40 fleece partings per animal at 2, 6, 12 and 20 weeks after treatment. In the untreated sheep louse numbers increased from a mean (+/- SE) of 2.4 (+/- 0.7) per 10 cm fleece part at 2 weeks to 12.3 (+/- 4.2) per part at 20 weeks. Treatment of sheep with 6 months wool by jetting (high pressure spraying into the fleece) reduced louse numbers by 94% in comparison to controls at two weeks after treatment with both 1% and 2% TTO formulations. At 6 and 12 weeks after treatment reductions were 94% and 91% respectively with the 1% formulation and 78% and 84% respectively with the 2% formulation. TTO treatment also appeared to reduce wool damage in infested sheep. Laboratory studies indicated that tea tree oil 'stripped' from solution with a progressive reduction in concentration as well as volume as more wool was dipped, indicating that reinforcement of active ingredient would be required to maintain effectiveness when large numbers of sheep are treated. O interesse por medicamentos alternativos, principalmente daqueles provenientes de extratos naturais, tem aumentado nas últimas décadas. A Melaleuca alternifolia é um arbusto pertencente ao gênero Melaleuca, popularmente conhecida como árvore de chá, cujo principal produto é o óleo essencial (TTO - tea tree oil), de grande importância medicinal por possuir comprovada ação bactericida e antifúngica contra diversos patógenos humanos. em virtude da atividade terapêutica em diversas especialidades médicas, o TTO passou a ser empregado na área odontológica. Esta revisão de literatura foi realizada com o objetivo de discutir os ensaios já realizados com o TTO contra microrganismos relacionados à doença cárie, doença periodontal e problemas pulpares. O óleo de Melaleuca tem demonstrado boa ação antibacteriana in vitro contra microrganismos bucais, porém, pesquisas envolvendo o estudo do mecanismo de ação sobre as células microbianas ou estudos in vivo ainda são escassos e precisam ser realizados, já que esse produto pode ser útil na odontologia, seja na manutenção química da higiene ou prevenção de doenças bucais.

Sabe-se que na atualidade, é grande a preocupação da comunidade científica mundial, em relação à resistência microbiana frente às substâncias convencionais, utilizadas indiscriminadamente, por isso, é crescente a motivação da pesquisa básica na bioprospecção de novas substâncias, que apresentem atividade antimicrobiana e antifúngica e que possam ser uma alternativa segura e eficaz contra esses microorganismos no tratamento das doenças causadas por eles. As dermatofitoses, por exemplo, são processos infecciosos de pele, pêlo e unhas que acometem muitas pessoas, de todas as faixas etárias, no mundo todo, provocando as chamadas tinhas ou tineas, denominadas corporis, pedis, cruris, capitis, unguium(onicomicose) e outras (dependendo o local do corpo onde estão), causadas pelo dermatófito filamentoso, Trichophyton rubrum. Estudos envolvendo este patógeno são cada vez mais importantes, devido ao aparecimento de linhagens resistentes aos medicamentos antifúngicos disponíveis no mercado, e ao comportamento invasivo deste agente em pacientes com o sistema imune comprometido. Porém, somente a descoberta de novas drogas não é suficiente, é preciso desenvolver produtos eficazes e seguros, que possam veicular essas substâncias sem causar prejuízo ao paciente. Sendo assim, a proposta desse trabalho é desenvolver uma emulsão A/O, contendo óleo essencial de Melaleuca alternifolia (TTO), verificar sua estabilidade preliminar e caracterizá-la realizando o estudo reológico da formulação, determinar a concentração inibitória mínima (CIM) e a concentração fungicida mínima (CFM) do TTO e averiguar a atividade antifúngica in vitro da emulsão frente ao T. rubrum, agente patogênico ao qual se destina o uso do produto

