São apresentados resultados em relação a infecção expertmental de Zygodontomys lasiurus (Rodentia-Cricetidae) com duas cepas de T. cruzi isoladas de casos humanos, Y e Berenice, e uma isolada de um triatomíneo silvestre, chamada costalimai. Foram realizados estudos em relação a evolução da parasitemia, duração da patência e prepatência da infecção. Com o objetivo de verificar a agressividade e tropismo tissular das cepas de T. cruzi nesta espécie de roedor, foram também realizados estudos histopatológicos. Os resultados obtidos mostraram que os níveis de parasitemias foram baixos para as três cepas estudadas. A patência da infecção variou de 14 a 16 dias nos animais inoculados com a cepa Y, 26 a 29 dias com a Berenice e 9 a 13 dias com a costalimai. O período prepatente variou de 3 a 5 dias nos animais inoculados com a cepa Y, de 2 a 6 dias com a cepa Berenice e de 6 a 8 dias com a costalimai. As três cepas apresentaram em Z. lasiurus, comprometimento nitidamente muscular, provocando reações leves, moderadas e intensas.

Cytogenetical studies based on conventional coloration by Giemsa, C-banding and Ag-NOR were performed on 2 species of bats from the vespertilionid family: Lasiurus cinereus (Beauvois, 1796) and Lasiurus ega (Gervais, 1856). The 2n was 28 and FN was 48 in both species. The constitutive heterochromatin is located in centromeric regions in the two species and in the short arm of the subtelocentric X chromosome in L. ega. NORs were observed in the secondary constriction of the smaller autosome in both species.

Studies on patterns of habitat use by mammals are necessary for understanding the mechanisms involved in their distribution and abundance. In this study, we used the spool-and-line method to investigate habitat utilization by two sigmodontine rodents from Brazilian Cerrado, Necromys lasiurus and Oryzomys scotti. We conducted the study in a Cerrado area in central Brazil (15 degrees 56'S and e 47 degrees 56'W) where the animals were caught in an area of 7.68 ha of Cerrado sensu stricto. Captured individuals were marked, equipped with a spool-and-Line device, and released at the same capture point. The next day we followed the thread to record their daily movements and find their nests. To investigate microhabitat selection we compared habitat characteristics along traits of each studied species with general habitat characteristics of the study area. Although the mean 24-h distance was greater for N. lasiurus (mean +/- SE: 41.9 +/- 42.2 m, N=3) than for O. scotti (28.7 +/- 14.2 m, N=6) this difference was not significant (Mann-Whitney test, U=26, P>0.6). We detected significant differences among observed microhabitats variables of both species and available microhabitat characteristics as determined by discriminant analysis (Wilks's lambda F=3.001; df=14, 116; P<0.001). Both species were associated to microhabitat characteristics whose values differed markedly from the overall available habitat. Along the first canonical discriminant function of the DFA both them were associated with greater grass height than the mean height available and along the second axis N. lasiurus selected areas with higher fruit availability and more shelters than those selected by 0. scotti. For stronger inferences regarding differential patterns of habitat utilization by Cerrado rodents we suggest the simultaneous use of both spool-and-line and standard trapping methods. We studied the development of the inverted yolk sac in a New World rodent, Necromys lasiurus during early placentation. Ten implantation sites were investigated by means of histology, immunohistochemistry and electron microscopy. The yolk sac was villous near its attachment to the placenta. Elsewhere it was non-villous and closely attached to the uterus. The uterine glands were shallow and wide mouthed. They were associated with vessels and filled with secretion, suggesting the release of histotroph. This feature was absent at later stages. The intimate association of the yolk sac with specialized glandular regions of the uterus may represent a derived character condition of Necromys and/or sigmodont rodents. (C) 2012 Elsevier Ltd. Background: Stereology is an established method to extrapolate three-dimensional quantities from two-dimensional images. It was applied to placentation in the mouse, but not yet for other rodents. Herein, we provide the first study on quantitative placental development in a sigmodontine rodent species with relatively similar gestational time. Placental structure was also compared to the mouse, in order to evaluate similarities and differences in developmental patterns at the end of gestation. Methods: Fetal and placental tissues of Necromys lasiurus were collected and weighed at 3 different stages of gestation (early, mid and late gestation) for placental stereology. The total and relative volumes of placenta and of its main layers were investigated. Volume fractions of labyrinth components were quantified by the One Stop method in 31 placentae collected from different individuals, using the Mercator® software. Data generated at the end of gestation from N. lasiurus placentae were compared to those of Mus musculus domesticus obtained at the same stage. Results: A significant increase in the total absolute volumes of the placenta and its main layers occurred from early to mid-gestation, followed by a reduction near term, with the labyrinth layer becoming the most prominent area. Moreover, at the end of gestation, the total volume of the mouse placenta was significantly increased compared to that of N. lasiurus although the proportions of the labyrinth layer and junctional zones were similar. Analysis of the volume fractions of the components in the labyrinth indicated a significant increase in fetal vessels and sinusoidal giant cells, a decrease in labyrinthine trophoblast whereas the proportion of maternal blood space remained stable in the course of gestation. On the other hand, in the mouse, volume fractions of fetal vessels and sinusoidal giant cells decreased whereas the volume fraction of labyrinthine trophoblast increased compared to N. lasiurus placenta. Conclusions: Placental development differed between N. lasiurus and M. musculus domesticus. In particular, the low placental efficiency in N. lasiurus seemed to induce morphological optimization of fetomaternal exchanges. In conclusion, despite similar structural aspects of placentation in these species, the quantitative dynamics showed important differences.

