Tissue destruction characterizes infection with Mycobacterium tuberculosis (Mtb). Type I collagen provides the lung's tensile strength, is extremely resistant to degradation, but is cleaved by matrix metalloproteinase (MMP)-1. Fibroblasts potentially secrete quantitatively more MMP-1 than other lung cells. We investigated mechanisms regulating Mtb-induced collagenolytic activity in fibroblasts in vitro and in patients. Lung fibroblasts were stimulated with conditioned media from Mtb-infected monocytes (CoMTb). CoMTb induced sustained increased MMP-1 (74 versus 16 ng/ml) and decreased tissue inhibitor of metalloproteinase (TIMP)-1 (8.6 versus 22.3 ng/ml) protein secretion. CoMTb induced a 2.7-fold increase in MMP-1 promoter activation and a 2.5-fold reduction in TIMP-1 promoter activation at 24 hours (P = 0.01). Consistent with this, TIMP-1 did not co-localize with fibroblasts in patient granulomas. MMP-1 up-regulation and TIMP-1 down-regulation were p38 (but not extracellular signal–regulated kinase or c-Jun N-terminal kinase) mitogen-activated protein kinase–dependent. STAT3 phosphorylation was detected in fibroblasts in vitro and in tuberculous granulomas.STAT3 inhibition reduced fibroblast MMP-1 secretion by 60% (P = 0.046). Deletion of the MMP-1 promoter NF-B–binding site abrogated promoter induction in response to CoMTb. TNF-, IL-1ß, or Oncostatin M inhibition in CoMTb decreased MMP-1 secretion by 65, 63, and 25%, respectively. This cytokine cocktail activated the same signaling pathways in fibroblasts and induced MMP-1 secretion similar to that induced by CoMTb. This study demonstrates in a cellular model and in patients with tuberculosis that in addition to p38 and NF-B, STAT3 has a key role in driving fibroblast-dependent unopposed MMP-1 production that may be key in tissue destruction in patients.

PURPOSE. Interleukin (IL)-17, which is responsible for the initial influx of leukocytes into the target tissue, was recently described as the main cytokine involved in autoimmune diseases. Vogt-Koyanagi-Harada (VKH) syndrome is a significant cause of noninfectious blindness in the world. Herein the authors aimed at unraveling the involvement of IL-17 in VKH and in experimental autoimmune uveitis, focusing on the signaling pathways involved in IL-17 synthesis. METHODS. Mice were immunized with 161-180 peptide and pertussis toxin. Draining lymph node cells, harvested 21 days after immunization, were cultured in the presence or absence of p38 alpha mitogen-activated protein kinase (MAPK) inhibitor (SB203580) and assayed for cytokine production and quantification of CD4(+)IL-17(+) cells. Mice received intraocular injections of SB203580, and disease severity was evaluated by histologic examination of the enucleated eyes at day 21. CD4(+) lymphocytes from MSK-1/2-deficient mice, human CD4(+) cells silenced with MSK1 siRNA, or peripheral blood mononuclear cells (PBMCs) from VKH patients were cultured in the presence or absence of p38 alpha MAPK inhibitor and then assayed for IL-17, IFN-gamma, and IL-4 production. RESULTS. The inhibition of p38 alpha MAPK fully blocked the synthesis of IL-17 by PBMCs from VKH patients and lymphocytes from EAU mice. The absence of the msk1/2 gene resulted in failure to produce IL-17 by murine and human lymphocytes. Interestingly, intraocular injections of SB203580 in EAU mice did not suppress development of the disease. CONCLUSIONS. These data show that p38 alpha MAPK-MSK1/2 is involved in the control of IL-17 synthesis by CD4(+) T cells and that inhibition of p38 alpha MAPK in vitro suppresses IL-17 synthesis but that inhibition of this kinase in vivo did not protect from EAU. BACKGROUND Angiogenesis and vascular remodelling are crucial events in tissue repair mechanisms promoted by cell transplantation. Current evidence underscores the importance of the soluble factors secreted by stem cells in tissue regeneration. In the present study we investigated the effects of paracrine factors derived from cultured endothelial progenitor cells (EPC) on rat brain endothelial cell properties and addressed the signaling pathways involved. METHODS Endothelial cells derived from rat brain (rBCEC4) were incubated with EPC-derived conditioned medium (EPC-CM). The angiogenic response of rBCEC4 to EPC-CM was assessed as effect on cell number, migration and tubular network formation. In addition, we have compared the outcome of the in vitro experiments with the effects on capillary sprouting from rat aortic rings. The specific PI3K/AKT inhibitor LY294002 and the MEK/ERK inhibitor PD98059 were used to study the involvement of these two signaling pathways in the transduction of the angiogenic effects of EPC-CM. RESULTS Viable cell number, migration and tubule network formation were significantly augmented upon incubation with EPC-CM. Similar findings were observed for aortic ring outgrowth with significantly longer sprouts. The EPC-CM-induced activities were significantly reduced by the blockage of the PI3K/AKT and MEK/ERK signaling pathways. Similarly to the outcome of the rBCEC4 experiments, inhibition of the PI3K/AKT and MEK/ERK pathways significantly interfered with capillary sprouting induced by EPC-CM. CONCLUSION The present study demonstrates that EPC-derived paracrine factors substantially promote the angiogenic response of brain microvascular endothelial cells. In addition, our findings identified the PI3K/AKT and MEK/ERK pathways to play a central role in mediating these effects.

