Several studies use only male rats in their experiments, even with all the evidence of differences between the sexes, not only in the reproductive behavior but also in the development of diseases. This absence of studies using female rats is due in part to the estrous cycle. It is known that one estrous cycle lasts four to 5 days and consists of four phases: proestrus, estrus, metestrus and diestrus. Ramos et al. (1999) identified and mapped a QTL in an F2 population derived from interbreeding of strains of SHR (spontaneously hypertensive rats) and Lewis (LEW) rats and found, on chromosome 4, the first QTL of the world related to anxiety in rats. After several backcrosses between SHR and LEW strains, it was developed a congenic strain called SLA16, to refine the study of this QTL. This QTL corresponds to a large genomic region, approximately 75 million basepairs (Mb), and was first called Ofil1 (inner open field locomotion 1). Currently it is called Anxrr16 (anxiety-related response region 16) according to the rat genome database (RGD). Izidio et al. (2011) suggested that the natural fluctuation of the hormones in the estrous cycle of female rats could influence the effects of this locus on behavior. That is, females in DP (diestrus-proestrus) present the QTL in the same genomic region that males, while females in EM (estrus-metestrus) do not present it. The effects of the estrous cycle in the SLA16 strain and its isogenic control SHR were analyzed on the elevated plus maze (EPM), black and white box and on the open field test (OF). In the open field test, the SLA16 strain had higher central locomotion (F=5.0, p <0.05) and peripheral locomotion (F=12.6, p <0.01) compared to the SHR strain. In the elevated plus maze test, the SLA16 strain had the higher number of entries (F=9.5, p <0.01) and time spent in the open arms (F=10.3, p <0.01) compared to the SHR strain. Behavioral adjustments may occur fast and with less cost than the physiological adaptations. Considering the social behavior is suggestive that the frequency and the intensity of aggressive interactions, the total social cohesion and the extent of vicious attitudes may be used to evaluate welfare. This research presents an analysis of the interactions between the experimental factors such as temperature, genetic and time of the day in the behavior of female broiler breeders under controlled environment in a climatic chamber in order to enhance the different reaction of the birds facing distinct environmental conditions. The results showed significant differences between the behaviors expressed by the studied genetics presenting the need of monitoring them in real-time in order to predict their welfare in commercial housing, due to the complexity of the environmental variables that interfere in the well being process. The research also concluded that the welfare evaluation of female broiler breeders needs to consider the time of the day during the observation of the behaviors.

A wild strain of Streptococcus thermophilus isolated from pasteurized milk was evaluated using an experimental model with respect to its adhesion onto stainless steel surfaces and its behaviour when submitted to cleansing and sanification. In milk, the adhesion of the microorganism on to stainless steel surfaces was studied after 6 hours of contact at 45°C with agitation, and after a cleansing process involving cleaning stages with alkaline and acid detergents followed by sanification, in order to evaluate the resistance of the adhered cells. The microorganism adhered to stainless steel surfaces producing a cell load of 10(4) CFU/cm². After alkaline cleansing, no adhered cells were detected but 6 CFU/cm² were still detected on the surfaces after acid cleansing. Cleansing, followed by sanification with sodium hypochlorite, was sufficient to reduce the load of wild S. thermophilus on the stainless steel surfaces to non-detectable levels. The experimental model proved adequate for the study indicating that the wild microorganism S. thermophilus produces biofilms on stainless steel surfaces. Alkaline cleansing remove more that 99.9% of the adhered cells. The few cells adhered on the surface are removed by acid cleansing demonstrating the need to use different steps and types of detergent for efficient cleansing. The best results for the removal of these biofilms are obtained by using alkaline cleansing followed by acid cleaning, this procedure being more efficient when complemented by sanification with sodium hypochlorite.

There is accumulating evidence that physical inactivity, associated with the modern sedentary lifestyle, is a major determinant of hypertension. It represents the most important modifiable risk factor for cardiovascular diseases, which are the leading cause of morbidity and mortality for both men and women. In addition to involving sympathetic overactivity that alters hemodynamic parameters, hypertension is accompanied by several abnormalities in the skeletal muscle circulation including vessel rarefaction and increased arteriole wall-to-lumen ratio, which contribute to increased total peripheral resistance. Low-intensity aerobic training is a promising tool for the prevention, treatment and control of high blood pressure, but its efficacy may differ between men and women and between male and female animals. This review focuses on peripheral training-induced adaptations that contribute to a blood pressure-lowering effect, with special attention to differential responses in male and female spontaneously hypertensive rats (SHR). Heart, diaphragm and skeletal muscle arterioles (but not kidney arterioles) undergo eutrophic outward remodeling in trained male SHR, which contributed to a reduction of peripheral resistance and to a pressure fall. In contrast, trained female SHR showed no change in arteriole wall-to-lumen ratio and no pressure fall. On the other hand, training-induced adaptive changes in capillaries and venules (increased density) were similar in male and female SHR, supporting a similar hyperemic response to exercise.

