Compression and section of the facial nerve were performed in 48 rats in order to study the anatomopathological alterations occurring after daily intraperitoneal injections of 100 mg of exogenous gangliosides (Sinaxial®) for 45, 90, 180 days. In groups submitted to nerve compression, the histopathological changes were discrete and in the 180-day subgroups the nerve was practically normal. In animals submitted to section and neurorrhaphy there was formation of an amputation neuroma, a granuloma around the suture, axonal unstructuration and inter and perineural fibrosis. No significant differences were observed between the groups submitted or not to injection of exogenous gangliosides, indicating that the major factors involved in the quality of nerve regeneration were the technique and the formation of fibrosis and of an amputation neuroma.

PURPOSE: Malignant ascites is debilitating for patients with advanced cancer. As shown previously, tumour cell production of vascular endothelial growth factor might be a major cause of the formation of malignant ascites. Intraperitoneal bevacizumab could therefore be an option for symptom control in refractory ascites. PATIENTS AND METHODS: Patients with advanced gastrointestinal cancer and malignant ascites who had undergone paracentesis at least twice within the past 4 weeks were randomly assigned in a 2:1 ratio to intraperitoneal bevacizumab (400 mg absolute) or placebo after paracentesis. During the 8-week treatment period, a minimum interval of 14 d was kept between the applications of the study drug. Primary end-point was paracentesis-free survival (ParFS). RESULTS: Fifty-three patients (median age 63 years) were randomised. Forty-nine patients received at least one study drug application and qualified for the main analysis. The proportion of patients with at least one common toxicity criteria grade III-V event was similar with 20/33 (61%) on bevacizumab and 11/16 (69%) on placebo. Median ParFS was 14 d (95% confidence interval [CI]: 11-17) in the bevacizumab arm and 10.5 d (95% CI: 7-21) on placebo (hazard ratio 0.74, 95% CI: 0.40-1.37; P = 0.16). The longest paracentesis-free period was 19 d on bevacizumab (range 6-66 d) and 17.5 d in the placebo arm (range 4-42) (P = 0.85). Median overall survival was 64 d (95% CI: 45-103) on bevacizumab compared to 31.5 d (95% CI: 20-117) on placebo (P = 0.31). CONCLUSION: Intraperitoneal bevacizumab was well tolerated. Overall, treatment did not result in a significantly better symptom control of malignant ascites. However, patients defined by specific immune characteristics may benefit.

Rapid ascent to high altitude causes illness and fatigue, and there is a demand for effective acute treatments to alleviate such effects. We hypothesized that increased oxygen delivery to the tissue using a combination of a hypertensive agent and an endothelin receptor A antagonist drugs would limit exercise-induced fatigue at simulated high altitude. Our data showed that the combination of 0.1 mg/kg ambrisentan with either 20 mg/kg ephedrine or 10 mg/kg methylphenidate significantly improved exercise duration in rats at simulated altitude of 4,267 m, whereas the individual compounds did not. In normoxic, anesthetized rats, ephedrine alone and in combination with ambrisentan increased heart rate, peripheral blood flow, carotid and pulmonary arterial pressures, breathing rate, and vastus lateralis muscle oxygenation, but under inspired hypoxia, only the combination treatment significantly enhanced muscle oxygenation. Our results suggest that sympathomimetic agents combined with endothelin-A receptor blockers offset altitude-induced fatigue in rats by synergistically increasing the delivery rate of oxygen to hypoxic muscle by concomitantly augmenting perfusion pressure and improving capillary conductance in the skeletal muscle. Our findings might therefore serve as a basis to develop an effective treatment to prevent high-altitude illness and fatigue in humans.

OBJECTIVE-We studied whether manganese-enhanced high-field magnetic resonance (MR) imaging (MEHFMRI) could quantitatively detect individual islets in situ and in vivo and evaluate changes in a model of experimental diabetes.RESEARCH DESIGN AND METHODS-Whole pancreata from untreated (n = 3), MnCl(2) and glucose-injected mice (n = 6), and mice injected with either streptozotocin (STZ; n = 4) or citrate buffer (n = 4) were imaged ex vivo for unambiguous evaluation of islets. Exteriorized pancreata of MnCl(2) and glucose-injected mice (n = 6) were imaged in vivo to directly visualize the gland and minimize movements. In all cases, MR images were acquired in a 14.1 Testa scanner and correlated with the corresponding (immuno)histological sections.RESULTS-In ex vivo experiments, MEHFMRI distinguished different pancreatic tissues and evaluated the relative abundance of islets in the pancreata of normoglycemic mice. MEHFMRI also detected a significant decrease in the numerical and volume density of islets in STZ-injected mice. However, in the latter measurements the loss of beta-cells was undervalued under the conditions tested. The experiments on the externalized pancreata confirmed that MEHFMRI could visualize native individual islets in living, anesthetized mice.CONCLUSIONS-Data show that MEHFMRI quantitatively visualizes individual islets in the intact mouse pancreas, both ex vivo and in vivo. In addition to being instrumental to the protection of mucosal epithelia, secretory IgA (SIgA) adheres to and is transported by intestinal Peyer's patch (PP) M cells. The possible functional reason for this transport is unknown. We have thus examined in mice the outcome of SIgA delivered from the intestinal lumen to the cells present in the underlying organized mucosa-associated lymphoreticular tissue. We show selective association of SIgA with dendritic cells and CD4(+) T and B lymphocytes recovered from PP in vitro. In vivo, exogenously delivered SIgA is able to enter into multiple PP lining the intestine. In PP, SIgA associates with and is internalized by dendritic cells in the subepithelial dome region, whereas the interaction with CD4(+) T cells is limited to surface binding. Interaction between cells and SIgA is mediated by the IgA moiety and occurs for polymeric and monomeric molecular forms. Thus, although immune exclusion represents the main function of SIgA, transport of the Ab by M cells might promote Ag sampling under neutralizing conditions essential to the homeostasis of mucosal surfaces.

