Guava (Psidium guajava L.) is a plant often employed in popular medicine. Recently several studies have alerted about the toxicity of substances present in medicinal plants, which can pose risks to the human health. In this sense, the present work aimed to investigate the phytotoxic, cytotoxic and genotoxic action of three guava varieties - Paluma, Pedro Sato and Roxa (purple) - on the plant test system Lactuca sativa L. Thus, macro- and microscopic evaluations were carried out for five infusion concentrations (2.5, 5.0, 10.0, 20.0 and 40.0 g.L-1) prepared from each variety. Distilled water was used as negative control. Chromatographic and spectroscopic analysis by HPLC-PAD indicated that the chemical composition of the infusion of Roxa is different than that of the infusions of the varieties Paluma and Pedro Sato. It was observed that seed germination and root growth in L. sativa exposed to infusions decreased with increasing infusion concentration, regardless of the tested cultivar. For the mitotic index, no statistical differences were observed. On the other hand, a significant increase in the frequency of cell cycle alterations was verified, especially for the highest concentrations tested. The cytogenotoxic was significant. Therefore, guava should not be used indiscriminately in popular medicine.

Objetivou-se avaliar os efeitos de extratos de quatro espécies de adubos verdes (Canavalia ensiformis, Crotalaria spectabilis, Mucuna aterrimae Sorghum bicolor cv. AG 2005) sobre a germinação de sementes e o crescimento inicial de plântulas deLactuca sativa e de Digitaria horizontalis. Foram realizados dois experimentos: no primeiro, avaliou-se a aplicação das soluções diluídas a 1%, 2%, 3%, 4%, 5% e10% de extratos aquosos da parte aérea e das raízes das espécies de adubos verdes sobre a germinação, índice de velocidade de germinação e o comprimento radicular de L. sativa; no segundo, avaliou-se a incorporação ao solo de 0, 10, 20, 30, 40 e 50 g dm-3 da biomassa dos adubos verdes sobre o acúmulo de massa seca (parte aérea e raízes) e o desenvolvimento radicular de plântulas de L. sativa e de D. horizontalis. Concluiu-se que os extratos aquosos das espécies C. ensiformis, C. spectabilis, M. aterrima e S. bicolor cv. AG 2005 promoveram efeitos negativos sobre a germinação e o IVG das sementes de L. sativa. A incorporação ao solo da biomassa dessas espécies de adubos verdes influenciou negativamente no desenvolvimento inicial das plântulas de L. sativa. Entretanto, verificou-se efeito positivo sobre o desenvolvimento inicial de D. horizontalis com a incorporação ao solo da biomassa de C. spectabilis.

Balimau Putih [an Indonesian cultivar tolerant to rice tungro bacilliform virus (RTBV)] was crossed with IR64 (RTBV, susceptible variety) to produce the three filial generations F1, F2 and F3. Agroinoculation was used to introduce RTBV into the test plants. RTBV tolerance was based on the RTBV level in plants by analysis of coat protein using enzyme-linked immunosorbent assay. The level of RTBV in cv. Balimau Putih was significantly lower than that of IR64 and the susceptible control, Taichung Native 1. Mean RTBV levels of the F1, F2 and F3 populations were comparable with one another and with the average of the parents. Results indicate that there was no dominance and an additive gene action may control the expression of tolerance to RTBV. Tolerance based on the level of RTBV coat protein was highly heritable (0.67) as estimated using the mean values of F3 lines, suggesting that selection for tolerance to RTBV can be performed in the early selfing generations using the technique employed in this study. The RTBV level had a negative correlation with plant height, but positive relationship with disease index value

Following microprojectile mediated delivery of a plasmid construct (pAHC-25) encoding bar (bialophos resistance) gene into five-day-old scutellar calli derived from mature embryos, the effectiveness of selection procedure for bar-gene expressing tissue was compared for two indica rice cultivars (IR-64 and Karnal Local). While IR-64 transformants could be selected through the generally used semi-solid selection medium, the same procedure was not effective in the basmati cultivar Karnal Local. In the latter case, while lower concentrations (2–4 mg 1−1) of the selective agent phosphinothricin (PPT) yielded only escapes, higher concentrations (6–8 mg l−1) inhibited proliferation of transformed as well as untransformed sectors. For Karnal Local, a liquid medium based selection system was successfully utilized for recovering transformed sectors and, eventually, regenerants. The study demonstrates the generation of transformants of two elite indica cultivars using the environment-independent system of mature embryos from seeds.

