O objetivo deste trabalho foi determinar os parâmetros biológicos e a tabela de vida de fertilidade da broca-do-morangueiro (Lobiopa insularis) criada em dieta artificial à base de frutos de morangueiro, em condições de laboratório. A duração e a viabilidade média dos períodos embrionário, larval, pupal e ovo-adulto foram de: 4,1±1,5 dias e 80,6%; 22,2±5,0 dias e 60%; 10,8±2,3 dias e 90%; e 37,1±8,8 dias e 43,5%, respectivamente. Os períodos de pré-oviposição, oviposição e pós-oviposição foram de 96±18,9, 133±27,5 e 77±16,3 dias. A longevidade de machos (271±20,7 dias) foi menor do que a de fêmeas (318±14,9 dias). Há potencial de utilização de dieta artificial à base de morangos para a multiplicação da broca-do-morangueiro em laboratório.

In a previous study, we observed no spatial genetic structure in Mexican populations of the parasitoids Chelonus insularis Cresson (Hymenoptera: Braconidae) and Campoletis sonorensis Cameron (Hymenoptera: Ichneumonidae) by using microsatellite markers In the current study, we Investigated whether for these important parasitoids of the fall armyworm (Lepidoptera: Noctuidae) there is any genetic structure at a larger scale Insects of both species were collected across the American continent and their phylogeography was Investigated using both nuclear and mitochondria] markers Our results suggest an ancient north-south migration of C insularis, whereas no clear pattern] could be determined for C sonorensis. Nonetheless, the resulting topology indicated the existence of a cryptic taxon within this later species. a few Canadian specimens determined as C. sonorensis branch outside a clack composed of the Argentinean Chelonus grioti Blanchard, the Brazilian Chelonus flavicincta Ashmead, and the rest of the C sonorensis individuals The individuals revealing the cryptic taxon were collected from Thichoplusia in (Hubner) (Lepidoptera. Noctuidae) on tomato (Lycopersicon spp) and may represent a biotype that has adapted to the early season phenology of its host. Overall, the loosely defined spatial genetic structure previously shown at a local fine scale also was found at the larger scale, for both species Dispersal of these insects may be partly driven by wind as suggested by genetic similarities between Individuals coming from very distant locations.

Sourgrass is a perennial weed infesting annual and perennial crops in Brazil. Three biotypes (R1, R2, and R3) of sourgrass suspected to be glyphosate-resistant (R) and another one (S) from a natural area without glyphosate application, in Brazil, were tested for resistance to glyphosate based on screening, dose-response, and shikimic acid assays. Both screening and dose-response assays confirmed glyphosate resistance in the three sourgrass biotypes. Dose-response assay indicated a resistance factor of 2.3 for biotype RI and 3.9 for biotypes R2 and R3. The hypothesis of a glyphosate resistance was corroborated on the basis of shikimic acid accumulation, where the S biotype accumulated 3.3, 5.0, and 5.7 times more shikimic acid than biotypes R1, R2, and R3, respectively, 168 h after treatment with 157.50 g ae ha(-1) of glyphosate. There were no differences in contact angle of spray droplets on leaves and spray retention, indicating that differential capture of herbicide by leaves was not responsible for resistance in these biotypes. The results confirmed resistance of sourgrass to glyphosate in Brazil.

