Twenty-two populations of seven species of Cremanthodium from high altitude regions of western China were observed karyologically. C. ellisii, C. microglossum, C. brunneo-pilosum, C. stenoglossum, C. discoideum and C. lineare all had the same chromosome number of 2n=58 whereas C. humile had 2n=60. All chromosome numbers of these species are documented here for the first time. The basic number of x=30 is new for this genus. The karyotypes of all species belong to 2A type according to Stebbins' asymmetry classification of karyotypes. Two basic chromosome numbers, x=30 and x=29 in Cremanthodium, correspond exactly to two branching patterns in this genus, sympodial versus monopodial. The systematic and taxonomic statuses of the sympodial species need further study. The karyomorphological data provide no support to the sectional subdivision in Cremanthodium. Four of the five members of the Dasyaceae found in the British Isles, Dasya corymbifera J. Agardh, Dasya hutchinsiae Harvey, Dasya punicea Meneghini ex Zanardini and Heterosiphonia plumosa (Ellis) Batters, appear to have Polysiphonia-type life histories on the basis of evidence from field collections of tetrasporophytes and gametophytes. In collections from the British Isles of the fifth species, Dasya ocellata (Grateloup) Harvey, only tetrasporophytes have ever been observed, but there are two reports of gametophytes in this species from further south in Europe. Dasya ocellata tetraspores were isolated into culture from populations in Strangford Lough, Northern Ireland, and Agadir, Morocco, where one female thallus was collected amongst tetrasporophytes. Dasya ocellata from Ireland underwent a direct tetraspore-to-tetrasporophyte life history, which was followed through two complete cycles. Karyological studies showed that meiosis does not occur during tetrasporangial development: tetrasporangia are mitotic, with c. 64 small chromosomes. Comparison with chromosome numbers in meiotic tetrasporangia of D. hutchinsiae (n = c. 32) showed that this is the diploid chromosome complement. Tetraspores from the Moroccan isolate, by contrast, gave rise to gametophytes (although only the males became fertile) and tetrasporophyte recycling did not occur. Thalli sampled from a population in southern Portugal consisted only of tetrasporophytes. Dasya ocellata, like many members of the Ceramiales, shows intraspecific life history variability; a sexual life history apparently occurs only in southern populations.

A single thallus of the rare red seaweed Tsengia bairdii (Farlow) K. Fan et Y.Fan( Platoma bairdii (Farlow) Kuckuck) (Nemastomataceae) was collected on a subtidal pebble on the west coast of Scotland. The terete gelatinous axes, which were only 7 mm high, were monoecious. They bore numerous cystocarps and a few spermatangia, which represent the first observation of male structures in this genus. Released carpospores grew into expanded basal discs that gave rise to erect axes bearing irregularly cruciate tetrasporangia. irregularly cruciate to zonate tetrasporangia were also formed on these basal discs. Karyological studies on dividing tetrasporocytes showed about 25 bodies, identified as paired meiotic chromosomes on the basis of their size in comparison to mitotic and meiotic chromosomes in other red algal species. These observations confirm the isomorphic life history inferred from early field collections and show that this species is monoecious. Tsengia bairdii is an extremely rare seaweed in Europe - it seems to be confined to sublittoral cobbles and has a temporally patchy distribution.

The present work deals with the development of primary cell culture and diploid cell lines from two fishes, such as Poecilia reticulata and Clarias gariepinus. The greatest difficulty experienced was the avoidance of bacterial and fungi contamination. Three types of cell cultures are commonly developed, primary cell culture, diploid cell lines and heteroploid cell lines. Primary cell culture obtained from the animal tissues that have been cultivated in vitro for the first time. They are characterized by the same chromosome number as parent tissue, cultivated in vitro for the first time, have wide range of virus susceptibility, usually not malignant, six chromatin retarded and do not grow as suspension cultures. Diploid cell lines arise from a primary cell culture at the time of subculturing. Diploid cell lines commercially used in virology are W1-38 (human embryonic lung), W1-26 (human embryonic lung) and HEX (Human embryonic kidney). Heteroploid cell lines have been subcultivated with less than 75% of the cells in the population having a diploid chromosome constitution. Tissue cultures have been extensively used in biomedical research. The main applications are in three areas, Karyological studies, Identification and study of hereditary metabolic disorders and Somatic cell genetics. Other applications are in virology and host-parasite relationships. In this study an attempt was made to preserve the ovarian tissue at low temperature in the presence of cryoprotectants so that the tissue can be retrieved at any time and a cell culture could be developed.

