Mammalian heparanase is an endo-β-glucuronidase associated with cell invasion in cancer metastasis, angiogenesis and inflammation. Heparanase cleaves heparan sulfate proteoglycans in the extracellular matrix and basement membrane, releasing heparin/heparan sulfate oligosaccharides of appreciable size. This in turn causes the release of growth factors, which accelerate tumor growth and metastasis. Heparanase has two glycosaminoglycan-binding domains; however, no three-dimensional structure information is available for human heparanase that can provide insights into how the two domains interact to degrade heparin fragments. We have constructed a new homology model of heparanase that takes into account the most recent structural and bioinformatics data available. Heparin analogs and glycosaminoglycan mimetics were computationally docked into the active site with energetically stable ring conformations and their interaction energies were compared. The resulting docked structures were used to propose a model for substrates and conformer selectivity based on the dimensions of the active site. The docking of substrates and inhibitors indicates the existence of a large binding site extending at least two saccharide units beyond the cleavage site (toward the nonreducing end) and at least three saccharides toward the reducing end (toward heparin-binding site 2). The docking of substrates suggests that heparanase recognizes the N-sulfated and O-sulfated glucosamines at subsite +1 and glucuronic acid at the cleavage site, whereas in the absence of 6-O-sulfation in glucosamine, glucuronic acid is docked at subsite +2. These findings will help us to focus on the rational design of heparanase-inhibiting molecules for anticancer drug development by targeting the two heparin/heparan sulfate recognition domains.

Iris Sloman, daughter of Albert and Josephine, is thought to be in the front row, centre. The location and date of the photo is unknown, however, it could be in the early 1920s. This photograph was in the possession of Rick Bell, of St. Catharines, son of Iris and Richard Bell. Relatives of the Sloman - Bell family are former Black slaves from the United States.

Mining seafloor massive sulfides for metals is an emergent industry faced with environmental management challenges. These revolve largely around limits to our current understanding of biological variability in marine systems, a challenge common to all marine environmental management. VentBase was established as a forum where academic, commercial, governmental, and non-governmental stakeholders can develop a consensus regarding the management of exploitative activities in the deep-sea. Participants advocate a precautionary approach with the incorporation of lessons learned from coastal studies. This workshop report from VentBase encourages the standardization of sampling methodologies for deep-sea environmental impact assessment. VentBase stresses the need for the collation of spatial data and importance of datasets amenable to robust statistical analyses. VentBase supports the identification of set-asides to prevent the local extirpation of vent-endemic communities and for the post-extraction recolonization of mine sites. © 2013.

Chiral N-heterocyclic carbene–borane complexes have been synthesised, and have been shown to reduce ketones with Lewis acid promotion. Chiral N-heterocyclic carbene–borane and diorganoborane complexes can reduce ketones with enantioselectivities up to 75% and 85% ee, respectively.

A dialkylborenium ion stabilized by an N-heterocyclic carbene has been prepared for the first time by reaction of IMes-9-BBN-H with triflic acid. The ion-separated nature of the borenium ion was confirmed by 1H and 19F diffusion ordered NMR spectroscopy.

When people rapidly judge the truth of claims presented with or without related but nonprobative photos, the photos tend to inflate the subjective truth of those claims-a "truthiness" effect (Newman et al., 2012). For example, people more often judged the claim "Macadamia nuts are in the same evolutionary family as peaches" to be true when the claim appeared with a photo of a bowl of macadamia nuts than when it appeared alone. We report several replications of that effect and 3 qualitatively new findings: (a) in a within-subjects design, when people judged claims paired with a mix of related, unrelated, or no photos, related photos produced truthiness but unrelated photos had no significant effect relative to no photos; (b) in a mixed design, when people judged claims paired with related (or unrelated) and no photos, related photos produced truthiness and unrelated photos produced "falseness;" and © in a fully between design, when people judged claims paired with either related, unrelated, or no photos, neither truthiness nor falsiness occurred. Our results suggest that photos influence people's judgments when a discrepancy arises in the expected ease of processing, and also support a mechanism in which-against a backdrop of an expected standard-related photos help people generate pseudoevidence to support claims. Fire is an important driver of nutrient cycling in savannas. Here, we determined the impact of fire frequency on total and soluble soil nitrogen (N) pools in tropical savanna. The study sites consisted of 1-ha experimental plots near Darwin, Australia, which remained unburnt for at least 14 years or were burnt at 1-, 2- or 5-year intervals over the past 6 years. Soil was analysed from patches underneath tree canopies and in inter-canopy patches at 1, 12, 28, 55 and 152 days after fire. Patch type had a significant effect on all soil N pools, with greater concentrations of total and soluble (nitrate, ammonium, amino acids) N under tree canopies than inter-canopy patches. The time since the last fire had no significant effect on N pools. Fire frequency similarly did not affect total soil N but it did influence soluble soil N. Soil amino acids were most prominent in burnt savanna, ammonium was highest in infrequently burnt (5-year interval) savanna and nitrate was highest in unburnt savanna. We suggest that the main effect of fire on soil N relations occurs indirectly through altered tree-grass dynamics. Previous studies have shown that high fire frequencies reduce tree cover by lowering recruitment and increasing mortality. Our findings suggest that these changes in tree cover could result in a 30% reduction in total soil N and 1060% reductions in soluble N pools. This finding is consistent with studies from savannas globally, providing further evidence for a general theory of patchiness as a key driver of nutrient cycling in the savanna biome.

Tropical marginal seas (TMSs) are natural subregions of tropical oceans containing biodiverse ecosystems with conspicuous, valued, and vulnerable biodiversity assets. They are focal points for global marine conservation because they occur in regions where human populations are rapidly expanding. Our review of 11 TMSs focuses on three key ecosystems—coral reefs and emergent atolls, deep benthic systems, and pelagic biomes—and synthesizes, illustrates, and contrasts knowledge of biodiversity, ecosystem function, interaction between adjacent habitats, and anthropogenic pressures. TMSs vary in the extent that they have been subject to human influence—from the nearly pristine Coral Sea to the heavily exploited South China and Caribbean Seas—but we predict that they will all be similarly complex to manage because most span multiple national jurisdictions. We conclude that developing a structured process to identify ecologically and biologically significant areas that uses a set of globally agreed criteria is a tractable first step toward effective multinational and transboundary ecosystem management of TMSs.