Snake venom C-type lectin-like proteins (CLPs) are ubiquitously found in Viperidae snake venoms and differ from the C-type lectins as they display different biological activities but no carbohydrate-binding activity. Previous analysis of the transcriptome obtained from the Bothrops insularis venom gland showed the presence of two clusters homologous to bothrojaracin (BJC) chains a and P. In an effort to identify a new BJC-like molecule, we used an approach associated with proteomic technologies to identify the presence of the expressed protein and then to purify and characterize a new thrombin inhibitor from B. insularis venom. We also constructed homology models of this protein and BJC, which were compared with other C-type lectin-like family members and revealed several conserved features of this intriguing snake venom toxin family. Digitaria insularis é uma espécie perene, que se reproduz por sementes e rizomas, sendo de difícil controle após a primeira floração. Visando definir técnicas para o manejo integrado dessa espécie, o seu crescimento foi avaliado em casa de vegetação, em recipiente plástico contendo 0,003 m³ de solo. Avaliações de altura, área foliar e massa seca foram realizadas em 15 épocas, entre 14 e 112 dias após a emergência (DAE), em intervalos regulares de sete dias. Foi utilizado o delineamento experimental inteiramente casualizado, com quatro repetições. As plantas foram fragmentadas em raiz + rizoma, colmo e folha. Posteriormente, determinou-se a área foliar e a massa seca das diferentes partes após secagem a 70 ºC em estufa com ventilação forçada, até massa constante. Os valores máximos de área foliar e a massa seca foram atingidos aos 98 e 105 DAE, respectivamente. As folhas apresentaram maior participação no acúmulo de massa seca total, seguida por raízes+rizomas, até os 105 DAE. O acúmulo de massa seca de Digitaria insularis foi lento até 45 DAE. A partir dessa época, verificou-se rápido acúmulo de massa seca nas raízes, o que pode ser atribuído à formação dos rizomas. Os valores da taxa de crescimento relativo (TCR) foram decrescentes com o tempo, devido à maior alocação de fotoassimilados para estruturas formadas com o desenvolvimento da planta. Digitaria insularis apresenta crescimento lento até 45 DAE, sendo este rápido a partir dos 45 até os 105 DAE, sugerindo a possibilidade de bom controle cultural dessa espécie por culturas que tenham crescimento inicial rápido, grande área foliar e que cubram rapidamente o solo.

A anatomia da folha, de colmo e do rizoma de Digitaria insularis foi investigada, visando identificar características que possam estar relacionadas à sua tolerância ao glyphosate. Sementes e rizomas de plantas adultas foram coletadas em campo, em área de plantio direto, onde o herbicida glyphosate vem sendo utilizado repetidamente há vários anos. As plantas provenientes dessas sementes e desses rizomas foram cultivadas em vasos com capacidade de 3 L, contendo solo de textura média, em casa de vegetação. Quando as plantas atingiram o estádio fenológico de pré-florescimento, foram coletadas três folhas totalmente expandidas por planta, entre o terceiro e o quinto nó. Simultaneamente, coletaram-se fragmentos dos rizomas e os entrenós recobertos pelas bainhas das folhas amostradas. As amostras foram fixadas em FAA50 e estocadas em etanol, para obtenção de cortes histológicos e montagem das lâminas, conforme metodologia usual. Para obtenção do índice estomático e da densidade estomática nas superfícies da epiderme, amostras das folhas foram diafanizadas, conforme método utilizado usualmente. Verificou-se que plantas provenientes de rizomas apresentavam maior índice estomático e maior número de estômatos por mm², maior espessura na epiderme das faces adaxial e abaxial e maior espessura da lâmina foliar. Foi observada coloração intensa nos rizomas submetidos ao Lugol, indicando presença de grande quantidade de amido, independentemente da origem do material.

Com a dessecação da vegetação espontânea, nem sempre se consegue obter controle total das espécies daninhas, o que leva à perenização e ao aumento da importância daquelas selecionadas. Em área de plantio direto com predominância de rebrotes de Digitaria insularis, foi testado nicosulfuron isolado (60 g ha-1) e em mistura em tanque com atrazine (40 + 1.500 g ha-1), foramsulfuron + iodosulfuron-methyl (45 + 3 g ha-1) e mesotrione em mistura em tanque com atrazine (144 + 1.500 g ha-1) sob delineamento de blocos ao acaso, com quatro repetições. Cada parcela apresentava 6 x 4 m, contendo seis linhas de milho. A aplicação dos herbicidas foi feita com pulverizador costal na pressão constante de 30 lbf pol-2, com gasto de 200L ha-1 de calda. Foram avaliadas a eficácia e a intoxicação dos herbicidas aos 7, 15 e 30 dias após a aplicação na cultura. Na época da colheita, realizouse levantamento da cobertura vegetal reinfestante e do seu grau de dificuldade de colheita mecanizada, além da produtividade da cultura. Pode-se inferir que o melhor resultado para o manejo de rebrotes de D. insularis foi com a utilização de nicosulfuron isolado a 60 g ha-1.

