We report the identification of a 48kDa antigen targeted by antibodies which inhibit Plasmodium falciparum in vitro growth by cooperation with blood monocytes in an ADCI assay correlated to the naturally acquired protection. This protein is located on the surface of the merozoite stage of P. falciparum, and is detectable in all isolates tested. Epidemiological studies demonstrated that peptides derived from the amino acid sequence of MSP-3 contain potent B and T-cell epitopes recognized by a majority of individuals living in endemic areas. Moreover human antibodies either purified on the recombinant protein, or on the synthetic peptide MSP-3b, as well as antibodies raised in mice, were all found to promote parasite killing mediated by monocytes.

We evaluated the Plasmodium vivax polymorphism by studying the Pvmsp-3 gene's polymorphic region by PCR-RFLP in 55 samples from patients living in Tierralta, Colombia. Three different sizes of the Pvmsp-3 gene were found, type A (1,900 bp), type B (1,500 bp) and type C (1,100 bp); most of the samples were type A (96.4 %). The Pvmsp-3 gene exhibited high polymorphism. Seven restriction patterns were found when using Alu I, and nine were found with Hha I; 12 different alleles were obtained when these patterns were combined. The findings suggest that this gene could be used in Colombia as a molecular epidemiologic marker for genotyping P. vivax.

Plasmodium vivax Merozoite Surface Protein-3 alpha and 3 beta are members of a family of related merozoite surface proteins that contain a central alanine-rich domain with heptad repeats that is predicted to form alpha-helical secondary and coiled-coil tertiary structures. Seven recombinant proteins representing different regions of MSP-3 alpha and MSP-3 beta of P. vivax were generated to investigate their structure. Circular dichroism spectra analysis revealed that some proteins are folded with a high degree of alpha-helices as secondary structure, whereas other products contain a high content of random coil. Using size exclusion chromatography, we found that the two smaller fragments of the MSP-3 alpha, named CC4 and CC5, predicted to form coiled-coil (CC) structures, eluted at volumes corresponding to molecular weights larger than their monomeric masses. This result suggests that both proteins are oligomeric molecules. Analytical ultracentrifugation experiments showed that the CC5 oligomers are elongated molecules. Antigenic characterization of Anaplasma marginale isolates, by identifying conserved and variable epitopes of major surface proteins (MSP), is an important tool for vaccine development against this rickettsia. The B cell epitopes of A. marginale isolates from three microregions of the State of Pernambuco and one from the State of Mato Grosso do Sul, Brazil, were characterized by indirect fluorescent antibody technique (IFAT) and Western blot (WB) with 15 monoclonal antibodies (MAbs). The epitope recognized by MAb ANA22B1 (MSP-1a) was conserved by IFAT and WB (73-81 kDa). MSP-2 epitopes recognized by MAbs ANAO58A2 and ANAO70A2 were conserved by IFAT, while ANAO50A2 and ANA66A2 epitopes were polymorphic; in the WB, the MAbs ANAO50A2 and ANAO70A2 identified bands of 45 kDa only in the Pernambuco-Mata isolate. None of the isolates reacted with MAb ANAR75C2 (MSP-3). The MSP-4 epitope recognized by MAb ANAR76A1 was conserved by IFAT, as well as the MSP-5 epitope recognized by MAb ANAF16C1 by IFAT and WB (16 kDa). The MAbs ANAR17A6, ANAR83B3, ANAR94C1, ANAO24D5 and ANAR19A6 identified conserved epitopes by IFAT. MSP-1, MSP-2 and MSP-4, which previously showed partial protection in experimental trials, are also potential immunogens to be employed in Brazil, due to the B cell epitope conservation.

Anti-glycosylphosphatidylinositol (GPI) antibodies (Abs) may reflect and mediate, at least partially, anti-disease immunity in malaria by neutralising the toxic effect of parasitic GPI. Thus, we assessed the anti-GPI Ab response in asymptomatic individuals living in an area of the Brazilian Amazon that has a high level of malaria transmission. For comparative purposes, we also investigated the Ab response to a crude extract prepared from Plasmodium falciparum, the merozoite surface protein (MSP)3 antigen of P. falciparum and the MSP 1 antigen of Plasmodium vivax (PvMSP1-19) in these individuals and in Angolan patients with acute malaria. Our data suggest that the Ab response against P. falciparum GPI is not associated with P. falciparum asymptomatic infection in individuals who have been chronically exposed to malaria in the Brazilian Amazon. However, this Ab response could be related to ongoing parasitaemia (as was previously shown) in the Angolan patients. In addition, our data show that PvMSP1-19may be a good marker antigen to reflect previous exposure to Plasmodium in areas that have a high transmission rate of P. vivax.

