The aim of this work is to obtain, purify and characterize biochemically a peroxidase from Copaifera langsdorffii leaves (COP). COP was obtained by acetone precipitation followed by ion-exchange chromatography. Purification yielded 3.5% of peroxidase with the purification factor of 46.86. The COP optimum pH is 6.0 and the temperature is 35 ºC. COP was stable in the pH range of 4.5 to 9.3 and at temperatures below 50.0 ºC. The apparent Michaelis-Menten constants (Km) for guaiacol and H2O2 were 0.04 mM and 0.39 mM respectively. Enzyme turnover was 0.075 s-1 for guaiacol and 0.28 s-1 for hydrogen peroxide. Copaifera langsdorffii leaves showed to be a rich source of active peroxidase (COP) during the whole year. COP could replace HRP, the most used peroxidase, in analytical determinations and treatment of industrial effluents at low cost.

The in vitro activity of the crude hydroalcoholic extract of the aerial parts of Miconia langsdorffii Cogn. was evaluated against the promastigote forms of L. amazonensis, the causative agent of cutaneous leishmaniasis in humans. The bioassay-guided fractionation of this extract led to identification of the triterpenes ursolic acid and oleanolic acid as the major compounds in the fraction that displayed the highest activity. Several ursolic acid semi-synthetic derivatives were prepared, to find out whether more active compounds could be obtained. Among these ursolic acid-derived substances, the C-28 methyl ester derivative exhibited the best antileishmanial activity.

Copaifera langsdorffii is a Neotropical tree with wide distribution in the Brazilian Atlantic rain forest and savanna. Although eight microsatellite loci (SSR) were developed in 2000 and have been widely used since then, there is yet no information about their inheritance, linkage and linkage disequilibrium (LD). Through the analysis of 28 open-pollinated (OP) progenies, the SSR loci revealed Mendelian inheritance and independent assortment. Using these progenies, young and adult trees LD was mainly detected in OP progenies. Our results show clear evidence that the eight SSR loci can be used without restriction in genetic diversity, mating system and parentage analysis.

In the present work, the anticariogenic activities of nine labdane type-diterpenes and four sesquiterpenes were investigated. Among these metabolites, (-)-copalic acid (CA) was the most active compound displaying MIC values very promising (ranging from 2.0 to 6.0 mu g/mL) against the main microorganisms responsible for dental caries: Streptococcus salivarius, S. sobrinus, S. mutans, S. mitis, S. sanguinis and Lactobacillus casei. Time kill assays performed with CA against the primary causative agent (S. mutans) revealed that, in the first 12 h, this compound only inhibits the growth of the inoculum (bacteriostatic effect). However, its bactericidal effect is clearly noted thereafter (between 12 and 24 h). Also, CA did not show a synergistic effect when combined with the anticariogenic gold standard (chlorhexidine, CHD) in the checkerboard assays against S. mutans. In conclusion, the results points out CA as an important metabolite in the search for new effective anticariogenic agents. Copyright (C) 2010 John Wiley & Sons, Ltd.

