Amb l’objectiu d’identificar els possibles efectes adversos d’una nova molècula antagonista del receptor de la histamina H4 (Compost A) en el sistema nerviós central (SNC), es va realitzar el test més àmpliament emprat per aquest fi: el test anomenat FOB (Functional Observational Battery test) en el qual es fa servir la rata com a model animal d’estudi. L’estudi FOB forma part dels d’estudis principals (Core Battery Studies) de la farmacologia de seguretat requerida per les autoritats reguladores per tal d’iniciar els estudis clínics en humans (ICH Topic S7A: Safety Pharmacology Studies for Human Pharmaceuticals; FDA & EMA). D’altra banda i per tal de complementar aquests estudis principals, es va avaluar el potencial efecte convulsivant del Compost A mitjançant el model de convulsions induïdes per pentilentetrazol en rata. Aquest efecte proconvulsionant ha estat associat de manera puntual als fàrmacs antagonistes dels receptor H1 i de manera particular en la població infantil. L’estudi de les convulsions forma part dels Follow Up Studies recomenats per la ICH S7A. El Compost A en cap de les dosis administrades, en cap dels assaigs realitzats, mostra una activitat depressora o estimulant en el sistema nerviós. Es caracteritza per tant, per primera vegada, part del perfil de seguretat a nivell del SNC d’un fàrmac antagonista selectiu del receptor H4 de la histamina.

Aquest treball es proposa analitzar els diferents mètodes existents en el camp assistencial per resoldre problemàtiques ètiques, per després proposar un mètode (nou o existent) de deliberació de problemàtiques ètiques, adreçat a professionals del camp del treball social. En el primer apartat de l’estudi s’exposa la definició del concepte de problema ètic en el camp del treball social. Seguidament es presenta una reflexió sobre la presència de problemes ètics en la professió de treball social. Posteriorment es realitza una justificació de la necessitat d’un mètode de resolució de problemes ètics per a professionals del treball social i d’una nova proposta, per exposar tot seguit els objectius del treball, amb les hipòtesis corresponents. En el segon apartat es realitza un repàs de les condicions necessàries per a la deliberació, entenent aquest concepte com a base per a aplicar qualsevol mètode d’anàlisi i/o resolució de problemes ètics. En el tercer apartat s’exposen i analitzen els mètodes de deliberació ètica més rellevants en medicina i treball social, després de dissenyar i aplicar ítems de valoració dels mateixos. En el quart apartat es proposa un mètode de deliberació sobre problemàtiques ètiques per a professionals del camp del treball social i es planteja una segona fase de l’estudi per implementar el mètode i aplicar-lo a casos pràctics. Per últim es presenten les conclusions del treball, i a continuació la bibliografia

L’objectiu d’aquest estudi, de caire interpretatiu - qualitatiu, consisteix en l’anàlisi de l’Atenció a la Diversitat a les tres escoles rurals de la ZER Tramuntana. Una recerca que pretén descriure, analitzar i interpretar els principals aspectes d’inclusió de l’alumnat en el grup - classe, la resposta educativa que donen els docents a la diversitat en les aules i els documents normatius que recullen aquests dos aspectes. Professors, alumnes i grup - classe són els actors que nosaltres hem estudiat detalladament, a partir d’entrevistes, tests sociomètrics i l’anàlisi dels documents vigents de les tres escoles de referència d’aquesta ZER que tenen present l’Atenció a la Diversitat. A partir de l’estudi dels tres casos reals, l’escola Puig Segalar de Viladamat, l’escola de Ventalló i l’escola Els Valentins de Vilamacolum, ens centrem en la perspectiva curricular i model inclusiu, per poder donar resposta a la nostra hipòtesi plantejada inicialment: El model d’escola rural afavoreix les estratègies d’Atenció a la Diversitat.

The senescence-accelerated SAMP8 mouse model displays features of cognitive decline and Alzheimer's disease. With the purpose of identifying potential epigenetic markers involved in aging and neurodegeneration, here we analyzed the expression of 84 mature miRNAs, the expression of histone-acetylation regulatory genes and the global histone acetylation in the hippocampus of 8-month-old SAMP8 mice, using SAMR1 mice as control. We also examined the modulation of these parameters by 8 weeks of voluntary exercise. Twenty-one miRNAs were differentially expressed between sedentary SAMP8 and SAMR1 mice and seven miRNAs were responsive to exercise in both strains. SAMP8 mice showed alterations in genes involved in protein acetylation homeostasis such as Sirt1 and Hdac6 and modulation of Hdac3 and Hdac5 gene expression by exercise. Global histone H3 acetylation levels were reduced in SAMP8 compared with SAMR1 mice and reached control levels in response to exercise. In sum, data presented here provide new candidate epigenetic markers for aging and neurodegeneration and suggest that exercise training may prevent or delay some epigenetic alterations associated with accelerated aging.

