A real-time polymerase chain reaction (PCR) test was developed on the basis of the Leishmania glucose-6-phosphate dehydrogenase locus that enables identification and quantification of parasites. Using two independent pairs of primers in SYBR-Green assays, the test identified etiologic agents of cutaneous leishmaniasis belonging to both subgenera, Leishmania (Viannia) and Leishmania (Leishmania) in the Americas. Furthermore, use of TaqMan probes enables distinction between L. (V.) braziliensis or L. (V.) peruviania from the other L. (Viannia) species. All assays were negative with DNA of related trypanosomatids, humans, and mice. The parasite burden was estimated by normalizing the number of organisms per total amount of DNA in the sample or per host glyceraldehyde-3-phosphate dehydrogenase copies. The real-time PCR assay for L. (Leishmania) subgenus showed a good linear correlation with quantification on the basis of a limiting dilution assay in experimentally infected mice. The test successfully identifies and quantifies Leishmania in human biopsy specimens and represents a new tool to study leishmaniasis.

We report the identification of two distinct homologues of the 70-kDa mitochondrial heat shock protein (mtHSP70) from Leishmania chagasi/Leishmania infantum (Lc2.1 and Lc2.2). in Leishmania species, multiple genes encoding Lc2.2 are present whilst single genes encode Lc2.1. Strikingly, genes encoding Lc2.1-like proteins are absent from Trypanosoma species. Lc2.2 is characterized by a poly-glutamine rich C-terminus, absent from Lc2.1 or mtHSP70 homologues outside the trypanosomatids. Lc2.1 displays unique substitutions within its peptide-binding domain which modify amino acids strictly conserved in cytoplasmic and mitochondrial HSP70 proteins alike. Affinity purified antibodies recognize mainly a single protein in extracts from promastigotes/epimastigotes of various Leishmania/Trypanosoma species. Upon differentiation of Leishmania amazonensis into amastigotes a second protein (presumably Lc2.1) is induced and becomes the predominant mtHSP70 homologue expressed. Subcellular localization of these proteins was investigated and ratified a distribution throughout the mitochondrial matrix. The first autochthonous case of American cutaneous leishmaniasis was reported in the Federal District in 1980, and the species involved in this type of leishmaniasis was unknown. This study aimed to identify the species that causes the disease in the Federal District and to investigate its clinical and epidemiological aspects. Between 2000 and 2007, 71 autochthonous cases of leishmaniasis were reported in the Federal District. Leishmania species were identified by means of direct immunofluorescence reactions using monoclonal antibodies and restriction fragment length polymorphism. The species of 40 (56.33%) out of 71 samples were identified. Thirty-six (90%) were identified as Leishmania (Viannia) braziliensis and four (10%) were identified as Leishmania (Leishmania) amazonensis. In this area, the disease had clinical and epidemiological characteristics similar to those found in other Brazilian regions.