The ecological and economic importance of oleoresin produced by Copaifera langsdorffii is well established. This study aims to investigate the ontogeny, anatomy and ultrastructure of the internal glands of C. langsdorffii during plant development. Samples were processed for light and electron microscopy and a specific technique was applied to impregnate endomembranes. Internal secretory glands were observed in the hypocotyl, epicotyl and eophylls of seedlings, and in the primary stem, pulvinus, petiole, rachis and leaf blade of adult plants. Canals and cavities show differential distribution. They arise from ground meristem cells, and the lumen is first formed by schizogenesis followed by later schizolysigenous development. The dense cytoplasm of epithelial cells shows mitochondria, plastids without thylakoids, polyribosomes and endoplasmic reticulum. A periplastidial reticulum was also observed. Secretion is released by eccrine, granulocrine and holocrine processes. Lipophilic and hydrophilic compounds were histochemically detected in both canals and cavities, whereas resin was detected only in canals. The presence of these substances has been associated with plants` defences against dehydration, as well as against attacks from herbivores and pathogens, from seedling stage onwards. This study focuses on the floral development of Copaifera langsdorffii to elucidate uncertain features in its floral morphology, such as the tetramerous calyx, lack of petals, blackened anthers and their supposed sterility, as well as polyembryony. Buds and flowers were dissected and prepared for examination under scanning electron and light microscopes. The floral apex initiates two bracteoles, five sepals, five petals, five outer stamens, five inner stamens, and one carpel. Order is helical for sepals, reversed unidirectional for the petals, and unidirectional for two whorls of stamens. The tetramerous calyx results from the union of two adaxial sepal primordia, which forms one large sepal and three other smaller sepals. Although the flower lacks petals, the petal primordia are initiated but do not elongate like the other floral organs, remaining as petal rudiments. Ten stamens are formed in two distinct whorls. Formation within each whorl is almost simultaneous, and the inner whorl is formed shortly after the outer. During organ elongation, the inner stamen primordia bases are reoriented outward, resulting in a single whorl of stamens. The darkened anthers have viable pollen grains. Thus, there is no relation between sterility and the dark coloration of the anthers. No signs of extranumerary embryos are observed; therefore, polyembryony is not confirmed. Although studies on floral development of Detarieae have been reported, few Neotropical genera of the tribe (such as Copaifera) have been studied.

The presence of saponins and the molluscicidal activity of the roots, leaves, seeds and fruits of Swartzia langsdorffii Raddi (Leguminosae) against Biomphalaria glabrata adults and eggs were investigated. The roots, seeds and fruits were macerated in 95% ethanol. These extracts exerted a significant molluscicidal activity against B. glabrata, up to a dilution of 100 mg/l. Four mixtures (A2, B2, C and D) of triterpenoid oleanane type saponins were chromatographically isolated from the seed and fruit extracts. Two known saponins (1 and 2) were identified as beta-D-glucopyranosyl-[alpha-L-rhamnopyranosyl-(1->3)- beta-D-glucuronopyranosyl-(1->3)]-3beta-hydroxyolean-12-ene-28 -oate, and beta-D-glucopyranosyl-(1->3)-beta-D-glucuronopyranosyl-(1 ->3)]-3beta-hydroxyolean-12-ene-28-oate, respectively. These two saponins were present in all the mixtures, together with other triterpenoid oleane type saponins, which were shown to be less polar, by reversed-phase HPLC. The saponin identifications were based on spectral evidence, including ¹H-¹H two-dimensional correlation spectroscopy, nuclear Overhauser and exchange spectroscopy, heteronuclear multiple quantum coherence, and heteronuclear multiple-bond connectivity experiments. The toxicity of S. langsdorffii saponins to non-target organisms was prescreened by the brine shrimp lethality test.

Chromatographic fractionation of the EtOH extract from the leaves of Swartzia langsdorffii afforded the pentacyclic triterpenes oleanolic acid and lupeol, and two saponins: oleanolic acid 3-sophoroside and the new ester 3-O-b-D-(6'-methyl)-glucopyranosyl-28-O-b-D-glucopyranosyl-oleanate. Their structures were elucidated from spectral data, including 2D NMR and HRESIMS experiments. Antifungal activity of all isolated compounds was evaluated, using phytopathogens Cladosporium cladosporioides and C. sphaerospermum, and human pathogens Candida albicans, C. krusei, C. parapsilosis and Cryptococcus neoformans.

Phytochemical investigation of the hexane extract of fruit shells of Copaifera langsdorffii Desf. (Caesalpinioideae) afforded ent-kaur-16-en-19-oic acid, polyalthic acid, nivenolide and the mixture of caryophyllene oxide and ent-kaur-16-en-19-oic acid. The chloroform extract of unripe seeds led to the isolation of coumarin and the GC/MS analysis of the extract allowed the identification of 81.8% of the fatty acid composition after hydrolysis followed by methylation. The main fatty acid identified was oleic acid (33.1%). The isolation of all secondary metabolites was accomplished by modern chromatographic methods and the structure determination was accomplished by spectrometric methods (IR, MS, NMR ¹H and 13C).

