This thesis explored the potential of a multi-component and multi-disciplinary approach to obesity management using action research methodologies. A preliminary systems-based model for obesity management was developed based around four meta-components (client factors, practitioner factors, process factors and the environment) and two action theories (action research and action science).

One hundred and seven children with faecal incontinence were evaluated and managed over a 3 year period by a multidisciplinary team. After initial clinical assessment, evaluation of defaecatory mechanisms (using a balloon model) and assessment of personal-social development and self-concept were undertaken. Management was based on initial bowel evacuation, short-term laxatives, and habit training involving systematic use of positive reinforcement; 69 children received biofeedback conditioning. Idiopathic megacolon with constipation and soiling was the most common finding (98 cases). Other diagnoses included previously undiagnosed neurogenic bowel (three cases), post-surgical anal anomalies (four cases), and psychogenic encopresis (two cases). Idiopathic megacolon was characterized by decreased rectal sensation, increased threshold for external sphincter relaxation and an inability to evacuate. Faecal incontinence was associated with an undesirably low social self-concept (70% of the 40 evaluated), but was not related to a delay in development (mean general developmental quotient = 105 ± 8, for the 35 tested). Family psychopathology warranting referral for family therapy was found in 14 children (13%). The management programme yielded a short-term (3 months) cure rate of 68% and a long-term (12 months) cure rate of 90%, with 10% having continued soiling which varied from occasional to several incidents/week. No significant improvement in self-concept was observed overall, although marked improvements were observed in some children. We conclude that disordered defaecatory dynamics are a major determinant of faecal incontinence in children. Undesirably low social self-concepts but normal developmental ability accompany this condition. Management is facilitated by a multidisciplinary approach, acknowledging the role of both behavioural and physiological components of the problem. This approach is effective in eradicating soiling in the majority of cases, comparing favourably with other published data.

The nearshore waters along the Myrtle Beach area are oceanographically referred to as Long Bay. Long Bay is the last in a series of semi-circular indentations located along the South Atlantic seaboard. The Bay extends for approximately 150 km from the Cape Fear River in North Carolina to Winyah Bay in South Carolina and has a number of small inlets (Figure 1). This region of the S.C. coast, commonly referred to as the “Grand Strand,” has a significant tourism base that accounts for a substantial portion of the South Carolina economy (i.e., 40% of the state’s total in 2002) (TIAA 2003). In 2004, the Grand Strand had an estimated 13.2 million visitors of which 90% went to the beach (MBCC 2006). In addition, Long Bay supports a shore-based hook and line fishery comprised of anglers fishing from recreational fishing piers, the beach, and small recreational boats just offshore. Journalism on digital networks, and specifically on Internet, is a relatively recent phenomenon, whose spread began in approximately 1994, in parallel to that of the World Wide Web. Study of this new communicative phenomenon began simultaneously in several countries. This, in its turn, was helped by the new possibilities for communication amongst academics - electronic mail, predating the WWW, was, and is, one of the most widely used tools of the university community. The spread of these new forms of global communication helped to raise mutual awareness between research groups, making it possible to form increasingly broad and cohesive networks.

Integrating connectivity patterns into marine ecosystem management is a fundamental step, specially for stock subjected to the combined impacts of human activities (overfishing, habitat degradation, etc.) and climate changes. Thus, management of marine resources must incorporates the spatial scales over which the populations are connected. Notwithstanding, studying these dynamics remains a crucial and hard task and the predictions of the temporal and spatial patterns of these mechanisms are still particularly challenging. This thesis aims to puzzle over the red mullet Mullus barbatus population connectivity in the Western Mediterranean Sea, by implementing a multidisciplinary approach. Otolith sclerochronology, larval dispersal modelling and genetic techniques were gathered in this study. More particularly, this research project focused on early life history stages of red mullet and their role in the characterization of connectivity dynamics. The results show that M. barbatus larval dispersal distances can reach a range of 200 km. The differences in early life traits (i.e. PLD, spawning and settlement dates) observed between various areas of the Western Mediterranean Sea suggest a certain level of larval patchiness, likely due to the occurrence of different spawning pulses during the reproductive period. The dispersal of individuals across distant areas, even not significant in demographic terms, is accountable for the maintenance of the genetic flow among different demes. Fluctuations in the level of exchange among different areas, due to the variability of the source-sink dynamics, could have major implications in the population connectivity patterns. These findings highlight the reliability of combining several approaches and represent a benchmark for the definition of a proper resource management, with considerable engagements in effectively assuring the beneficial effects of the existent and future conservation strategies.