As origens e distribuições das artérias que vascularizaram os lobos torácicos do timo foram estudadas em fetos de 30 suínos da linhagem C40, sendo 12 machos e 18 fêmeas. Os exemplares tiveram o sistema arterial preenchido com solução aquosa a 50% de Neoprene Látex corado e, em seguida, foram submetidos à fixação em solução aquosa a 10% de formaldeído. Os lobos torácicos do timo foram vascularizados por ramos diretos das artérias torácica interna direita (63,33%) e esquerda (53,33%), subclávia esquerda (3,33%), vertebral esquerda (3,33%), cervical superficial direita (3,33%) e esquerda (3,33%), carótida comum esquerda (3,33%), coronária direita (3,33%) e pelos troncos braquiocefálico (33,33%) e costocervical (3,33%). Observaram-se ainda os ramos indiretos das artérias torácica interna direita (70%) e esquerda (76,67%), subclávia esquerda (23,33%), cervical superficial esquerda (3,33%) e do arco aórtico (6,67%).

A vascularização arterial do timo (número, origem e ordenação) bem como a distribuição parenquimal dos ramos penetrantes foram estudadas em 40 aves da linhagem Paraíso Pedrês. Trinta aves foram injetadas com látex e dissecadas, enquanto 10 aves tiveram seus sistemas arteriais injetados com resina (metil metacrilato e mercox) para a preparação de moldes vasculares. A principal fonte de irrigação encontrada foi a associação dos ramos oriundos das artérias comuns do nervo vago, tireóideas e ingluviais, sendo que seus ramos penetravam o parênquima dos lobos, principalmente pelas suas extremidades cranial e caudal. A partir da penetração, os ramos tímicos apresentavam distribuição predominante para a periferia do lobo, formando uma trama capilar poligonal, com espaços irregulares, característica de um órgão linforreticular.

Congenic strains provide evidence that four mapped loci in chromosomes 2, 4, and 16 influence hypertension in the SHR. Physiol Genomics 37: 52-57, 2009. First published January 6, 2009; doi: 10.1152/physiolgenomics.90299.2008. - To dissect the genetic architecture controlling blood pressure (BP) regulation in the spontaneously hypertensive rat (SHR) we derived congenic rat strains for four previously mapped BP quantitative trait loci (QTLs) in chromosomes 2, 4, and 16. Target chromosomal regions from the Brown Norway rat (BN) averaging 13 - 29 cM were introgressed by marker-assisted breeding onto the SHR genome in 12 or 13 generations. Under normal salt intake, QTLs on chromosomes 2a, 2c, and 4 were associated with significant changes in systolic BP (13, 20, and 15 mmHg, respectively), whereas the QTL on chromosome 16 had no measurable effect. On high salt intake (1% NaCl in drinking water for 2 wk), the chromosome 16 QTL had a marked impact on SBP, as did the QTLs on chromosome 2a and 2c (18, 17, and 19 mmHg, respectively), but not the QTL on chromosome 4. Thus these four QTLs affected BP phenotypes differently: 1) in the presence of high salt intake (chromosome 16), 2) only associated with normal salt intake (chromosome 4), and 3) regardless of salt intake (chromosome 2c and 2a). Moreover, salt sensitivity was abrogated in congenics SHR. BN2a and SHR. BN16. Finally, we provide evidence for the influence of genetic background on the expression of the mapped QTLs individually or as a group. Collectively, these data reveal previously unsuspected nuances of the physiological roles of each of the four mapped BP QTLs in the SHR under basal and/or salt loading conditions unforeseen by the analysis of the F2 cross.

