This thesis reports the application of metabolomics to human tissues and biofluids (blood plasma and urine) to unveil the metabolic signature of primary lung cancer. In Chapter 1, a brief introduction on lung cancer epidemiology and pathogenesis, together with a review of the main metabolic dysregulations known to be associated with cancer, is presented. The metabolomics approach is also described, addressing the analytical and statistical methods employed, as well as the current state of the art on its application to clinical lung cancer studies. Chapter 2 provides the experimental details of this work, in regard to the subjects enrolled, sample collection and analysis, and data processing. In Chapter 3, the metabolic characterization of intact lung tissues (from 56 patients) by proton High Resolution Magic Angle Spinning (HRMAS) Nuclear Magnetic Resonance (NMR) spectroscopy is described. After careful assessment of acquisition conditions and thorough spectral assignment (over 50 metabolites identified), the metabolic profiles of tumour and adjacent control tissues were compared through multivariate analysis. The two tissue classes could be discriminated with 97% accuracy, with 13 metabolites significantly accounting for this discrimination: glucose and acetate (depleted in tumours), together with lactate, alanine, glutamate, GSH, taurine, creatine, phosphocholine, glycerophosphocholine, phosphoethanolamine, uracil nucleotides and peptides (increased in tumours). Some of these variations corroborated typical features of cancer metabolism (e.g., upregulated glycolysis and glutaminolysis), while others suggested less known pathways (e.g., antioxidant protection, protein degradation) to play important roles. Another major and novel finding described in this chapter was the dependence of this metabolic signature on tumour histological subtype. While main alterations in adenocarcinomas (AdC) related to phospholipid and protein metabolisms, squamous cell carcinomas (SqCC) were found to have stronger glycolytic and glutaminolytic profiles, making it possible to build a valid classification model to discriminate these two subtypes. Chapter 4 reports the NMR metabolomic study of blood plasma from over 100 patients and near 100 healthy controls, the multivariate model built having afforded a classification rate of 87%. The two groups were found to differ significantly in the levels of lactate, pyruvate, acetoacetate, LDL+VLDL lipoproteins and glycoproteins (increased in patients), together with glutamine, histidine, valine, methanol, HDL lipoproteins and two unassigned compounds (decreased in patients). Interestingly, these variations were detected from initial disease stages and the magnitude of some of them depended on the histological type, although not allowing AdC vs. SqCC discrimination. Moreover, it is shown in this chapter that age mismatch between control and cancer groups could not be ruled out as a possible confounding factor, and exploratory external validation afforded a classification rate of 85%. The NMR profiling of urine from lung cancer patients and healthy controls is presented in Chapter 5. Compared to plasma, the classification model built with urinary profiles resulted in a superior classification rate (97%). After careful assessment of possible bias from gender, age and smoking habits, a set of 19 metabolites was proposed to be cancer-related (out of which 3 were unknowns and 6 were partially identified as N-acetylated metabolites). As for plasma, these variations were detected regardless of disease stage and showed some dependency on histological subtype, the AdC vs. SqCC model built showing modest predictive power. In addition, preliminary external validation of the urine-based classification model afforded 100% sensitivity and 90% specificity, which are exciting results in terms of potential for future clinical application. Chapter 6 describes the analysis of urine from a subset of patients by a different profiling technique, namely, Ultra-Performance Liquid Chromatography coupled to Mass Spectrometry (UPLC-MS). Although the identification of discriminant metabolites was very limited, multivariate models showed high classification rate and predictive power, thus reinforcing the value of urine in the context of lung cancer diagnosis. Finally, the main conclusions of this thesis are presented in Chapter 7, highlighting the potential of integrated metabolomics of tissues and biofluids to improve current understanding of lung cancer altered metabolism and to reveal new marker profiles with diagnostic value.

