Coastal lagoons where salinity varies within a wide range during the year are colonized by euryhaline macrophytes which can develop extensive beds. Seasonal changes in biomass of Ruppia cirrhosa and Potamogeton pectinatus were studied in Tancada Lagoon (Ebro Delta, NE Spain) in order to reveal the environmental factors controlling their population development. Ruppia cirrhosa occupy a larger area of the lagoon than Potarnogeton pectinatus. Their maximum above ground biomasses are also different (495 g m-2 and 351 g m-2 ash free dry weight, respectively). Below ground biomass of Ruppia cirrhosa is between 9 and 53 % of the above ground biomass, while it is 3-40 % for Potamogeton pectinatus. Chlorophyll a contents show fluctuations similar to biomass. Low salinity and high turbidity caused by freshwater inflows favour Potamogeton expansion, while Ruppia development is favoured by high salinity and transparent water.

Dans le cadre de la recherche de nouveaux composés naturels, les métabolites secondaires de plantes aquatiques indigènes, les potamots Potamogeton pectinatus L., P. lucens L., P. perfoliatus L. et P. crispus L. (Potamogetonaceae), ont été étudiés. Par leur position écologique et évolutive particulière entre environnement terrestre et aquatique, les plantes aquatiques ou macrophytes pourraient en effet avoir sélectionné des composés avec des caractéristiques originales. Les extraits dichlorométhaniques (apolaires) des potamots ont été analysés par HPLCUV, HPLC-MS, HPLC-RMN et GC-MS, et testés contre diverses cibles biologiques. Sur la base de ces résultats, les extraits apolaires de P. pectinatus et P. lucens ont été étudiés de manière plus approfondie. Ils ont été fractionnés sur des colonnes ouvertes et par VLC, LPLC, MPLC, CPC et HPLC semi-préparative. Une partie de leurs constituants ont été isolés et leurs structures déterminées par des méthodes spectroscopiques, en particulier par RMN et par MS. Quinze composés ont été ainsi isolés de P. pectinatus et P. lucens, dont sept sont des nouveaux produits naturels. Parmi ces quinze produits, neuf sont des diterpènes ent-labdanes contenant un noyau furane ou un groupe lactonique, dont six sont décrits ici pour la première fois. Certains de ces diterpènes ont montré une activité algicide, ce qui indique une de leurs fonctions possible dans les potamots, et un de ces labdanes, le méthyl-15,16-époxy-12-oxo-8(17),13(16),14-ent-labatrièn-19-oate, a également des propriétés anti-inflammatoires. Les composés présents dans les extraits méthanoliques (polaires) n?ont pas été isolés, mais quatorze d?entre eux ont pu être identifiés par HPLC-UV, HPLC-MS et HPLCRMN. Une majorité de ces constituants sont des flavonoïdes connus, des dérivés glycosylés de l?apigénine, la lutéoline et le chrysoériol, également présents en tant qu?aglycones. Plusieurs ent-labdanes glycosylés ont pu être également identifiés dans ces extraits, parmi lesquels un nouveau composé dont la structure a pu être partiellement déterminée. En conclusion, ce travail a permis de mieux connaître la phytochimie de plusieurs plantes aquatiques de Suisse, et d?isoler de nouveaux produits naturels avec des propriétés biologiques et pharmacologiques intéressantes.<br/><br/>The secondary metabolites of Swiss freshwater plants, the pondweeds Potamogeton pectinatus L., P. lucens L., P. perfoliatus L. and P. crispus L. (Potamogetonaceae), were investigated. Because of their peculiar habitat, in-between aquatic and terrestrial life, aquatic plants should produce secondary metabolites with original chemical or biological features. Their apolar extracts were analysed by HPLC-UV, HPLC-MS, HPLC-NMR and GCMS, and were tested with different bioassays. Based on these results, the apolar extracts of P. pectinatus and P. lucens were investigated more extensively. They were fractionated on open columns, and by VLC, LPLC, MPLC, CPC and semi-preparative HPLC. Their constituents were isolated and their structures elucidated by spectroscopic methods as MS and NMR. Fifteen compounds could be isolated from P. pectinatus and P. lucens, and seven were new natural products. Nine of them were ent-labdane diterpenes with a furan moiety or a lactone group, and six of these labdanes were reported here for the first time as natural products. Some of these diterpenes showed an algaecide effect. This activity indicated their potential ecological function in pondweeds. One compound, methyl-15,16-epoxy-12-oxo-8(17),13(16),14-ent-labatrien-19-oate, revealed also some anti-inflammatory properties. The constituents of polar extracts were not isolated, but fourteen of them could be identified by HPLC-UV, HPLC-MS and HPLC-NMR. The major part of these compounds was known flavonoids as apigenin, lutolin, chrysoeriol and their glycosylated derivatives. Several glycosylated ent-labdanes were also identified, and the structure of a new labdane dihexoside was partially elucidated. In conclusion this study allowed a better knowledge of the phytochemistry of Swiss aquatic plants, and the isolation of new natural products with interesting biological and pharmacological properties.

