Confocal and two-photon microcopy have become essential tools in biological research and today many investigations are not possible without their help. The valuable advantage that these two techniques offer is the ability of optical sectioning. Optical sectioning makes it possible to obtain 3D visuahzation of the structiu-es, and hence, valuable information of the structural relationships, the geometrical, and the morphological aspects of the specimen. The achievable lateral and axial resolutions by confocal and two-photon microscopy, similar to other optical imaging systems, are both defined by the diffraction theorem. Any aberration and imperfection present during the imaging results in broadening of the calculated theoretical resolution, blurring, geometrical distortions in the acquired images that interfere with the analysis of the structures, and lower the collected fluorescence from the specimen. The aberrations may have different causes and they can be classified by their sources such as specimen-induced aberrations, optics-induced aberrations, illumination aberrations, and misalignment aberrations. This thesis presents an investigation and study of image enhancement. The goal of this thesis was approached in two different directions. Initially, we investigated the sources of the imperfections. We propose methods to eliminate or minimize aberrations introduced during the image acquisition by optimizing the acquisition conditions. The impact on the resolution as a result of using a coverslip the thickness of which is mismatched with the one that the objective lens is designed for was shown and a novel technique was introduced in order to define the proper value on the correction collar of the lens. The amoimt of spherical aberration with regard to t he numerical aperture of the objective lens was investigated and it was shown that, based on the purpose of our imaging tasks, different numerical apertures must be used. The deformed beam cross section of the single-photon excitation source was corrected and the enhancement of the resolution and image quaUty was shown. Furthermore, the dependency of the scattered light on the excitation wavelength was shown empirically. In the second part, we continued the study of the image enhancement process by deconvolution techniques. Although deconvolution algorithms are used widely to improve the quality of the images, how well a deconvolution algorithm responds highly depends on the point spread function (PSF) of the imaging system applied to the algorithm and the level of its accuracy. We investigated approaches that can be done in order to obtain more precise PSF. Novel methods to improve the pattern of the PSF and reduce the noise are proposed. Furthermore, multiple soiu'ces to extract the PSFs of the imaging system are introduced and the empirical deconvolution results by using each of these PSFs are compared together. The results confirm that a greater improvement attained by applying the in situ PSF during the deconvolution process.

In this paper an adaptive approach for color image enhancement is proposed. In this approach, the saturation feedback technique is used as a means of supplementing color image shmpness and contrast. This technique of the saturation feedback can serve to bring out image details that have low luminance contrast. In the technique, the feedback parameters are the key component and are usually determined manually. In order to realize the adaptive color image enhancement, the genetic algorithm is employed to search global optimal parameters for saturation feedback automatically. The detailed procedures are described in the paper. Experimental results on color images show the feasibility of the proposed method.

Facial nerve segmentation plays an important role in surgical planning of cochlear implantation. Clinically available CBCT images are used for surgical planning. However, its relatively low resolution renders the identification of the facial nerve difficult. In this work, we present a supervised learning approach to enhance facial nerve image information from CBCT. A supervised learning approach based on multi-output random forest was employed to learn the mapping between CBCT and micro-CT images. Evaluation was performed qualitatively and quantitatively by using the predicted image as input for a previously published dedicated facial nerve segmentation, and cochlear implantation surgical planning software, OtoPlan. Results show the potential of the proposed approach to improve facial nerve image quality as imaged by CBCT and to leverage its segmentation using OtoPlan.

