Foram caracterizados/identificados por eletroforese de isoenzimas 23 isolados de Leishmania sp de pacientes dos municípios de Rio Preto da Eva e Manaus, analisando-se o grau de similaridade entre os organismos. Os resultados indicaram ocorrência de Leishmania guyanensis e Leishmania naiffi nestes dois ambientes e a heterogeneidade das amostras de Leishmania naiffi.

Durante o período de 2002 a 2003 foram realizadas coletas de flebotomíneos em duas áreas do estado do Amazonas (Base de treinamento militar - BI1 e Tarumã Mirim). Nessas coletas foram capturadas um total de 1.440 fêmeas de Lutzomyia (Nyssomyia) umbratilis. Lu.umbratilis é a principal responsável pela transmissão da Leishmaniose Tegumentar Americana (LTA) ao norte do Rio Amazonas. Do total coletado apenas 15 espécimens (ou 1,04%) apresentaram infecção natural por tripanosomatídeos, sendo 12 na BI1 e 3 em Tarumã-Mirim. Isso representou uma taxa de infecção de 1,66% (12 dos 720 capturados em BI1) e 0,42% (3 dos 720 em Tarumã-Mirim). Estes resultados confirmam as informações prévias por outros autores de reduzidos valores de infecção natural por tripanosomatídeos em flebotomíneos, mesmo em áreas altamente endêmicas para leishmaniose.

O teste de intradermorreação de Montenegro é utilizado para detectar infecção por Leishmania em humanos. A técnica se baseia numa reação de hipersensibilidade tardia. Os antígenos de Montenegro utilizados no experimento são soluções de antígenos homólogos brutos de Leishmania (Viannia) braziliensis e L.(V)guyanensis. Este experimento demonstrou que os animais inoculados com as espécies de Leishmania inoculadas desenvolveram enduração no local do teste mais acentuada que os animais controle. Os resultados sugerem que o teste cutâneo pode vir a ser indicado como método auxiliar de diagnóstico em cães infectados por Leishmania sp.

Monogenetic insect trypanosomatids of the genera Crithidia, Leptomonas and Herpetomonas, multiplied as in axenic cultures, for many months, in the lumen of the scent glands of the opossum Didelphis marsupialis. Specific antibodies were detected in the serum of the animals but there was no evidence of invasion of their tissues by the parasites.

Descreve-se a espécie Trypanosoma guairaensis sp. n. parasita de Megaloancistrus aculeatus do rio Paraná (reservatório de Itaipu). Características morfométricas como comprimento e largura do núcleo e comprimento da parte livre do flagelo, bem como os valores do índice nuclear, diferenciam T. guairaensis de todas as espécies de tripanosomas descritas para peixes de água doce do Brasil.

It is described Trypanosoma nupelianus sp. n. parasitizing Rhinelepis aspera taken from Paraná river (Itaipu reservoir, Brazil). Morphometric characters such as body length, width of the body and nucleus dimensions show that T. nupelianus sp. n. is different from other trypanosomes described for fishes from tropical regions.

Characterization is given of a new parasite, Leishmania equatoriensis sp.n. wich was isolated from the viscera of a sloth (Choloepus hoffmanni) and a squirrel (Sciurus granatensis), captured in humid tropical forest onthe Pacific Coast of Ecuador. Data based on biological and molecular criteria, as well as numerical zymotaxonomical analysis, indicate that this parasite is a new species of the L. brasiliensis complex. L. equatoriensis is cleary distinguishable form all other known species within this complex, using the following molecular criteria: reactivity patterns with specific monoclonal antibodies, isoenzyme electrophoresis, and restriction-endonuclease fragment patterns of kinetoplast DNA (k-DNA).

On few occasions, Phytomonas davidi (McGhee & Postell isolate) cultures in LIT (liver infusion-tryptose) medium around 27oC presented, as seen in Giemsa-stained smears, a set of peculiar morphological features, among them being noticeable the pairs of apposed cells attached by their posterior ends, where occurred a stained line and/or a dilatation, usually bulb-like in shape; sometimes this dilatation could occupy one of the cells or hold both together. In some pairs, the nucleus of each parasite seemed migrating towards the other, entering into such dilatation; in others, both nuclei were inside it, sometimes in close proximity or seeming fused; peculiar chromatin arrangements involving both nuclei were occasionally observed. Several mono or binucleate round forms bearing one or two flagella, as well as flagellate slender cells without nucleus were concomitantly seen there. In some instances, an intriguing small stained body occurred beside a single large nucleus, either in pairs presenting the bulb-like structure or in round cells. These cytological findings seemed steps of a dynamic process suggesting sexuality, since in several of them nuclear interactions following fusion of two parasites appeared to occur

This article reviews current concepts of the biology of Endotrypanum spp. Data summarized here on parasite classification and taxonomic divergence found among these haemoflagellates come from our studies of molecular characterization of Endotrypanum stocks (representing an heterogenous population of reference strains and isolates from the Brazilian Amazon region) and from scientific literature. Using numerical zymotaxonomy we have demonstrated genetic diversity among these parasites. The molecular trees obtained revealed that there are, at least, three groups (distinct species?) of Endotrypanum, which are distributed in Central and South America. In concordance with this classification of the parasites there are further newer molecular data obtained using distinct markers. Moreover, comparative studies (based on the molecular genetics of the organisms) have shown the phylogenetic relationships between some Endotrypanum and related kinetoplastid lineages.

From August 1997 to August 1998, 334 specimens of Triatoma longipennis and 62 of T. picturata were collected in four groups of localities placed in the zone from Guadalajara, Jalisco to Tepic, Nayarit, in the West Coast of Mexico. Most T. longipennis were collected outdoors (69.2%) while most T. picturata (58.1%) were collected indoors. All collected specimens were examined for Trypanosoma cruzi infection, which was detected on 98 (29.3%) T. longipennis and 17 (27.4%) T. picturata. This study confirms the role of T. longipennis and T. picturata as some of the main T. cruzi vectors to humans in Mexico. Habitation Infestation Rate with T. longipennis was of 0.09 and with T. picturata was of 0.03 and the predominating ecotopes were pile of blocks, chicken coops, pigsties, wall crawls and beds.

To further investigate phylogeny of kinetoplastid protozoa, the sequences of small subunit (18S) ribosomal RNA of nine bodonid isolates and ten isolates of insect trypanosomatids have been determined. The root of the kinetoplastid tree was attached to the branch of Bodo designis and/or Cruzella marina. The suborder Trypanosomatina appeared as a monophyletic group, while the suborder Bodonina was paraphyletic. Among bodonid lineages, parasitic organisms were intermingled with free-living ones, implying multiple transitions to parasitism and supporting the `vertebrate-first hypothesis'. The tree indicated that the genera Cryptobia and Bodo are artificial taxa. Separation of fish cryptobias and Trypanoplasma borreli as different genera was not supported. In trypanosomatids, the genera Leptomonas and Blastocrithidia were polyphyletic, similar to the genera Herpetomonas and Crithidia and in contrast to the monophyletic genera Trypanosoma and Phytomonas. This analysis has shown that the morphological classification of kinetoplastids does not in general reflect their genetic affinities and needs a revision.