Background: Methotrexate alone or in combination with other agents is the standard treatment for moderate-to-severe rheumatoid arthritis. As the biological agents are expensive, they are not usually used until methotrexate has failed to give a good response. Thus, there is scope for the development of cheaper drugs that can be used instead of methotrexate or in addition to methotrexate. Objectives/methods: Pamapimod is a p38α inhibitor being developed for use in the treatment of rheumatoid arthritis. The objective was to evaluate the recent clinical trials of pamapimod in subjects with rheumatoid arthritis. Results: There is no clear cut evidence that pamapimod alone or in the presence of methotrexate is efficacious in subjects with rheumatoid arthritis, but it does cause adverse effects. Conclusion: It is unlikely that pamapimod will be useful in the treatment of rheumatoid arthritis.

Cellular signalling events are at the core of every adaptive response. Signalling events link environmental changes to physiological responses, consequently allowing cellular and organismal sustenance and survival. Classical approaches to study cellular signalling have relied on a variety of cell disruptive techniques which yield limited kinetic information, while the underlying events are much more complex. In this article, we discuss how modern live cell imaging microscopy has found increasing utilization in revealing spatio temporal dynamics of various signalling pathways. Utilizing the well studied mitogen-activated protein kinase (MAPK) signalling cascade as a template, the design, construction and utilization of `mobile' (translocation proficient) biosensors, suitable for studying MAPK signalling in living cells are described in detail. Experimental setup and results obtained from these biosensors, based on different proteins involved in the MAPK signalling cascade, have been described along with the setup of a microscope optimal for live cell imaging applications. Utilizing the ability to activate or deactivate signalling pathways using defined activators and specific pharmacological inhibitors, we also show how these sensors can yield unique spatial and temporal kinetic information of signalling in living cells.

The current treatment for glioblastoma includes temozolomide (TMZ) chemotherapy, yet the mechanism of action of TMZ is not thoroughly understood. Here, we investigated the TMZ-induced changes in the proteome of the glioma-derived cell line (U251) by 2D DIGE. We found 95 protein spots to be significantly altered in their expression after TMZ treatment. MS identified four upregulated spots: aspartyl tRNA synthetase glutathione synthetase, interleukin-1 receptor-associated kinase-4 (IRAK4), and breast carcinoma amplified sequence-1 and one downregulated spot: optineurin. TMZ-induced regulation of these five genes was validated by RT-qPCR andWestern blot analysis. RNAi-mediated knockdown of IRAK4, an important mediator of Toll-like receptors signaling and chemoresistance, rendered the glioma cells resistant to TMZ. High levels of IRAK4 induced upon TMZ treatment resulted in IRAK1 downregulation and inhibition of NFkB pathway. Endogenous IRAK4 protein, but not transcript levels in glioma cell lines, correlated with TMZ sensitivity. Thus, we have identified several TMZ-modulated proteins and discovered an important novel role for IRAK4 in determining TMZ sensitivity of glioma cells through its ability to inhibit Toll-like receptor signaling and NFkB pathway.

