Cytogenetic and random amplified polymorphic DNA analyses carried out in the species Leptodactylus podicipinus, L. ocellatus, L. labyrinthicus, and L. fuscus from rural and urban habitats of the northwest region of São Paulo State, Brazil, showed that the karyotypes (2n = 22), constitutive heterochromatin distribution and nucleolus organizer region (NOR) location did not differ between the populations from the two environments. The in situ hybridization with an rDNA probe confirmed the location of the NORs on chromosome 8 revealing an in tandem duplication of that region in one of the chromosomes of L. fuscus. DAPI showed that part of the C-band-positive heterochromatin is rich in AT, including that in the proximity the NORs in L. podicipinus and L. ocellatus. The molecular analyses showed that the two populations (urban and rural) of L. podicipinus and L. fuscus are similar from a genetic point of view. The urban and rural populations of species L. ocellatus and L. labyrinthicus showed differences in genetic structures, probably due to urbanization which interferes with the dispersion of those frogs. The marked differences observed between the two populations of L. ocellatus can be representing the cryptic condition of the species. Unweighted pair-group method of analysis and genetic distance analysis detected the genetic proximity between L. ocellatus and L. fuscus. The results indicate that there was no reduction in the genetic diversity in the populations from the urban environment; however, the survival of these frogs would not be guaranteed in the case of an increase in human impact especially for populations of L. labyrinthicus and L. ocellatus. ©FUNPEC-RP.

Foram construídas seis estufas climatizadas, instaladas inicialmente no Ranário Experimental da Universidade Federal de Viçosa (UFV) e, posteriormente, no Ranário Experimental da Fundação Universidade Federal do Rio Grande, com o objetivo de realizar experimentos para avaliar os efeitos do ambiente sobre o desempenho de rãs em gaiolas de fibra de vidro. Ambientes com temperaturas de 25ºC e fotoperíodo de 12/12 horas de luz/horas de escuridão (h L/E) serviram para adaptação das rãs por 15 dias antes de cada experimento. Os tratamentos consistiram em simular ambientes com temperaturas variando de 20 a 35ºC e fotoperíodos de 8/16, 12/12 e 16/8 h L/E. Foram realizados experimentos com rã-touro (Rana catesbeiana Shaw, 1802) e rã-manteiga (Leptodactylus ocellatus Linnaeus, 1758). Nessas estufas foi possível estimar que: a) os maiores ganhos de peso de rã-touro foram obtidos entre 27,6 e 29,7ºC, com melhor crescimento entre 28,2 e 30,1ºC; para rã-manteiga os melhores ganhos e conversão alimentar foram observados a 28,6 e 28ºC, respectivamente; b) a temperatura interage com fotoperíodo sobre o desempenho das rãs e seu desenvolvimento gonadal; c) a 27,7ºC (temperatura de conforto térmico) haverá menos rãs dentro d'água; d) a maior temperatura cloacal de rã-touro, 32,1ºC no seco e 33,8ºC dentro d'água, a 35ºC, evidenciou que as rãs se termorregulam; e) os níveis de tetraiodotironina (T4) no plasma decrescem na temperatura de conforto térmico; f) rã-manteiga condiciona-se ao manejo de rotina, reunindo-se ao redor do cocho na hora da alimentação.

Cytogenetic studies were carried out on five species of Leptodactylus, namely L. fuscus, L, notoaktites, L. labyrinthicus, L. ocellatus, and L. podicipinus, after standard staining, Ag-NOR and C-banding as well as BrdU incorporation for three of them. The species had 2n = 22 chromosomes and two basic karyotype patterns. Chromosome 8 was a marker bearing a secondary constriction. In all species, this secondary constriction corresponded to the Ag-NOR site. The species had centromeric C-bands in all chromosomes of the complement, but some interstitial or telomeric bands seemed to differentiate some karyotypes, either at the species or the population level. In L. ocellatus, the C-banding pattern confirmed the occurrence of a heteromorphic pericentric inversion in chromosome 8 in specimens from one of the populations. The BrdU incorporation technique showed no detectable difference in the replication patterns of the major bands in the chromosomes of L. noroaktites, L. labyrinthicus, and L. ocellatus.