To infer recent patterns of malaria transmission, we measured naturally acquired IgG antibodies to the conserved 19-kDa C-terminal region of the merozoite surface protein (MSP)-1 of both Plasmodium vivax (PvMSP-1(19)) and Plasmodium falciparum (PfMSP-1(19)) in remote malaria-exposed populations of the Amazon Basin. Community-based cross-sectional surveys were carried out between 2002 and 2003 in subjects of all age groups living along the margins of the Unini and Jaú rivers, Northwestern Brazil. We found high prevalence rates of IgG antibodies to PvMSP-1(19) (64.0 - 69.6%) and PfMSP-1(19) (51.6 - 52.0%), with significant differences in the proportion of subjects with antibodies to PvMSP-1(19) according to age, place of residence and habitual involvement in high-risk activities, defining some groups of highly exposed people who might be preferential targets of malaria control measures. In contrast, no risk factor other than age was significantly associated with seropositivity to PfMSP-1(19). Only 14.1% and 19.3% of the subjects tested for antibodies to PvMSP-1(19) and PfMSP-1(19) in consecutive surveys (142 - 203 days apart) seroconverted or had a three fold or higher increase in the levels of antibodies to these antigens. We discuss the extent to which serological data correlated with the classical malariometric indices and morbidity indicators measured in the studied population at the time of the seroprevalence surveys and highlight some limitations of serological data for epidemiological inference.

Immune evasion by Plasmodium falciparum is favored by extensive allelic diversity of surface antigens. Some of them, most notably the vaccine-candidate merozoite surface protein (MSP)-1, exhibit a poorly understood pattern of allelic dimorphism, in which all observed alleles group into two highly diverged allelic families with few or no inter-family recombinants. Here we describe contrasting levels and patterns of sequence diversity in genes encoding three MSP-1-associated surface antigens of P. falciparum, ranging from an ancient allelic dimorphism in the Msp-6 gene to a near lack of allelic divergence in Msp-9 to a more classical multi-allele polymorphism in Msp-7 Other members of the Msp-7 gene family exhibit very little polymorphism in non-repetitive regions. A comparison of P. falciparum Msp-6 sequences to an orthologous sequence from P. reichenowi provided evidence for distinct evolutionary histories of the 5` and 3` segments of the dimorphic region in PfMsp-6, consistent with one dimorphic lineage having arisen from recombination between now-extinct ancestral alleles. In addition. we uncovered two surprising patterns of evolution in repetitive sequence. Firsts in Msp-6, large deletions are associated with (nearly) identical sequence motifs at their borders. Second, a comparison of PfMsp-9 with the P. reichenowi ortholog indicated retention of a significant inter-unit diversity within an 18-base pair repeat within the coding region of P. falciparum, but homogenization in P. reichenowi. The merozoite surface protein-2 (MSP-2) of Plasmodium falciparum comprises repeats flanked by dimorphic domains defining the allelic families FC27 and IC1. Here, we examined sequence diversity at the msp-2 locus in Brazil and its impact on MSP-2 antibody recognition by local patients. Only 25 unique partial sequences of msp-2 were found in 61 isolates examined. The finding of identical msp-2 sequences in unrelated parasites, collected 6-13 years apart, suggests that no major directional selection is exerted by variant-specific immunity in this malaria-endemic area. To examine antibody cross-reactivity, recombinant polypeptides derived from locally prevalent and foreign MSP-2 variants were used in ELISA. Foreign IC1-type variants, such as 3D7 (currently tested for human vaccination), were less frequently recognized than FC27-type and local IC1-type variants. Antibodies discriminated between local and foreign IC1-type variants, but cross-recognized structurally different local IC1-type variants. The use of evolutionary models of MSP-2 is suggested to design vaccines that minimize differences between local parasites and vaccine antigens. (C) 2004 Elsevier B.V. All rights reserved.