FGF/Erk MAP Kinase Signaling is a central regulator of mouse embryonic stem cell (mESC) self-renewal, pluripotency and differentiation. However, the mechanistic connection between this signaling pathway activity and the gene circuits stabilizing mESCs in vitro remain unclear. Here we show that FGF signaling post-transcriptionally regulates the mESC transcription factor network by controlling the expression of Brf1 (zfp36l1), an AU-rich element mRNA binding protein. Changes in Brf1 level disrupts the expression of core pluripotency-associated genes and attenuates mESC self-renewal without inducing differentiation. These regulatory effects are mediated by rapid and direct destabilization of Brf1 targets, such as Nanog mRNA. Interestingly, enhancing Brf1 expression does not compromise mESC pluripotency, but does preferentially regulate differentiation to mesendoderm by accelerating the expression of primitive streak markers. Together, these studies demonstrate that FGF signals utilize targeted mRNA degradation by Brf1 to enable rapid post-transcriptional control of gene expression.

Mammary epithelial cells in primary cell culture require both growth factors and specific extracellular matrix (ECM)-attachment for survival. Here we demonstrate for the first time that inhibition of the ECM-induced Erk 1/Erk 2 (p42/44 MAPK) pathway, by PD 98059, leads to apoptosis in these cells. Associated with this cell death is a possible compensatory signalling through the p38 MAP kinase pathway the inhibition of which, by SB 203580, leads to a more rapid onset of apoptosis. This provides evidence for a hitherto undescribed Erk 1/Erk 2 to p38 MAP kinase pathway 'cross-talk' that is essential for the survival of these cells. The cell death associated with inhibition of these two MAP kinase pathways however, occurred in the presence of insulin that activates the classical PI-3 kinase-dependent Akt/PKB survival signals and Akt phosphorylation. Cell death induced by inhibition of the MAP kinase pathways did not affect Akt phosphorylation and may, thus, be independent of PI-3 kinase signalling.

The pyruvate dehydrogenase (PDH) complex regulates the oxidation of carbohydrates in mammals. Decreased activation of PDH following exhaustive exercise may aid the resynthesis of glycogen through increased activity of PDH kinase-4 (PDK4), one of four kinases that decrease the activity of the PDH complex. The purpose of this study was to examine the role of PDK4 in post-exercise glycogen resynthesis. Wild-type (WT) and PDK4-knockout (PDK4-KO mice) were exercised to exhaustion and were sampled at rest (Rest), at exercise exhaustion (Exh), and after two-hours post-exercise (Rec). Differences in feeding post-exercise led to the addition of a PDK4-KO group, pair-fed (PF) with WT mice. Glycogen fully recovered in all Rec groups in muscle however remained low in the PF group in liver. Flux through PDH was elevated in PDK4-KO muscle with feeding and low in the PF group in both tissues. This suggests PDK4 may fine-tune flux through PDH during exercise recovery.