O presente trabalho teve por objetivo o aproveitamento da água residuária proveniente de um sistema biológico de tratamento de esgotos domésticos como alternativa à fertirrigação convencional de viveiros florestais, visando a produção de mudas de Croton floribundus (capixingui) e Copaifera langsdorffii (Copaíba) via subirrigação. As variáveis avaliadas mensalmente foram altura e diâmetro do coleto da muda e, ao final do experimento, peso da matéria seca da parte aérea e da raiz, área foliar, razão parte aérea/ raiz, análise nutricional foliar, análises químicas do substrato antes e após o ensaio e análises nutricionais periódicas da água residuária. O crescimento das plantas, em geral, foi superior no tratamento convencional com adubos minerais. O desenvolvimento do sistema radicular foi favorecido no tratamento com água residuária, o que pode ser característica desejável para maior sobrevivência das mudas no campo. Os resultados mostraram que a água residuária pode ser utilizada na fertirrigação de viveiros para produção das espécies estudadas, pois todas as plantas se mostraram vigorosas, com bom desenvolvimento, sem mortalidade, deficiência ou toxidez. Entretanto, constatou-se que essas mudas necessitarão de um maior tempo no viveiro, quando comparadas com as produzidas com fertilizantes minerais.

A eletroforese vem sendo utilizada com a finalidade de auxiliar a avaliação da qualidade fisiológica de sementes de espécies florestais, uma vez que respostas mais rápidas podem ser obtidas usando-se marcadores isoenzimáticos. Sementes de copaíba (Copaifera langsdorffii) foram submetidas a diferentes tempos de envelhecimento artificial (0, 24, 48, 72 e 96 horas), em germinador (42 ºC e 100% de UR), tendo como objetivo avaliar as possíveis mudanças nos padrões eletroforéticos de proteínas e isoenzimas, decorrentes do envelhecimento artificial. O envelhecimento artificial ocasionou redução na germinação e vigor de sementes de C. langsdorffii, sendo o vigor afetado mais rapidamente. Em relação aos padrões eletroforéticos, não houve diferença significativa no conteúdo de proteínas solúveis e de armazenamento, mas, nas enzimas esterase e peroxidase, verificou-se redução na atividade destas, à medida que as sementes foram envelhecidas.

Copaifera langsdorffii é uma espécie em perigo de extinção, com dormência múltipla em suas sementes e baixa resistência ao armazenamento. Nesse sentido, é importante saber se as sementes perdem viabilidade a partir do ponto de maturidade fisiológica e se resistem à perda de água durante o armazenamento. Quatro experimentos foram instalados com sementes coletadas em julho (matriz 1) e agosto de 2005 (matriz 2), no Vale do Rio Araguari, MG, dois com sementes recém-colhidas e dois com sementes armazenadas em câmara fria. A perda de 31% de água das sementes coletadas no solo, em relação às colhidas de frutos aderidos à árvore, reduziu em aproximadamente 10% o porcentual de emergência. A presença do arilo nas sementes recém-colhidas causou aumento do coeficiente de variação de emergência, indicando que essa estrutura possui substâncias inibidoras de germinação. Com o armazenamento, as sementes perderam viabilidade, diminuindo significativamente os porcentuais de emergência, o que indica que as sementes dessa espécie possuem certo grau de recalcitrância, com baixa resistência à perda de água após a maturidade fisiológica. Para a obtenção de maior número de mudas, em menor tempo e com maior sincronia, recomenda-se a utilização de sementes oriundas de frutos recém-abertos, ainda aderidos à árvore, com semeadura sem arilo e com escarificação.

