Microsatellites or simple sequence repeats (SSRs) are ubiquitous in eukaryotic genomes. Single-locus SSR markers have been developed for a number of species, although there is a major bottleneck in developing SSR markers whereby flanking sequences must be known to design 5'-anchors for polymerase chain reaction (PCR) primers. Inter SSR (ISSR) fingerprinting was developed such that no sequence knowledge was required. Primers based on a repeat sequence, such as (CA)(n), can be made with a degenerate 3'-anchor, such as (CA)(8)RG or (AGC)(6)TY. The resultant PCR reaction amplifies the sequence between two SSRs, yielding a multilocus marker system useful for fingerprinting, diversity analysis and genome mapping. PCR products are radiolabelled with P-32 or P-33 via end-labelling or PCR incorporation, and separated on a polyacrylamide sequencing gel prior to autoradiographic visualisation. A typical reaction yields 20-100 bands per lane depending on the species and primer. We have used ISSR fingerprinting in a number of plant species, and report here some results on two important tropical species, sorghum and banana. Previous investigators have demonstrated that ISSR analysis usually detects a higher level of polymorphism than that detected with restriction fragment length polymorphism (RFLP) or random amplified polymorphic DNA (RAPD) analyses. Our data indicate that this is not a result of greater polymorphism genetically, but rather technical reasons related to the detection methodology used for ISSR analysis.

Polyembryonic seeds are characterized by the development of over one embryo in the same seed, which can be zygotic and nucellar. The objective of this work was to identify the genetic origin, whether zygotic or nucellar, of seedlings of polyembryonic seeds of 'Ubá' mango tree using ISSR markers, and relating them with the vigor of the seedlings. Thus, mangos were harvested in Visconde do Rio Branco (accession 102) and Ubá (accessions 112, 138, 152 and 159), whose seeds were germinated in plastic trays filled with washed sand. Fifty days after sowing, seedlings from five seeds of each one of the accessions 102, 112, 138, 159 and from 10 seeds of the accession 152, were analyzed. These sseedlings were characterized and evaluated for plant height, stem circumference and mass of fresh aerial part and the most vigorous seedling was the one displaying at least two of these traits higher than the other seedlings from seed. Leaves were collected for genomic DNA extraction, which was amplified using seven ISSR primers previously selected based on the amplification profile and considering the number and resolution of fragments. Zygotic seedlings were found in 18 seeds, which were the most vigorous in six seeds. The results evidenced the existence of genetic variability in orchards using seedlings grown from seeds, because the farmer usually uses the most vigorous ones, assuming that this is of nucellar origin. These results also indicate that the most vigorous seedling are not always nucellar, inasmuch as of 20% of the total seeds evaluated, the zygotic seedling was the most vigorous.

Distinct genetic structure in populations of Chrysoperla externa (Hagen) (Neuroptera, Chrysopidae) shown by genetic markers ISSR and COI gene. Green lacewings are generalist predators, and the species Chrysoperla externa presents a great potential for use in biological control of agricultural pests due to its high predation and reproduction capacities, as well as its easy mass rearing in the laboratory. The adaptive success of a species is related to genetic variability, so that population genetic studies are extremely important in order to maximize success of the biological control. Thus, the present study used nuclear (Inter Simple Sequence Repeat - ISSR) and mitochondrial (Cytochrome Oxidase I - COI) molecular markers to estimate the genetic variability of 12 populations in the São Paulo State, Brazil, as well as the genetic relationships between populations. High levels of genetic diversity were observed for both markers, and the highest values of genetic diversity appear associated with municipalities that have the greatest areas of native vegetation. There was high haplotype sharing, and there was no correlation between the markers and the geographic distribution of the populations. The AMOVA indicated absence of genetic structure for the COI gene, suggesting that the sampled areas formed a single population unit. However, the great genetic differentiation among populations showed by ISSR demonstrates that these have been under differentiation after their expansion or may also reflect distinct dispersal behavior between males and females.

O objetivo deste trabalho foi estimar a diversidade genética de populações de Zabrotes subfasciatus, por meio de marcadores moleculares ISSR. Foram avaliadas 12 populações, provenientes de oito estados brasileiros, no total de 269 indivíduos. Cinco iniciadores ISSR permitiram a obtenção do total de 51 fragmentos polimórficos. A percentagem média de locos polimórficos, dentro de cada população, foi de 83,8%. A heterozigosidade corrigida de Nei esperada variou de 0,23 a 0,33, com média de 0,29, e o índice de diversidade genética de Shannon e Weaver variou de 0,29 a 0,48, com média de 0,42. No nível de espécie, estes dois índices apresentaram valores de 0,36 e 0,54, respectivamente. A análise de variância molecular mostrou que 66% da variância molecular total pode ser atribuída a diferenças intrapopulacionais e que os 34% restantes podem ser atribuídos a diferenças interpopulacionais. O teste de Mantel mostrou baixa correlação entre: distância geográfica e diferenciação genética, identidade genética e diferenciação genética e, distância genética de Nei e diferenciação genética. As populações brasileiras de Z. subfasciatus possuem baixa diferenciação genética e fraca estruturação geográfica.

