The dorsal striatum (DS) is involved in various forms of learning and memory such as procedural learning, habit learning, reward-association and emotional learning. We have previously reported that bilateral DS lesions disrupt tone fear conditioning (TFC), but not contextual fear conditioning (CFC) [Ferreira TL, Moreira KM, Ikeda DC, Bueno OFA, Oliveira MGM (2003) Effects of dorsal striatum lesions in tone fear conditioning and contextual fear conditioning. Brain Res 987:17-24]. To further elucidate the participation of DS in emotional learning, in the present study, we investigated the effects of bilateral pretest (postraining) electrolytic DS lesions on TFC. Given the well-acknowledged role of the amygdala in emotional learning, we also examined a possible cooperation between DS and the amygdala in TFC, by using asymmetrical electrolytic lesions, consisting of a unilateral lesion of the central amygdaloid nucleus (CeA) combined to a contralateral DS lesion. The results show that pre-test bilateral DS lesions disrupt TFC responses, suggesting that DS plays a role in the expression of TFC. More importantly, rats with asymmetrical pre-training lesions were impaired in TFC, but not in CFC tasks. This result was confirmed with muscimol asymmetrical microinjections in DS and CeA, which reversibly inactivate these structures. On the other hand, similar pretest lesions as well as unilateral electrolytic lesions of CeA and DS in the same hemisphere did not affect TFC. Possible anatomical substrates underlying the observed effects are proposed. Overall, the present results underscore that other routes, aside from the well-established CeA projections to the periaqueductal gray, may contribute to the acquisition/consolidation of the freezing response associated to a TFC task. It is suggested that CeA may presumably influence DS processing via a synaptic relay on dopaminergic neurons of the substantia nigra compacta and retrorubral nucleus. The present observations are also in line with other studies showing that TFC and CFC responses are mediated by different anatomical networks. (C) 2008 IBRO. Published by Elsevier Ltd. The ventral tegmental area (VTA) is a nodal link in reward circuitry. Based on its striatal output, it has been subdivided in a caudomedial part which targets the ventromedial striatum, and a lateral part which targets the ventrolateral striatum [Ikemoto S (2007) Dopamine reward circuitry: two projection systems from the ventral midbrain to the nucleus accumbens-olfactory tubercle complex. Brain Res Rev 56:27-78]. Whether these two VTA parts are interconnected and to what extent the VTA innervates the substantia nigra compacta (SNc) and retrorubral nucleus (RR) are critical issues for understanding information processing in the basal ganglia. Here, VTA projections to the VTA-nigral complex were examined in rats, using Phaseolus vulgaris leucoagglutinin (PHA-L) as anterograde tracer. The results show that the dorsolateral VTA projects to itself, as well as to the dorsal tier of the SNc and RR, largely avoiding the caudomedial VTA. The ventrolateral VTA innervates mainly the interfascicular nucleus. The components of the caudomedial VTA (the interfascicular, paranigral and caudal linear nuclei) are connected with each other. In addition, the caudomedial VTA (especially the paranigral and caudal linear nuclei) innervates the lateral VTA, and, to a lesser degree, the SNc and RR. The caudal pole of the VTA sends robust, bilateral projections to virtually all the VTA-nigral complex, which terminate in the dorsal and ventral tiers. Modest inputs from the medial supramammillary nucleus to ventromedial parts of the VTA-nigral complex were also identified. In double-immunostained sections, PHA-L-labeled varicosities were sometimes found apposed to tyrosine hydroxylase-positive neurons in the ventral mesencephalon. Overall, the results underscore that VTA projections to the VTA-nigral complex are substantial and topically organized. In general, these projections, like the spiralated striato-nigro-striatal loops, display a medial-to-lateral organization. This anatomical arrangement conceivably permits the ventromedial striatum to influence the activity of the lateral striatum. The mesopontine rostromedial tegmental nucleus (RMTg) is a mostly ?