O presente trabalho teve como principal objetivo caracterizar os metabolitos secundários, compostos lipofílicos e compostos mais polares, de três macroalgas existentes na costa portuguesa, nomeadamente da H. elongata, L. ochroleuca e U. pinnatifida, de forma a contribuir para a sua valorização. Foram realizadas extrações Soxhlet com diclorometano para extrair os compostos lipofílicos, enquanto as frações mais polares foram obtidas por extrações convencionais sólido-líquido, usando diferentes misturas de solventes (acetona:H2O e metanol:H2O:AcOH). Os extratos foram analisados por GC-MS e HPLC-MS. Os extratos polares foram ainda avaliados quanto à sua atividade antioxidante e quanto ao teor de fenóis totais (método de Folin-Ciocâlteau) e florotaninos (método DMBA). A fração lipofílica das três macroalgas estudadas é composta principalmente por ácidos gordos, álcoois alifáticos de cadeia longa e esteróis. O ácido hexadecanóico mostrou ser o composto maioritário das três espécies de algas, seguido dos ácidos octadeca-9-enóico e tetradecanóico. O fucosterol foi o esterol mais abundante encontrado para a H. elongata, enquanto que na L. ochroleuca e na U. pinnatifida foi o 24-metilenocolesterol. Os extratos polares obtidos a partir das duas metodologias de extração apresentaram rendimentos de extração elevados, tendo os extratos acetona:H2O apresentado rendimentos de extração superiores (88.73-92.33 %). Estes extratos mostraram ainda teores de fenóis e florotaninos totais mais elevados, com valores entre 524.03-635.69 g EAG/kg de peso seco e 1.48-1.55 g EFG/kg de peso seco, respetivamente. Os extratos de acetona:H2O no ensaio DPPH apresentaram valores de IC50 entre 6.57-7.64 μg/mL. Estes valores, apesar de serem inferiores ao IC50 do ácido ascórbico, são superiores ao determinado para o antioxidante sintético butil-hidroxitolueno (BHT). O extrato da acetona:H2O da L. ochroleuca foi o que apresentou melhor atividade antioxidante, com um IC50 de 6.57 ± 0.71 μg/mL. A análise por HPLC-DAD-MSn não permitiu até ao momento detetar compostos fenólicos nos extratos obtidos. Estes resultados são uma contribuição relevante para a valorização destas espécies de macroalgas como fonte de fitoquímicos valiosos.

Kelp forests dominate temperate and polar rocky coastlines and represent critical marine habitats because they support elevated rates of primary and secondary production and high biodiversity. A major threat to the stability of these ecosystems is the proliferation of non-native species, such as the Japanese kelp Undariapinnatifida (‘Wakame’), which has recently colonised natural habitats in the UK. We quantified the abundance and biomass of U. pinnatifida on a natural rocky reef habitat over 10 months to make comparisons with three native canopy-forming brown algae (Laminaria ochroleuca, Saccharina latissima, and Saccorhiza polyschides). We also examined the biogenic habitat structure provided by, and epibiotic assemblages associated with, U. pinnatifida in comparison to native macroalgae. Surveys conducted within the Plymouth Sound Special Area of Conservation indicated that U. pinnatifida is now a dominant and conspicuous member of kelp-dominated communities on natural substrata. Crucially, U. pinnatifida supported a structurally dissimilar and less diverse epibiotic assemblage than the native perennial kelp species. However, U. pinnatifida-associated assemblages were similar to those associated with Saccorhiza polyschides, which has a similar life history and growth strategy. Our results suggest that a shift towards U. pinnatifida dominated reefs could result in impoverished epibiotic assemblages and lower local biodiversity, although this could be offset, to some extent, by the climate-driven proliferation of L. ochroleuca at the poleward range edge, which provides complex biogenic habitat and harbours relatively high biodiversity. Clearly, greater understanding of the long-term dynamics and competitive interactions between these habitat-forming species is needed to accurately predict future biodiversity patterns.

The North-east Atlantic storm season of 2013–14 was unusually severe; the south coast of the UK was subjected to 6 of the 12 most intense storms recorded in the past 5 years. Inshore significant wave heights and periods exceeded 7 m and 13 s with two storms classified as ‘1-in-30 year’ events. We examined the impacts of the storm season on kelp canopies at three study sites. Monospecific canopies comprising Laminaria hyperborea were unaffected by storm disturbance. However, at one study site a mixed canopy comprising Laminaria ochroleuca, Saccharina latissima and L. hyperborea was significantly altered by the storms, due to decreased abundances of the former two species. Quantification of freshly severed stipes suggested that the ‘warm water’ kelp L. ochroleuca was more susceptible to storm damage than L. hyperborea. Overall, kelp canopies were highly resistant to storm disturbance because of the low vulnerability of L. hyperborea to intense wave action. However, if climate-driven shifts in kelp species distributions result in more mixed canopies, as predicted, then resistance to storm disturbance may be eroded.