O cobre é um metal essencial para as plantas, porém considerado tóxico quando em elevadas concentrações na água. No caso de macrófitas aquáticas já foi demonstrado que este metal inibe o processo fotossintético e provoca alterações pigmentares. Neste contexto, expusemos (96h) a macrófita aquática Potamogeton pectinatus (L.) à diferentes concentrações de cobre 1, 10, 100 e 1000 µM para avaliar o potencial bioacumulador da planta, e às concentrações de 1, 10 e 100 µM de Cu para verificar os possíveis efeitos do metal na taxa fotossintética (24 e 96h) em diferentes intensidades luminosas (17, 100, 300 e 500 µmol/m2 /s), no teor pigmentar (96h) e no crescimento das macrófitas (30 dias). Para os experimentos de bioacumulação mantivemos um grupo de plantas controle (sem adição de cobre no meio), enquanto que para os outros testes mantivemos um grupo controle e um grupo em solução nutritiva de Hoagland 100%, que contem cobre e outros micronutrientes em concentrações ideais para sobrevivência e crescimento de P. pectinatus. Nossos resultados mostram que a macrófita P. pectinatus é capaz de acumular altas concentrações de cobre, sendo que este acúmulo aumenta com a elevação dos níveis do metal na água. Apesar de basicamente não haver diferença estatística entre a concentração do metal nos diferentes órgãos da planta, as raízes mostraram-se capazes de acumular mais cobre que as folhas e caule com base no fator de bioconcentração. Com relação aos teores de clorofila “a”, “b” e carotenoides, estes foram menores nas folhas das plantas controle em comparação com as plantas em solução de Hogland, mas esta diferença só foi significativa nas plantas expostas ao cobre, que apresentaram menor concentração dos teores pigmentares já à 1 µM de Cu. Quanto à fotossíntese, em 24h de exposição, novamente observamos um efeito negativo da ausência e presença de cobre nas concentrações de 1, 10 e 100 µM, bem como, um efeito da luminosidade, de forma que as plantas em solução de Hoagland apresentaram maior taxa fotossintética quando em 100 µmol/m2 /s. Em virtude de um aumento na respiração em 96h, a fotossíntese, quando ocorreu, foi menor que em 24h e não diferiu entre os grupos e luminosidade. Em relação ao crescimento, as plantas perderam biomassa, mas mantiveram seus comprimentos e apenas aquelas em solução de Hoagland aumentaram seu número de folhas. Ainda, verificou-se clorose e necrose nas plantas controle e expostas ao cobre. Diante do exposto, concluímos que a macrófita P. pectinatus acumula altas concentrações de cobre, principalmente na raiz, sendo capaz e refletir as concentrações do metal no meio. Esta condição, sugere seu uso no biomonitoramento e fitorremediação de locais contaminados por cobre. Por outro lado, elas mostram-se sensíveis ao metal pela redução no teor pigmentar e fotossíntese, sugerindo estes como mecanismos toxicidade do cobre.

Gammarus aequicauda and Sphaeroma hookeri are the most abundant macroinvertebrates on Ruppia cirrhosa (1275 and 1290 individuals.m-2) and Potamogeton pectinatus (140 and 680 individuals. m-2) in Tancada lagoon, a Mediterranean coastal lagoon in the Ebro Delta (NE Spain). Consumption and assimilation efficiencies were calculated from bell jar experiments. Gammarus grazing effects are higher than Sphaeroma on both Ruppia cirrhosa and Potamogeton pectinatus. Green Ruppia leaves lost 0.3 mg per mg Gammarus per day, while Potamogeton leaves lost 0.2 mg per mg Gammarus per day. Decomposing Ruppia leaves lost 0.35-0.54 mg per mg Gammarus per day. Losses of weight by both Ruppia and Potamogeton due to Sphaeroma feeding were less than half those by Gammarus. Assimilation efficiencies are higher for Gammarus (44-78% feeding on Ruppia 2% feeding on Potamogeton) than for Sphaeroma (26-48%) feeding on Ruppia). These assimilation efficiencies were higher than those reported by other authors working at higher latitudes perhaps because of the higher temperature under which the experiments were carried out.