O objetivo deste trabalho foi avaliar a diversidade genética intra e interespecífica de acessos de Manihot por meio de marcadores ISSR. Foram analisadas cinco espécies e duas variedades de Manihot, além de duas espécies do gênero Croton, utilizadas como grupo externo, por meio de 20 oligonucleotídeos iniciadores (Olii) ISSR UBC. Para análise do índice de similaridade entre as espécies e os acessos foram utilizados os coeficientes de Jaccard e de 'simple matching'. Os 20 Olii testados foram altamente polimórficos para todas as espécies analisadas, e 89,7% dos locus foram polimórficos. Há maior similaridade genética entre espécies diferentes de Manihot, como M. dichotoma var. undulata e M. caerulescens, do que entre indivíduos da mesma espécie, como M. dichotoma e M. dichotoma var. undulata.

A umbu-cajazeira (Spondias sp.) é uma frutífera nativa do Semiárido brasileiro, ainda em fase de domesticação, cujos frutos apresentam excelentes perspectivas de aproveitamento comercial. O objetivo deste trabalho foi caracterizar a variabilidade genética entre acessos de umbu-cajazeira pertencentes ao Banco Ativo de Germoplasma de Fruteiras Tropicais da Embrapa Mandioca e Fruticultura, por meio de marcadores moleculares ISSR (Inter Simple Sequence Repeat). Foram analisados 17 acessos de umbu-cajazeira, com 25 marcadores ISSR, os quais produziram um total de 249 bandas, sendo 201 bandas polimórficas e 48 monomórficas. As dissimilaridades genéticas entre os acessos variaram de 0,247 a 0,665, com base no coeficiente de Jaccard. O método UPGMA (Unweighted Pair-Group Method Average) agrupou os acessos em cinco grupos, sendo que 'Preciosa' e 'Suprema' foram os acessos mais similares. A maior dissimilaridade foi observada entre os acessos 'Esperança' e 'Pomar'. O alto grau de polimorfismo encontrado demonstrou a eficiência dos marcadores ISSR, indicando que estes podem ser utilizados com sucesso na caracterização molecular de germoplasma e em futuros trabalhos de melhoramento genético dessa frutífera. Existe considerável variabilidade genética entre os acessos de umbu-cajazeira presentes no BAG Fruteiras Tropicais, que pode ser explorada para a conservação e o melhoramento da espécie.

O objetivo deste trabalho foi avaliar a distância genética entre 37 acessos da espécie cultivada Psidium guajava, L. (goiaba) e de araçás do gênero Psidium do banco de germoplasma da Universidade Estadual do Norte Fluminense (UENF), via marcadores moleculares ISSR. Nos 17 marcadores selecionados, foram obtidas 216 bandas polimórficas. Pelo método de agrupamento UPGMA, houve a formação de cinco principais grupos. Os acessos de araçá da espécie P. cattleyanum Sabine , ficaram alocados nos grupos I e II. No grupo II, foi observada, dentro da espécie P cattleyanum, maior proximidade com a goiabeira. No grupo III, ficou alocado o acesso da espécie P. guineense Sw (araçá-do-campo) e dentre os araçás, foi o que ficou mais próximo da goiaba. Os genótipos de goiabeira ficaram alocados do grupo IV e V, confirmando sua alta divergência. Os marcadores moleculares foram eficientes em estimar a distância genética intra e interespecífica.

O biribazeiro é uma planta frutífera nativa das matas Atlântica e Amazônica. Seus frutos têm grande aceitação popular para consumo in natura. Objetivou-se com este estudo a avaliação da diversidade genética de acessos de biribazeiro (Rollinia mucosa [Jacq.] Baill) com a utilização de marcadores moleculares ISSR. Foram analisados 16 acessos com 20 primers ISSR, os quais produziram um total de 118 bandas, sendo 96 polimórficas e 22 monomórficas. Os valores de dissimilaridade genética, calculados de acordo com o complemento do índice de Jaccard, variaram de 0,0909 a 0,5147. O método UPGMA (Unweighted Pair Group Method Average) agrupou os acessos em seis grupos. Os acessos 1 e 5 foram mais dissimilares e 11 e 12 os menos dissimilares. Os marcadores ISSR utilizados neste estudo demonstraram eficiência na detecção de polimorfismos moleculares, revelando variabilidade genética entre os 16 acessos. Diante dos resultados obtidos neste trabalho, é possível inferir que existe considerável variabilidade genética entre os acessos de biribazeiro, demonstrando a importância dos marcadores na análise de variabilidade de espécies pouco estudadas, como Rollinia mucosa [Jacq.]Baill.