Células Mononucleares do sangue periférico (CMSP) oriundas de 28 pacientes que residem em áreas endêmicas para Leishmania braziliensis foram estudados. Destes, 10 encontravam-se na fase ativa da doença e 18 curados clinicamente para as lesões de leishmaniose. As células foram cultivadas frente à antígenos de 6 espécies de Leishmania: Leishmania: L. (V.) braziliensis (LbAg), L. (V.) guyanensis (LgAg), L. (V.) lainsoni (LlAg), L. (V.) naiffi (LnAg), L. (L.) amazonensis (LaAg), L. (L.) chagasi (LcAg) e antígeno de concanavalina A como mitógeno. Os resultados foram expressos em índices de estimulação (IE). Temendo uma possível degradação protéica, os antígenos foram preparados com PBS e com PBS tampão antiproteolítico e o perfil eletroforético foi analisado através de SDS-PAGE. Os sobrenadantes das culturas foram estocados para os ensaios de ELISA para detecção de IFN-γ. A análise do perfil eletroforético dos antígenos de diferentes espécies de Leishmania demonstrou um padrão distinto e heterogêneo e aparentemente, não foi observada degradação assim que os antígenos foram preparados e 10 meses de estoque. Entretanto bandas parecem ser compartilhadas entre as espécies, que podem estar associadas a uma conservação de antígenos entre as espécies de Leishmania. A resposta proliferativa dos linfócitos (RPL) dos pacientes da forma ativa foi mais intensa para LbAg (1710,3), seguido de LnAg (177,6), LaAg (16,89,8) e LlAg (1410,1), e menos intenso para LgAg (11,46,6). Quando os IE obtidos para LbAg foram comparadas com outras espécies observou-se diferenças para LgAg (p<0.004) e LlAg (p<0.03). Não foram encontrados diferenças no perfil protéico dos extratos antigênicos produzidos com e sem inibidores de protease. Os estímulos obtidos de CMSP de indivíduos curados clinicamente não demonstraram diferenças quando comparados com aqueles da fase ativa da doença. O índice de estimulação (médiadesvio padrão) frente estímulos antigênicos pouco variou entre as espécies: LbAg - 22,222,2, LnAg - 15,911,5, LgAg - 20,720,6, LlAg - 14,710,1, LaAg - 15,1114,5 e L. chagasi - 13,78). A produção de IFN-γ quantificada nos sobrenadantes das culturas frente aos antígenos totais e solúvel mostrou níveis da citocina mais elevados quando utilizou-se antígenos total de L. guyanensis (568,1544,5; mediana: 518,5 pg/mL) quando comparadas às outras espécies analisadas. Porém quando analisadas o antígeno solúvel, podemos observar que tanto para os antígenos da espécie L. braziliensis quanto para L. naiffi, a fração total obteve os maiores índices de estímulos quando comparadas com a fração solúvel. Estes estudos sugerem que há uma reatividade cruzada entre as espécies dos subgêneros Viannia e Leishmania.

The META cluster of Leishmania amazonensis contains both META1 and META2 genes, which are upregulated in metacyclic promastigotes and encode proteins containing the META domain. Previous studies defined META2 as a 48.0-kDa protein, which is conserved in other Leishmania species and in Trypanosoma brucei. In this work, we demonstrate that META2 protein expression is regulated during the Leishmania life cycle but constitutive in T. brucei. META2 protein is present in the cytoplasm and flagellum of L amazonensis promastigotes. Leishmania META2-null replacement mutants are more sensitive to oxidative stress and, upon heat shock, assume rounded morphology with shortened flagella. The increased susceptibility of null parasites to heat shock is reversed by extra-chromosomal expression of the META2 gene. Defective Leishmania promastigotes exhibit decreased ability to survive in macrophages. By contrast, META2 expression is decreased by 80% in RNAi-induced T. brucei bloodstream forms with no measurable effect on survival or resistance to heat shock. (C) 2010 Elsevier Inc. Limonene is a monoterpene that has antitumoral, antibiotic and antiprotozoal activity. In this study we demonstrate the activity of limonene against Leishmania species in vitro and in vivo. Limonene killed Leishmania amazonensis promastigotes and amastigotes with 50% inhibitory concentrations of 252.0 +/- 49.0 and 147.0 +/- 46.0 mu M, respectively. Limonene was also effective against Leishmania major, Leishmania braziliensis and Leishmania chagasi promastigotes. The treatment of L. amazonensis-infected macrophages with 300 mu M limonene resulted in 78% reduction in infection