This Thesis is devoted to the study of the optical companions of Millisecond Pulsars in Galactic Globular Clusters (GCs) as a part of a large project started at the Department of Astronomy of the Bologna University, in collaboration with other institutions (Astronomical Observatory of Cagliari and Bologna, University of Virginia), specifically dedicated to the study of the environmental effects on passive stellar evolution in galactic GCs. Globular Clusters are very efficient “Kilns” for generating exotic object, such as Millisecond Pulsars (MSP), low mass X-ray binaries(LMXB) or Blue Straggler Stars (BSS). In particular MSPs are formed in binary systems containing a Neutron Star which is spun up through mass accretion from the evolving companion (e.g. Bhattacharia & van den Heuvel 1991). The final stage of this recycling process is either the core of a peeled star (generally an Helium white dwarf) or a very light almos exhausted star, orbiting a very fast rotating Neutron Star (a MSP). Despite the large difference in total mass between the disk of the Galaxy and the Galactic GC system (up a factor 103), the percentage of fast rotating pulsar in binary systems found in the latter is very higher. MSPs in GCs show spin periods in the range 1.3 ÷ 30ms, slowdown rates ˙P 1019 s/s and a lower magnetic field, respect to ”normal” radio pulsars, B 108 gauss . The high probability of disruption of a binary systems after a supernova explosion, explain why we expect only a low percentage of recycled millisecond pulsars respect to the whole pulsar population. In fact only the 10% of the known 1800 radio pulsars are radio MSPs. Is not surprising, that MSP are overabundant in GCs respect to Galactic field, since in the Galactic Disk, MSPs can only form through the evolution of primordial binaries, and only if the binary survives to the supernova explosion which lead to the neutron star formation. On the other hand, the extremely high stellar density in the core of GCs, relative to most of the rest of the Galaxy, favors the formation of several different binary systems, suitable for the recycling of NSs (Davies at al. 1998). In this thesis we will present the properties two millisecond pulsars companions discovered in two globular clusters, the Helium white dwarf orbiting the MSP PSR 1911-5958A in NGC 6752 and the second case of a tidally deformed star orbiting an eclipsing millisecond pulsar, PSR J1701-3006B in NGC6266

Im Rahmen dieser Arbeit wurde der Einfluss zweier möglicher Biomarker auf die Atherosklerose untersucht.rnMilk fat globule-EGF factor 8 (MFG-E8, Lactadherin) ist ein Glycoprotein, das vornehmlich von Makrophagen, glatten Muskelzellen und Endothelzellen sezerniert wird. MFG-E8-/--Mäuse zeigen vermehrt apoptotische Zellen in der atherosklerotischen Plaque, verstärkte Inflammationszeichen und vergrößerte Läsionen. In situ-Hybridisierung und Immunfluoreszenz zeigen eine starke Lactadherin-Expression in den Schaumzellen atherosklerotischer Plaques von Apo E-/-, Apo E-/-/GPx 1-/-und LDLR-/- Mäusen, vor allem in der Nähe des Lipid Core. Dort kolokalisiert Lactadherin mit dem Makrophagenmarker CD 68 und dem Chemokin Fraktalkin, das die MFG-E8 Sekretion stimuliert und so die Phagocytose forciert. Untersuchungen mittels RTD-PCR ergaben, dass Peritonealmakrophagen der Genotypen Apo E-/-, Apo E-/-/GPx 1-/- und GPx 1-/-, deren Gemeinsamkeit eine höhere Empfindlichkeit gegenüberrnoxidativem Stress ist, mehr Lactadherin exprimieren als andere Genotypen (B6, LDLR-/-). Die Inkubation muriner oder humaner Makrophagen mit oxLDL und eLDL hat keinen Einfluss auf die Expression der MFG-E8 mRNA. Der Kontakt mit apoptotischer Zellen hingegen erhöht die Expression signifikant. Lactadherin ist entscheidend für die effektive Phagozytose apoptotischer Zellen in der atherosklerotischen Läsion. Seine Expression wird vermutlich durch die Apoptose in der Nähe liegender Zellen und das verstärkte Vorkommen von ROS reguliert. Macrophage stimulating protein (MSP) übt Einfluss auf Migration, Proliferation und Phagocytose von Makrophagen aus. Seine Beteiligung an inflammatorischen Vorgängen und der Karzinogenese ist intensiv untersucht worden, nicht jedoch der Einfluss auf die Atherosklerose. Es ist bekannt, dass der SNP rs3197999 mit chronisch entzündlichen Darmerkrankungen (CED) assoziiert ist. Zudem geht er vermutlich mit einem erniedrigten Atheroskleroserisiko einher. Der Polymorphismus c2078t hat den Aminosäureaustausch R689C zur Folge. Rekombinant erzeugtes, mutantes und wildtypisches MSP induziert Migration und Proliferation bei THP-1-Makrophagen. MSPmut vermittelt dies jedoch wesentliche effektiver als MSPwt. Apoptose hingegen wird durch keine der Formen induziert. R689C führt zu einem “gain of function” des MSP-Proteins in Bezug auf die Proliferations- und Migrationsfähigkeit von Makrophagen und verändert vermutlich deren Cytokinfreisetzung. Dies führt möglicherweise zu einer erhöhten Phagocytoseeffizienz in der atherosklerotischen Läsion (erniedrigtes Atherosklerose-Risiko), und zu einer aberranten immunologischen Reaktion im Rahmen der CED (erhöhtes CED-Risiko).