We have mapped and identified DNA markers linked to morphology, yield, and yield components of lucerne, using a backcross population derived from winter-active parents. The high-yielding and recurrent parent, D, produced individual markers that accounted for up to 18% of total yield over 6 harvests, at Gatton, south-eastern Queensland. The same marker, AC/TT8, was consistently identified at each individual harvest, and in individual harvests accounted for up to 26% of the phenotypic variation for yield. This marker was located in linkage group 2 of the D map, and several other markers positively associated with yield were consistently identified in this linkage group. Similarly, markers negatively associated with yield were consistently identified in the W116 map, W116 being the low-yielding parent. Highly significant positive correlations were observed between total yield and yield for harvests 1-6, and between total yield and stem length, tiller number, leaf yield/plant, leaf yield/5 stems, stem yield/plant, and stem yield/5 stems. Highly significant QTL were located for all these characters as well as for leaf shape and pubescence.

Flax and hemp have traditionally been used mainly for textiles, but recently interest has also been focused on non-textile applications. Microbial quality throughout the whole processing chain of bast fibres has not previously been studied. This study concentrates on the microbial quality and possible microbial risks in the production chain of hemp and flax fibres and fibrous thermal insulations. In order to be able to utilize hemp and flax fibres, the bast fibres must be separated from the rest of the plant. Non-cellulosic components can be removed with various pretreatment processes, which are associated with a certain risk of microbial contamination. In this study enzymatic retting and steam explosion (STEX) were examined as pretreatment processes. On the basis of the results obtained in this study, the microbial contents on stalks of both plants studied increased at the end of the growing season and during the winter. However, by processing and mechanical separation it is possible to produce fibres containing less moulds and bacteria than the whole stem. Enzymatic treatment encouraged the growth of moulds in fibres. Steam explosion reduced the amount of moulds in fibres. Dry thermal treatment used in this study did not markedly reduce the amount of microbes. In this project an emission measurement chamber was developed which was suitable for measurements of emissions from both mat type and loose fill type insulations, and capable of interdisciplinary sampling. In this study, the highest amounts of fungal emissions were in the range of 10^3 10^5 cfu/m^3 from the flax and hemp insulations at 90% RH of air. The fungal emissions from stone wool, glass wool and recycled paper insulations were below 10^2 cfu/m^3 even at 90% RH. Equally low values were obtained from bast fibrous materials in lower humidities (at 30% and 80% RH of air). After drying of moulded insulations at 30% RH, the amounts of emitted moulds were in all cases higher compared to the emissions at 90% RH before drying. The most common fungi in bast fibres were Penicillium and Rhizopus. The widest variety of different fungi was in the untreated hemp and linseed fibres and in the commercial loose-fill flax insulation. Penicillium, Rhizopus and Paecilomyces were the most tolerant to steam explosion. According to the literature, the most common fungi in building materials and indoor air are Penicillium, Aspergillus and Cladosporium, which were all found in some of the bast fibre materials in this study. As organic materials, hemp and flax fibres contain high levels of nutrients for microbial growth. The amount of microbes can be controlled and somewhat decreased by the processing methods presented.