The European Nature Information System (EUNIS) has been implemented for the establishment of a marine European habitats inventory. Its hierarchical classification is defined and relies on environmental variables which primarily constrain biological communities (e.g. substrate types, sea energy level, depth and light penetration). The EUNIS habitat classification scheme relies on thresholds (e.g. fraction of light and energy) which are based on expert judgment or on the empirical analysis of the above environmental data. The present paper proposes to establish and validate an appropriate threshold for energy classes (high, moderate and low) and for subtidal biological zonation (infralittoral and circalittoral) suitable for EUNIS habitat classification of the Western Iberian coast. Kineticwave-induced energy and the fraction of photosynthetically available light exerted on the marine bottom were respectively assigned to the presence of kelp (Saccorhiza polyschides, Laminaria hyperborea and Laminaria ochroleuca) and seaweed species in general. Both data were statistically described, ordered fromthe largest to the smallest and percentile analyseswere independently performed. The threshold between infralittoral and circalittoral was based on the first quartile while the ‘moderate energy’ class was established between the 12.5 and 87.5 percentiles. To avoid data dependence on sampling locations and assess the confidence interval a bootstrap technique was applied. According to this analysis,more than 75% of seaweeds are present at locations where more than 3.65% of the surface light reaches the sea bottom. The range of energy levels estimated using S. polyschides data, indicate that on the IberianWest coast the ‘moderate energy’ areas are between 0.00303 and 0.04385 N/m2 of wave-induced energy. The lack of agreement between different studies in different regions of Europe suggests the need for more standardization in the future. However, the obtained thresholds in the present study will be very useful in the near future to implement and establish the Iberian EUNIS habitats inventory.

The co-cultivation of fungi has recently been described as a promising strategy to induce the production of novel metabolites through possible gene activation. A large screening of fungal co-cultures in solid media has identified an unusual long-distance growth inhibition between Trichophyton rubrum and Bionectria ochroleuca. To study metabolite induction in this particular fungal interaction, differential LC-MS-based metabolomics was performed on pure strain cultures and on their co-cultures. The comparison of the resulting fingerprints highlighted five de novo induced compounds, which were purified using software-oriented semipreparative HPLC-MS. One metabolite was successfully identified as 4″-hydroxysulfoxy-2,2″-dimethylthielavin P (a substituted trimer of 3,5-dimethylorsellinic acid). The nonsulfated form, as well as three other related compounds, were found in the pure strain culture of B. ochroleuca.

The biological control of the plants diseases, caused by fungi, is carried out using others organisms (predator, parasite or pathogen). Among the possible agents of biocontrol, a fungus has been highlighting as promising and it is known as Clonostachys rosea, asexual form of Bionectria ochroleuca. For that, it is necessary the in vitro production of spores of this fungus. In this study were tested several culture media to select those with better conidia production. The study was conducted at Plant Protection Division, in the Plant Production Department, FCA - UNESP, Botucatu campus, São Paulo state, Brazil. We used the isolated CCR64 (Empresa Brasileira de Pesquisa Agropecuária (EMBRAPA)-CNPMA). The means of crops were: BDA; Oats-Agar; Mazeina-Agar; Rice-Agar; V8-5%; V8-10%; V8-20%; TJ-5 %; TJ-10, TJ-20%. The sporulation of the fungus in different culture media was estimated at 8 days after of the incubation. The data were analyzed using method of comparing averages, using the Tukey test, at 5% probability, and the data processed, using (X + 1) 0.5 transformation. All culture media tested were able to produce conidia. It was found that the best culture media for production of conidia of Bionectria ochroleuca is the TJ-5%, followed by TJ-20%, with an sporulation average of 3,5 x 106 conidia / ml.

Our objective for this study was to evaluate the influence of preindustrial and expected future atmospheric CO2 concentrations (280 µatm and 700 µatm pCO2, respectively) on different life-cycle stages of the kelp Laminaria hyperborea from Helgoland (Germany, North Sea). Zoospore germination, gametogenesis, vegetative growth, sorus formation and photosynthetic performance of vegetative and fertile tissue were examined. The contribution of external carbonic anhydrase (exCA) to C-supply for net-photosynthesis (net-PS) and the Chla- and phlorotannin content were investigated. Female gametogenesis and vegetative growth of sporophytes were significantly enhanced under the expected future pCO2. rETR(max) and net-PS of young vegetative sporophytes tended to increase performance at higher pCO2. The trend towards elevated net-PS vanished after inhibition of exCA. In vegetative sporophytes, phlorotannin content and Chla content were not significantly affected by pCO2.