We compute the E-polynomial of the character variety of representations of a rank r free group in SL(3,C). Expanding upon techniques of Logares, Muñoz and Newstead (Rev. Mat. Complut. 26:2 (2013), 635-703), we stratify the space of representations and compute the E-polynomial of each geometrically described stratum using fibrations. Consequently, we also determine the E-polynomial of its smooth, singular, and abelian loci and the corresponding Euler characteristic in each case. Along the way, we give a new proof of results of Cavazos and Lawton (Int. J. Math. 25:6 (2014), 1450058).

A moratorium on further bivalve leasing was established in 1999–2000 in Prince Edward Island (Canada). Recently, a marine spatial planning process was initiated explore potential mussel culture expansion in Malpeque Bay. This study focuses on the effects of a projected expansion scenario on productivity of existing leases and available suspended food resources. The aim is to provide a robust scientific assessment using available datasets and three modelling approaches ranging in complexity: (1) a connectivity analysis among culture areas; (2) a scenario analysis of organic seston dynamics based on a simplified biogeochemical model; and (3) a scenario analysis of phytoplankton dynamics based on an ecosystem model. These complementary approaches suggest (1) new leases can affect existing culture both through direct connectivity and through bay-scale effects driven by the overall increase in mussel biomass, and (2) a net reduction of phytoplankton within the bounds of its natural variation in the area.

ANTECEDENTES: La OMS recomienda adoptar un estilo de vida saludable a lo largo de todo el ciclo vital. En estudios previos se obtuvo que 4 de cada 5 médicos tienen una vida poco saludable. OBJETIVO GENERAL: Determinar los estilos de vida y los factores individuales asociados a los médicos que trabajan en ambiente hospitalario frente los que trabajan a nivel comunitario del MSP en la provincia del Azuay en el 2015. METODOLOGÍA DE ESTUDIO: Se realizó un estudio analítico transversal. Con un universo de 630 médicos, mediante el software Epi-Info; y una frecuencia esperada del 16% con peor resultado del 10% y con nivel de confianza del 95%, se obtuvo una muestra estadística de 120 médicos, divididos en un grupo de médicos que trabajan en medio hospitalario y otro grupo perteneciente al medio comunitario. Los datos fueron recolectados directamente y analizados en Excel y SPPS en tablas para determinar asociación y significancia respecto a la prevalencia de buenos estilos de vida. RESULTADOS: La prevalencia de estilos de vida saludable corresponde al 67,5%, seguido del estilo poco saludable con el 32,5%. A nivel comunitario 85% tiene estilo de vida saludable, mientras que al estilo de vida poco saludable pertenecen el 15%. En el ambiente hospitalario el 50% tienen estilo de vida saludable y el otro 50% el estilo poco saludable. CONCLUSIÓN: Los médicos que laboran a nivel comunitario tiene mejor estilo de vida que los médicos que trabajan en el nivel hospitalario ya que el primero obtuvo 85% en el estilo de vida saludable, mientras el segundo solo el 50%