Nowadays, the emergence of resistance to the current available chemotherapeutic drugs by cancer cells makes the development of new agents imperative. The skin secretion of amphibians is a natural rich source of antimicrobial peptides (AMP), and researchers have shown that some of these wide spectrum molecules are also toxic to cancer cells. The aim of this study was to verify a putative anticancer activity of the AMP pentadactylin isolated for the first time from the skin secretion of the frog Leptodactylus labyrinthicus and also to study its cytotoxic mechanism to the murine melanoma cell line B16F10. The results have shown that pentadactylin reduces the cell viability of B16F10 cells in a dose-dependent manner. It was also cytotoxic to normal human fibroblast cells; nevertheless, pentadactylin was more potent in the first case. The studies of action mechanism revealed that pentadactylin causes cell morphology alterations (e.g., round shape and shrinkage morphology), membrane disruption, DNA fragmentation, cell cycle arrest at the S phase, and alteration of mitochondrial membrane potential, suggesting that B16F10 cells die by apoptosis. The exact mechanism that causes reduction of cell viability and cytotoxicity after treatment with pentadactylin is still unknown. In conclusion, as cancer cells become resilient to death, it is worthwhile the discovery of new drugs such as pentadactylin that induces apoptosis.

Peptidases are ubiquitous enzymes involved in diverse biological processes. Fragments from bioactive peptides have been found in skin secretions from frogs, and their presence suggests processing by peptidases. Thus, the aim of this work was to characterize the peptidase activity present in the skin secretion of Leptodactylus labyrinthicus. Zymography revealed the presence of three bands of gelatinase activity of approximately 60 kDa, 66 kDa, and 80 kDa, which the first two were calcium-dependent. These three bands were inhibited either by ethylenediaminetetraacetic acid (EDTA) and phenathroline; thus, they were characterized as metallopeptidases. Furthermore, the proteolytic enzymes identified were active only at pH 6.0–10.0, and their activity increased in the presence of CHAPS or NaCl. Experiments with fluorogenic substrates incubated with skin secretions identified aminopeptidase activity, with cleavage after leucine, proline, and alanine residues. This activity was directly proportional to the protein concentration, and it was inhibited in the presence of metallo and serine peptidase inhibitors. Besides, the optimal pH for substrate cleavage was determined to be 7.0–8.0. The results of the in gel activity assay showed that all substrates were hydrolyzed by a 45 kDa peptidase. Gly-Pro-AMC was also cleaved by a peptidase greater than 97 kDa. The data suggest the presence of dipeptidyl peptidases (DPPs) and metallopeptidases; however, further research is necessary. In conclusion, our work will help to elucidate the implication of these enzymatic activities in the processing of the bioactive peptides present in frog venom, expanding the knowledge of amphibian biology.