This work aims to characterise the current autotrophic compartment of the Albufera des Grau coastal lagoon (Menorca, Balearic Islands) and to assess the relationship between the submerged macrophytes and the limnological parameters of the lagoon. During the study period the submerged vegetation was dominated by the macrophyte Ruppia cirrhosa, which formed dense extensive meadows covering 79% of the surface. Another macrophyte species, Potamogeton pectinatus, was also observed but only forming small stands near the rushing streams. Macroalgae were only occasionally observed. Macrophyte biomass showed a clear seasonal trend, with maximum values in July. The biomass of R. cirrhosa achieved 1760 g DW m-2, the highest biomass ever reported for this species in the literature. The seasonal production-decomposition cycle of the macrophyte meadows appears to drive the nutrient dynamics and carbon fluxes in the lagoon. Despite the significant biomass accumulation and the absence of a washout of nutrients and organic matter to the sea, the lagoon did not experience a dystrophic collapse. These results indicate that internal metabolism is more important than exchange processes in the lagoon.

The biological variation in nature is called biodiversity. Anthropogenic pressures have led to a loss of biodiversity, alarming scientists as to what consequences declining diversity has for ecosystem functioning. The general consensus is that diversity (e.g. species richness or identity) affects functioning and provides services from which humans benefit. The aim of this thesis was to investigate how aquatic plant species richness and identity affect ecosystem functioning in terms of processes such as primary production, nutrient availability, epifaunal colonization and properties e.g. stability of Zostera marina subjected to shading. The main work was carried out in the field and ranged temporally from weeklong to 3.5 months-long experiments. The experimental plants used frequently co-occur in submerged meadows in the northern Baltic Sea and consist of eelgrass (Z. marina), perfoliate pondweed (Potamogeton perfoliatus), sago pondweed (P. pectinatus), slender-leaved pondweed (P. filiformis) and horned pondweed (Zannichellia palustris). The results showed that plant richness affected epifaunal community variables weakly, but had a strong positive effect on infaunal species number and functional diversity, while plant identity had strong effects on amphipods (Gammarus spp.), of which abundances were higher in plant assemblages consisting of P. perfoliatus. Depending on the starting standardizing unit, plant richness showed varying effects on primary production. In shoot density-standardized plots, plant richness increased the shoot densities of three out of four species and enhanced the plant biomass production. Both positive complementarity and selection effects were found to underpin the positive biodiversity effects. In shoot biomass-standardized plots, richness effects only affected biomass production of one species. Negative selection was prevalent, counteracting positive complementarity, which resulted in no significant biodiversity effect. The stability of Z. marina was affected by plant richness in such that Z. marina growing in polycultures lost proportionally less biomass than Z. marina in monocultures and thus had a higher resistance to shading. Monoculture plants in turn gained biomass faster, and thereby had a faster recovery than Z. marina growing in polycultures. These results indicate that positive interspecific interactions occurred during shading, while the faster recovery of monocultures suggests that the change from shading stress to recovery resulted in a shift from positive interactions to resource competition between species. The results derived from this thesis show that plant diversity affects ecosystem functioning and contribute to the growing knowledge of plant diversity being an important component of aquatic ecosystems. Diverse plant communities sustain higher primary productivity than comparable monocultures, affect faunal communities positively and enhance stability. Richness and identity effects vary, and identity has generally stronger effects on more variables than richness. However, species-rich communities are likely to contain several species with differing effects on functions, which renders species richness important for functioning. Mixed meadows add to coastal ecosystem functioning in the northern Baltic Sea and may provide with services essential for human well-being.

Intestinal microbial community is involved in the pathogenesis of Crohn's disease, but knowledge of its potential abnormalities has been limited by the impossibility to grow many dominant intestinal bacteria. Using sequence analysis of randomly cloned bacterial 16S ribosomal DNA, the dominant faecal species from four Crolin's disease patients and four controls were compared. Whereas marked inter-individual differences were observed in the faecal microflora of patients, three remained distantly related to controls on the basis of their operational taxonomic unit composition. Bacteroides vidgatus and closely related organisms represented the only molecular species shared by all patients and exhibited an unusually high rate of occurrence. Escherichia coli clones were isolated only in two patients with ileocolonic Crohn's disease. Moreover, numerous clones belonged to phylogenetic groups or species that are commonly not dominant in the faecal microflora of healthy subjects: Pectinatus, Sutterella, Verritcomicrobium, Fusobacterium, Clostridium disporicum, clostridium glycolicum, Clostridium ramosum, Clostridium innocuum and Clostridium perfringens. The subfamily Tetragonopterinae is composed by a large number of species distributed in South and Central America. This subfamily has many taxonomic and phylogenetic problems, being considered by several authors as an artificial group. With the objective to better understanding the relationships among the components of this fish group, cytogenetic studies were conduced on five species of Tetragonopterinae. Astyanax janeiroensis had 2n=50 chromosomes (6M+14SM+14ST+16A), Hyphessobrycon reticulatus had 2n=50 chromosomes (14M+20SM+16ST), Hollandichthys multifasciatus had 2n=50 chromosomes (10M+12SM+28ST), Ctenobrycon hauxwellianus had 2n=50 chromosomes (10M+6SM+34ST), and Phenacogaster cf. pectinatus had 2n=46 chromosomes (12M+2ST+32A). Only A. janeiroensis had multiple NORs, while all other species had simple NORs. Small heterochromatic blocks were observed in the chromosomes of all species in a pericentromeric position. A. janeiroensis also had some chromosomes with large heterochromatic blocks in a terminal position and a pair with an interstitial block. The karyotypic evolution of each genus is discussed.