Objective: Hantaviruses are rodent-borne RNA viruses that have caused hantavirus cardiopulmonary syndrome in several Brazilian regions. In the present study, geographical distribution, seroprevalence, natural host range, and phylogenetic relations of rodent-associated hantaviruses collected from seven counties of Southeastern Brazil were evaluated. Methods: ELISA, RT-PCR and phylogenetic analysis were used in this study. Results: Antibodies to hantavirus were detected in Bolomys lasiurus, Akodon sp. and Oligoryzomys sp., performing an overall seroprevalence of 5.17%. All seropositive rodents were associated with grasslands or woods surrounded by sugar cane fields. Phylogenetic analysis of partial S- and M-segment sequences showed that viral sequences isolated from B. lasiurus specimens clustered with Araraquara virus. However, a sequence from Akodon sp. shared 100% similarity with Argentinian/Chilean viruses based on the partial S- segment amino acid sequence. Conclusion: These results indicate that there are associations between rodent reservoirs and hantaviruses in some regions of Southeastern Brazil, and suggest the existence of additional hantavirus genetic diversity and host ecology in these areas. Insectivorous bats are the main reservoirs of rabies virus (RABV) in various regions of the world. The aims of this study were to (a) establish genealogies for RABV strains from different species of Brazilian insectivorous bats based on the nucleoprotein (N) and glycoprotein (G) genes, (b) investigate specific RABV lineages associated with certain genera of bats and (c) identify molecular markers that can distinguish between these lineages. The genealogic analysis of N and G from 57 RABV strains revealed seven genus-specific clusters related to the insectivorous bats Myotis, Eptesicus, Nyctinomops, Molossus, Tadarida, Histiotus and Lasiurus. Molecular markers in the amino acid sequences were identified which were specific to the seven clusters. These results, which constitute a novel finding for this pathogen, show that there are at least seven independent epidemiological rabies cycles maintained by seven genera of insectivorous bats in Brazil. Com o intuito de averiguar a importância de alguns roedores como possíveis reservatórios do S. mansoni, na ausência do homem parasitado, foi realizada pesquisa, visando contribuir para o esclarecimento de aspectos ligados à cadeia epidemiológica da esquistossomose, bem como conhecer alguns parâmetros da biologia de certos roedores, em seu habitat semi-natural. O experimento foi realizado num viveiro de 952 m², no município de Taubaté, São Paulo (Brasil), numa área endêmica de esquistossomose mansônica humana, e teve a duração de três anos e seis meses (agosto de 1973 a dezembro de 1976). Foram utilizados como hospedeiros definitivos, Holochilus brasiliensis leucogaster, Zygodontomys lasiurus, Oryzomys nigripes eliurus e Cavia aperea aperea; como hospeideiro intermediário, Biomphalaria tenagophila e posteriormente B. glabrata. Entre agosto de 1973 e janeiro de 1976, não houve encontro de B. tenagophila eliminando cercárias de S. mansoni; não se verificou, também, infecção natural de roedores. Em agosto de 1975, houve introdução acidental de desovas da B. glabrata, cujos adultos, em 1976, apresentaram infecção por S. mansoni em três ocasiões, com índices de 2,0; 1,6 e 0,8%. No mesmo ano de 1976, dois Holochilus, nascidos no Viveiro, eliminaram ovos viáveis de S. mansoni. Foi possível obter dados de 41 H. b. leucogaster, 28 introduzidos e 14 nascidos no local. O exemplar que sobreviveu mais tempo completou 346 dias. Os animais nascidos no Viveiro e capturados pela primeira vez pesavam, em média, 20 a 50 g. Notou-se que o peso corporal aumentou com o tempo e parece não estacionar até a morte do animal. Z. lasiurus e C. a aperea não procriaram e nem adquiriram infecção ao S. mansoni. O. n. eliurus, procriou e permaneceu vivo, em média, menos de 100 dias; não foi observada eliminação de ovos do parasita. É pouco provável que H. b. leucogaster e B. tenagophila mantenham o ciclo da esquistossomose na ausência da contaminação humana, na natureza. Porém, é possível que, futuramente, H. b. leucogaster na presença de B. glabrata, possa servir de reservatório da esquistossomose, na natureza, quando encontrados em abundância e desde que adaptados com cepas adequadas do parasita.