Red drum is one ofthe most popular species sought by anglers in Florida Bay, yet juveniles are rarely encountered. We evaluated Florida Bay as a nursery area for red drum by sampling for recently-settled late larvae in basin areas within the bay with an epi-benthic sled at six stations in November 2000, and at seven stations during December 2000 through February 2001. In November 2000 we surveyed potential sampling sites in quiet backwaters adjacent to mangroves for juvenile red drum. A total of 202 sites were sampled mainly in northern Florida Bay and adjacent waters with a cast net. We collected only one recently-settled red drum larvae and no juveniles. Obviously the sites that we sampled in Florida Bay and adjacent waters are not nursery habitat for this valuable species. Sled collections were dominated by bay anchovy, Anchoa mitchilli, but densities were biased by one collection. Five small resident species were among the dominant species: rainwater killifish, Lucania parva; dusky pipefish, Syngnathus floridae; dwarf seahorse, Hippocampus zosterae; and clown goby, Microgobius gulosus. Three species that spawn outside Florida Bay in the GulfofMexico were common: pinfish, Lagodon rhomboides; pigfish, Orthopristis chrysoptera; and silver perch, Bairdiella chrysoura. Twenty-seven species were collected with the cast net. Hardhead silversides (Atherinomorus stipes), bay anchovy, tidewater mojarra (Eucinostomus harengulus), silver jenny (Eucinostomus gula), and goldspotted killifish (Floridichthys carpio) were the most common in cast net collections. Although only one red drum was collected, we were able to: (1) identify mesohaline waters from our cast net sites to test our preliminary assessment that mesohaline habitat might be limited in Florida Bay, (2) document the distribution and abundance of fishes collected by cast net that should enhance our understanding of ichthyofauna in the Northern Subdivision ofFlorida Bay and adjacent waters, and (3) from epibenthic sled collections, describe the habitats, abundance and distribution of recently settled larvae/small juveniles/small resident fishes during late fall and winter. We used 25 years of conventional tagging data (n= 6173 recoveries) and 3 years of ultrasonic telemetry data (n=105 transmitters deployed) to examine movement rates and directional preferences of four age classes of red drum (Sciaenops ocellatus) in estuarine and coastal waters of North Carolina. Movement rates of conventionally tagged red drum were dependent on the age, region, and season of tagging. Age-1 and age-2 red drum tagged along the coast generally moved along the coast, whereas fish tagged in oligohaline waters far from the coast were primarily recovered in coastal regions in fall months. Adult (age-4+) red drum moved from overwintering grounds on the continental shelf through inlets into Pamlico Sound in spring and summer months and departed in fall. Few tagged red drum were recovered in adjacent states (0.6% of all recoveries); however, some adult red drum migrated seasonally from overwintering grounds in coastal North Carolina northward to Virginia in spring, returning in fall. Age-2 transmitter-tracked red drum displayed seasonal emigration from a small tributary, but upstream and downstream movements within the tributary were correlated with fluctuating salinity regimes and not season. Large-scale conventional tagging and ultrasonic telemetry programs can provide valuable insights into the complex movement patterns of estuarine fish.

In this note, we document polymerase-chain-reaction (PCR) primer pairs for 101 nuclear-encoded microsatellites designed and developed from a genomic library for red drum (Sciaenops ocellatus). Details of the genomic library construction, the sequencing of positive clones, primer design, and PCR protocols may be found in Karlsson et al. (2008). The 101 microsatellites (GENBA NK Accession Numbers EU015882-EU015982) were amplified successfully and used to genotype 24 red drum obtained from Galveston Bay, Texas (Table 1). A total of 69 of the microsatellites had an uninterrupted (perfect) dinucleotide motif, and 30 had an imperfect dinucleotide motif; one microsatellite had an imperfect tetranucleotide motif, and one had an imperfect and compound motif (Table 1 ). Sizes of the cloned alleles ranged from 84 to 252 base pairs. A ‘blast’ search of the GENBANK database indicated that all of the primers and the cloned alleles were unique (i.e., not duplicated).

Because of a lack of fishery-dependent data, assessment of the recovery of fish stocks that undergo the most aggressive form of management, namely harvest moratoriums, remains a challenge. Large schools of red drum (Sciaenops ocellatus) were common along the northern Gulf of Mexico until the late 1980s when increased fishing effort quickly depleted the stock. After 24 years of harvest moratorium on red drum in federal waters, the stock is in need of reassessment; however, fisherydependent data are not available in federal waters and fishery-independent data are limited. We document the distribution, age composition, growth, and condition of red drum in coastal waters of the north central Gulf of Mexico, using data collected from a nearshore, randomized, bottom longline survey. Age composition of the fishery-independent catch indicates low mortality of fish age 6 and above and confirms the effectiveness of the federal fishing moratorium. Bottom longline surveys may be a cost-effective method for developing fishery-independent indices for red drum provided additional effort can be added to nearshore waters (<20 m depth). As with most stocks under harvest bans, effective monitoring of the recovery of red drum will require the development of fishery-independent indices. With limited economic incentive to evaluate non-exploited stocks, the most cost-effective approach to developing such monitoring is expansion of existing fishery independent surveys. We examine this possibility for red drum in the Gulf of Mexico and recommend the bottom longline survey conducted by the National Marine Fisheries Service expand effort in nearshore areas to allow for the development of long-term abundance indices for red drum.
