In Italy, National Law (281/1991) prohibits euthanasia of shelter dogs if they are not dangerous or suffering seriously. Adoption rates in rescue shelters are often lower than entrance rates, leading inevitably to overcrowded facilities where animals are likely to spend the rest of their lives in kennels. In this situation, housing conditions (i.e. space provided, environmental, and social stimulation) may have an impact on canine welfare. In this research project, the effects of two different forms of housing (group- and pair housing) on long-term shelter dogs were compared using behavioural and physiological parameters. Observational data and saliva samples were collected from dogs exposed to both experimental settings; behaviour and cortisol concentration levels were used as welfare indicators. Pair housing offered fewer social and environmental stimuli and behavioural analysis showed a significant decrease in locomotor, exploratory, and social behaviour. Cortisol levels show that this parameter varied independently of housing conditions. Although this study found no evidence suggesting that one form of confinement reduced animal welfare more than the other (e.g. in terms of abnormal behaviour, or higher cortisol concentrations), the type of confinement did affect the expression of a variety of behaviours and these variations should not be ignored with respect to housing decisions for long-term shelter dogs.

A pair of primers directed to 16S-23S rDNA interspacer (ITS) was designed directed to Brucella genetic sequences in order to develop a polymerase chain reaction (PCR) putatively capable of amplifying DNA from any Brucella species. Nucleic acid extracts from whole-blood from naive dogs were spiked with decreasing amounts of Brucella canis RM6/66 DNA and the resulting solutions were tested by PCR. In addition, the ability of PCR to amplify Brucella spp. genetic sequences from naturally infected dogs was evaluated using 210 whole-blood samples of dogs from 19 kennels. The whole-blood samples collected were subjected to blood culture and PCR. Serodiagnosis was performed using the rapid slide agglutination test with and without 2-mercaptoethanol. The DNA from whole blood was extracted using proteinase-K, sodium dodecyl sulphate and cetyl trimethyl ammonium bromide followed by phenol-chloroform purification. The PCR was capable of detecting as little as 3.8 fg of Brucella DNA mixed with 450 ng of host DNA. Theoretically, 3.8 fg of Brucella DNA represents the total genomic mass of fewer than two bacterial cells. The PCR diagnostic sensitivity and specificity were 100%. From the results observed in the present study, we conclude that PCR could be used as confirmatory test for diagnosis of B. canis infection.

Objective Dogs play an important role as infection source of human cryptosporidiosis. The objective of the study was to determine the prevalence of Cryptosporidium sp. in dogs as well as to compare two techniques of fecal analysis. Methods Four-hundred and fifty canine fecal samples from the city of São Paulo were analyzed between 2003 and 2004. Fecal samples were randomly selected from dogs housed in a university veterinary hospital (group 1, n=200) and private kennels (group 2, n=250). The detection of Cryptosporidium was performed using modified Ziehl-Neelsen staining and Polymerase Chain Reaction (PCR) technique. Statistical analysis was performed using the two-tailed test of significance at 5% confidence interval (z critical=+/- 1.645).Results Only Cryptosporidium parvum oocysts were found the prevalences found by light microscopy examination and PCR techniques were 8.8% and 9.5%, respectively. Young animals showed a lower frequency (5.5%) compared to adults (10.1%). There was no statistically significant difference in Cryptosporidium prevalence between males and females. Conclusions the prevalence of C. parvum in the canine population studied was similar to that one found in the literature and affects equally males and females. The use of PCR allowed the detection of more positive cases than light microscopy.

The epidemiologic frameworks of zoonosis periodically change in some of its aspects, involving the emergence or reemergence of men and animals' worsening of health. In this scenario, in which rapid changes can happen, the analysis of the prevalence of diseases, as the filariasis diagnosed in dogs from the coastline of Parana, has significant relevance. This study has been carried out with 422 dogs and aimed to determine the prevalence of circulating microfilariae of Acanthocheilonema reconditum, both in animals accommodated at the municipal kennels, as those that live in the district COHAPAR II, both places located in the metropolitan area of the city of Guaratuba-Parana, through two blood tests: evaluation of fresh blood, by the technique of the 'thick drop' and method of Knott modified by Newton and Wright. Thirty of the evaluated animals (7.10%), twenty two males and eight females, showed the presence of microfilariae of A. reconditum as result.
