Social organization is an important component of the population biology of a species that influences gene flow, the spatial pattern and scale of movements, and the effects of predation or exploitation by humans. An important element of social structure in mammals is group fidelity, which can be quantified through association indices. To describe the social organization of marine tucuxi dolphins (Sotalia guianensis) found in the Cananeia estuary, southeastern Brazil, association indices were applied to photo-identification data to characterize the temporal stability of relationships among members of this population. Eighty-seven days of fieldwork were conducted from May 2000 to July 2003, resulting in direct observations of 374 distinct groups. A total of 138 dolphins were identified on 1-38 distinct field days. Lone dolphins were rarely seen, whereas groups were composed of up to 60 individuals (mean +/- 1 SD = 12.4 +/- 11.4 individuals per group). A total of 29,327 photographs were analyzed, of which 6,312 (21.5%) were considered useful for identifying individuals. Half-weight and simple ratio indices were used to investigate associations among S. guianensis as revealed by the entire data set, data from the core study site, and data from groups composed of <= 10 individuals. Monte Carlo methods indicated that only 3 (9.3%) of 32 association matrices differed significantly from expectations based on random association. Thus, our study suggests that stable associations are not characteristic of S. guianensis in the Cananeia estuary.

A comunidade de tubarões (Selachii:Galea) proveniente da Formação Pirabas pode ser considerada como uma das mais representativas e de maior diversidade entre as unidades do Neógeno da América do Sul. A presença de 8 gêneros permitiu elaborar hipóteses sobre a reconstrução da cadeia trófica envolvendo outros elementos da paleofauna de vertebrados, assim como também serviu como um indicador paleoecológico adicional que corrobora dados anteriores sobre a reconstituição dos parâmetros paleoambientais da unidade geológica. Foi realizado uma revisão taxonômica das espécies previamente conhecidos para a Formação Pirabas, cuja composição da paleocomun idade de tubarões foi a seguinte: Carcharhinus spl Carcharhinus sp2, Carcharhinus sp3, Carcharhinus priscus, Sphyma magna, Hemipristis serra, Carcharodon megalodon, Isurus sp, Ginglyrnostoma serra, Ginglymostoma obliquum. Novas coletas possibilitaram a expansão de 31 exemplares depositados em museus brasileiros para 231 novos indivíduos, incluindo material microscópico. Esta coleta serviu para aumentar o conhecimento sobre a diversidade e aspectos paleoecológicos dos grupos representados.

The annual return, seasonal occurrence, and site fidelity of Korean-Okhotsk or western gray whales on their feeding grounds off northeastern Sakhalin Island, Russia, were assessed by boat-based photo-identification studies in 1994-1998. A total of 262 pods were observed, ranging in size from 1 to 9 whales with an overall mean of 2.0'. Sixty-nine whales were individually identified, and a majority of all whales (71.0%) were observed in multiple years. Annual sighting frequencies ranged from 1 to 18 d, with a mean of 5.4 d. The percentage of whales re-identified from previous years showed a continuous annual increase, reaching 87.0% by the end of the study. Time between first and last sighting of identified individuals within a given year was 1-85 d, with an overall mean of 40.6 d. Annual calf proportions ranged from 4.3% (1997) to 13.2% (1998), and mother-calf separations generally occurred between July and September. The seasonal site fidelity and annual return of whales to this part of the Okhotsk Sea emphasize its importance as a primary feeding ground for this endangered population.