The yerba mate (Ilex paraguariensis) is composed of bioactive that interfere with lipid metabolism. The objective was to evaluate if daily consumption of mate tea (MT) change the lipid deposits and dyslipidemia caused by excessive consumption of sucrose. Thirty male Wistar rats (40 days old) were divided into four groups: Group C - free access to commercial chow and deionized water; S - free access to commercial food, water and sucrose solution 30% (w/v) in water; MTS and deionized - free access to commercial feed solution, water, 30% sucrose (w/v) and treated with daily infusion of MT (soluble mate Leão Júnior®) via orogastric tube at a dose of 100 mg/kg/mc for 8 weeks. After the trial period the lipid profile was evaluated by the following parameters: a) direct weighing of the retroperitoneal (RP) and epididymal (EPI) adipose tissues; b) determination of plasma total cholesterol, triglycerides and HDL cholesterol concentration. The MT promoted the reduction of 1.4 times in both tissues RP and EPI in MTS group compared to group S. Treatment with MT decreased 2.7 times triglyceride in the MTS group compared with the group S. The sucrose consumption did not alter the plasma cholesterol concentration, however the consumption of MT significantly reduced total cholesterol circulating. The HDL cholesterol concentration, in the MTS group, showed higher concentration than in group S (1.3 times). MT prevents in young male rats the increase of lipid deposits and dyslipidemia caused by excessive consumption of sucrose.

Dislipidemia são alterações nas concentrações dos lipídios no organismo, e são divididos em hipo e hiperlipidemias. As hiperlipidemias possuem maior importância clínica, frequentemente associadas com um aumento no risco em desenvolver outras doenças, como doenças coronarianas, pancreatites, etc. Altas concentrações dos lipídios plasmáticos, principalmente colesterol total, estão associados com doenças coronarianas. Altos níveis de triglicerídios e baixos níveis de HDL-colesterol, também são fatores que predispõem a doenças cardiovasculares. Os níveis de triglicerídios podem ter várias causas: como a dieta ou alterações relacionadas ao metabolismo lipídico. Este estudo foi composto por 96 indivíduos descendentes de japoneses residindo no Sul do Brasil (Cascavel-PR). O objetivo deste trabalho foi analisar alterações nos lipídios plasmáticos em uma população específica – indivíduos descendentes de japoneses residentes na região de Cascavel (PR) . Foram determinados os níveis de triglicerídios, colesterol total, HDL-colesterol, LDL-colesterol, VLDL-colesterol, apolipoproteína A-I e B e o padrão eletroforético de lipoproteínas em 96 indivíduos japoneses entre 10 a 89 anos. Para os níveis de triglicerídios, 18,7% dos indivíduos descendentes de japoneses obtiveram valores acima de 200 mg/dL.. Quando relacionamos os níveis de colesterol total, 69,7% dos indivíduos descendentes de japoneses possuem níveis acima da normalidade. Observamos dois padrões eletroforéticos de lipoproteínas na população japonesa; 10,4% desses indivíduos obtiveram alterações na banda pré-beta relacionada ao VLDL-colesterol e 8,3% obtiveram alterações nas bandas da origem (Quilomicra) e na banda pré-beta.

Despite the observation of an increase in life expectancy in individuals with Spinal cord injury (SCI), it is lower than that of the general population. Studies have shown that affected individuals have a sedentary lifestyle that reflects negatively on health and quality of life. Studies have demonstrated that HDL cholesterol (HDL-C) levels, a high-density lipoprotein and important predictor of cardiovascular disease, are lower in this population exposing these people to a greater incidence of heart disease from atherosclerotic process In the general population, exercise increases HDL-C serum levels, but this phenomenon is not very clear in people with spinal cord injury (SCI). The present study examined the effect of both swimming and wheelchair basketball in the lipid profile of eleven men and seven women with SCI. The subjects included in regular exercise programs showed increases in HDL-C levels and decreases in CT/HDL-C and LDL-C/HDL-C ratios. We found better results mainly in men with lower levels of SCI and in those that sustained exercise intensities above 60% of the heart rate reserve. The duration of training sessions can be an essential factor in these results. The results suggest that both the exercise prescription and the personal characteristics of people with SCI influence changes in the lipid profile mediated through exercise. The elaboration of this work is an attempt to clarify uncertainties about health and the longevity of people with SCI generated in discussion of all members of the interdisciplinary rehabilitation team, especially the physiotherapists, nutritionists, nurses and physicians that contributed considerably in all phases of the research

