In the work described by this paper, we studied the development of a selective potassium ion sensor constituted of a carbon paste electrode modified (CPEM) with a novel KSr(2)Nb(2)O(15). The material KSr(2)Nb(2)O(15) is an oxide with the tetragonal tungsten bronze structure (TTB) type are in forefront both in the area of research as well as in industrial applications. The sensor response to potassium ions was linear in the concentration range 1.26 x 10(-5) at 1.62 x 10(-3) mol L(-1) (E (mV) = 32.7 + 51.1 log [K(+)]). The sensor based KSr(2)Nb(2)O(15), of the TTB-type presented very good potentiometric response, with a slope of 51.1 mV/dec (at 25 degrees C) and detection limit for the potassium ions of 7.27 x 10(-5) mol.L(-1)

Kinase suppressor of Ras (KSR) is an evolutionarily conserved component of Ras-dependent signaling pathways. Here, we find that murine KSR (mKSR1) translocates from the cytoplasm to the plasma membrane in the presence of activated Ras. At the membrane, mKSR1 modulates Ras signaling by enhancing Raf-1 activity in a kinase-independent manner. The activation of Raf-1 is mediated by the mKSR1 cysteine-rich CA3 domain and involves a detergent labile cofactor that is not ceramide. These findings reveal another point of regulation for Ras-mediated signal transduction and further define a noncatalytic role for mKSR1 in the multistep process of Raf-1 activation.

Armas Einar Leopold Lönnbohm f. 6.7.1878 på Hövelö gård i Paltamo d. 10.1.1926 i Nuppulinna, Tusby Som diktare representerar Eino Leino den nationella nyromantiken, och hans lyrik har blivit gemensam egendom bland finländarna. Finsk folkdiktning och europeiska litterära strömningar möts framför allt i hans två samlingar av Helkavirsiä (sv. Helkasånger) från 1903 och 1916. I dem smälter en ny, levande bildlighet ihop med en mytisk värld, som bottnar i folkdiktningen. Från år 1906 skriver Leino även prosa. Hans trilogi från ofärdsåren, bestående av romanerna Tuomas Vitikka, Jaana Rönty och Olli Suurpää (utgivna mellan 1906 och 1908), representerar ett brytningsskede i skildringen av folket. i sin dramatik behandlar Leino framför allt historiska ämnen. Catalytic activity and selectivity of niobate-based nanostructured materials were investigated. Dry methane reforming (DMR) and ethylene homologation reaction (EHR) were selected as test reactions. KSr 2Nb5O15, Sr2NaNb5O 15 and NaSr2(NiNb4)O15 δ niobate powders were prepared by the high energy ball milling method and calcined in a reductor atmosphere. N2 adsorption isotherms, X-ray diffraction and infrared spectroscopy characterization was performed. Hydrogen pretreated niobates showed from low to moderate catalytic initial activity in DMR's test, nevertheless the materials were deactivated rapidly and the kinetic parameters associated to deactivation were estimated. Otherwise, non-treated catalysts showed a high initial activity in EHR's test and KSr2Nb 5O15 catalyst requires 24 h to the total deactivation with a high selectivity to form propylene. A reaction mechanism to the propylene formation is discussed. © 2012 Elsevier Ltd. All rights reserved.

Connector enhancer of KSR (CNK) is a multidomain protein required for RAS signaling. Its C-terminal portion (CNKC-term) directly binds to RAF. Herein, we show that the N-terminal portion of CNK (CNKN-term) strongly cooperates with RAS, whereas CNKC-term efficiently blocks RAS- and RAF-dependent signaling when overexpressed in the Drosophila eye. Two effector loop mutants of RASV12, S35 and C40, which selectively activate the mitogen-activated protein kinase (MAPK) and phosphatidylinositol-3-kinase pathways, respectively, do not cooperate with CNK. However, a strong cooperation is observed between CNK and RASV12G37, an effector loop mutant known in mammals to activate specifically the RAL pathway. We have identified two domains in CNKN-term that are critical for cooperation with RAS. Our results suggest that CNK functions in more than one pathway downstream of RAS. CNKc-term seems to regulate RAF, a component of the MAPK pathway, whereas CNKN-term seems to be involved in a MAPK-independent pathway.

Using microarrays to probe protein-protein interactions is becoming increasingly attractive due to their compatibility with highly sensitive detection techniques, selectivity of interaction, robustness and capacity for examining multiple proteins simultaneously. The major drawback to using this approach is the relatively large volumes and high concentrations necessary. Reducing the protein array spot size should allow for smaller volumes and lower concentrations to be used as well as opening the way for combination with more sensitive detection technologies. Dip-Pen Nanolithography (DPN) is a recently developed technique for structure creation on the nano to microscale with the capacity to create biological architectures. Here we describe the creation of miniaturised microarrays, 'mesoarrays', using DPN with protein spots 400× smaller by area compared to conventional microarrays. The mesoarrays were then used to probe the ERK2-KSR binding event of the Ras/Raf/MEK/ERK signalling pathway at a physical scale below that previously reported. Whilst the overall assay efficiency was determined to be low, the mesoarrays could detect KSR binding to ERK2 repeatedly and with low non-specific binding. This study serves as a first step towards an approach that can be used for analysis of proteins at a concentration level comparable to that found in the cellular environment.