Les voies de signalisation des MAP kinases (MAPK) conventionnelles jouent des rôles essentiels pendant le développement des lymphocytes T (LT) ainsi que lors de leur activation suite à la reconnaissance antigénique. En raison de ses différences structurelles ainsi que de son mode de régulation, ERK3 fait partie des MAPK dites non-conventionnelles. Encore aujourd’hui, les événements menant à l’activation de ERK3, ses substrats ou partenaires ainsi que sa fonction physiologique demeurent peu caractérisés. Nous avons entrepris dans cette thèse d’étudier le rôle de ERK3 lors du développement et de l’activation des LT en utilisant un modèle de souris déficient pour l’expression de ERK3. Nous avons premièrement établi que ERK3 est exprimée chez les thymocytes. Ensuite, nous avons évalué le développement thymique chez la souris ERK3-déficiente et nous avons observé une diminution significative de la cellularité aux étapes DN1, DP et SP CD4+ du développement des LT. La création de chimères hématopoïétiques ERK3-déficientes nous a permis de démontrer que la diminution du nombre de cellules observée aux étapes DN1 et DP est autonome aux thymocytes alors que le phénotype observé à l’étape SP CD4+ est dépendant de l’abolition simultanée de ERK3 dans l’épithélium thymique et dans les thymocytes. Une étude plus approfondie de l’étape DP nous a permis de démontrer qu’en absence de ERK3, les cellules DP meurent plus abondamment et accumulent des cassures doubles brins (DSB) dans leur ADN. De plus, nous avons démontré que ces cassures dans l’ADN sont réalisées par les enzymes RAG et qu’en absence de ces dernières, la cellularité thymique est presque rétablie chez la souris ERK3-déficiente. Ces résultats suggèrent que ERK3 est impliquée dans un mécanisme essentiel à la régulation des DSB pendant le réarrangement V(D)J de la chaîne  du récepteur des cellules T (RCT). Dans le deuxième article présenté dans cette thèse, nous avons montré que ERK3 est exprimé chez les LT périphériques, mais seulement suite à leur activation via le RCT. Une fois activés in vitro les LT ERK3-déficients présentent une diminution marquée de leur prolifération et dans la production de cytokines. De plus, les LT ERK3-déficients survivent de façon équivalente aux LT normaux, mais étonnamment, ils expriment des niveaux plus faibles de la molécule anti-apoptotique Bcl-2. Ces résultats suggèrent que la prolifération réduite des LT ERK3-déficients est la conséquence d’une altération majeure de leur activation. Ainsi, nos résultats établissent que ERK3 est une MAPK qui joue des rôles essentiels et uniques dans le développement thymique et dans l’activation des lymphocytes T périphériques. Grâce à ces travaux, nous attribuons pour la toute première fois une fonction in vivo pour ERK3 au cours de deux différentes étapes de la vie d’un LT.

Les MAP kinases sont des enzymes essentielles impliquées dans 7 voies de signalisation distinctes qui permettent à la cellule de répondre de manière adéquate aux stimuli extra-cellulaires. Chez les mammifères, les MAP kinases les mieux caractérisées sont Erk1/2, Jnk, p38 et Erk5. Ces enzymes jouent un rôle important dans l’embryogenèse, la prolifération et la différenciation cellulaire ainsi que dans la réponse au stress. Erk4 est un membre atypique de la famille MAP kinase. D’une part, la boucle d’activation de Erk4 possède un motif SEG au lieu du motif TXY, très conservé chez les MAP kinases. D’autre part, Erk4 possède une extension en C-terminal du domaine kinase qui n’est pas présente chez les MAP kinases classiques. Jusqu’à présent aucune fonction n’a été attribuée à Erk4. De plus, la voie de signalisation ainsi que le mode de régulation conduisant à l’activation de Erk4 ne sont pas connus. Le seul substrat de Erk4 identifié jusqu’à maintenant est la MAPKAP kinase MK5. L’impact fonctionnel de cette interaction n’est également pas connu. Afin d’en apprendre davantage sur la MAP kinase atypique Erk4, nous avons étudié le mécanisme d’activation de cette kinase ainsi que sa fonction physiologique par une approche de délétion génique chez la souris. En ce qui concerne l’activation de Erk4, nous avons montré que la boucle d’activation de Erk4 (S186EG) est constitutivement phosphorylée in vivo et que cette phosphorylation n’est pas modulée par les stimuli classiques des MAP kinases dont le sérum et le sorbitol. Cependant, nous avons observé que la phosphorylation de la S186 augmente en présence de MK5 et que cette augmentation est indépendante de l’activité kinase de l’une ou l’autre de ces kinases. De plus, nous avons établi que la phosphorylation de la boucle d’activation de Erk4 est requise pour l’interaction stable entre Erk4 et MK5 ainsi que pour l’activation, et la relocalisation cytoplasmique de MK5. Ainsi, notre étude a permis de révéler que Erk4 est régulée de manière différente des MAP kinases classiques et que la phosphorylation de la boucle d’activation de Erk4 joue un rôle essentiel dans la régulation de l’activité de MK5. Parallèlement, nos résultats mettent en évidence l’existence d’une “Erk4 kinase”, dont le recrutement et/ou l’activation semble être facilité par MK5. Afin identifier la fonction physiologique de Erk4, nous avons généré des souris Erk4-déficientes. L’inactivation génique de Erk4 est viable et les souris ne présentent aucune anomalie apparente. Dans le but d’expliquer l’absence de phénotype, nous avons regardé si l’expression de Erk3, le paralogue de Erk4, pouvait compenser la perte de Erk4. Notre analyse a révélé que l’expression de Erk3 dans les souris Erk4-/- n’augmente pas au cours du développement embryonnaire ou dans les tissus adultes afin de compenser pour la perte de Erk4. Par la suite, nous avons adressé la question de redondance entre Erk4 et Erk3. Dans notre laboratoire, les souris Erk3-déficientes ont également été générées et le phénotype de ces souris a récemment été analysé. Cette étude a révélé que l’inactivation génique de Erk3 entraîne un retard de croissance intra-utérin, un défaut de maturation pulmonaire et la mort néo-natale des souriceaux. Nous avons donc regardé la contribution de Erk4 dans ces phénotypes. L’analyse des souris Erk4-/- a révélé que l’inactivation de Erk4 n’entraîne pas de retard de croissance ou de maturation du poumon. De plus, nous avons montré que l’inactivation additionnelle de Erk4 dans les souris Erk3-/- n’accentue pas le phénotype des souris Erk3-déficientes. Ainsi, notre étude a révélé que contrairement à Erk3, Erk4 n’est pas essentielle au développement murin dans des conditions physiologiques. Parallèlement, nous avons montré que Erk4 et Erk3 possèdent des fonctions non-redondantes in vivo.