The loss of large areas of Cerrado (Brazilian savanna) in Brazil can lead to reduced biodiversity and to the extinction of species. Therefore, the present study aimed to investigate the genetic fragility of populations of Copaifera langsdorffii Desf exposed to different anthropic conditions in fragments of Cerrado in the state of São Paulo. The study was carried out in two Experimental Stations operated by the Forest Institute (Assis and Itirapina), in one fully protected conservation unit (Pedregulho) and in one private property (Brotas). Analyses were conducted using leaf samples from 353 adult specimens and eight pairs of microsatellite loci. The number of alleles per locus ranged from 13 to 15 in all populations, but the mean number of effective alleles was approximately half this value (7.2 to 9-1). Observed heterozygosity was significant and lower than the expected in all populations. Consequently, all populations deviated from Hardy-Weinberg expected frequencies. Fixation indexes were significant for all populations, with the Pedregulho population having the lowest value (0.189) and Itirapina having the highest (0.283). The analysis of spatial genetic structure detected family structures at distance classes of 20 to 65 m in the populations studied. No clones were detected in the populations. Estimates of effective population size were low, but the area occupied by each population studied was large enough for conservation, medium and long term. Recent reductions or bottlenecks were detected in all four populations. Mean Gst’ (genetic divergence) indicated that most of the variation was within populations. Cluster structure analysis based on the genotypes detected K= 4 clusters with distinct allele frequencies patterns. The genetic differentiation observed among populations is consistent with the hypothesis of genetic and geographic isolation. Therefore, it is essential to adopt conservation strategies that raise the gene flow between fragments.

A Copaifera langsdorffii é uma leguminosa nativa do Brasil, da qual pode ser extraído um óleo, popularmente conhecido como óleo de copaíba. Este óleo é amplamente utilizado para tratamento de feridas cutâneas por ser reconhecido como antiinflamatório e cicatrizante. Apesar disso, poucas comprovações científicas do verdadeiro efeito terapêutico desta planta medicinal foram produzidas. O objetivo desse trabalho foi avaliar o efeito do tratamento tópico com C. langsdorffii no processo de reparo quando um corpo estranho é o indutor da inflamação. Para isso, 60 camundongos da linhagem BALB/c foram submetidos à incisão cirúrgica linear de 1cm no dorso para realizar o implante de uma lamínula de vidro com 12mm de diâmetro no tecido subcutâneo. Quatro tratamentos para a ferida foram estabelecidos: controle (C) tratado com solução salina estéril, controle veículo (CV) tratado com óleo mineral estéril, tratamento 1 (T1) tratado com diluição (v:v) de óleo mineral estéril e óleo de C. langsdorffii, tratamento 2 (T2) tratado com óleo de C. langsdorffii puro. As avaliações foram realizadas em períodos de tempo pré-determinados (1, 3, 5, 7 e 14 dias). Era possível perceber características da fase proliferativa como a reepitelização, a presença de fibroblastos e a neovascularização, porém os grupos tratados com o óleo (T1 e T2) não apresentavam reepitelização aos três dias. Esses grupos aos 5 e 7 dias apresentavam no exame macroscópico maior intensidade de edema, hiperemia e permanência de crostas. Na microscopia, a reepitelização ainda não estava completa e a crosta era serocelular. Nos grupos C e CV, apesar de discreta, era predominante a presença de mononucleares, enquanto nos grupos T1 e T2 o infiltrado inflamatório era misto e com maior intensidade que nos outros grupos. Quatorze dias depois da incisão cirúrgica, os aspectos macroscópicos dos grupos C e CV eram semelhantes e os grupos T1 e T2, apesar de a ferida estar completamente fechada e sem crostas, a pele desses animais estava espessada e na derme do grupo T2 parecia ter fibrose mais acentuada do que nos outros grupos. Esses resultados demonstraram que o tratamento tópico com óleo de C. langsdorffii prejudicou o processo de reparação normal de ferida na presença de corpo estranho.