Objetivou-se com estse trabalho avaliar o controle químico de diferentes populações de capim-amargoso (Digitaria insularis) pelo herbicida glyphosate por meio de curva de dose-resposta, além de propor tratamentos alternativos para as populações mais tolerantes. O delineamento experimental foi o de blocos ao acaso, com quatro repetições, em esquema fatorial 5 x 9. As sementes de capim-amargoso foram coletadas em cinco locais: área de produção de grãos da Fazenda de Ensino, Pesquisa e Produção da UNESP, Jaboticabal (SP); área de produção comercial de grãos, localizada nos municípios de Campo Florido-MG e Rio Verde-GO; pomar de laranja, localizado no município de Matão (SP); e área não agrícola sem histórico da aplicação de glyphosate (Jaboticabal-SP). O glyphosate (0D, 1/4D, 1/2D, D, 2D, 4D e 8D, em que D é a dose recomendada de 1,5 kg ha-1 de equivalente ácido) e as suas associações [glyphosate + fluazifop-p-butil (1,5 + 0,25 kg ha-1) e glyphosate (1,5 kg ha-1) com sequencial de diuron + paraquat (0,20 + 0,40 kg ha-1 + 0,2% de surfatante)] foram pulverizados em plantas de sete a oito perfilhos e altura média de 20 cm. As populações de capim-amargoso de Campo Florido e Rio Verde foram consideradas suscetíveis; as de Jaboticabal e Matão, tolerantes; e a da área não agrícola, de sensibilidade intermediária. A associação de glyphosate ao fluazifop ou a sua aplicação com sequencial de diuron + paraquat foram eficazes no controle das populações mais tolerantes de capim-amargoso.

This study aimed to control different populations of Digitaria insularisby glyphosate herbicide, isolated and mixed, besides the combination of methods (chemical and mechanical) to manage resistant adult plants. Three experiments were conducted, one in pots which were maintained under non-controlled conditions and two under field conditions. In the experiment in pots, twelve populations of D. insularis were sprayed with isolated glyphosate (1.44 and 2.16 kg a.e. ha-1) and mixed (1.44 and 2.16 kg a.e. ha-1) with quizalofop-p tefuryl (0.12 kg i.a. ha-1). The treatment of 1.44 kg a.e. ha-1 of glyphosate plus 0.12 kg a.i. ha-1 of quizalofop was sufficient for adequate control (>95%) of all populations. Population 11 (area of grain production in Itumbiara, GO) was considered sensitive to glyphosate. Others populations were moderately sensitive or tolerant to the herbicide. In the field, the plants of D. insularis of one of the experiments were mowed and, in the other, there were not. Eight treatments with herbicides [isolated glyphosate (1.44 and 2.16 kg a.e. ha-1) and mixed (1.44 and 2.16 kg a.e. ha-1) with quizalofop-p-tefuryl at 0.12 kg a.i. ha-1), clethodim at 0.108 kg a.i. ha-1) or nicosulfuron at 0.06 kg a.i. ha-1)] were assessed, in combination with or without sequential application of the standard treatment, sprayed 15 days after the first application. The combination of the mechanic control with the application of glyphosate (2.16 and 1.44 kg a.e. ha-1) plus quizalofop-p-tefuryl (0.12 kg a.i. ha-1) or clethodim (0.108 kg a.i. ha-1), associated to the sequential application, was the most effective strategy for the management of adult plants of resistant D. insularis.

A joint transcriptomic and proteomic approach employing two-dimensional electrophoresis, liquid chromatography and mass spectrometry was carried out to identify peptides and proteins expressed by the venom gland of the snake Bothrops insularis, an endemic species of Queimada Grande Island, Brazil. Four protein families were mainly represented in processed spots, namely metalloproteinase, serine proteinase, phospholipase A(2) and lectin. Other represented families were growth factors, the developmental protein G10, a disintegrin and putative novel bradykinin-potentiating peptides. The enzymes were present in several isoforms. Most of the experimental data agreed with predicted values for isoelectric point and M(r) of proteins found in the transcriptome of the venom gland. The results also support the existence of posttranslational modifications and of proteolytic processing of precursor molecules which could lead to diverse multifunctional proteins. This study provides a preliminary reference map for proteins and peptides present in Bothrops insularis whole venom establishing the basis for comparative studies of other venom proteomes which could help the search for new drugs and the improvement of venom therapeutics. Altogether, our data point to the influence of transcriptional and post-translational events on the final venom composition and stress the need for a multivariate approach to snake venomics studies. Bothrops insularis venom contains a variety of substances presumably responsible for several pharmacological effects. We investigated the biochemical and biological effects of phospholipase A(2) protein isolated from B. insularis venom and the chromatographic profile showed 7 main fractions and the main phospholipase A(2) (PLA(2)) enzymatic activity was detected in fractions IV and V. Fraction IV was submitted to a new chromatographic procedure on ion exchange chromatography, which allowed the elution of 5 main fractions designated as lV-1 to IV-5, from which lV-4 constituted the main fraction. The molecular homogeneity of this fraction was characterized by high-performance liquid chromatography (HPLC) and demonstrated by mass spectrometry (MS), which showed a molecular mass of 13984.20 Da; its N-terminal sequence presented a high amino acid identity (up to 95%) with the PLA(2) of Bothrops jararaca and Bothrops asper. Phospholipase A(2) isolated from B. insularis (Bi PLA(2)) venom (10 mu g/mL) was also studied as to its effect on the renal function of isolated perfused kidneys of Wistar rats (n = 6). Bi PLA(2) increased perfusion pressure (PP), renal vascular resistance (RVR), urinary flow (UF) and glomerular filtration rate (GFR). Sodium (%TNa+) and chloride tubular reabsorption (%TCl-) decreased at 120 min, without alteration in potassium transport. In conclusion, PLA(2) isolated from B. insularis venom promoted renal alterations in the isolated perfused rat kidney. (c) 2007 Elsevier Ltd. All rights reserved.