rates. L. amazonensis-infected mice treated topically or intrarectally with limonene had significant reduction of lesion sizes. A significant decrease in the parasite load was shown in the lesions treated topically with limonene by histopathological examination. The intrarectal treatment was highly effective in decreasing the parasite burden, healing established lesions and suppressing the dissemination of ulcers. Limonene presents low toxicity in humans and has been shown to be effective as an agent for enhancing the percutaneous permeation of drugs. Control of human visceral leishmaniasis in endemic regions is hampered in part by the lack of knowledge with respect of the role reservoirs and vector. In addition, there is not yet an understanding of how non-symptomatic subclinical infection might influence the maintenance of infection in a particular locality. Of worrisome is the limited accessibility to medical care in places with emerging drug resistance. There is still no available protective vaccine either for humans or other reservoirs. Leishmania species are protozoa that express multiple antigens which are recognized by the vertebrate immune system. Since there is not one immunodominant epitope recognized by most hosts, strategies must be developed to optimize selection of antigens for prevention and immunodiagnosis. For this reason, we generated a cDNA library from the intracellular amastigote form of Leishmania chagasi, the causative agent of South American visceral leishmaniasis. We employed a two-step expression screen of the library to systematically identify T and T-dependent B cell antigens. The first step was aimed at identifying the largest possible number of clones producing an epitope-containing polypeptide with a pool of sera from Brazilians with documented visceral leishmaniasis. After removal of clones encoding heat shock proteins, positive clones underwent a second step screen for their ability to cause proliferation and IFN-γ responses of T cells from immune mice. Six unique clones were selected from the second screen for further analysis. The clones encoded part of the coding sequence of glutamine synthetase, transitional endoplasmic reticulum ATPase, elongation factor 1γ, kinesin K-39, repetitive protein A2, and a hypothetical conserved protein. Humans naturally infected with L. chagasi mounted both cellular and antibody responses to these protein Preparations containing multiple antigens may be optimal for immunodiagnosis and protective vaccines against Leishmania

The aim of this work was to molecularly detect Leishmania species in 52 cats from Andradina Municipality, São Paulo State, Brazil. The direct parasitological test was performed by using imprints of poplited lymph node, bone marrow and spleen to verify amastigote forms of Leishmania spp. The samples that were positive parasitological tests were subjected to molecular analysis (PCR) and sequencing. Infection was detected for 5.76% (3/52) of the examined cats and two had presence of amastigote forms of Leishmania spp. in lymph nodes. Polymerase chain reaction (PCR) of kinetoplast minicircle DNA, indicated positive amplification for samples of spleen and lymph nodes and the sequencing resulted in 97% similarity with Leishmania (L) chagasi. This study proved the occurrence of infection with Leishmania (L) chagasi in felines from Andradina municipality, São Paulo State. (C) 2010 Elsevier B.V. All rights reserved.

A new parasite species of Leishmania is described, L. (Leishmania) forattinii sp. n., which was isolated from a pooled triturate of liver and spleen of a opossum (Didelphis marsupialis aurita) and from skin samples from a rodent (Proechmys iheringi denigratus), captured in primary forest on the Atlantic Cost of Brazil. Our results on the basis of biological and molecular criteria indicate that this taxonomically distinct parasite ias a new species of the L. mexicana complex, but closely related to L. (L.) aristidesi Laison & shaw, 1979, as revelated by phenetic and phylogenetic numerical analyses of the enzyme data. L. forattinii was clearly distinguishable from other Leishmania species of the genus usisng enzyme electrophoresis, monoclonal antibodies, molecular karyotypes, analysis of restriction enzyme digestion patterns of kinetoplast DNA (kDNA), as well as the use of kDNA hybridization procedures.