To break the yield ceiling of rice production, a super rice project was developed in 1996 to breed rice varieties with super high yield. A two-year experiment was conducted to evaluate yield and nitrogen (N)-use response of super rice to different planting methods in the single cropping season. A total of 17 rice varieties, including 13 super rice and four non-super checks (CK), were grown under three N levels [0 (N0), 150 (N150), and 225 (N225) kg ha−1] and two planting methods [transplanting (TP) and direct-seeding in wet conditions (WDS)]. Grain yield under WDS (7.69 t ha−1) was generally lower than TP (8.58 t ha−1). However, grain yield under different planting methods was affected by N rates as well as variety groups. In both years, there was no difference in grain yield between super and CK varieties at N150, irrespective of planting methods. However, grain yield difference was dramatic in japonica groups at N225, that is, there was an 11.3% and 14.1% average increase in super rice than in CK varieties in WDS and TP, respectively. This suggests that high N input contributes to narrowing the yield gap in super rice varieties, which also indicates that super rice was bred for high fertility conditions. In the japonica group, more N was accumulated in super rice than in CK at N225, but no difference was found between super and CK varieties at N0 and N150. Similar results were also found for N agronomic efficiency. The results suggest that super rice varieties have an advantage for N-use efficiency when high N is applied. The response of super rice was greater under TP than under WDS. The results suggest that the need to further improve agronomic and other management practices to achieve high yield and N-use efficiency for super rice varieties in WDS.

A field study was established to evaluate oxadiargyl and pendimethalin during the wet seasons in Bangladesh in 2012 and 2013. The study evaluated the following treatments: oxadiargyl applied at 80, 120, and 160 g ai ha−1; pendimethalin at 800, 1200, and 1600 g ai ha−1; partial weedy; and weed-free. Rice plant density was greatly affected by weed control treatment. Lower density and lower uniformity of the rice plant stand occurred as a result of increased rates of herbicides. Increased rates of pendimethalin were more toxic than increased rates of oxadiargyl. Both herbicides effectively controlled Digitaria ciliaris, Echinochloa colona, and Phyllanthus niruri; however, they were unable to control Murdannia nudiflora. Oxadiargyl controlled Cyperus rotundus across rates by 31–55%, but pendimethalin was completely ineffective on it, and higher rates of both herbicides had no effect in controlling this weed. Both herbicides at higher rates reduced total weed biomass significantly. Among herbicide treatments, the highest yield (3.7–4.0 t ha−1) was recorded in plots treated with oxadiargyl at 160 g ai ha−1 and the lowest yield (2.4–2.8 t ha−1) was in plots treated with pendimethalin at 1600 g ai ha−1. Results from our study suggest that a higher rate of oxadiargyl can increase yield by suppressing weeds in dry-seeded rice systems. Similar to the results of oxadiargyl, pendimethalin at higher rates also greatly suppressed weeds; however, yield decreased due to phytotoxicity to rice seedlings.

An investigation was conducted to study the levels of nitrogen fixation on the leaf or sheath surfaces of four cultivars of paddy plants by using acetylene reduction technique. Varying levels of positive nitrogenase activity were observed on all the leaf surfaces. Sheath of IET 1991 cultivar showed a higher rate of fixation than the leaf surface. All the nitrogen-fixing organisms on the leaf or sheath surfaces belonged to the genus Beijerinckia. There was no correlation between the bacterial density and the level of fixation. Scanning electron microscopic data revealed that the upper surface of IET 1991 leaf was highly silicified and the microflora was either scanty or nil while the lower surface appeared quite different and harboured more micro-organisms. Similarly, the inner surface of sheath was devoid of silicification and showed the presence of micro-organisms.