-aminobutyric acid (GABA)ergic structure believed to be a node for signaling aversive events to dopamine (DA) neurons in the ventral tegmental area (VTA). The RMTg receives glutamatergic inputs from the lateral habenula (LHb) and sends substantial GABAergic projections to the VTA, which also receives direct projections from the LHb. To further specify the topography of LHb projections to the RMTg and VTA, small focal injections of the anterograde tracer Phaseolus vulgaris leucoagglutinin were aimed at different subdivisions of the LHb. The subnuclear origin of LHb inputs to the VTA and RMTg was then confirmed by injections of the retrograde tracer cholera toxin subunit b into the VTA or RMTg. Furthermore, we compared the topographic position of retrogradely labeled neurons in the RMTg resulting from VTA injections with that of anterogradely labeled axons emerging from the LHb. As revealed by anterograde and retrograde tracing, LHb projections were organized in a strikingly topographic manner, with inputs to the RMTg mostly arising from the lateral division of the LHb (LHbL), whereas inputs to the VTA mainly emerged from the medial division of the LHb (LHbM). In the RMTg, profusely branched LHb axons were found in close register with VTA projecting neurons and were frequently apposed to the latter. Overall, our findings demonstrate that LHb inputs to the RMTg and VTA arise from different divisions of the LHb and provide direct evidence for a disynaptic pathway that links the LHbL to the VTA via the RMTg. As implicações ambientais dos plásticos convencionais, produzidos através de combustíveis fósseis, não permitem a sua utilização de forma sustentável pela sociedade devido aos seus problemas de degradabilidade e à sua dependência de matérias-primas não renováveis. Surgem então os polihidroxialcanoatos (PHA) como uma alternativa ambientalmente sustentável, já que conseguem reproduzir grande parte das aplicações dos plásticos ditos normais, tendo as grandes vantagens de serem biodegradáveis e produzidos a partir de uma fonte de carbono renovável. Mais recentemente, de forma a tornar a produção do PHA mais sustentável, tem-se desenvolvido o conceito de associar a produção destes polímeros ao tratamento de efluentes líquidos através do processo de lamas ativadas. Apesar destas vantagens todo o processo que envolve a produção do PHA é ainda dispendioso. Esta dissertação insere-se na estratégia de tentar minorar esses custos. Uma das formas de o fazer será na otimização de técnicas de quantificação do PHA em culturas mistas. Neste trabalho o principal objetivo reside na avaliação da capacidade da metodologia de análise de imagem, acoplada a microscopia de fluorescência, como método de quantificação de PHA. Pretende-se também explorar a produção de PHA a partir da simulação de um sistema de tratamento de águas residuais implementado num reator descontínuo sequencial (SBR) em regimes cíclicos de feast/famine, aliando assim a função de tratamento de efluentes líquidos com a produção de um produto de valor acrescentado como é o PHA. Deste modo, foi montada uma instalação com um sistema de lamas ativadas num SBR cujo principal objetivo era produzir PHA. Este objetivo de produzir PHA através do regime de alimentação dinâmica aeróbia com uma alimentação com acetato como fonte de carbono, e sem controlo do pH foi conseguido, atingindo-se um máximo de concentração mássica de 0,37 mg PHA/mg SST e de concentração volumétrica de 1785 mg PHA/L. Em relação à capacidade de tratamento, comprovou-se a adequabilidade do sistema utilizado para a remoção da carga orgânica devido a uma elevada captação de nutrientes do afluente pela biomassa. Foram encontradas algumas limitações na metodologia de análise de imagem ao nível da transformação da bidimensionalidade da área projetada dos grânulos de PHA nas imagens para a sua tridimensionalidade real, na gama de concentração de PHA na qual a metodologia é válida e na amostragem relativamente curta. Contudo, foi possível comprovar a potencialidade deste método obtendo-se um R2 de 0,80 para o modelo de previsão da concentração mássica de PHA. Deste modo, pode-se concluir que esta é uma metodologia promissora para quantificar PHA em amostras de licor misto.