Social networks are static illustrations of dynamic societies, within which social interactions are constantly changing. Fundamental sources of variation include ranging behaviour and temporal demographic changes. Spatiotemporal dynamics can favour or limit opportunities for individuals to interact, and then a network may not essentially represent social processes. We examined whether a social network can embed such nonsocial effects in its topology, whereby emerging modules depict spatially or temporally segregated individuals. To this end, we applied a combination of spatial, temporal and demographic analyses to a long-term study of the association patterns of Guiana dolphins, Sotalia guianensis. We found that association patterns are organized into a modular social network. Space use was unlikely to reflect these modules, since dolphins' ranging behaviour clearly overlapped. However, a temporal demographic turnover, caused by the exit/entrance of individuals (most likely emigration/immigration), defined three modules of associations occurring at different times. Although this factor could mask real social processes, we identified the temporal scale that allowed us to account for these demographic effects. By looking within this turnover period (32 months), we assessed fission-fusion dynamics of the poorly known social organization of Guiana dolphins. We highlight that spatiotemporal dynamics can strongly influence the structure of social networks. Our findings show that hypothetical social units can emerge due to the temporal opportunities for individuals to interact. Therefore, a thorough search for a satisfactory spatiotemporal scale that removes such nonsocial noise is critical when analysing a social system. Li-Fraumeni Syndrome (LFS) is a hereditary cancer syndrome which predisposes individuals to cancer beginning in childhood. These risks are spread across a lifetime, from early childhood to adulthood. Mutations in the p53 tumor suppressor gene are known to cause the majority of cases of LFS. The risk for early onset cancer in individuals with Li-Fraumeni Syndrome is high. Studies have shown that individuals with LFS have a 90% lifetime cancer risk. Children under 18 have up to a 15% chance of cancer development. Effectiveness of cancer screening and management in individuals with Li-Fraumeni Syndrome is unclear. Screening for LFS-associated cancers has not been shown to reduce mortality. Due to the lack of effective screening techniques for childhood cancers, institutions vary with regard to their policies on testing children for LFS. There are currently no national guidelines regarding predictive testing of children who are at risk of inheriting LFS. No studies have looked at parental attitudes towards predictive p53 genetic testing in their children. This was a cross-sectional pilot study aimed at describing these attitudes. We identified individuals whose children were at risk for inheriting p53 genetic mutations. These individuals were provided with surveys which included validated measures addressing attitudes and beliefs towards genetic testing. The questionnaire included qualitative and quantitative measures. Six individuals completed and returned the questionnaire with a response rate of 28.57%. In general, respondents agreed that parents should have the opportunity to obtain p53 genetic testing for their child. Parents vary in regard to their attitudes towards who should be involved in the decision making process and at what time and under what considerations testing should occur. Testing motivations cited most important by respondents included family history, planning for the future and health management. Concern for insurance genetic discrimination was cited as the most important “con” to genetic testing. Although limited by a poor response rate, this study can give health care practitioners insight into testing attitudes and beliefs of families considering pediatric genetic testing.

We previously reported the presence of a novel variant (β-T594M) of the amiloride-sensitive Na+ channel (ASSC) in which the threonine residue at position 594 in the β-subunit has been replaced by a methionine residue. Electrophysiological studies of the ASSC on Epstein–Barr virus (EBV)-transformed lymphocytes carrying this variant showed that the 8-(4-chlorophenylthio) adenosine 3′:5′-cyclic monophosphate (8cpt-cAMP)-induced responses were enhanced when compared to wild-type EBV-transformed lymphocytes. Furthermore, in wild-type EBV-transformed cells, the 8cpt-cAMP-induced response was totally blocked by the phorbol ester, phorbol 12-myristate 13-acetate (PMA). This inhibitory effect of PMA was blocked by a protein kinase C inhibitor, chelerythrine. We now have identified individuals who are homozygous for this variant, and showed that PMA had no effect on the 8cpt-cAMP-induced responses in the EBV-transformed lymphocytes from such individuals. Cells heterozygous for this variant showed mixed responses to PMA, with the majority of cells partially inhibited by PMA. Our results demonstrate that an alteration in a single amino acid residue in the β-subunit of the ASSC can lead to a total loss of inhibition to PMA, and establish the β-subunit as having an important role in conferring a regulatory effect on the ASSC of lymphocytes.