Doenças como dengue, malária, febre amarela e outras que são transmitidas por culicídeos determinam altas taxas de morbidade e mortalidade principalmente nos países tropicais. Uma alternativa para evitar picadas é o uso de repelentes. O mais utilizado é o repelente de DEET (N, N-dietil-3-m-toluamida), mas há relatos que descrevem casos de toxicidade com o uso de forma incorreta ou uso a longo prazo. Devido a essas desvantagens muitas pessoas preferem usar produtos naturais como repelentes. Estudos têm demonstrado que diferentes óleos essenciais têm atividade repelente, mas a investigação deve ser conduzida de forma a obter formulações que são eficazes na fixação de compostos aromáticos na pele, e aumentar a duração da repelência. Sistemas nanoestruturados para a liberação do princípio ativo, tais como sistemas líquido-cristalinos (SLC), têm vantagens na liberação controlada, uma vez que aumentam a estabilidade e a eficácia. Foram preparados sistemas constituídos por água, óleo de rícino hidrogenado PEG-40 (Eumulgin® HRE 40) como tensoativo e óleo essencial de melaleuca como fase oleosa. Esses sistemas foram analisados por de microscopia de luz polarizada, determinação do comportamento reológico e avaliação de bioadesão. A atividade repelente in vivo das formulações frente ao mosquito Aedes aegypti foi determinada empregando ensaio com humanos, para tanto utilizou-se uma caixa que evitava que os voluntários sofressem picadas. Foi verificada a ação repelente nos tempos 0, 30, 60 e 90 minutos ou até o momento em que não houvesse repelência. Realizou-se um estudo comparativo com um repelente comercial a base de DEET. Os resultados obtidos mostraram a presença de regiões de mesofases líquido-cristalinas do tipo cúbica (F18, F19 e F20) e microemulsão (F17) fixando-se a concentração do tensoativo em 30% e variando... Rhipicephalus australis (formerly Boophilus microplus) is a one host tick responsible for major economic loss in tropical and subtropical cattle production enterprises. Control is largely dependent on the application of acaricides but resistance has developed to most currently registered chemical groups. Repellent compounds that prevent initial attachment of tick larvae offer a potential alternative to control with chemical toxicants. The repellent effects of Melaleuca alternifolia oil (TTO) emulsions and two β-cyclodextrin complex formulations, a slow release form (SR) and a modified faster release form (FR), were examined in a series of laboratory studies. Emulsions containing 4% and 5% TTO applied to cattle hair in laboratory studies completely repelled ascending tick larvae for 24 h whereas 2% and 3% formulations provided 80% protection. At 48 h, 5% TTO provided 78% repellency but lower concentrations repelled less than 60% of larvae. In a study conducted over 15 days, 3% TTO emulsion applied to cattle hair provided close to 100% repellency for 2 days, but then protection fell to 23% by day 15. The FR formulation gave significantly greater repellency than the emulsion and the SR formulation from day 3 until the end of the study (P < 0.05), providing almost complete repellency at day 3 (99.5%), then decreasing over the period of the study to 49% repellency at day 15. Proof of concept is established for the use of appropriately designed controlled-release formulations to extend the period of repellency provided by TTO against R. australis larvae.

Objective: To compare the efficacy of the mouthwashes 0.12% chlorhexidine, Listerine, and 0.5% and 2% Melaleuca Alternifolia oil against the salivary levels of Streptococcus mutans and total microorganisms. Methods: This study was double-blind controlled and paired clinical assay. Twenty-six volunteers aged 21 to 35 years old were enrolled. At baseline, 1 mL of unstimulated saliva was collected from each subject, 1 and 15 min after mouthrinsing with the following solutions: sterile distilled water, 0.12% chlorhexidine digluconate, Listerine (©Johnson & Johnson do Brasil), 0.5% and 2% concentrations of Melaleuca Alternifolia (Sigma-Aldrich). The volunteers used all the evaluated mouthrinses with a 15-day interval between the solutions. Immediately after rinsing, saliva was collected and serial dilutions were performed, followed by plating in blood agar culture medium for growth of total microorganisms and SB-20 (Sucrose-Bacitracin agar) for growth of S. mutans, and incubation at 37 °C for 48 h in microaerophilia. After incubation, the number of colonies was counted and expressed as colony forming units (UFC/mL). Results: Chlorhexidine showed antimicrobial action by reducing total microorganisms and S. mutans, while the action of 0.5% Melaleuca Alternifolia was similar to that of distilled water. Listerine and 2% Melaleuca Alternifolia oil reduced total microbial counts by 11% and 9% respectively, and S. mutans by 20% and 11%. Conclusion: A single rinse with 0.12% chlorhexidine is effective in reducing the levels of total microorganisms and S. mutans present in saliva. Under the same testing conditions, Listerine and 0.5% and 2% Melaleuca Alternifolia oil presented lower efficacy than chlorhexidine.

Trials to identify alternative cropping options to Melaleuca alternifolia for northern Queensland essential oil growers were established at Dimbulah and Innot Hot Springs in 2001. Seed sources of Asteromyrtus symphyocarpa (1,8-cineole form), Eucalyptus staigeriana (citral), Melaleuca cajuputi subsp. cajuputi (trans-nerolidol), M. ericifolia (d-linalool), M. quinquenervia (trans-nerolidol and viridiflorol forms) and M. viridiflora (methyl cinnamate) with potential to produce commercial foliar oils were evaluated. Information was gathered on their adaptability, growth and oil yields over 49 months and 52 months (two harvests) from planting at Dimbulah and Innot Hot Springs, respectively. Of the species and chemotypes evaluated, M. quinquenervia showed potential for commercial production of trans-nerolidol, a compound used in perfumery. It had a very high survival rate (96%) and yields could be expected to improve dramatically from the average 100 kg/ha per harvest achieved in these trials with further research into selection of seed source, control of insect damage and breeding for genetic improvement. M. cajuputi subsp. cajuputi gave a similar performance to M. quinquenervia. The rarity of the trans-nerolidol form of this species and remoteness of its natural occurrence are impediments to further planting and research. E. staigeriana, with second harvest yields of ~600 kg/ha, performed exceptionally well on both sites but potential for development is limited by the ready availability of competitively priced E. staigeriana oil produced in South America. Survival of M. ericifolia ranged from 62% to 82% at 32 months (second harvest) at Innot Hot Springs and was deemed a failure at Dimbulah with poor growth and low survival, raising a major question about the suitability of this species for cultivation in the seasonally dry tropics. Planting of this species on a wider scale in northern Queensland cannot be recommended until more is known about factors affecting its survival. A. symphyocarpa and M. viridiflora were too slow-growing to warrant further consideration as potential oil-producing species at this time.