Previous studies using morphine-treated dams reported a role for the rostral lateral periaqueductal gray (rIPAG) in the behavioral switching between nursing and insect hunting, likely to depend on an enhanced seeking response to the presence of an appetitive rewarding cue (i.e., the roach). To elucidate the neural mechanisms mediating such responses, in the present study, we first observed how the rIPAG influences predatory hunting in male rats. Our behavioral observations indicated that bilateral rIPAG NMDA lesions dramatically interfere with prey hunting, leaving the animal without chasing or attacking the prey, but do not seem to affect the general levels of arousal, locomotor activity and regular feeding. Next, using Phaseolus vulgaris-leucoagglutinin (PHA-L), we have reviewed the rIPAG connection pattern, and pointed out a particularly dense projection to the hypothalamic orexinergic cell group. Double labeled PHA-L and orexin sections showed an extensive overlap between PHA-L labeled fibers and orexin cells, revealing that both the medial/perifornical and lateral hypothalamic orexinergic cell groups receive a substantial innervation from the rIPAG. We have further observed that both the medial/perifornical and lateral hypothalamic orexinergic cell groups up-regulate Fos expression during prey hunting, and that rIPAG lesions blunted this Fos increase only in the lateral hypothalamic, but not in the medial/perifornical, orexinergic group, a finding supposedly associated with the lack of motivational drive to actively pursue the prey. Overall, the present results suggest that the rIPAG should exert a critical influence on reward seeking by activating the lateral hypothalamic orexinergic cell group. (C) 2011 Elsevier B.V. We showed that when CA3 pyramidal neurons in the caudal 80% of the dorsal hippocampus had almost disappeared completely, the efferent pathway of CA3 was rarely detectable. We used the mouse pilocarpine model of temporal lobe epilepsy (TLE), and injected iontophoretically the anterograde tracer phaseolus vulgaris leucoagglutinin (PHA-L) into gliotic CA3, medial septum and the nucleus of diagonal band of Broca, median raphe, and lateral supramammillary nuclei, or the retrograde tracer cholera toxin B subunit (CTB) into gliotic CA3 area of hippocampus. In the afferent pathway, the number of neurons projecting to CA3 from medial septum and the nucleus of diagonal band of Broca, median raphe, and lateral supramammillary nuclei increased significantly. In the hippocampus, where CA3 pyramidal neurons were partially lost, calbindin, calretinin, parvalbumin immunopositive back-projection neurons from CA1-CA3 area were observed. Sprouting of Schaffer collaterals with increased number of large boutons in both sides of CA1 area, particularly in the stratum pyramidale, was found. When CA3 pyramidal neurons in caudal 80% of the dorsal hippocampus have almost disappeared completely, surviving CA3 neurons in the rostral 20% of the dorsal hippocampus may play an important role in transmitting hyperactivity of granule cells to surviving CA1 neurons or to dorsal part of the lateral septum. We concluded that reorganization of CA3 area with its downstream or upstream nuclei may be involved in the occurrence of epilepsy.