Nowadays, a lot of applications use digital images. For example in face recognition to detect and tag persons in photograph, for security control, and a lot of applications that can be found in smart cities, as speed control in roads or highways and cameras in traffic lights to detect drivers ignoring red light. Also in medicine digital images are used, such as x-ray, scanners, etc. These applications depend on the quality of the image obtained. A good camera is expensive, and the image obtained depends also on external factor as light. To make these applications work properly, image enhancement is as important as, for example, a good face detection algorithm. Image enhancement also can be used in normal photograph, for pictures done in bad light conditions, or just to improve the contrast of an image. There are some applications for smartphones that allow users apply filters or change the bright, colour or contrast on the pictures. This project compares four different techniques to use in image enhancement. After applying one of these techniques to an image, it will use better the whole available dynamic range. Some of the algorithms are designed for grey scale images and others for colour images. It is used Matlab software to develop and present the final results. These algorithms are Successive Means Quantization Transform (SMQT), Histogram Equalization, using Matlab function and own implemented function, and V transform. Finally, as conclusions, we can prove that Histogram equalization algorithm is the simplest of all, it has a wide variability of grey levels and it is not suitable for colour images. V transform algorithm is a good option for colour images. The algorithm is linear and requires low computational power. SMQT algorithm is non-linear, insensitive to gain and bias and it can extract structure of the data. RESUMEN. Hoy en día incontable número de aplicaciones usan imágenes digitales. Por ejemplo, para el control de la seguridad se usa el reconocimiento de rostros para detectar y etiquetar personas en fotografías o vídeos, para distintos usos de las ciudades inteligentes, como control de velocidad en carreteras o autopistas, cámaras en los semáforos para detectar a conductores haciendo caso omiso de un semáforo en rojo, etc. También en la medicina se utilizan imágenes digitales, como por ejemplo, rayos X, escáneres, etc. Todas estas aplicaciones dependen de la calidad de la imagen obtenida. Una buena cámara es cara, y la imagen obtenida depende también de factores externos como la luz. Para hacer que estas aplicaciones funciones correctamente, el tratamiento de imagen es tan importante como, por ejemplo, un buen algoritmo de detección de rostros. La mejora de la imagen también se puede utilizar en la fotografía no profesional o de consumo, para las fotos realizadas en malas condiciones de luz, o simplemente para mejorar el contraste de una imagen. Existen aplicaciones para teléfonos móviles que permiten a los usuarios aplicar filtros y cambiar el brillo, el color o el contraste en las imágenes. Este proyecto compara cuatro técnicas diferentes para utilizar el tratamiento de imagen. Se utiliza la herramienta de software matemático Matlab para desarrollar y presentar los resultados finales. Estos algoritmos son Successive Means Quantization Transform (SMQT), Ecualización del histograma, usando la propia función de Matlab y una nueva función que se desarrolla en este proyecto y, por último, una función de transformada V. Finalmente, como conclusión, podemos comprobar que el algoritmo de Ecualización del histograma es el más simple de todos, tiene una amplia variabilidad de niveles de gris y no es adecuado para imágenes en color. El algoritmo de transformada V es una buena opción para imágenes en color, es lineal y requiere baja potencia de cálculo. El algoritmo SMQT no es lineal, insensible a la ganancia y polarización y, gracias a él, se puede extraer la estructura de los datos.

Purpose: To examine the use of real-time, generic edge detection, image processing techniques to enhance the television viewing of the visually impaired. Design: Prospective, clinical experimental study. Method: One hundred and two sequential visually impaired (average age 73.8 ± 14.8 years; 59% female) in a single center optimized a dynamic television image with respect to edge detection filter (Prewitt, Sobel, or the two combined), color (red, green, blue, or white), and intensity (one to 15 times) of the overlaid edges. They then rated the original television footage compared with a black-and-white image displaying the edges detected and the original television image with the detected edges overlaid in the chosen color and at the intensity selected. Footage of news, an advertisement, and the end of program credits were subjectively assessed in a random order. Results: A Prewitt filter was preferred (44%) compared with the Sobel filter (27%) or a combination of the two (28%). Green and white were equally popular for displaying the detected edges (32%), with blue (22%) and red (14%) less so. The average preferred edge intensity was 3.5 ± 1.7 times. The image-enhanced television was significantly preferred to the original (P < .001), which in turn was preferred to viewing the detected edges alone (P < .001) for each of the footage clips. Preference was not dependent on the condition causing visual impairment. Seventy percent were definitely willing to buy a set-top box that could achieve these effects for a reasonable price. Conclusions: Simple generic edge detection image enhancement options can be performed on television in real-time and significantly enhance the viewing of the visually impaired. © 2007 Elsevier Inc. Digital Image Processing is a rapidly evolving eld with growing applications in Science and Engineering. It involves changing the nature of an image in order to either improve its pictorial information for human interpretation or render it more suitable for autonomous machine perception. One of the major areas of image processing for human vision applications is image enhancement. The principal goal of image enhancement is to improve visual quality of an image, typically by taking advantage of the response of human visual system. Image enhancement methods are carried out usually in the pixel domain. Transform domain methods can often provide another way to interpret and understand image contents. A suitable transform, thus selected, should have less computational complexity. Sequency ordered arrangement of unique MRT (Mapped Real Transform) coe cients can give rise to an integer-to-integer transform, named Sequency based unique MRT (SMRT), suitable for image processing applications. The development of the SMRT from UMRT (Unique MRT), forward & inverse SMRT algorithms and the basis functions are introduced. A few properties of the SMRT are explored and its scope in lossless text compression is presented.

