The estrogenic activity of the Chinese herb kudzu root was investigated by a recombinant yeast screening assay (YES). Isoflavones are the main components in the plant, of which puerarin is the most abundant one. The kudzu root extract was separated into four fractions according to the polarity. The crude extract and its sub-fractions, except the water fraction, showed clear estrogenic activity and the potencies were in the range of 10(-3) to 10(-1) g/l. The ligand potency was used to compare the estrogenic activity of these fractions. The crude extract and its sub-fractions were further analyzed by high performance liquid chromatography (HPLC) to correlate the activity and the active components. Bioassay and chemical analysis showed that theoretical estrogenic activity expressed as equivalent 17 beta-estradiol concentration or the cumulative effects are comparable to that experimentally determined by YES. The results showed that the high content of isoflavones as well as the high estrogenic activity could make kudzu root extract an interesting candidate for hormone replacement therapy. High-performance liquid chromatography-tandem mass spectrometry has been used to identify isoflavone aglycones and glycosides in kudzu root. Fourteen isoflavones were detected. Among these, six were identified by comparison with authentic standards. Tentative identifications of the other isoflavones are based on UV spectra, mass spectra of protonated and deprotonated molecules, and MS-MS data. Several are reported for the first time in kudzu root. The bioactivity and bioavailability of isoflavone aglycones are usually greater than those of their glycosides. To improve the bioavailability of kudzu root isoflavones, crude beta-glycosidases prepared from microbes were used to hydrolyze the isoflavone glycosides. Several MS modes are combined not only to identify the isoflavones in kudzu root, but also to describe the biotransformation of kudzu root isoflavone glycosides. It is also proved that crude beta-glycosidases have high selectivity toward the O-glycosides of isoflavones.

This study assesses the use of dried (5% w/w moisture) kudzu (Peuraria lobata ohwi) as an adsorbent medium for the removal of two basic dyes, Basic Yellow 21 and Basic Red 22, from aqueous solutions. The extent of adsorption was measured through equilibrium sorption isotherms for the single component systems. Equilibrium was achieved after 21 days. The experimental isotherm data were analysed using Langmuir, Freundlich, Redlich-Peterson, Temkin and Toth isotherm equations. A detailed error analysis was undertaken to investigate the effect of using different error criteria for the determination of the single component isotherm parameters. The performance of the kudzu was compared with an activated carbon (Chemviron F-400). Kudzu was found to be an effective adsorbent for basic dye colour removal, though its capacity for colour removal was not as high as an activated carbon, the potential appeared to exist to use it as an alternative to activated carbon where carbon cost was prohibitive. Banana is one of the most consumed fruits in the world, which is grown in most tropical countries. The objective of this work was to evaluate the main attributes of soil fertility in a banana crop under two cover crops and two root development locations. The work was conducted in Curaçá, BA, Brazil, between October 2011 and May 2013, using a randomized block design in split plot with five repetitions. Two cover crops were assessed in the plots, the cover 1 consisting of Pueraria phaseoloid es, and the cover 2 consisting of a crop mix with Sorghum bicolor, Ricinus commun is L., Canavalia ensiform is, Mucuna aterrima and Zea mays, and two soil sampling locations in the subplots, between plants in the banana rows (location 1) and between the banana rows (location 2). There were significant and independent effects for the cover crop and sampling location factors for the variables organic matter, Ca and P, and significant effects for the interaction between cover crops and sampling locations for the variables potassium, magnesium and total exchangeable bases. The cover crop mix and the between-row location presented the highest organic matter content. Potassium was the nutrient with the highest negative variation from the initial content and its leaf content was below the reference value, however not reducing the crop yield. The banana crop associated with crop cover using the crop mix provided greater availability of nutrients in the soil compared to the coverage with tropical kudzu.