Cysteine is a thiol containing amino acid that readily undergoes oxidation by reactive oxygen species (ROS) to form sulphenic (R-SOH) sulphinic (RSO2H) and sulphonic (RSO3H) acids. Thiol modifications of cysteine have been implicated as modulators of cellular processes and represent significant biological modifications that occur during oxidative stress and cell signalling. However, the different oxidation states are difficult to monitor in a physiological setting due to the limited availability of experimental tools. Therefore it is of interest to synthesise and use a chemical probe that selectively recognises the reversible oxidation state of cysteine sulphenic acid to understand more about oxidative signalling. The aim of this thesis was to investigate a synthetic approach for novel fluorescent probe synthesis, for the specific detection of cysteine sulphenic acids by fluorescence spectroscopy and confocal microscopy. N-[2-(Anthracen-2-ylamino)-2-oxoethyl]-3,5-dioxocyclohexanecarboxamide was synthesised in a multistep synthesis and characterised by nuclear magnetic resonance spectroscopy. The optimisation of conditions needed for sulphenic acid formation in a purified protein using human serum albumin (HSA) and the commercially available biotin tagged probe 3-(2,4-dioxocyclohexyl)propyl-5-((3aR,6S,6aS)-hexahydro-2-oxo-1H-thieno[3,4-d]imidazol-6-yl)pentanoate (DCP-Bio1) were identified. This approach was extended to detect sulphenic acids in Jurkat T cells and CD4+ T cells pre- and post-stimulus. Buthionine sulfoximine (BSO) was used to manipulate the endogenous antioxidant glutathione (GSH) in human CD4+ T cells. Then the surface protein thiol levels and sulphenic acid formation was examined. T cells were also activated by the lectin phytohaemagglutinin-L (PHA-L) and formation of sulphenic acid was investigated using SDS-PAGE, western blotting and confocal microscopy. Resting Jurkat cells have two prominent protein bands that have sulphenic acid modifications whereas resting CD4+ T cells have an additional band present. When cells were treated with BSO the number of bands increased whereas activation reduced the number of proteins that were modified. The identities of the protein bands containing sulphenic acids were explored by mass spectrometry. Cysteine oxidation was observed in redox, metabolic and cytoskeletal proteins. In summary, a novel fluorescent probe for detection of cysteine sulphenic acids has been synthesised alongside a model system that introduces cysteine sulphenic acid in primary T cells. This probe has potential application in the subcellular localisation of cysteine oxidation during T cell signalling.

Plusieurs agents anticancéreux très puissants sont caractérisés par une solubilité aqueuse limitée et une toxicité systémique importante. Cette dernière serait liée d’une part à la solubilisation des agents anticancéreux à l’aide de surfactifs de bas poids moléculaire, connus pour leur toxicité intrinsèque, et d’autre part, par le manque de spécificité tissulaire des anticancéreux. Les vecteurs colloïdaux à base de polymères permettraient de résoudre certains défis liés à la formulation d’agents anticancéreux hydrophobes. D’abord, les polymères peuvent être sélectionnés afin de répondre à des critères précis de compatibilité, de dégradation et d’affinité pour le médicament à formuler. Ensuite, le fait d’encapsuler l’agent anticancéreux dans un vecteur peut améliorer son efficacité thérapeutique en favorisant son accumulation au niveau du tissu cible, i.e. la tumeur, et ainsi limiter sa distribution au niveau des tissus sains. Des travaux antérieurs menés au sein de notre laboratoire ont mené à la mise au point de micelles à base de poly(N-vinyl-pyrrolidone)-bloc-poly(D,L-lactide) (PVP-b-PDLLA) capables de solubiliser des agents anticancéreux faiblement hydrosolubles dont le PTX. Ce dernier est commercialisé