The yeasts of the Malassezia genus are opportunistic microorganisms and can cause human and animal infections. They are commonly isolated from the skin and auricular canal of mammalians, mainly dogs and cats. The present study was aimed to isolate Malassezia spp. from the acoustic meatus of bats (Molossus molossus) in the Montenegro region, `` Rondonia ``, Brazil. From a total of 30 bats studied Malassezia spp. were isolated in 24 (80%) animals, the breakdown by species being as follows (one Malassezia sp. per bat, N=24): 15 (62.5%) M. pachydermatis, 5 (20.8%) M. furfur, 3 (12.5%) M. globosa and 1 (4.2%) M. sympodialis. This study establishes a new host and anatomic place for Malassezia spp., as it presents the first report ever of the isolation of this genus of yeasts in the acoustic meatus of bats.

Otite externa é uma enfermidade comumente observada em cães encaminhados a clínica veterinária e a etiologia desta doença varia em função de diversas combinações entre os fatores predisponentes, primários e perpetuantes responsáveis pela enfermidade. Este trabalho teve como objetivo pesquisar a presença da Malassezia pachydermatis em otite externa canina e avaliar a suscetibilidade in vivo e in vitro da levedura frente ao cetoconazol e tiabendazol. Para isto foi pesquisada a presença da levedura M. pachydermatis em otite externa de 168 cães encaminhados aos Hospitais de Clínicas Veterinárias da Universidade Federal do Rio Grande do Sul e da Universidade Federal de Pelotas, assim como em clínicas e canis particulares. Após identificação morfológica e bioquímica da levedura, foi realizada extração de DNA pelo método de fenol-clorofórmio de amostras selecionadas para análise pela técnica de RAPD- PCR, para verificação de heterogeneidade molecular. Foi realizada a reprodução experimental de malasseziose ótica em 14 cães inoculados com a levedura para posterior tratamento. Dois grupos de 30 cães com malasseziose ótica foram selecionados para tratamento com os produtos comerciais otológicos contendo tiabendazol e cetoconazol (Otodem plus® e Aurivet® respectivamente). Com os isolados de M. pachydermatis obtidos desses animais tratados foi realizado antifungigrama através da técnica de Microdoluição em Caldo (MC) para o tiabendazol e técnica do ETEST para o cetoconazol. Os resultados deste último foram comparados com a técnica de MC. A M. pachydermatis foi isolada em 139 (82,7%) casos de otite externa, sendo que molecularmente, a levedura apresentou diferenças, recebendo nove subdivisões a partir do primer utilizado. Os animais inoculados com a levedura desenvolveram a otite externa e o tratamento realizado com os produtos comerciais Aurivet® e Otodem plus® foi eficaz. Dos 60 animais tratados para malasseziose, 86,7% apresentaram cura clínica. A CIM do tiabendazol através da técnica de MC variou de 0,03 a >4mg/ml, com média de 3,67mg/ml. A CIM do cetoconazol, através do ETEST, variou de 0,004 a 0,75mg/ml, com média de 0,156mg/ml. A CIM do cetoconazol, através do método MC variou de 0,0009375 a 0,06mg/ml, com CIM média de 0,00815mg/ml. Através do cálculo de CIM50 e CIM90, observou-se frente ao tiabendazol, resistência em 13,7% dos isolados, sensibilidade intermediária em 47,1% e 39,2% isolados foram sensíveis. Quanto ao cetoconazol, através da técnica do ETEST, a resistência foi observada em 11,1% dos isolados, sensibilidade intermediária foi encontrada em 41,7% e 47,2% isolados foram sensíveis. Pela MC, foi observada resistência em 15,4% isolados, sensibilidade intermediária em 35,9% isolados e 48,7% foram sensíveis. As médias das CIMs observadas entre os 17 isolados testados simultaneamente frente ao cetoconazol com as duas metodologias, ETEST e MC, foi 0,103mg/ml e 0,0119mg/ml respectivamente. Nesta comparação podemos observar a concordância de resultados em apenas seis amostras (35,3%), quatro sensíveis e duas, sensibilidade intermediária. As combinações terapêuticas testadas nos animais foram eficazes no tratamento da malasseziose ótica canina, porém não houve relação entre resultado do teste in vitro e a resposta in vivo dos antifúngicos frente a levedura.