The extracellularly-responsive kinase (ERK) subfamily of mitogen-activated protein kinases (MAPKs) has been implicated in the regulation of cell growth and differentiation. Activation of ERKs involves a two-step protein kinase cascade lying upstream from ERK, in which the Raf family are the MAPK kinase kinases and the MEK1/MEK2 isoforms are the MAPK kinases. The linear sequence of Raf --> MEK --> ERK constitutes the ERK cascade. Although the ERK cascade is activated through growth factor-regulated receptor protein tyrosine kinases, they are also modulated through G protein-coupled receptors (GPCRs). All four G protein subfamilies (Gq/11 Gi/o, Gs and G12/13) influence the activation state of ERKs. In this review, we describe the ERK cascade and characteristics of its activation through GPCRs. We also discuss the identity of the intervening steps that may couple agonist binding at GPCRs to activation of the ERK cascade.

The mitogen-activated protein (MAP) kinase phosphatase (MKP) family plays an important function in regulating the pro-inflammatory cytokines by deactivating MAP kinases. MKP-1 is essential for the dephosphorylation of p38 MAP kinase that regulates expression of IL-6, TNF-alpha, and IL-1 beta. We hypothesized that MKP-1 regulates inflammatory bone loss in experimental periodontitis. Wild-type and Mkp-1(-/-) mice received A. actinomycetemcomitans LPS injection in the palatal region or PBS control 3 times/wk for 30 days. Mice were killed, and maxillae were assessed by microcomputed tomography, histological analysis, and TRAP staining for measurement of bone loss, extent of inflammation, and degree of osteoclastogenesis. Results indicated that, in LPS-injected Mkp-1(-/-) mice, significantly greater bone loss occurred with more inflammatory infiltrate and a significant increase in osteoclastogenesis compared with Mkp-1(-/-) control sites or either wild-type group. Analysis of these data indicates that MKP-1 plays a key role in the regulation of inflammatory bone loss.

Matrix metal loprotease-13 (MMP-13) is induced by pro-inflammatory cytokines and increased expression is associated with a number of pathological conditions such as tumor metastasis, osteoarthritis, rheumatoid arthritis and periodontal diseases. MMP-13 gene regulation and the signal transduction pathways activated in response to bacterial LPS are largely unknown. In these studies, the role of the mitogen-activated protein kinase (MAPK) pathways in the regulation of MMP-13 induced by lipopolysaccharide was investigated. Lipopolysaccharide from Escherichia coli and Actinobacillus actinomycetemcomitans significantly (P < 0.05) increased MMP-13 steady-state mRNA (average of 27% and 46% increase, respectively) in murine periodontal ligament fibroblasts. MMP-13 mRNA induction was significantly reduced by inhibition of p38 MAP kinase. Immunoblot analysis indicated that p38 signaling was required for LPS-induced MMP-13 expression. Lipopolysaccharide induced proximal promoter reporter (-660/+32 mMMP-13) gene activity required p38 signaling. Collectively, these results indicate that lipopolysaccharide-induced murine MMP-13 is regulated by p38 signaling through a transcriptional mechanism.