RESUMO O conhecimento da diversidade genética de espécies nativas é de grande valia quando se objetiva o melhoramento e a conservação de populações naturais. Neste sentido, o objetivodeste trabalho foi selecionar iniciadores ISSR (inter repetições de sequências simples) para Hancornia speciosa (Apocynaceae), assim como quantificar a variabilidade genética em uma população natural. Foramamostrados 15 indivíduos de uma população localizada em Natal-RN. Amostras de caule foram coletadas para a posterior extração do DNA. DNA. Para a seleção, 19 primers ISSR foram testados, dos quais seis foram eficientes, apresentando locos nítidos e em maior número (UBC 808; UBC 810; UBC 826; UBC 827; UBC 841 e UBC 842), totalizando 63 locos. Desses, apenas 30 (47,62%) apresentaram polimorfismo. O valor de PIC (conteúdo de informações polimórficas) para os primers selecionados atingiu a média de 0,37, variando de 0,26 a 0,44. A diversidade genética foi considerada baixa dentro da população, com o número de alelos observados (na =1,48), número de alelos efetivos (ne = 1,32), índice de diversidade de Nei (He = 0,18) e índice de Shannon (I = 0,26). Os padrões de diversidade alélica encontrados indicam a ocorrência de um gargalo populacional recente. A utilização de marcadores ISSR para Hancornia speciosa mostrou-se eficaz para a quantificação da diversidade genética dos indivíduos, servindo como aporte para estratégias e planos que visem à conservação e à manutenção da espécie.

Fragaria vesca is a short-lived perennial with a seasonal-flowering habit. Seasonality of flowering is widespread in the Rosaceae and is also found in the majority of temperate polycarpic perennials. Genetic analysis has shown that seasonal flowering is controlled by a single gene in F. vesca, the SEASONAL FLOWERING LOCUS (SFL). Here, we report progress towards the marker-assisted selection and positional cloning of SFL, in which three ISSR markers linked to SFL were converted to locus-specific sequence-characterized amplified region (SCAR1–SCAR3) markers to allow large-scale screening of mapping progenies. We believe this is the first study describing the development of SCAR markers from ISSR profiles. The work also provides useful insight into the nature of polymorphisms generated by the ISSR marker system. Our results indicate that the ISSR polymorphisms originally detected were probably caused by point mutations in the positions targeted by primer anchors (causing differential PCR failure), by indels within the amplicon (leading to variation in amplicon size) and by internal sequence differences (leading to variation in DNA folding and so in band mobility). The cause of the original ISSR polymorphism was important in the selection of appropriate strategies for SCAR-marker development. The SCAR markers produced were mapped using a F. vesca f. vesca × F. vesca f. semperflorens testcross population. Marker SCAR2 was inseparable from the SFL, whereas SCAR1 mapped 3.0 cM to the north of the gene and SCAR3 1.7 cM to its south.

The genus Eigenmannia (Teleostei: Gymnotiformes), a widely distributed fish genus from the Neotropical region, presents very complex morphological patterns and many taxonomic problems. It is suggested that this genus harbors a species complex that is hard to differentiate using only morphological characteristics. As a result, many species of Eigenmannia may be currently gathered under a common name. With the objective of providing new tools for species characterization in this group, an analysis of the polymorphism of DNA inter-simple sequence repeats (ISSR), obtained by single primer amplification reaction (SPAR), combined with karyotype identification, was carried out in specimens sampled from populations of the Upper Parana, So Francisco and Amazon river basins (Brazil). Specific ISSR patterns generated by primers (AAGC)(4) and (GGAC)(4) were found to characterize the ten cytotypes analyzed, even though the cytotypes 2n = 38 and 2n = 38 XX:XY, from the Upper Parana basin, share some ISSR amplification patterns. The geographical distribution of all Eigenmannia specimens sampled was inferred, showing the cytotype 2n = 31/2n = 32 as the most frequent and largely distributed in the Upper Parana basin. The cytotype 2n = 34 was reported for the first time in the genus Eigenmania, restricted to the So Francisco basin. Polymorphic ISSR patterns were also detected for each cytotype. Considering our results and the data reported previously in the literature, it is suggested that many of the forms of Eigenmannia herein analyzed might be regarded as different species. This work reinforces the importance of employing diverse approaches, such as molecular and cytogenetic characterization, to address taxonomic and evolutionary issues.