sous le nom de Taxol® et formulé à l’aide du Crémophor EL (CrEL), un surfactif de bas poids moléculaire pouvant provoquer, entre autres, des réactions d’hypersensibilité sévères. Bien que les micelles de PVP-b-PDLLA chargées de PTX aient démontré une meilleure tolérance comparée au Taxol®, leur potentiel de ciblage tumoral et leur efficacité thérapeutique étaient similaires à la forme commerciale à doses égales. Ceci était possiblement dû au fait que les micelles étaient rapidement déstabilisées et ne pouvaient retenir leur cargo suite à leur administration intraveineuse. Nous avons donc décidé de poursuivre les travaux avec un autre type de vecteur, soit des nanoparticules, qui possèdent une stabilité intrinsèque supérieure aux micelles. L’objectif principal de cette thèse de doctorat était donc de mettre au point des nanoparticules polymères pour l’administration parentérale d’agents anticancéreux faiblement solubles dans l’eau. Les nanoparticules devaient permettre d’encapsuler des agents anticancéreux hydrophobes et de les libérer de manière contrôlée sur plusieurs jours. De plus, elles devaient démontrer un temps de circulation plasmatique prolongée afin de favoriser l’accumulation passive du médicament encapsulé au niveau de la tumeur. La première partie du travail visait à employer pour la première fois le copolymère amphiphile PVP-b-PDLLA comme émulsifiant dans la préparation de nanoparticules polymères. Ainsi, une méthode de fabrication des nanoparticules par émulsion huile-dans-eau a été appliquée afin de produire des nanoparticules à base de PDLLA de taille inférieure à 250 nm. Grâce aux propriétés lyoprotectrices de la couronne de PVP présente à la surface des nanoparticules, celles-ci pouvaient retrouver leur distribution de taille initiale après lyophilisation et redispersion en milieu aqueux. Deux anticancéreux hydrophobes, soit le PTX et l’étoposide (ETO), ont été encapsulés dans les nanoparticules et libérés de ces dernières de façon contrôlée sur plusieurs jours in vitro. Une procédure de « salting-out » a été appliquée afin d’améliorer le taux d’incorporation de l’ETO initialement faible étant donnée sa solubilité aqueuse légèrement supérieure à celle du PTX. Le second volet des travaux visait à comparer le PVP comme polymère de surface des nanoparticules au PEG, le polymère le plus fréquemment employé à cette fin en vectorisation. Par le biais d’études d’adsorption de protéines, de capture par les macrophages et de biodistribution chez le rat, nous avons établi une corrélation in vitro/in vivo démontrant que le PVP n’était pas un agent de surface aussi efficace que le PEG. Ainsi, malgré la présence du PVP à la surface des nanoparticules de PDLLA, ces dernières étaient rapidement éliminées de la circulation sanguine suite à leur capture par le système des phagocytes mononucléés. Par conséquent, dans le troisième volet de cette thèse, le PEG a été retenu comme agent de surface, tandis que différents polymères biodégradables de la famille des polyesters, certains synthétiques (PDLLA et copolymères d’acide lactique/acide glycolique), d’autres de source naturelle (poly(hydroxyalkanoates)(PHAs)), ont été investiguées comme matériaux formant le cœur des nanoparticules. Il en est ressorti que les propriétés physicochimiques des polyesters avaient un impact majeur sur l’efficacité d’encapsulation du PTX et son profil de libération des nanoparticules in vitro. Contrairement aux PHAs, les polymères synthétiques ont démontré des taux d’incorporation élevés ainsi qu’une libération contrôlée de leur cargo. Des études de pharmacocinétique et de biodistribution ont démontré que les nanoparticules de PDLLA dotées d’une couronne de PEG conféraient un temps de circulation plasmatique prolongé au PTX et favorisaient son accumulation tumorale. Les nanoparticules polymères représentent donc une alternative intéressante au Taxol®.