The lipid profile is a group of lab tests that include triglycerides, total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C). However, serum non-HDL-C, Apo A-I and Apo B levels, as well as the lipids ratios (TC/HDL-C, LDL-C/HDL-C and Apo B/Apo A-I), have been described as better predictors of cardiovascular diseases. Reference intervals are tools often used to help the evaluation of the people s health state. These days, Brazilian studies still use the reference intervals of lipids and lipoproteins from other countries, ignoring differences between the populations. Therefore, this study aimed to establish reference intervals for lipids, lipoproteins and apolipoproteins in adults of Rio Grande do Norte/Brazil. Healthy individuals (96 men and 283 women) between 18 and 59years old formed the reference sample group. The samples were collected after fasting 12 to 14 hours. Information on lifestyle and dietary habits of the participants were obtained through questionnaire. The serum glucose level and renal and liver activity were evaluated by laboratory testing. The results of lipid profile were analyzed according to sex, age and mesoregion of Rio Grande do Norte, with significance level of 5% (p < 0,05). The lower and upper reference limits were identified by the 2.5 percentile and 97.5 percentile, respectively, and assurance intervals of 90% was calculated for each of these limits. Among the determinants of lipid profile analyzed, only a few significant differences were observed according to sex, but in terms of age, the groups of smaller and older ages were most likely different. When evaluated by region, the means of West region shown the most significant variations. Not many studies were useful to compare the reference intervals determined in this study. Thus, it becomes necessary to carry out similar studies in other regions of Brazil and of the world given the clinical importance of reference intervals

AIMS An independent, powerful coronary heart disease (CHD) predictor is a low level of high-density lipoprotein cholesterol (HDL-C). Discoidal preβ-HDL particles and large HDL2 particles are the primary cholesterol acceptors in reverse cholesterol transport, a key anti-atherogenic HDL mechanism. The quality of HDL subspecies may provide better markers of HDL functionality than does HDL-C alone. We aimed I) to study whether alterations in the HDL subspecies profile exist in low-HDL-C subjects II) to explore the relationship of any changes in HDL subspecies profile in relation to atherosclerosis and metabolic syndrome; III) to elucidate the impact of genetics and acquired obesity on HDL subspecies distribution. SUBJECTS The study consisted of 3 cohorts: A) Finnish families with low HDL-C and premature CHD (Study I: 67 subjects with familial low HDL-C and 64 controls; Study II: 83 subjects with familial low HDL-C, 65 family members with normal HDL-C, and 133 controls); B) a cohort of 113 low- and 133 high-HDL-C subjects from the Health 2000 Health Examination Survey carried out in Finland (Study III); and C) a Finnish cohort of healthy young adult twins (52 monozygotic and 89 dizygotic pairs) (Study IV). RESULTS AND CONCLUSIONS The subjects with familial low HDL-C had a lower preβ-HDL concentration than did controls, and the low-HDL-C subjects displayed a dramatic reduction (50-70%) in the proportion of large HDL2b particles. The subjects with familial low HDL-C had increased carotid atherosclerosis measured as intima-media-thickness (IMT), and HDL2b particles correlated negatively with IMT. The reduction in both key cholesterol acceptors, preβ-HDL and HDL2 particles, supports the concept of impaired reverse cholesterol transport contributing to the higher CHD risk in low-HDL-C subjects. The family members with normal HDL-C and the young adult twins with acquired obesity showed a reduction in large HDL2 particles and an increase in small HDL3 particles, which may be the first changes leading to the lowering of HDL-C. The low-HDL-C subjects had a higher serum apolipoprotein E (apoE) concentration, which correlated positively with the metabolic syndrome components (waist circumference, TG, and glucose), highlighting the need for a better understanding of apoE metabolism in human atherosclerosis. In the twin study, the increase in small HDL3b particles was associated with obesity independent of genetic effects. The heritability estimate, of 73% for HDL-C and 46 to 63% for HDL subspecies, however, demonstrated a strong genetic influence. These results suggest that the relationship between obesity and lipoproteins depends on different elements in each subject. Finally, instead of merely elevating HDL-C, large HDL2 particles and discoidal preβ-HDL particles may provide beneficial targets for HDL-targeted therapy.

[es]En sus habitas naturales, los microorganismos están en un estado constante de adaptación a cambios tanto bióticos como abióticos. Ante situaciones de estré s, como por ejemplo cambios en nutriente s, temperatura o de osmolar idad , la s estrategias de supervivencia o adapta ción se puede n manifestar como cambios fenotípicos y genotípicos . En este estudio se analizaron algunos mecanismos de cambio asociados a la supervivencia y la composición proteica de membrana en Escherichia coli (bact eria mesófila), al ser expuesta a condiciones de ayuno y a temperaturas subó ptimas (4 y 20ºC). Al realizar un análisis comparativ o del subproteoma de membrana entre estas dos temperaturas, se observó que ante la ausencia de nutrientes, E. coli respondía de forma diferen te en la expresió n de proteí nas as ociadas a estructura (lipoproteínas), conservación de la energía y transporte, con un aumento en el nú mero de proteí nas expresadas a 20 o C. Se observó, además, una importante diferencia en la supervivencia a estas dos temperaturas, donde el número de células en el estado viable no cultivable (VNC) representaron un porcentaje importante a 20ºC