Immunity against the intracellular protozoan Leishmania species is highly dependent on Th1 development. We have previously shown that IL-12 is a powerful and probably obligatory factor for Th1 cell generation and proliferation. We also observed that the experimental infection of C3H and BALB/c mice with Leishmania major is associated with IL-12 production in vivo. Now we demonstrate that metacyclic L. major promastigotes are poor inducers of IL-12 in vitro in fresh human PBMC and monocytes. In addition, we show that the ability of this pathogen to induce IL-12 and other cytokines is modulated by the metacyclogenic process, which had previously not been recognized. In contrast to the infective parasites (metacyclic promastigotes), the procyclic promastigotes collected at the logarithmic phase of the culture displayed a striking ability to induce IL-12, IFN-γ, TNF-α, and IL-10. Despite this differential effect of procyclic and metacyclic parasites in terms of IL-12 induction, both stages were inhibitory for IL-12 production induced by Staphylococcus aureus. The ability of procyclic promastigotes and, to a much lesser extent, that of metacyclic promastigotes to induce IL-12 were enhanced by pretreatment of monocytes in a cytokine milieu containing IFN-γ, IL-4, IL-13, or granulocyte-macrophage CSF or. by neutralization of endogenous IL-10. Our results suggest the development of an evasion mechanism as the Leishmania promastigotes mature to infectious forms and the possi-bility of using Ags derived from procyclic promastigotes for immunization procedures. A leishmaniose tegumentar americana (LTA) constitui uma doença infecciosa causada por protozoários do gênero Leishmania com elevada incidência na região Amazônica. Uma variedade de espécies de leishmania é responsável por esta patologia. Desta forma, dependendo da espécie e da resposta imunológica do hospedeiro vertebrado, a doença pode apresentar diferentes formas clínicas, como a leishmaniose cutânea localizada (LCL) e a leishmaniose mucocutânea (LMC). A principal espécie responsável pela LTA é a Leishmania (Viannia) braziliensis. Contudo, devido à existência de uma multiplicidade de cepas desta espécie e ao reduzido número de estudos relacionados, torna-se importante o conhecimento dos aspectos metabólicos básicos do protozoário, como o metabolismo lipídico, na tentativa de caracterizar vias ou componentes fundamentais para seu desenvolvimento e infectividade. Desta forma, este trabalho teve como objetivo analisar distribuição de corpos lipídicos (CLs) e o perfil lipídico de duas cepas de L. (V.) braziliensis, isolada de diferentes casos clínicos, em diferentes períodos da fase estacionária do crescimento celular. As formas promastigotas das cepas M17593 (LCL) e M17323 (LMC) de L. (V.) braziliensis foram utilizadas na fase estacionária inicial (EST-I) e estacionária tardia (EST-T) de crescimento. Inicialmente, foi realizada análise ultraestrutural das formas promastigotas por microscopia eletrônica de transmissão (MET) e foram observadas estruturas sugestivas de CLs distribuídos no citoplasma do parasito, confirmados pela técnica citoquímica ósmio-imidazol, organelas necessárias para o metabolismo energético do parasito. Para quantificar a distribuição de CLs entre os dias de cultivo e entre as cepas, foi realizada análise por citometria de fluxo com Bodipy® 493/503. Os resultados indicaram que a cepa responsável pela LMC apresentou maior quantidade de CLs durante a fase estacionária tardia. Na cepa LCL não foi observado diferença significativa entre as fases estudadas. Assim, pode ser sugerido que a exacerbada resposta inflamatória que ocorre em pacientes com LMC, esteja relacionada com o acúmulo de CLs no parasito, fonte de energia e eicosanoides, como prostaglandinas. Outra hipótese é a possível correlação de CLs com a baixa exposição do fosfolipídio fosfatidilserina para a superfície externa da membrana, importante para a infectividade do parasito. Para análise dos lipídios totais, os parasitos foram submetidos à extração lipídica, seguido da técnica de HPTLC, onde foram encontrados predominantemente fosfolipídios, esterol esterificado, esteróis, triglicerídeos e ácidos graxos compondo o parasito, com variações entre as cepas e entre as fases estudadas. A cepa LCL na fase estacionária tardia possui maior quantidade de lipídios totais, que pode ser justificado por já ser conhecida como a cepa mais infectiva e possivelmente apresentar maior quantidade de glicoconjugados associados com subdomínios lipídicos importantes para o reconhecimento de fagócitos. É importante ressaltar que a maior infectividade da cepa LCL quando comparada à cepa LMC, resulta em um menor processo inflamatório. Estes resultados indicam que há uma variação no perfil lipídico e na distribuição de CLs entre as diferentes cepas de L. (V.) braziliensis, que pode estar relacionado com a infectividade do parasito e com a manifestação clinica da doença.