OBJETIVO: Identificar as espécies de morcegos envolvidas na manutenção do ciclo da raiva, verificar a distribuição do vírus da raiva em tecidos e órgãos de morcegos e os períodos de mortalidade dos camundongos inoculados. MÉTODOS: A positividade para o vírus da raiva foi avaliada por imunofluorescência direta em morcegos de municípios do Estado de São Paulo, de abril de 2002 a novembro de 2003. A distribuição do vírus nos morcegos foi avaliada pela inoculação de camundongos e infecção de células N2A, com suspensões a 20% preparadas a partir de fragmentos de diversos órgãos e tecidos, além de cérebro e glândula salivar. A mortalidade dos camundongos foi observada diariamente, após inoculação intracerebral. RESULTADOS: Dos 4.393 morcegos pesquisados, 1,9% foram positivos para o vírus da raiva, pertencentes a dez gêneros, com predomínio de insetívoros. A média do período máximo de mortalidade dos camundongos pós-inoculação a partir de cérebros e glândulas salivares de morcegos hematófagos foi de 15,33±2,08 dias e 11,33±2,30 dias; insetívoros, 16,45±4,48 dias e 18,91±6,12 dias; e frugívoros, 12,60±2,13 dias e 15,67±4,82 dias, respectivamente. CONCLUSÕES: As espécies infectadas com o vírus da raiva foram: Artibeus lituratus, Artibeus sp., Myotis nigricans, Myotis sp., Eptesicus sp., Lasiurus ega, Lasiurus cinereus, Nyctinomops laticaudatus, Tadarida brasiliensis, Histiotus velatus, Molossus rufus, Eumops sp. e Desmodus rotundus. A pesquisa de vírus em diferentes tecidos e órgãos mostrou-se que os mais apropriados para o isolamento foram cérebro e glândulas salivares.

No período de 1966 a 1982 foram isoladas 861 cepas de Yersinia pestis sendo 471 originadas de material de roedores e outros pequenos mamíferos, 236 de lotes de pulgas, 2 de lotes de Ornithodorus e 152 de seres humanos dos focos pestosos do Nordeste do Brasil. Entre os roedores, a espécie que concorreu para o maior número de isolamentos foi o Zygodontomys lasiurus pixuna que, também, forneceu o maior número de lotes de pulgas naturalmente infectados, principalmente do gênero Polygenis. O isolamento da Yersinia pestis de material proveniente de 13 Municípios do Estado de Pernambuco, 7 do Ceará, 3 da Paraíba, 1 do Piauí e 1 da Bahia, evidencia que o problema da peste nos focos brasileiros é bastante atual e merecedor de atenção. O maior número de cepas isoladas e de localidades afetadas, registradas no Estado de Pernambuco não significa maior incidência da peste no mesmo, mas é conseqüência da pesquisa mais intensa da Yersinia pestis e da existência de laboratórios melhor preparados para o seu diagnóstico neste Estado.

Animal and human rabies samples isolated between 1989 and 2000 were typified by means of a monoclonal antibody panel against the viral nucleoprotein. The panel had been previously established to study the molecular epidemiology of rabies virus in the Americas. Samples were isolated in the Diagnostic Laboratory of the Pasteur Institute and in other rabies diagnostic centers in Brazil. In addition to the fixed virus samples CVS-31/96-IP, preserved in mouse brain, and PV-BHK/97, preserved in cell culture, a total of 330 rabies virus samples were isolated from dogs, cats, cattle, horses, bats, sheep, goat, swine, foxes, marmosets, coati and humans. Six antigenic variants that were compatible with the pre-established monoclonal antibodies panel were defined: numbers 2 (dog), 3 (Desmodus rotundus), 4 (Tadarida brasiliensis), 5 (vampire bat from Venezuela), 6 (Lasiurus cinereus) and Lab (reacted to all used antibodies). Six unknown profiles, not compatible with the panel, were also found. Samples isolated from insectivore bats showed the greatest variability and the most commonly isolated variant was variant-3 (Desmodus rotundus). These findings may be related to the existence of multiple independent transmission cycles, involving different bat species.