Octopus insularis, target species in this study, is the dominant benthic octopus of the North and Northeast Brazil. Studies on behavior and ecology of the species have been conducted primarily on oceanic islands, with little information on the continental populations. In this study, two regions of the coast of RN, Rio do Fogo and Pirangi, were chosen for the characterization of the niche by O. insularis populations. The dietary niche, habitat and distribution of O. insularis of oceanic islands and the mainland, were compared. In addition, individual characteristics of feeding behavior in a population at Atol das Rocas was studied, taking into account the size of individuals, the proximity of the dens and characteristics of their "personality". The diet of the Rio do Fogo population was composed mainly of bivalve molluscs (82%), unlike Pirangi population that has a diet consisting mainly of crustaceans Decapoda (68%), similar to that described for the populations of the islands. Consequently, the feeding niches of the island populations were more similar, with greater overlap, but the niche breadth of the continent was larger. The habitats of occurrence on the coast includes reefs, rocks, gravel and an environment called Restinga, a plateau composed of biogenic gravel, sand, sponges and algae, showed a high density of animals. Similarly to that found in the islands, O. insularis in the continent, had a clumped distribution, and a bathymetric segregation between small and large individuals. The differences in diet composition among populations were explained by differences in habitat and coverage of the substrate, which may be directly influencing the diversity and prey availability in these environments. The individual analyzes of the population at Atol das Rocas, showed no relationship between the degree of individual specialization and the different personalities, or the distance between dens. The results suggest that the foraging strategy with greater availability of prey in the environment has an influence on diet octopuses over preferences or personalities

In northeastern Brazil, Octopus insularis is the most commercially important cephalopod species and its capture has been performed for several years by the lobster fishermen in the region. In order to obtain information about the reproductive biology, 1108 specimens were collected between November 2009 and September 2011 in the landings and fish markets of Rio do Fogo (RN). For each specimen the mantle length (CM) and total fresh weight (PT) were recorded. Gonads of 264 males and 295 females were examined macroscopically and histologically to assess sexual maturation and determine reproductive indices. Four reproductive stages were determined for males (immature, maturing, mature and post-mature) and females (immature, early maturing, late maturing and mature). The average of eggs recorded in the female s gonads was 93.820 and 39 was the average of spermatophores found in male Needham s complex. Spermathecae with sperm were found in females with 69 mm CM (immature). Males and females become sexually mature at 64.41 and 98.50 mm of CM, respectively. The weight at sexual maturity was 270 g for males and 630 g for females. The values of the size and weight at sexual maturity found in this study show that males mature at smaller sizes than females. For both sexes the maturation peaks occurred in February and November 2010 and also in September 2011. The periods of maximum reproductive activity lasted about 3 months and it seems to occur every 7 10 months. Only one spent female (stage V) was found and the number of mature females was low. Presumably, mature females migrate to deep waters with complex habitat to protect the offspring, indicating that fishery by snorkeling with maximum depth of 15 meters is not reaches this part of the stock. Finally, it is noticeable the importance of the establishment of management strategies for the exploitation of O. insularis different of the ones used for O. vulgaris, once the species have distinct biological features