Sea level rise (SLR) is a primary factor responsible for inundation of low-lying coastal regions across the world, which in turn governs the agricultural productivity. In this study, rice (Oryza sativa L.) cultivated seasonally in the Kuttanad Wetland, a SLR prone region on the southwest coast of India, were analysed for oxygen, hydrogen and carbon isotopic ratios (delta O-18, delta H-2 and delta C-13) to distinguish the seasonal environmental conditions prevalent during rice cultivation. The region receives high rainfall during the wet season which promotes large supply of fresh water to the local water bodies via the rivers. In contrast, during the dry season reduced river discharge favours sea water incursion which adversely affects the rice cultivation. The water for rice cultivation is derived from regional water bodies that are characterised by seasonal salinity variation which co-varies with the delta O-18 and delta H-2 values. Rice cultivated during the wet and the dry season bears the isotopic imprints of this water. We explored the utility of a mechanistic model to quantify the contribution of two prominent factors, namely relative humidity and source water composition in governing the seasonal variation in oxygen isotopic composition of rice grain OM. delta C-13 values of rice grain OM were used to deduce the stress level by estimating the intrinsic water use efficiency (WUEi) of the crop during the two seasons. 1.3 times higher WUE, was exhibited by the same genotype during the dry season. The approach can be extended to other low lying coastal agro-ecosystems to infer the growth conditions of cultivated crops and can further be utilised for retrieving paleo-environmental information from well preserved archaeological plant remains. Con el objeto de evaluar el comportamiento adaptativo de cuatro líneas de alfalfa (Medicago sativa L.) en condiciones de campo y de vivero, se realizó un estudio en la Hacienda Las Mercedes, propiedad de la Universidad Nacional Agraria, ubicada en la ciudad de Managua Km 11 carretera norte, , entrada al CARNIC 2 km al lago. Teniendo su ubicación geográfica en un cuadrante con las siguientes coordenadas: 12°10'14"a 12"08'05" en latitud Norte y 86.10'22" a 86"09'44" longitud Oeste. El estudio se realizó en dos fuses 1) de campo y 2) en vivero. En ambos se determinó el grado de adaptación de cuatro líneas de alfalfa (Medicago sativa L.), tres procedentes de Texas-EE UU (8L418, 105916 y 9818) y una de Sébaco-Nicaragua (l3-A50) donde se ha establecido por más de tres años. El ensayo de campo se estableció en un área que anteriormente fue utilizada para la siembra de sorgo forrajero y el de vivero se realizó en el vivero de la UNA, en la misma finca. Se consideró cada una de las líneas como tratamiento. El Diseño experimental usado para ambas fases fue de bloques completos al azar (BCA), con 4 repeticiones. En campo con parcelas experimentales fueron de 4 m2 (2 m x 2 m), para un área total de 120m2 Se sembraron 6 surcos a una distancia de 30 cm entre surco y 14 plantas por surcos distanciados a 15 cm. Se realizó análisis de varianza utilizando programa SAS versión 99, cuando se encontró diferencias significativas o altamente significativas para tratamientos se realizaron pruebas de medias según Duncan. El terreno se preparó de forma convencional, con una chapea inicial, un paso de arado y gradeo de forma mecanizada, posteriormente se realizó la estructuración del diseño de campo. Las variables evaluadas según las condiciones de campo fueron: germinación, altura de la plantas, daños por plaga y enfermedades y ramificación, en las condiciones de vivero fueron: germinación, sobrevivencia, altura (cm), daños por plagas y enfermedades. Como resultado se obtuvo que 3 de las líneas presentaron buena germinación en condiciones de campo y vivero siendo la de mejor comportamiento la línea 13A-50 con un promedio del 97%. Para altura la línea 9818 presentó el mejor comportamiento en condiciones de campo con rangos de 48cm - 58 cm manteniendo superioridad durante el estudio en comparación con el resto de las líneas evaluadas. En daños por plagas la línea 8L418 la de menor afectación, y la más afectada fue la línea l3A-50. En daños por enfermedades la línea 9818 obtuvo los mayores daños en los niveles de moderado a muy grave y la línea l3A-50 fue la de menor incidencia. Todas las líneas presentaron una ramificación media de 30%, siendo la línea 105916 la de mejor comportamiento. En vivero la línea l3A-50 presentó la mejor altura. La línea 9818 fue la de mejor adaptabilidad en condiciones de campo, seguida de la línea BA-50. Pero en condiciones de vivero la línea 13A-50 fue la de mejor adaptabilidad, seguida de la línea 8L418.en resumen la línea l3A-50 fue la de mejor comportamiento adaptativo.