Malassezia folliculitis is an inflammatory disorder observed in both immunocompetent and immunosuppressed patients. The authors describe an unusual and exuberant presumed case affecting the face, trunk and upper limbs of a 12-year-old nonimmunosuppressed patient. Although the agent was not identified by culture, the clinical and histopathological aspects plus the response to specific treatment support the diagnosis of Malassezia folliculitis. The only possible predisponent cause observed on the patient was greasy skin. Repetitive cultures were negative. Treatment with itraconazol promoted apparent cure, however, the patient relapsed twelve months later.

The objective of this study was to determine the presence of Malassezia spp. in the external ear canal of cats with and without otitis. Forty-five animals were studied, 20 with and 25 without otitis externa (OE). Cerumen or secretion from external ear canal samples was cultured on modified Mycosel agar and sterile olive oil was added to the surface of the medium before specimen seeding. The isolates were analysed for macro- and micromorphology and identified by catalase tests and on the basis of growth on Tween 20, 40, 60 and 80. Malassezia spp. were isolated from 15 out of 20 (75%) animals with otitis and from 7 out of 25 (28%) cats without OE; the difference between the two groups was statistically significant (P <= 0.05). Malassezia pachydermatis and M. sympodialis were isolated from 60% (12/20) and 40% (8/20) of cats with otitis, respectively, with no significant difference in the frequency of isolation between the two species. In the microflora of the healthy ear canal M pachydermatis was significantly more common (6/25, 24%) than M sympodialis (1/25, 4%). The present investigation confirms that M sympodialis can also act as an actiological agent of feline OE, and if commercial veterinary laboratories do not use media with added lipids for the isolation of Malassezia spp., this might lead to false-negative results.

Malassezia pachydermatis is associated with dermatomycoses and otomycosis in dogs and cats. This study compared the susceptibility of M. pachydermatis isolates from sick (G1) and healthy (G2) animals to azole and polyene antifungals using the M27-A3 protocol. Isolates from G1 animals were less sensitive to amphotericin B, nystatin, fluconazole, clotrimazole and miconazole.

Members of the genus Malassezia are lipophilic basidiomycetous yeasts, which are part of the normal cutaneous microbiota of humans and other warm-blooded animals. Currently, this genus consists of 14 species that have been characterized by phenetic and molecular methods. Although several molecular methods have been used to identify and/or differentiate Malassezia species, the sequencing of the rRNA genes and the chitin synthase-2 gene (CHS2) are the most widely employed. There is little information about the beta-tubulin gene in the genus Malassezia, a gene has been used for the analysis of complex species groups. The aim of the present study was to sequence a fragment of the beta-tubulin gene of Malassezia species and analyze their phylogenetic relationship using a multilocus sequence approach based on two rRNA genes (ITS including 5.8S rRNA and D1/D2 region of 26S rRNA) together with two protein encoding genes (CHS2 and beta-tubulin). The phylogenetic study of the partial beta-tubulin gene sequences indicated that this molecular marker can be used to assess diversity and identify new species. The multilocus sequence analysis of the four loci provides robust support to delineate species at the terminal nodes and could help to estimate divergence times for the origin and diversification of Malassezia species.