A adesão ao tratamento ocorre quando o conselho médico ou de saúde coincide com o comportamento do indivíduo, ao uso de medicamentos, cumprimento da dieta e mudanças no estilo de vida, não sendo, portanto, um ato não passivo do paciente. Em pacientes com hipertensão arterial sistêmica a adesão ao tratamento pode ser definida como o grau de cumprimento das medidas terapêuticas indicadas, sejam elas medicamentosas ou não, com o objetivo de manter a pressão arterial em níveis pressóricos normais. A não adesão em pacientes com doenças crônicas em tratamento a longo prazo em países desenvolvidos é em média de 50%, revelando a importância de serem avaliados os motivos que levam a esse comportamento. O estudo teve como objetivo avaliar a não adesão em idosos hipertensos de uma unidade pública de saúde de Ribeirão Preto - SP. Trata-se de um estudo de corte transversal, desenvolvido com uma amostra de 196 pessoas. A coleta de dados ocorreu entre agosto de 2014 até junho de 2015, após aprovação do Comitê de Ética em Pesquisa. Para essa etapa foram utilizados os instrumentos Brief Medication Questionnaire, Medical Outcomes Studies 36-item Short Form Survey, Escore de Risco Global e Escore de Risco pelo Tempo de Vida. Após a coleta dos dados, as entrevistas foram codificadas, os dados foram tabulados e foi realizada a análise estatística descritiva e de correlação. Como resultado, constatou-se que houve predomínio de mulheres, com idade média de 69,4 anos, casados/união estável, não moravam sozinhos, com 1,85 pessoas na casa em média, de cor branca, com ensino fundamental incompleto, renda de até dois salários mínimos e aposentados/pensionistas, atendidos pelo SUS. Apresentaram hábitos de vida razoáveis, sem predomínio de consumo de bebidas alcoólicas, tabagismo, uso excessivo de sal e sedentarismo. A mais frequente comorbidade associada à HAS foi a dislipidemia. Foi observado elevado predomínio de fatores de risco cardiovasculares como obesidade abdominal, obesidade geral, comorbidades, razão de lipídeos e fatores agravantes como proteína c reativa ultrassensível, microalbuminúria e síndrome metabólica. A maioria da amostra foi classificada como sendo portador de risco cardiovascular alto após estratificação do risco. A percepção da qualidade de vida relacionada à saúde foi considerada baixa na maioria principalmente devido a limitações emocionais. A não adesão esteve presente em quase metade dos idosos, relacionada principalmente à complexidade da farmacoterapia e dificuldade em lembrar sobre o uso de seus medicamentos. Não foi observada correlação entre a não adesão e as variáveis estudadas. Conclui-se que o comportamento de não adesão observado não esteve relacionada às variáveis estudadas nessa amostra e que são necessárias intervenções urgentes para reduzir o risco cardiovascular e prevenir doenças cardiovasculares e mortalidade, bem como melhora da percepção da qualidade de vida relacionada à saúde.

OBJETIVO: Determinar las características socio-demográficas asociadas a la presencia de dolor músculo-esquelético (MSQ) en personas mayores de 18 años que viven en el Cantón Cuenca. PACIENTES Y MÉTODOS: Estudio transversal analítico, realizado en sujetos del área rural y urbana, mayores de 18 años, utilizando la metodología COPCORD (Community Oriented Program for the Control of Rheumatic Diseases), adaptada para Ecuador. Se identificaron a los individuos con síntomas reumáticos, casa por casa. La revisión para confirmar el diagnóstico realizó un médico Reumatólogo. El análisis estadístico fue univariado y multivariado. RESULTADOS: Participaron 2500 personas encuestadas, el 59,6% correspondió al sexo femenino, con una edad promedio de 42,8 años, el 25,5% de los participantes realizaban actividades domésticas. El dolor MSQ se presentó en el 32,2% de las personas, el cual se asoció significativamente a vivir en el área rural, tener menor escolaridad, ingresos económicos por debajo de 340 dólares, carga física mayor de 8 libras, mayor discapacidad funcional y el cocinar con leña en el sector rural. CONCLUSIÓN: El dolor MSQ tiene un alta prevalencia en la población de Cuenca, Ecuador; produciendo discapacidad especialmente en el área rural. se asocia con menor nivel de escolaridad, ingresos económicos bajos y mayor carga física