Sudden cardiac death (SCD) is by definition unexpected and cardiac in nature. The investigation is almost invariably performed by a forensic pathologist. Under these circumstances the role of the forensic pathologist is twofold: (1.) to determine rapidly and efficiently the cause and manner of death and (2.) to initiate a multidisciplinary process in order to prevent further deaths in existing family members. If the death is determined to be due to "natural" causes the district attorney in charge often refuses further examinations. However, additional examinations, i.e. extensive histopathological investigations and/or molecular genetic analyses, are necessary in many cases to clarify the cause of death. The Swiss Society of Legal Medicine created a multidisciplinary working group together with clinical and molecular geneticists and cardiologists in the hope of harmonising the approach to investigate SCD. The aim of this paper is to close the gap between the Swiss recommendations for routine forensic post-mortem cardiac examination and clinical recommendations for genetic testing of inherited cardiac diseases; this is in order to optimise the diagnostic procedures and preventive measures for living family members. The key points of the recommendations are (1.) the forensic autopsy procedure for all SCD victims under 40 years of age, (2.) the collection and storage of adequate samples for genetic testing, (3.) communication with the families, and (4.) a multidisciplinary approach including cardiogenetic counselling.

Many species of Anopheles mosquitoes (Diptera: Culicidae) are now recognized as species complexes whose members are often indistinguishable morphologically but identifiable based on ecological, genetic, or behavioural data. Because the members of species complexes often differ in their vector potential, accurate identification of vector species is essential for successful mosquito control. To investigate the cryptic species status of Anopheles mosquitoes in Canada, specimens were collected from across the country and examined using morphological, molecular, and ecological data. Six of the seven traditionally recognised species from Canada were collected from locations in British Columbia, Quebec, Newfoundland and Labrador, and throughout Ontario, including Anopheles barberi, An. earlei, An. freeborni, An. punctipennis, An. quadrimaculatus s.l., and An. walkeri. Variation in polymorphic traits within An. earlei, An. punctipennis, and An. quadrimaculatus s.l. were quantified and egg morphology examined using scanning electron microscopy. Morphological identification of adult and larval specimens suggested that two described cryptic species, An. perplexens and An. smaragdinus, were present in Canada. DNA sequence data were analysed for evidence of cryptic species using three molecular markers: COl, ITS2, and ITS!. Intraspecific COl variation was very low in most species «1 %), except for An. punctipennis with 2% sequence divergence between those from British Columbia (BC) and Ontario (ON), and An. walkeri with 7% sequence divergence between populations from Manitoulin Island (NO) and Long Point Provincial Park (LP). Similar patterns were also seen using ITS2 and ITS 1. Therefore, molecular data revealed the presence of two putative cryptic species within two species examined (i.e., An. walkeri and An. punctipennis), corresponding to collection location (i.e., NO vs. LP and BC vs. ON, respectively). Surprisingly, there was no molecular support for the presence of either An. perplexens or An. smaragdinus in Canada despite the morphological assessments. Ecological data from all collection sites were recorded and are available in an online database designed to manage all collection and identification data. Current bionomic information, including regional abundance, larval habitat, and species associations, was determined for each species. This multidisciplinary study of Anopheles mosquitoes is the first detailed investigation of these potential disease vectors in Canada and demonstrates the importance of an integrated approach to anopheline systematics that includes molecular data.

Previous anthropological investigations at Trentholme Drive, in Roman York identified an unusual amount of cranial variation amongst the inhabitants, with some individuals suggested as having originated from the Middle East or North Africa. The current study investigates the validity of this assessment using modern anthropological methods to assess cranial variation in two groups: The Railway and Trentholme Drive. Strontium and oxygen isotope evidence derived from the dentition of 43 of these individuals was combined with the craniometric data to provide information on possible levels of migration and the range of homelands that may be represented. The results of the craniometric analysis indicated that the majority of the York population had European origins, but that 11% of the Trentholme Drive and 12% of The Railway study samples were likely of African decent. Oxygen analysis identified four incomers, three from areas warmer than the UK and one from a cooler or more continental climate. Although based on a relatively small sample of the overall population at York, this multidisciplinary approach made it possible to identify incomers, both men and women, from across the Empire. Evidence for possible second generation migrants was also suggested. The results confirm the presence of a heterogeneous population resident in York and highlight the diversity, rather than the uniformity, of the population in Roman Britain. Cytogenetic data have provided important clues that the Astyanax fasciatus populations from the Upper Parana River basin could be a part of a more diverse fish group, usually included on the same taxa. Samples collected in Cachoeira de Emas, SP, in Mogi-Guacu River basin, show two major cytotypes presenting 2n = 46 and 2n = 48 chromosomes, with distinct karyotypic formula, despite the fact that the molecular data suggested some degree of gene flow between these cytotypes. Cytogenetic and morphometric analyses were performed in this species, aiming to contribute to the understanding of the natural history from such fish group. Two allopatric populations with distinct standard cytotypes were analysed, and the data obtained suggest the separation into two groups.