Schizomeris Kützing é uma alga filamentosa sem ramificações e com sistema basal de fixação diferenciado. O gênero contém a espécie-tipo Schizomeris leibleinii Kützing e duas espécies pouco conhecidas, S. irregularis Fritsch & Rich e S. indicum (Ghose) Fritsch & Rich. As espécies são diferenciadas, principalmente, através do tipo de sistema basal de fixação. Contudo, a taxonomia específica do gênero é confusa. Estudos morfométricos e morfológicos foram realizados em populações identificadas como S. leibleinii de acordo com duas abordagens. Na primeira, foram analisadas 11 populações de diferentes localidades e na segunda, duas populações distintas através de coletas mensais durante 11 meses. O diâmetro de filamentos multisseriados, diâmetro e comprimento de células de filamentos multi e unisseriados, presença de constrição, ápice dos filamentos e tipo de sistema basal foram analisados em ambas abordagens. Além disso, estudos cariológicos foram efetuados para complementar as informações taxonômicas. Os resultados revelaram ampla variação morfológica, sendo mais acentuada entre as populações de localidades diferentes que nas mesmas populações ao longo do tempo, mas as características mostraram um gradiente contínuo. Os diferentes tipos de sistemas basais descritos para as espécies do gênero (claviforme, apressório, lobado e rizóides) foram encontrados nas populações estudadas. Esses resultados sugerem que a distinção entre as três espécies é muito frágil e não pode ser apoiada pela diferenciação do sistema basal ou por características métricas. Acredita-se que, por essas razões, o gênero Schizomeris seja composto por uma única espécie. A análise cariológica resultou em n = 12, diferindo de estudos anteriores e algumas possíveis explicações são apresentadas.

As treze espécies de Phaneropterinae estudadas neste trabalho podem ser organizadas em quatro diferentes grupos tomando como referência suas características cariotípicas. Todas possuem sistema cromossômico de determinação sexual do tipo X0(masculino), XX(feminino). O cromossomo X é sempre heteropicnótico durante a prófase I, tem dimensões e morfologias variáveis nas diferentes espécies mas é sempre o maior elemento do cariótipo, além de apresentar segregação precoce durante a anáfase I. O número cromossômico fundamental (NF) varia de 21 a 32. Neste trabalho, são discutidos os significados evolutivos das variações cariotípicas encontradas e suas correlações filogenéticas com outros grupos de espécies pertencentes à mesma subfamília.

The karyotypes of 12 species of Psittaciformes new to cytology are described: Lorius hypoinochrous, L. lory and Phigys solitarius of the Loriidae, and Amazona autumnallis, Aratinga jandaya, Eclectus roratus, Pionus maximiliani, P. menstruus, P. senilis, P. seniloides, Poicephalus senegalus and Polytelis alexandrae of the Psittacidae. The karyotypes of Amazona ochrocephala, Ara ararauna, Ara macao, Psittacula krameri, Psittacus erithacus and Pyrrhura molinae of the Psittacidae have been previously described. For reasons of comparison the karyotypes of Aratinga aurea, Forpus xanthopterygius, Brotogeris sanctithomae and B. versicolorus of the Psittacidae are also described. These karyotypes are compared to those in the literature and the karyological relationships in the Psittaciformes are briefly discussed. Microchromosome fusions and translocations and pericentric inversions probably are responsible for the heterogeneity of karyotypes in the Psittaciformes.

In this present work attempts have been made to study the glass transition temperature of alternative mould materials by using both microwave heating and conventional oven heating. In this present work three epoxy resins, namely R2512, R2515 and R2516, which are commonly used for making injection moulds have been used in combination with two hardeners H2403 and H2409. The magnetron microwave generator used in this research is operating at a frequency of 2.45 GHz with a hollow rectangular waveguide. In order to distinguish the effects between the microwave and conventional heating, a number of experiments were performed to test their mechanical properties such as tensile and flexural strengths. Additionally, differential scanning calorimeter technique was implemented to measure the glass transition temperature on both microwave and conventional heating. This study provided necessary evidences to establish that microwave heated mould materials resulted with higher glass transition temperature than the conventional heating. Finally, attempts were also made to study the microstructure of microwave-cured materials by using a scanning electron microscope in order to analyze the morphology of cured specimens.