Most manufactured foods contain chemicals added as a deliberate part of the manufacturing process. The aims of the present study were to evaluate the mutagenicity and antimutagenicity of annatto, a natural pigment extracted from the Bixa orellana L. and widely used as a colorant in foods. The micronucleus test was performed in bone marrow cells from Swiss male mice treated with one of the three concentrations of annatto (1330, 5330 and 10,670 ppm), incorporated into the diet. The animals were fed with the diets for 7 days and sacrificed 24 h after the last treatment. For the evaluation of the antimutagenic potential of annatto, at day 7, the animals received an intraperitoneal injection of cyclophosphamide (50 mg/kg body weight). Under the concentrations tested annatto did not present mutagenic or antimutagenic activities on the mice bone marrow cells. However, an increased frequency of micronucleated cells was observed when the highest concentration (10,670 ppm) was administered simultaneously with cyclophosphamide. In conclusion, the data indicate that annatto colour, for the conditions used, is neither mutagenic nor an inhibitor of induced mutations, although it should be used carefully since high doses may increase the effect of a mutagen. © 2003 Elsevier Science Ltd. Recombinant glucagon-like peptide-1 (7–36)amide (rGLP-1) was recently shown to cause significant weight loss in type 2 diabetics when administered for 6 weeks as a continuous subcutaneous infusion. The mechanisms responsible for the weight loss are not clarified. In the present study, rGLP-1 was given for 5d by prandial subcutaneous injections (PSI) (76nmol 30min before meals, four times daily; a total of 302·4nmol/24h) or by continuous subcutaneous infusion (CSI) (12·7nmol/h; a total of 304·8nmol/24h). This was performed in nineteen healthy obese subjects (mean age 44·2 (sem 2·5) years; BMI 39·0 (sem 1·2)kg/m2) in a prospective randomised, double-blind, placebo-controlled, cross-over study. Compared with the placebo, rGLP-1 administered as PSI and by CSI generated a 15% reduction in mean food intake per meal (P=0·02) after 5d treatment. A weight loss of 0·55 (sem 0·2) kg (P<0·05) was registered after 5d with PSI of rGLP-1. Gastric emptying rate was reduced during both PSI (P<0·001) and CSI (P<0·05) treatment, but more rapidly and to a greater extent with PSI of rGLP-1. To conclude, a 5d treatment of rGLP-1 at high doses by PSI, but not CSI, promptly slowed gastric emptying as a probable mechanism of action of increased satiety, decreased hunger and, hence, reduced food intake with an ensuing weight loss.

Cancer-associated proteases promote peritoneal dissemination and chemoresistance in malignant progression. In this study, kallikrein-related peptidases 4, 5, 6, and 7 (KLK4-7)-cotransfected OV-MZ-6 ovarian cancer cells were embedded in a bioengineered three-dimensional (3D) microenvironment that contains RGD motifs for integrin engagement to analyze their spheroid growth and survival after chemotreatment. KLK4-7-cotransfected cells formed larger spheroids and proliferated more than controls in 3D, particularly within RGD-functionalized matrices, which was reduced upon integrin inhibition. In contrast, KLK4-7-expressing cell monolayers proliferated less than controls, emphasizing the relevance of the 3D microenvironment and integrin engagement. In a spheroid-based animal model, KLK4-7-overexpression induced tumor growth after 4 weeks and intraperitoneal spread after 8 weeks. Upon paclitaxel administration, KLK4-7-expressing tumors declined in size by 91% (controls: 87%) and showed 90% less metastatic outgrowth (controls: 33%, P<0.001). KLK4-7-expressing spheroids showed 53% survival upon paclitaxel treatment (controls: 51%), accompanied by enhanced chemoresistance-related factors, and their survival was further reduced by combination treatment of paclitaxel with KLK4/5/7 (22%, P=0.007) or MAPK (6%, P=0.006) inhibition. The concomitant presence of KLK4-7 in ovarian cancer cells together with integrin activation drives spheroid formation and proliferation. Combinatorial approaches of paclitaxel and KLK/MAPK inhibition may be more efficient for late-stage disease than chemotherapeutics alone as these inhibitory regimens reduced cancer spheroid growth to a greater extent than paclitaxel alone.