La fibrose kystique (FK) est la maladie autosomique récessive la plus fréquente chez les individus de race caucasienne. Elle est secondaire à la mutation du gène Cystic Fibrosis Transmembrane Regulator (CFTR). Grâce à des traitements plus agressifs, la médiane de l’espérance de vie des individus atteints de la FK a augmenté et cette augmentation est associée à l’émergence du diabète secondaire ou associé à la FK (DAFK), une complication associée à une augmentation du taux de mortalité. La pathophysiologie du DAFK n’est pas parfaitement comprise. Par exemple, la cause de l’accélération de la perte de la fonction pulmonaire, qui débute des années avant l’apparition du DAFK, n'est pas élucidée. Tous les patients atteints de la FK, même ceux sans le DAFK, présentent de l’hyperglycémie et des fluctuations glycémiques. D’ailleurs, une étude a démontré que la réactivité immunitaire est affectée par l’hyperglycémie dans un modèle animal de la FK et il y a des évidences que les lymphocytes sans CFTR fonctionnel ou en présence d’un excès de glucose ont des réactions inflammatoires anormales. Donc, nous avons émis l’hypothèse que les patients atteints de la FK, surtout ceux non-diabétiques et pré-diabétiques, auront une plus grande proportion de lymphocytes Th17 et Treg produisant la cytokine pro-inflammatoire IL-17A comparativement aux sujets sains et que l’augmentation de cette cytokine pourrait influencer la chute accélérée des fonctions pulmonaires avant l’apparition du DAFK. Des niveaux élevés d’IL-17A sont retrouvés dans les poumons des patients atteints de la FK et dans le sang périphérique des patients avec le diabète de type 1 (DT1) et de type 2 (DT2). L’IL-17A peut aussi être produite par les lymphocytes Treg dysfonctionnels. Habituellement, ces lymphocytes atténuent les réponses inflammatoires excessives, mais lorsqu’ils sont dysfonctionnels, ils peuvent produire de l’IL-17A, contribuant ainsi à l’état inflammatoire. De plus, nous avons supposé que les proportions de Th17 et Treg produisant de l’IL-17A seront associées aux fonctions pulmonaires des patients atteints de la FK et que l’alimentation, l’activité physique et la composition corporelle influenceraient ces relations. Les résultats de cette thèse ont montré que, malgré une association entre la proportion de lymphocytes dans le sang périphérique et les indices de fluctuations glycémiques, celles-ci n’influençaient pas les proportions de lymphocytes Th17 et Treg produisant de l’IL-17A lorsqu’ils étaient mis en culture pour 24 ou 48 heures dans des milieux contenant soit 5 mM ou 25 mM de glucose et stimulés par le phorbol 12-myristate 13-acetate (PMA) et le phytohemagglutinine (PHA) ou, encore, non stimulés. De plus, ces proportions étaient semblables entre les patients atteints de la FK et les individus en santé. Toutefois, les proportions de lymphocytes Treg stimulés produisant de l’IL-17A des sujets sains étaient plus élevées que les proportions de lymphocytes Treg non stimulés de tous les participants (patients atteints de la FK et individus en santé). Tout ceci suggérant donc que les Treg des sujets sains et atteints de la FK ne réagissaient pas de la même façon à la stimulation. D’ailleurs, la durée d’incubation affectait les proportions de Th17 produisant de l’IL-17A, mais elle n’avait aucun effet sur les proportions de Treg produisant cette cytokine. Donc, ces types cellulaires réagissaient différemment dans les mêmes milieux de culture. De plus, nous avons observé que seulement l’énergie provenant des glucides affectait modestement les indices de fluctuations glycémiques et que les proportions de Th17 et Treg produisant de l’IL-17A n’étaient pas associées aux fonctions pulmonaires des patients atteints de la FK. En conclusion, les patients atteints de la FK avaient plus d’hyperglycémie et de fluctuations glycémiques, mais elles n’influençaient pas les proportions de lymphocytes Th17 et Treg produisant de l’IL-17A ex vivo. Dans des études futures, il faudrait étudier le rôle de l’IL-17A dans les poumons des patients avec et sans le DAFK et réaliser une étude prospective pour déterminer si une augmentation des niveaux d’IL-17A chez les patients sans le DAFK se traduit par une chute accélérée des fonctions pulmonaires avant l’apparition de cette complication.