Gordon E. Moore, co-fundador de Intel, predijo en una publicación del año 1965 que aproximadamente cada dos años se duplicaría el número de transistores presentes en un circuito integrado, debido a las cada vez mejores tecnologías presentes en el proceso de elaboración. A esta ley se la conoce como Ley de Moore y su cumplimiento se ha podido constatar hasta hoy en día. Gracias a ello, con el paso del tiempo cada vez se presentan en el mercado circuitos integrados más potentes, con mayores prestaciones para realizar tareas cada vez más complejas. Un tipo de circuitos integrados que han podido evolucionar de forma importante por dicho motivo, son los dispositivos de lógica programable, circuitos integrados que permiten implementar sobre ellos las funciones lógicas que desee implementar el usuario. Hasta hace no muchos años, dichos dispositivos eran capaces de implementar circuitos compuestos por unas pocas funciones lógicas, pero gracias al proceso de miniaturización predicho por la Ley de Moore, hoy en día son capaces de implementar circuitos tan complejos como puede ser un microprocesador; dichos dispositivos reciben el nombre de FPGA, siglas de Field Programmable Gate Array. Debido a la mayor capacidad y por lo tanto a diseños más complejos implementados sobre las FPGA, en los últimos años han aparecido herramientas cuyo objetivo es hacer más fácil el proceso de ingeniería dentro de un desarrollo en este tipo de dispositivos, como es la herramienta HDL Coder de la compañía MathWorks, creadores también Matlab y Simulink, unas potentes herramientas usadas ampliamente en diferentes ramas de la ingeniería. El presente proyecto tiene como objetivo evaluar el uso de dicha herramienta para el procesado digital de señales, usando para ello una FPGA Cyclone II de la casa Altera. Para ello, se empezará analizando la herramienta escogida comparándola con herramientas de la misma índole, para a continuación seleccionar una aplicación de procesado digital de señal a implementar. Tras diseñar e implementar la aplicación escogida, se deberá simular en PC para finalmente integrarla en la placa de evaluación seleccionada y comprobar su correcto funcionamiento. Tras analizar los resultados de la aplicación de implementada, concretamente un analizador de la frecuencia fundamental de una señal de audio, se ha comprobado que la herramienta HDL Coder, es adecuada para este tipo de desarrollos, facilitando enormemente los procesos tanto de implementación como de validación gracias al mayor nivel de abstracción que aporta.

Durante a temporada de nidação, fêmeas de tartarugas marinhas costumam reduzir ou cessar por completo a ingestão de alimentos. Este fato sugere que o armazenamento de energia e nutrientes para a reprodução ocorra durante o período que antecede a migração para os sítios reprodutivos, enquanto estes animais ainda se encontram nas áreas de alimentação. Do ponto de vista fisiológico, tartarugas em atividade reprodutiva são capazes de permanecer longos períodos em jejum. Fatores neuroendócrinos vêm sendo recentemente apontados como os mais relevantes para a manutenção da homeostase energética de todos os vertebrados; entre eles, a leptina (hormônio anorexígeno) e a grelina (peptídeo orexígeno). Com o objetivo de compreender o mecanismo de fome e saciedade nas tartarugas marinhas, investigamos os níveis séricos destes hormônios e de outros indicadores nutricionais em fêmeas de Eretmochelys imbricata desovando no litoral do Rio Grande do Norte, Brasil. Foram coletadas amostras de sangue de 41 tartarugas durante as temporadas reprodutivas de 2010/2011 e 2011/2012. Os níveis séricos de leptina diminuíram significativamente ao longo do período de nidação, de modo a explicar a busca por alimentos ao término da temporada. Ao mesmo tempo, registramos uma tendência crescente nos níveis séricos de grelina, fator este que também justifica a remigração para as áreas de alimentação no fim do período. Não foram observadas tendências lineares para alguns dos parâmetros avaliados, entre eles: hematócrito, alanina aminotransferase (ALT), aspartato aminotransferase (AST), fosfatase alcalina (FA), gama glutamil transferase (GGT), lipoproteínas de baixa densidade (LDL) e lipoproteínas de alta densidade (HDL). É possível que a maior parte dos indicadores nutricionais tenha apresentado redução gradativa devido ao estresse fisiológico decorrente da vitelogênese e de repetidas oviposições. No entanto, é valido ressaltar que o quadro de restrição calórica por tempo prolongado é o principal responsável pelas alterações em índice de massa corpórea e padrões bioquímicos nestes animais.