Ocean Drilling Program (ODP) Leg 114 recovered nannofossil-bearing sediments from seven sites in the high latitudes of the South Atlantic Ocean. Cretaceous sections were recovered from Sites 698 and 700, located on the Northeast Georgia Rise and its lower flanks, respectively. These contain distinctive high-latitude nannofossil floras similar to those from high-latitude areas of the Northern Hemisphere. Most of the biostratigraphic datums used to date the upper Campanian to Maestrichtian interval appear to lie at approximately the same level in both hemispheres. The FAD of Nephrolithus frequens is confirmed to be diachronous with an earlier occurrence in high latitudes. The LAD of Monomarginatus primus n. sp. also appears to be diachronous with a later LAD in the high latitudes of the Southern Hemisphere. Fossiliferous Paleocene to lowermost Miocene sediments were recovered at all seven sites, from the Northeast Georgia Rise in the west to the Meteor Rise in the east. These nannofossil floras, although restricted in diversity and only poorly preserved, are sufficiently distinctive to allow the recognition of 19 zones and three subzones, which are used to date and correlate the cores recovered. Only Site 704 on the Meteor Rise yielded a substantial section of Miocene to Quaternary nannofossil-rich sediments. The nannofossil floras of this section are of very low diversity, with usually fewer than eight species present. Some stratigraphic ranges of important biostratigraphic datum species are observed to be different in the high-latitude sections from those recorded from low-latitude areas. The LAD of Reticulofenestra bisecta, when calibrated by magnetostratigraphy, appears to occur earlier in Hole 699A (within Chron C6CR) than in Hole 703A and possibly Hole 704B and in other published accounts of lower latitude sites in the South Atlantic. The FAD of Nannotetrina fulgens/N. cristata appears to occur later in Hole 702B (Chron C20R) than it does in other published accounts of lower latitude sites in the South Atlantic. Diachroneity is also suspected in the stratigraphic ranges of Chiasmolithus solitus and Chiasmolithus oamaruensis, although poor magnetostratigraphic results through the critical interval prevent confirmation of this. Differences in the relative stratigraphic ranges of lsthmolithus recurvus and Cribrocentrum coenurumlC. reticulatum at Sites 699 and 703 are noted. These possibly suggest warmer surface waters on the eastern side (Site 703) of the middle to late Eocene South Atlantic than those on the western side (Site 699). The diversities of the nannofossil floras and the presence of the warm-water genera Discoaster, Sphenolithus, Helicosphaera, and Amaurolithus reflect the changing surface water temperatures throughout the Cenozoic. Warmer periods are inferred for the late Paleocene to early middle Eocene, late middle Eocene to late Eocene, latest Oligocene to earliest Miocene, and possibly the Pliocene. Colder periods are inferred for the middle Eocene, most of the Oligocene, and the Miocene. Dramatic changes in the nannofossil floras of the Pleistocene of Site 704 are thought to reflect a rapidly changing environment. Monomarginatus primus, a new species from the Upper Cretaceous strata of Hole 700B, is described.

In der Döberitzer Heide nördlich von Potsdam wurden vegetationsgeschichtliche Untersuchungen durchgeführt. Das Untersuchungsgebiet befindet sich im östlichen Teil der Nauener Platte, die bisher vegetationsgeschichtlich weitgehend unerforscht war. In sechs verschiedenen Mooren wurden acht Bohrungen niedergebracht. Die Bohrkerne wurden stratigraphisch und pollenanalytisch untersucht und für die Radiocarbondatierung beprobt. Die Pollendiagramme ermöglichen die Rekonstruktion der Vegetationsentwicklung der terrestrischen Standorte und der Moore in der Döberitzer Heide in den letzten 14.000 Jahren. Neben einer Revision der Gliederungsprinzipien der spätglazialen Vegetationsentwicklung Brandenburgs und einer vergleichenden Betrachtung der Moorentwicklung in der Döberitzer Heide wurde besonderes Augenmerk auf die Geschichte des Döberitzer Lindenwaldes gerichtet, der einen Sonderfall in der brandenburgischen Vegetation darstellt. Die Untersuchungen boten die Möglichkeit, die Ursachen seiner Entstehung zu klären, Aussagen zu den Perspektiven seiner Entwicklung zu treffen und mögliche Entwicklungspotentiale von Lindenwäldern im Land Brandenburg aufzuzeigen.