The production of PHA from plant oils by Pseudomonas species soil isolated from a sugarcane crop was evaluated. Out of 22 bacterial strains three were able to use efficiently plant oils to grow and to accumulate PHA. Pseudomonas putida and Pseudomonas aeruginosa strains produced PHA presenting differences on monomer composition compatible with variability on monomer specificity of their PHA biosynthesis system. The molar fraction of 3-hydroxydodecanoate detected in the PHA was linearly correlated to the oleic acid supplied. A non-linear relationship between the molar fractions of 3-hydroxy-6-dodecenoate (3HDd Delta(6)) detected in PHA and the linoleic acid supplied was observed, compatible with saturation in the biosynthesis system capability to channel intermediate of P-oxidation to PHA synthesis. Although P. putida showed a higher 3HDd Delta(6) yield from linoleic acid when compared to P. aeruginosa, in both species it was less than 10% of the maximum theoretical value. These results contribute to the knowledge about the biosynthesis of PHA with a controlled composition from plant oils allowing in the future establishing the production of these polyesters as tailor-made polymers. A lectin-like protein from the seeds of Acacia farnesiana was isolated from the albumin fraction, characterized, and sequenced by tandem mass spectrometry. The albumin fraction was extracted with 0.5 M NaCl, and the lectin-like protein of A. farnesiana (AFAL) was purified by ion-exchange chromatography (Mono-Q) followed by chromatofocusing. AFAL agglutinated rabbit erythrocytes and did not agglutinate human ABO erythrocytes either native or treated with proteolytic enzymes. In sodium dodecyl sulfate gel electrophoresis under reducing and nonreducing conditions, AFAL separated into two bands with a subunit molecular mass of 35 and 50 kDa. The homogeneity of purified protein was confirmed by chromatofocusing with a pI=4.0+/-0.5. Molecular exclusion chromatography confirmed time-dependent oligomerization in AFAL, in accordance with mass spectrometry analysis, which confers an alteration in AFAL affinity for chitin. The protein sequence was obtained by a liquid chromatography quadrupole time-of-flight experiment and showed that AFAL has 68% and 63% sequence similarity with lectins of Phaseolus vulgaris and Dolichos biflorus, respectively.

I processi microbiologici rivestono una grande importanza nello sviluppo di combustibili e sostanze chimiche da fonti rinnovabili (biomassa), quali: il biometano e le bioplastiche, come i poli(idrossialcanoati), PHA. I PHA sono poliesteri ottenuti per condensazione di 3-idrossiacidi, il più comune dei quali è il poli(3-idrossibutirrato, PHB). Gli alti costi di produzione del PHA da colture microbiche singole ha portato a valutare nuove strategie, tra cui l’utilizzo di colture microbiche miste (MMC) e substrati di scarto. Il bio-olio ottenuto dalla pirolisi di biomassa potrebbe essere un substrato interessante per la produzione di biogas e PHA. Gli acidi a catena corta (acidi grassi volatili, VFA), infatti, sono i prodotti intermedi nella produzione sia di biogas che di PHA. L’analisi di questi ultimi viene normalmente effettuata tramite GC-FID o GC-MS. La degradazione termica (pirolisi, Py) di PHA produce acidi alchenoici caratteristici, come l’acido crotonico. Il metodo tradizionale per la determinazione del PHA è la metanolisi seguita da un’analisi GC-MS, una procedura laboriosa con uso di solventi clorurati e sostanze corrosive. Lo scopo principale di questa tesi è stato quello di sviluppare un nuovo metodo di analisi dei PHA. Il metodo studiato si basa sulla pirolisi diretta della biomassa e determinazione degli acidi alchenoici prodotti dalla degradazione del PHA in GC-FID. La pirolisi analitica è stata studiata tramite analisi di polimeri puri (per la calibrazione) e poi è stata applicata a campioni batterici derivanti da MMC e a ceppi selezionati. Il confronto con il metodo convenzionale ha dimostrato che la Py/GC-FID è un metodo valido per l’identificazione dei PHA e per la loro quantificazione nelle matrici batteriche. Il pre-trattamento del campione è minimo e non richiede l’uso di solventi e reagenti chimici. Inoltre, è stata applicata una tecnica di analisi dei VFA nei test di biometanazione basata sull’estrazione con una microquantità di dimetilcarbonato.