Au cours des dernières décennies, l’effort sur les applications de capteurs infrarouges a largement progressé dans le monde. Mais, une certaine difficulté demeure, en ce qui concerne le fait que les objets ne sont pas assez clairs ou ne peuvent pas toujours être distingués facilement dans l’image obtenue pour la scène observée. L’amélioration de l’image infrarouge a joué un rôle important dans le développement de technologies de la vision infrarouge de l’ordinateur, le traitement de l’image et les essais non destructifs, etc. Cette thèse traite de la question des techniques d’amélioration de l’image infrarouge en deux aspects, y compris le traitement d’une seule image infrarouge dans le domaine hybride espacefréquence, et la fusion d’images infrarouges et visibles employant la technique du nonsubsampled Contourlet transformer (NSCT). La fusion d’images peut être considérée comme étant la poursuite de l’exploration du modèle d’amélioration de l’image unique infrarouge, alors qu’il combine les images infrarouges et visibles en une seule image pour représenter et améliorer toutes les informations utiles et les caractéristiques des images sources, car une seule image ne pouvait contenir tous les renseignements pertinents ou disponibles en raison de restrictions découlant de tout capteur unique de l’imagerie. Nous examinons et faisons une enquête concernant le développement de techniques d’amélioration d’images infrarouges, et ensuite nous nous consacrons à l’amélioration de l’image unique infrarouge, et nous proposons un schéma d’amélioration de domaine hybride avec une méthode d’évaluation floue de seuil amélioré, qui permet d’obtenir une qualité d’image supérieure et améliore la perception visuelle humaine. Les techniques de fusion d’images infrarouges et visibles sont établies à l’aide de la mise en oeuvre d’une mise en registre précise des images sources acquises par différents capteurs. L’algorithme SURF-RANSAC est appliqué pour la mise en registre tout au long des travaux de recherche, ce qui conduit à des images mises en registre de façon très précise et des bénéfices accrus pour le traitement de fusion. Pour les questions de fusion d’images infrarouges et visibles, une série d’approches avancées et efficaces sont proposés. Une méthode standard de fusion à base de NSCT multi-canal est présente comme référence pour les approches de fusion proposées suivantes. Une approche conjointe de fusion, impliquant l’Adaptive-Gaussian NSCT et la transformée en ondelettes (Wavelet Transform, WT) est propose, ce qui conduit à des résultats de fusion qui sont meilleurs que ceux obtenus avec les méthodes non-adaptatives générales. Une approche de fusion basée sur le NSCT employant la détection comprime (CS, compressed sensing) et de la variation totale (TV) à des coefficients d’échantillons clairsemés et effectuant la reconstruction de coefficients fusionnés de façon précise est proposée, qui obtient de bien meilleurs résultats de fusion par le biais d’une pré-amélioration de l’image infrarouge et en diminuant les informations redondantes des coefficients de fusion. Une procédure de fusion basée sur le NSCT utilisant une technique de détection rapide de rétrécissement itératif comprimé (fast iterative-shrinking compressed sensing, FISCS) est proposée pour compresser les coefficients décomposés et reconstruire les coefficients fusionnés dans le processus de fusion, qui conduit à de meilleurs résultats plus rapidement et d’une manière efficace.

In this thesis, an image enhancement application is developed for low-vision patients when they use iPhones to see images/watch videos. The thesis has two contributions. The first contribution is the new image enhancement algorithm which combines human vision features. The new image enhancement algorithm is modified from a wavelet transform based image enhancement algorithm developed by Dr. Jinshan Tang. Different from the original algorithm, the new image enhancement algorithm combines human visual feature into the algorithm and thus can make the new algorithm more effective. Experimental simulation results show that the proposed algorithm has better visual results than the algorithm without combining visual features. The second contribution of this thesis is the development of a mobile image enhancement application. In this application, users with low-vision can see clearer images on an iPhone which is installed with the application I have developed.