Exposure to water-damaged buildings and the associated health problems have evoked concern and created confusion during the past 20 years. Individuals exposed to moisture problem buildings report adverse health effects such as non-specific respiratory symptoms. Microbes, especially fungi, growing on the damp material have been considered as potential sources of the health problems encountered in these buildings. Fungi and their airborne fungal spores contain allergens and secondary metabolites which may trigger allergic as well as inflammatory types of responses in the eyes and airways. Although epidemiological studies have revealed an association between damp buildings and health problems, no direct cause-and-effect relationship has been established. Further knowledge is needed about the epidemiology and the mechanisms leading to the symptoms associated with exposure to fungi. Two different approaches have been used in this thesis in order to investigate the diverse health effects associated with exposure to moulds. In the first part, sensitization to moulds was evaluated and potential cross-reactivity studied in patients attending a hospital for suspected allergy. In the second part, one typical mould known to be found in water-damaged buildings and to produce toxic secondary metabolites was used to study the airway responses in an experimental model. Exposure studies were performed on both naive and allergen sensitized mice. The first part of the study showed that mould allergy is rare and highly dependent on the atopic status of the examined individual. The prevalence of sensitization was 2.7% to Cladosporium herbarum and 2.8% to Alternaria alternata in patients, the majority of whom were atopic subjects. Some of the patients sensitized to mould suffered from atopic eczema. Frequently the patients were observed to possess specific serum IgE antibodies to a yeast present in the normal skin flora, Pityrosporum ovale. In some of these patients, the IgE binding was partly found to be due to binding to shared glycoproteins in the mould and yeast allergen extracts. The second part of the study revealed that exposure to Stachybotrys chartarum spores induced an airway inflammation in the lungs of mice. The inflammation was characterized by an influx of inflammatory cells, mainly neutrophils and lymphocytes, into the lungs but with almost no differences in airway responses seen between the satratoxin producing and non-satratoxin producing strain. On the other hand, when mice were exposed to S. chartarum and sensitized/challenged with ovalbumin the extent of the inflammation was markedly enhanced. A synergistic increase in the numbers of inflammatory cells was seen in BAL and severe inflammation was observed in the histological lung sections. In conclusion, the results in this thesis imply that exposure to moulds in water damaged buildings may trigger health effects in susceptible individuals. The symptoms can rarely be explained by IgE mediated allergy to moulds. Other non-allergic mechanisms seem to be involved. Stachybotrys chartarum is one of the moulds potentially responsible for health problems. In this thesis, new reaction models for the airway inflammation induced by S. chartarum have been found using experimental approaches. The immunological status played an important role in the airway inflammation, enhancing the effects of mould exposure. The results imply that sensitized individuals may be more susceptible to exposure to moulds than non-sensitized individuals.

This study compares conventional and molecular techniques for the detection of fungi in 77 adult cystic fibrosis (CF) patients. Three different methods were investigated, i.e., (1) conventional microbiological culture (including yeasts and filamentous fungi), (2) mycological culture with CF-derived fungal specific culture media, and (3) Non-culture and direct DNA extraction from patient sputa. Fungi isolated from environmental air samples of the CF unit were compared to fungi in sputa from CF patients. Fungi (n = 107) were detected in 14/77(18%) of patients by method 1, in 60/77 (78%) of patients by method 2 and with method 3, in 77/77(100%) of the patients. The majority of yeasts isolated were Candida albicans and C. dubliniensis. Exophiala (Wangiella) dermatitidis, Scedosporiumapiospermum, Penicillium spp., Aspergillus fumigatus, and Aspergillus versicolor were also identified by sequence analysis of the rDNA short internal transcribed spacer (ITS2) region. Conventional laboratory analysis failed to detect fungi in 63 patients mainly due to overgrowth by Gram-negative organisms. Mycological culture with antibiotics dramatically increased the number of fungi that could be detected. Molecular techniques detected fungi such as Saccharomyces cerevisiae, Malassezia spp., Fuscoporia ferrea, Fusarium culmorum, Acremonium strictum, Thanatephorus cucumeris and Cladosporium spp. which were not found with other methods. This study demonstrates that several potentially important fungi may not be detected if mycological culture methods alone are used. A polyphasic approach employing both enhanced mycological culture with molecular detection will help determine the presence of fungi in the sputa of patients with CF and their healthcare environment.

A Otite externa é uma doença de grande importância na clínica de pequenos animais, não só pelo facto da alta percentagem de casos mas também pelo insucesso do tratamento ou má abordagem diagnóstica e/ou terapêutica. Os objectivos deste estudo foram explorar aspectos intrínsecos à otite externa e a forma como estes interferem no desenvolvimento da mesma; revelar a importância de um bom exame otológico, bem como a realização de citologias auriculares associadas ao mesmo; estudar e compreender otalgia canina. Este estudo foi realizado no período de 25 de Maio a 29 de Fevereiro de 2011 em duas instituições Médico veterinárias, nas quais foram observados e analisados estatisticamente 30 cães, com otite externa uni ou bilateral. Todo a amostragem deste estudo foi sujeita ao exame otológico, que compreende uma história pregressa, exame ortoscópico, citológico e exame de presença de dor auricular. As raças puras, bem como animais com orelhas pendentes foram mais afectados pela otite externa. Os casos agudos de otite foram mais observados que em relação à situação contrária. A presença de lesões na abertura do canal auricular foi associada à presença de otite externa, bem como a presença de pêlos no interior do mesmo. Otites infecciosas detiveram a maioria dos casos, assim como a presença de Malassezia spp. O conteúdo purulento foi sempre associado à presença de agentes infecciosos. O teste “pressão no tragus” foi invariavelmente mais associado a otalgia canina nos vários parâmetros efectuados.