Tumor cells in ascites are a major source of disease recurrence in ovarian cancer patients. In an attempt to identify and profile the population of ascites cells obtained from ovarian cancer patients, a novel method was developed to separate adherent (AD) and non-adherent (NAD) cells in culture. Twenty-five patients were recruited to this study; 11 chemonaive (CN) and 14 chemoresistant (CR). AD cells from both CN and CR patients exhibited mesenchymal morphology with an antigen profile of mesenchymal stem cells and fibroblasts. Conversely, NAD cells had an epithelial morphology with enhanced expression of cancer antigen 125 (CA125), epithelial cell adhesion molecule (EpCAM) and cytokeratin 7. NAD cells developed infiltrating tumors and ascites within 12-14 weeks after intraperitoneal (i.p.) injections into nude mice, whereas AD cells remained non-tumorigenic for up to 20 weeks. Subsequent comparison of selective epithelial, mesenchymal and cancer stem cell (CSC) markers between AD and NAD populations of CN and CR patients demonstrated an enhanced trend in mRNA expression of E-cadherin, EpCAM, STAT3 and Oct4 in the NAD population of CR patients. A similar trend of enhanced mRNA expression of CD44, MMP9 and Oct4 was observed in the AD population of CR patients. Hence, using a novel purification method we demonstrate for the first time a distinct separation of ascites cells into epithelial tumorigenic and mesenchymal non-tumorigenic populations. We also demonstrate that cells from the ascites of CR patients are predominantly epithelial and show a trend towards increased mRNA expression of genes associated with CSCs, compared to cells isolated from the ascites of CN patients. As the tumor cells in the ascites of ovarian cancer patients play a dominant role in disease recurrence, a thorough understanding of the biology of the ascites microenvironment from CR and CN patients is essential for effective therapeutic interventions.

Perfusion of liver with plasmid DNA-lipofectin complexes via the portal vein results in efficient accumulation of the vector in hepatocytes. Such hepatocytes, when administered intraperitoneally into a hepatectomized rat, repopulate the liver and express the transgene efficiently. This procedure obviates the need for large-scale hepatocyte culture for ex vivo gene transfer. Further, intraperitoneal transplantation is a simple and cost-effective strategy of introducing genetically modified hepatocytes into liver. Thus, in situ lipofection of liver and intraperitoneal transfer of hepatocytes can be developed into a novel method of non-viral ex vivo gene transfer technique that has applications in the treatment of metabolic disorders of liver and hepatic gene therapy.

O objetivo deste trabalho foi analisar as alterações estruturais e bioquímicas na parede da bexiga resultante do tratamento crônico de ratos pré-púberes com altas doses de corticosterona. Foram estudados 26 ratos Wistar divididos em quatro grupos: T30 foi tratado com corticosterona até 29 dias de idade e morto no dia 30. T65 recebeu o mesmo tratamento, mas foi morto com 65 dias. Cada grupo tinha seu próprio controle (C30 e C65). Os animais foram tratados com injeções intraperitoneais diárias de corticosterona (2 mg/100g peso corporal) entre o 7 e 29 dias de vida. A bexigas foram removidas e processadas para inclusão em parafina. Foram estudados os seguintes elementos da parede vesical: Colágeno, músculo liso, fibras do sistema elástico, densidade vascular e do epitélio. Estes parâmetros foram analisados por métodos morfométricos, imunofluorescência e bioquímica. A densidade vascular na lâmina foi reduzida em 40% (p <0,05) no grupo T65. A organização do colágeno foi alterada em T30 e T65, apesar da concentração de colágeno total não ser alterada. O grupo T65 teve um aumento de fibras do sistema elástico. Não houve diferença na altura e na densidade de células epiteliais entre os grupos. Quanto à densidade de fibras musculares lisas, observamos um aumento de 19% (p <0,05) no grupo T65. A administração de corticosterona na fase pré-púbere provoca modificações estruturais na bexiga de ratos afetando de modo significativo o substrato morfológico sob qual repousa a fisiologia vesical. Foi observado também que estas modificações normalmente aparecem num tempo mais longo após o término do tratamento