Segundo O`Kane et al, em 2008, foi demonstrado que 88,9% dos erros laboratoriais são realizados na fase pré-analítica. No laboratório de análises clínicas, o sistema de controle da qualidade pode ser definido como toda a ação sistemática necessária para dar confiança e segurança em todos os exames e prevenir a ocorrência de erros. O tempo de armazenamento pode variar de dias a meses ou mesmo anos, influenciando na definição da temperatura de estocagem. O armazenamento de longo prazo pode resultar na criopreservação inadequada para determinados analitos e pode desnaturar as lipoproteínas. Objetivos: Este trabalho teve como objetivo avaliar a fase pré-analítica, controle da qualidade interno e também avaliar o efeito do congelamento (- 80C) quanto ao tempo de armazenamento do soro e plasma de sangue colhido com ácido etilenodiamino tetra-acético (EDTA). As dosagens foram realizadas no Laboratório de Lípides LabLip, e estocadas em - 80 C por três anos, as mesmas foram redosadas no Serviço de Patologia Clínica da Policlínica Piquet Carneiro da UERJ com metodologias iguais e realizada a comparabilidade dos resultados. Foram analisados o perfil lipídico (CT, HDLc e TG) e PCR-US de 103 amostras, 73 no soro e 30 no plasma em dois laboratórios altamente qualificados. Discussão: Após redosagem foram encontrados nas dosagens de HDLc e CT resultados diminuídos respectivamente (correlação coeficiente no soro 0,48 e 0,62) teste t pareado no soro (CT p 0,0012 e HDLc p 0,0001). Conclusões: Os dados obtidos nas avaliações dos resultados de diferentes laboratórios e tempo de estocagem revelaram que as amostras quando armazenadas por um longo período após redosagem no soro, apresentaram diferenças em certos analitos, tais como CT e HDLc, no qual obteve-se resultados significativamente diminuídos, diferentemente no plasma, que após três anos de estocagem a -80C foram redosados e aplicados no teste t pareado, os analitos CT e PCR-US mantiveram a estabilidade.

This paper proposes a novel and innovative scheme for 10Gb/s parallel Very Short Reach (VSR) optical communication system. The optimized scheme properly manages the SDH/SONET redundant bytes and adjusts the position of error detecting bytes and error correction bytes. Compared with the OIF-VSR4-01.0 proposal, the scheme has a coding process module. The SDH/SONET frames in transmission direction are disposed as follows: (1) The Framer-Serdes Interface (FSI) gets 16x622.08Mb/s STM-64 frame. (2) The STM-64 frame is byte-wise stripped across 12 channels, all channels are data channels. During this process, the parity bytes and CRC bytes are generated in the similar way as OIF-VSR4-01.0 and stored in the code process module. (3) The code process module will regularly convey the additional parity bytes and CRC bytes to all 12 data channels. (4) After the 8B/10B coding, the 12 channels is transmitted to the parallel VCSEL array. The receive process approximately in reverse order of transmission process. By applying this scheme to 10Gb/s VSR system, the frame size in VSR system is reduced from 15552x12 bytes to 14040x12 bytes, the system redundancy is reduced obviously.

本文主要阐述了CSR数字高频分析系统的同步采样器的研究、设计和实现,该分析系统是基于软件无线电技术构建的。 本文的创新点主要在于提出了一种很好的正交同步欠采样方法,这种基于软件无线电原理的I、Q两路正交同步的采样方法,主要利用了平方律部件、一阶环路滤波器和NCO来对I、Q两路采样触发脉冲上升沿的时间差进行闭环控制,从而实现对高频信号的数据采集和处理。这种方法降低了对AD芯片采样速率要求,同时也为后续的基带信号处理提供了方便。 在整个同步采样系统的实现过程中我采用了功能强大的Matlab7.0作为通信算法的仿真平台,选用了AD6645、StratixIIEP2S60和DSP6416作为主要的硬件平台,并在SynaptiCAD和ModelSim中做出了FPGA的HDL程序设计及波形实现