L’infection au VIH s’accompagne souvent de dérégulations du compartiment des lymphocytes B qui nuisent à la génération de réponses efficaces. En effet, détectées tôt après l’infection, ces dérégulations perdurent, ne sont pas totalement restaurées par la thérapie, et mènent souvent à des manifestations auto-immunes et lymphomes. Une étude longitudinale de notre groupe, effectuée avec des cellules mononucléées du sang circulant provenant de patients VIH+ avec différents types de progression clinique, a démontré qu’un niveau élevé de BLyS chez des individus VIH+ progresseurs était associé à une dérégulation des fréquences de populations de cellules B avec augmentation de cellules innées de la zone marginale (MZ) présentant des caractéristiques d’immaturité et d’activation. Au contraire, chez des individus VIH+ non-progresseurs avirémiques ou contrôleurs d’élite, les niveaux de BLyS étaient dans la normale et ce sont les fréquences de cellules B MZ plus matures qui étaient diminuées. La résistance au VIH pourrait aussi impliquer le contrôle de BLyS et son impact sur les cellules B. De ce fait, nous avons préalablement recruté une cohorte de travailleuses du sexe (TS) à Cotonou (Bénin) dans laquelle nous avons identifié des femmes qui demeurent séronégatives malgré une exposition soutenue au virus. Nous avons mesuré les niveaux de BLyS dans le sang et dans les lavages cervico-vaginaux (CVL) de TS VIH- et les avons comparés à ceux mesurés chez des TS VIH+ et un groupe contrôle de non-TS VIH- . Nous avons trouvé que les niveaux de BLyS dans le sang et le CVL des TS VIH- étaient inférieurs à ceux des TS VIH+ et des non-TS VIH-. Le niveau d’expression de BLyS à la surface des lymphocytes T, monocytes et cellules dendritiques de TS VIH- était augmenté, mais à un niveau moindre que les TS VIH+. Chez les TS VIH+, les hauts niveaux de BLyS étaient concomitants avec une dérégulation du compartiment B caractérisée par une hyperglobulinémie, une augmentation de la fréquence de populations avec un profil immature/inné et une plus grande proportion de plasmablastes IgG vs IgA. Au contraire, les niveaux inférieurs de BLyS dans le sang des TS VIH- coïncident avec un compartiment B préservé, révélant que les lymphocytes B MZ peuvent être impliqués dans l’immunité naturelle au VIH. Ces résultats démontrent l’importance du contrôle des niveaux de BLyS et du maintien de l’intégrité du compartiment B dans la résistance au VIH.

Congenital hypogonadotropic hypogonadism (CHH) and its anosmia-associated form (Kallmann syndrome [KS]) are genetically heterogeneous. Among the >15 genes implicated in these conditions, mutations in FGF8 and FGFR1 account for ∼12% of cases; notably, KAL1 and HS6ST1 are also involved in FGFR1 signaling and can be mutated in CHH. We therefore hypothesized that mutations in genes encoding a broader range of modulators of the FGFR1 pathway might contribute to the genetics of CHH as causal or modifier mutations. Thus, we aimed to (1) investigate whether CHH individuals harbor mutations in members of the so-called "FGF8 synexpression" group and (2) validate the ability of a bioinformatics algorithm on the basis of protein-protein interactome data (interactome-based affiliation scoring [IBAS]) to identify high-quality candidate genes. On the basis of sequence homology, expression, and structural and functional data, seven genes were selected and sequenced in 386 unrelated CHH individuals and 155 controls. Except for FGF18 and SPRY2, all other genes were found to be mutated in CHH individuals: FGF17 (n = 3 individuals), IL17RD (n = 8), DUSP6 (n = 5), SPRY4 (n = 14), and FLRT3 (n = 3). Independently, IBAS predicted FGF17 and IL17RD as the two top candidates in the entire proteome on the basis of a statistical test of their protein-protein interaction patterns to proteins known to be altered in CHH. Most of the FGF17 and IL17RD mutations altered protein function in vitro. IL17RD mutations were found only in KS individuals and were strongly linked to hearing loss (6/8 individuals). Mutations in genes encoding components of the FGF pathway are associated with complex modes of CHH inheritance and act primarily as contributors to an oligogenic genetic architecture underlying CHH.

An interdisciplinary study was conducted at Qijurittuq (IbGk-3), an archaeological site located on Drayton Island along the eastern shore of Hudson Bay, Nunavik. Local Inuit made important contributions to the research. High school students participated in the field school, and elders shared their traditional knowledge. The elders expressed an interest in the source of the wood used to construct Qijurittuq's semi-subterranean dwellings, and this inspired us to expand our research in that direction. This interdisciplinary study included a reconstruction of the geomorphological and environmental history of Drayton Island, wood provenance and dendrochronology studies, research on house architecture and settlement patterns, and a zooarchaeological analysis. This paper synthesizes the preliminary results of this interdisciplinary investigation within the context of climate change. We discuss the persistence of semi-subterranean dwellings in eastern Hudson Bay long after they had been abandoned elsewhere. At Qijurittuq, their abandonment corresponds with the end of Little Ice Age. However, at the same time, the development of more permanent contact with Euro-Canadians was having a strong impact upon Inuit culture.