BACKGROUND: Aging is characterized by a low-grade systemic inflammation that contributes to the pathogenesis of neurodegenerative disorders such as Alzheimer's disease (AD). However, little knowledge is currently available on the molecular processes leading to chronic neuroinflammation. In this context, recent studies have described the role of chromatin regulators in inflammation and longevity including the REST corepressor (Rcor)-2 factor, which seems to be involved in an inflammatory suppressive program. METHODS: To assess the impact of Rcor2 in age-related inflammation, gene expression levels were quantified in different tissues and ages of the spontaneous senescence-accelerated P8 mouse (P8) using the SAMR1 mouse (R1) as a control. Specific siRNA transfection in P8 and R1 astrocyte cultures was used to determine Rcor2 involvement in the modulation of neuroinflammation. The effect of lipopolysaccharide (LPS) treatment on Rcor2 levels and neuroinflammation was analyzed both in vivo and in vitro. RESULTS: P8 mice presented a dramatic decrease in Rcor2 gene expression compared with R1 controls in splenocytes, an alteration also observed in the brain cortex, hippocampus and primary astrocytes of these mice. Rcor2 reduction in astrocytes was accompanied by an increased basal expression of the interleukin (Il)-6 gene. Strikingly, intraperitoneal LPS injection in R1 mice downregulated Rcor2 in the hippocampus, with a concomitant upregulation of tumor necrosis factor (Tnf-α), Il1-β and Il6 genes. A negative correlation between Rcor2 and Il6 gene expression was also verified in LPS-treated C6 glioma cells. Knock down of Rcor2 by siRNA transfection (siRcor2) in R1 astrocytes upregulated Il6 gene expression while siRcor2 further increased Il6 expression in P8 astrocytes. Moreover, LPS activation provoked a further downregulation of Rcor2 and an amplified induction of Il6 in siRcor2-tranfected astrocytes. CONCLUSIONS: Data presented here show interplay between Rcor2 downregulation and increased inflammation and suggest that Rcor2 may be a key regulator of inflammaging

The visual system is a potential target for methylmercury (MeHg) intoxication. Nevertheless, there are few studies about the cellular mechanisms of toxicity induced by MeHg in retinal cells. Various reports have indicated a critical role for nitric oxide synthase (NOS) activation in modulating MeHg neurotoxicity in cerebellar and cortical regions. The aim of the present study is to describe the effects of MeHg on cell viability and NOS activation in chick retinal cell cultures. For this purpose, primary cultures were prepared from 7-day-old chick embryos: retinas were aseptically dissected and dissociated and cells were grown at 37ºC for 7-8 days. Cultures were exposed to MeHg (10 µM, 100 µM, and 1 mM) for 2, 4, and 6 h. Cell viability was measured by MTT method and NOS activity by monitoring the conversion of L-[H3]-arginine to L-[H3]-citrulline. The incubation of cultured retina cells with 10 and 100 µM MeHg promoted an increase of NOS activity compared to control (P < 0.05). Maximum values (P < 0.05) were reached after 4 h of MeHg incubation: increases of 81.6 ± 5.3 and 91.3 ± 3.7%, respectively (data are reported as mean ± SEM for 4 replicates). MeHg also promoted a concentration- and time-dependent decrease in cell viability, with the highest toxicity (a reduction of about 80% in cell viability) being observed at the concentration of 1 mM and after 4-6 h of incubation. The present study demonstrates for the first time the modulation of MeHg neurotoxicity in retinal cells by the nitrergic system

Four populations in the Amazon area were selected for a comparative study of mercury-exposed and non-exposed populations: São Luiz do Tapajós, Barreiras, Panacauera, and Pindobal Grande. The highest mercury levels in human hair samples were found in São Luiz do Tapajós and Barreiras, greatly exceeding the limits established by the World Health Organization. Panacauera showed an intermediate level below 9 µg/g. This was the first comparative and simultaneous evaluation of mercury exposure in the Amazon area. Also, thanks to this type of monitoring, we were able to eliminate the uncertainties about the reference dose. On the basis of these data, we can conclude that the mercury levels detected in exposed populations of the Tapajós River basin may be dangerous not only because they are above the World Health Organization limits, but also because the simultaneous mercury detection in non-exposed populations with similar characteristics provided a valid control and revealed lower mercury levels. Our results support the importance of continuous monitoring in both exposed and non-exposed populations.