First published July 14, 2010; doi:10.1152/ajprenal.00654.2009.-Abnormalities in renal proximal tubular (PT) sodium transport play an important role in the pathophysiology of essential hypertension. The Na(+)/H(+) exchanger isoform 3 (NHE3) represents the major route for sodium entry across the apical membrane of renal PT cells. We therefore aimed to assess in vivo NHE3 transport activity and to define the molecular mechanisms underlying NHE3 regulation before and after development of hypertension in the spontaneously hypertensive rat (SHR). NHE3 function was measured as the rate of bicarbonate reabsorption by means of in vivo stationary microperfusion in PT from young prehypertensive SHR (Y-SHR; 5-wk-old), adult SHR (A-SHR; 14-wk-old), and age-matched Wistar Kyoto (WKY) rats. We found that NHE3-mediated PT bicarbonate reabsorption was reduced with age in the SHR (1.08 +/- 0.10 vs. 0.41 +/- 0.04 nmol/cm(2)xs), while it was increased in the transition from youth to adulthood in the WKY rat (0.59 +/- 0.05 vs. 1.26 +/- 0.11 nmol/cm(2)xs). Higher NHE3 activity in the Y-SHR compared with A-SHR was associated with a predominant microvilli confinement and a lower ratio of phosphorylated NHE3 at serine-552 to total NHE3 (P-NHE3/total). After development of hypertension, P-NHE3/total increased and NHE3 was retracted out of the microvillar microdomain along with the regulator dipeptidyl peptidase IV (DPPIV). Collectively, our data suggest that the PT is playing a role in adapting to the hypertension in the SHR. The molecular mechanisms of this adaptation possibly include an increase of P-NHE3/total and a redistribution of the NHE3-DPPIV complex from the body to the base of the PT microvilli, both predicted to decrease sodium reabsorption.

Estudou-se comparativamente a viabilidade, em moluscos hospedeiros, de miracídios de Schistosoma mansoni de duas origens: de ovos eliminados com as fezes e de granulomas hepáticos. Procurou-se saber se havia efeito da origem sobre o número total de cercárias obtidas e sobre o período de eliminação de cercárias. Foram também verificados o período pré-patente da infecção dos moluscos, a sobrevivência do caramujo após a infecção e o sexo das cercárias eliminadas. Concluiu-se que: a) o período pré-patente foi maior nos moluscos infectados por miracídios provenientes de granulomas hepáticos; b) a sobrevivência dos moluscos após a infecção foi maior naqueles infectados por miracídios provenientes de granulomas hepáticos; c) o número de cercadas eliminadas por molusco infectado com miracídios provenientes de granulomas hepáticos foi três vezes maior que o número de cercárias eliminadas por moluscos infectados com miracídios provenientes de ovos eliminados com as fezes.

Utilizando-se um artefato de vidro composto de duas câmaras unidas por um canal, testamos a atração miraxonal exercida por Biomphalaria straminea, Lymnaea columella e Physa sp, sobre larvas de Schistosoma mansoni da linhagem BH. Os moluscos, ou suas águas de condicionamento (SCW), foram colocados aleatoriamente em uma das câmaras, contendo o restante do artefato somente água declorada. Dez miracídios foram depositados no centro do canal. O comportamento dos miracídios foi observado por 15 min. Foram feitas dez replicações de cada experimento, utilizando-se espécimens de moluscos e miracídios diferentes. Verificou-se que todos os moluscos e suas SCW exerceram atração miraxonal, sendo que Physa sp atraiu mais que Lymnaea columella. Foi isolado um único exemplar de B. straminea que exerceu efeito repulsivo sobre os miracídios de S. mansoni.

Uma linhagem celular de rim de coelho (denominada RC-IAL), que foi isolada em 1976, e que atualmente está na 150ª passagem, teve suas características analisadas. As células apresentaram morfologia semelhante aos fibroblastos desde o início de seu cultivo. A proporção de crescimento celular não se alterou desde seu isolamento, com uma eficiência de clonagem ao redor de 9%. A linhagem mostrou crescimento dependente de ancoragem, e a análise cromossômica apresentou o número modal da espécie com pequenas variações para mais ou menos um cromossomo, resultando uma somatória de 50%. Sua espécie de origem foi comprovada através da reação de imunofluorescência indireta e a susceptibilidade da linhagem a alguns vírus, com demonstração do efeito citopático, foi verificada com os vírus da vacínia, cowpox, herpes simples tipo 1 e 2 e da rubéola. Esse substrato celular está livre de contaminantes, satisfazendo assim, as condições para seu uso em trabalhos científicos, principalmente os relacionadas à saúde pública.