Sono stati studiati ed applicati metodi “green” per l’estrazione di poliidrossialcanoati (PHA) da colture microbiche singole (Cupriavidus necator) e miste. Sono stati effettuati esperimenti che prevedono l’utilizzo di surfattanti anionici switchable (SAS) per l’estrazione del polimero, al fine di arrivare ad un protocollo ottimale per l’estrazione di PHB da C.necator che prevede l’uso di NH4 laurato come SAS (100 wt% rispetto al peso dei batteri) per tre ore a 90°C. La sostanza ambifilica agisce distruggendo la membrana cellulare esponendo così i granuli di PHB. Il SAS utilizzato si caratterizza per la possibilità di essere recuperabile tramite aggiunta di CO2 al sistema con una resa del 98%. Per le colture microbiche miste, più refrattarie all’impiego dei surfattanti, sono stati utilizzati dei pre-trattamenti, meccanici e fotocatalitici, al fine di indebolire la membrana batterica. Per il trattamento meccanico sono stati impiegati un omogeneizzatore e un sonicatore ad ultrasuoni; mentre per il trattamento fotocatalitico è stata sperimentata l’azione del nano biossido di titanio (sospensione 1% nano-TiO2) in miscela con i batteri, esposti a radiazioni UV. Dai risultati ottenuti sia il trattamento meccanico che quello fotocatalitico non sono risultati adeguati per i batteri misti. Infine il PHB estratto dalla biomassa batterica è stato purificato con un trattamento fotocatalitico che prevede l’utilizzo di nano-TiO2 1% supportato in tessuti, esposti a luce UV. I risultati di questo post-trattamento mostrano che, anche utilizzando diverse tipologie di tessuto, il PHB viene in parte adsorbito dal tessuto, con la seguente perdita di campione. In conclusione il trattamento del C. necator con i SAS risulta essere un ottimo metodo di estrazione di PHB considerando sia la qualità del polimero estratto sia il riciclo e il riutilizzo del surfattante, mentre per il trattamento delle colture microbiche miste ancora non è stato trovato un protocollo soddisfacente.

One of the several factors that contribute to the low efficiency of mammalian somatic cloning is poor fusion between the small somatic donor cell and the large recipient oocyte. This study was designed to test phytohemagglutinin (PHA) agglutination activity on fusion rate, and subsequent developmental potential of cloned bovine embryos. The toxicity of PHA was established by examining its effects on the development of parthenogenetic bovine oocytes treated with different doses (Experiment 1), and for different durations (Experiment 2). The effective dose and duration of PHA treatment (150 microg/mL, 20 min incubation) was selected and used to compare membrane fusion efficiency and embryo development following somatic cell nuclear transfer (Experiment 3). Cloning with somatic donor fibroblasts versus cumulus cells was also compared, both with and without PHA treatment (150 microg/mL, 20 min). Fusion rate of nuclear donor fibroblasts, after phytohemagglutinin treatment, was increased from 33 to 61% (P < 0.05), and from 59 to 88% (P < 0.05) with cumulus cell nuclear donors. The nuclear transfer (NT) efficiency per oocyte used was improved following PHA treatment, for both fibroblast (13% versus 22%) as well as cumulus cells (17% versus 34%; P < 0.05). The cloned embryos, both with and without PHA treatment, were subjected to vitrification and embryo transfer testing, and resulted in similar survival (approximately 90% hatching) and pregnancy rates (17-25%). Three calves were born following vitrification and embryo transfer of these embryos; two from the PHA-treated group, and one from non-PHA control group. We concluded that PHA treatment significantly improved the fusion efficiency of somatic NT in cattle, and therefore, increased the development of cloned blastocysts. Furthermore, within a determined range of dose and duration, PHA had no detrimental effect on embryo survival post-vitrification, nor on pregnancy or calving rates following embryo transfer.