O consumo de tabaco está freqüentemente associado ao consumo de álcool e a maioria dos usuários inicia o uso e abuso destas drogas na adolescência. Como a ansiedade pode ser um fator de risco para a etiologia do uso e da retirada destas drogas, no presente estudo investigamos os efeitos da fumaça do cigarro e/ou exposição ao etanol durante a adolescência nos níveis de ansiedade ao final do período de exposição e em período curto e longo após retirada. Do 30 ao 45 dia de vida pós-natal (PN30-PN45), camundongos Suíços de ambos os sexos foram expostos a fumaça do cigarro gerada a partir da queima de cigarros para pesquisa contendo 1,7mg de nicotina 8 h/dia e/ou ao etanol (2g/kg, 25%, v/v) através de injeção intraperitoneal em dias alternados. Assim foram utilizados 4 grupos experimentais: 1) SMK+ETOH (exposição a fumaça de cigarro e injeção i.p. de etanol); 2) SMK (exposição a fumaça de cigarro e a salina i.p.); 3) ETOH (exposição a ar e injeção i.p. de etanol); 4) VEH (exposição a ar e injeção i.p. salina). Ao final do período de exposição (PN44-45) e durante a retirada de curta (PN49-50) e longa duração (PN74-75) utilizamos o Labirinto em Cruz Elevado (LCE) para avaliar os níveis de ansiedade e o Teste de Campo Aberto (CA) para avaliar os níveis de ansiedade e a atividade locomotora. Ao final do período de exposição, as fêmeas do grupo SMK apresentaram uma resposta ansiolítica no LCE. Esta resposta foi intensificada pela co-exposição ao etanol. A retirada de curto prazo da fumaça do cigarro provocou um efeito ansiogênico. Os machos do grupo ETOH apresentaram um efeito ansiogênico no LCE ao final do período de exposição, no entanto, este efeito não persistiu em um período curto de retirada. Embora nem os efeitos da fumaça de cigarro, nem os efeitos do etanol tenham persistido após um período longo de retirada, os animais do grupo SMK+ETOH apresentaram uma resposta ansiogênica no CA. Assim, nossos resultados sugerem que fêmeas adolescentes são mais susceptíveis aos efeitos ansiolíticos do fumo do tabaco. O efeito mais intenso no grupo SMK+ETOH sugere que as fêmeas co-usam estas drogas para chegar a níveis mais baixos de ansiedade. Além disso, um aumento da ansiedade após um período longo da retirada conjunta do cigarro e do etanol, pode facilitar as recaídas ao uso destas drogas.

A cicatrização constitui processo complexo envolvendo diferentes sistemas biológicos e imunológicos , sendo essencial para manter a integridade do organismo. Três fases bem definidas ocorrem, a inflamatória, a proliferativa e a de maturação. A falha ou prolongamento em uma fase pode resultar em retardo da cicatrização tecidual. A sutura dos tecidos e sua cicatrização é um dos fundamentos básicos da cirurgia e a procura de substâncias que melhorem este processo é um desafio constante. O uso de substâncias de plantas têm sido testados por vários autores. Objetivo - Analisar comparativamente as alterações macroscópicas e histológicas proporcionadas pelo uso do extrato bruto da Jatropha gossypiifolia intraperitonial, na cicatrização de suturas realizadas na bexiga urinária de ratos. Material e método Sessenta ratos da linhagem Wistar, adultos, machos foram distribuídos em 2 grupos animais. O procedimento experimental constituiu-se em laparotomia mediana infraumbelical, incisão longitudinal de 1cm na parede ventral da bexiga e síntese em plano único com pontos separados de poliglactina 910 5-0 (Ethicon). O procedimento nos animais do grupo controle (ratos 1 a 30) instilou-se na cavidade peritonial água destilada na proporção de 1ml por kg de peso e no grupo Jatropha (ratos 31 a 60) utiilizou-se o extrato bruto de Jatropha gossypiifolia na proporção de 1ml por kg de peso, que representava 200mg do fototerápico, intraperitoneal. Cada grupo foi dividido em 3 subgrupos de 10 animais sendo estes submetidos à eutanásia no 7 e 14 pós-operatório. Foi feita análise macroscópica e histológica comparativa entre os subgrupos . Resultados - No 7 dia foi observado diferença estatisticamente significativa nas variáveis inflamação aguda, neoformação vascular e colagenização, sendo a primeira maior no grupo controle e as duas últimas no grupo Jatropha; no 14 variáveis inflamação aguda e proliferação fibroblástica apresentaram-se mais intensas com significado estatístico no grupo controle. No 21 dia as variantes se alinharam não havendo diferença estatística na sua comparação. Conclusão - Foi observado homogeneidade na cicatrização nos 2 grupos, contudo, a mesma foi mais intensa no grupo controle. Não se observou, portanto, efeito favorecedor cicatrizante do extrato bruto da Jatropha gossypiifolia intraperitonial, em dose única, na bexiga urinária de rato.
